  • In order to micropropagate uniform plantlets of Philodendron cannifolium in vitro, the shoot tips were cultured on MS media supplemented with $0.5{\sim}10.0$ mg/L BA or $0.05{\sim}0.1$ mg/L thidiazuron(TDZ). The adventitious multi-bud clusters from basal part of shoots were formed on MS media containing $2.0{\sim}5.0$ mg/L BA or $0.05{\sim}0.1$ mg/L TDZ. But the shoots grown on MS media with TDZ showed necrosis by the lack of chlorophyll. The adventitious multi-bus clusters were cut into $5{\sim}7$ mm sections and cultured on MS media containing BA and TDZ for shoot proliferation. Shoots were proliferated vigorously on MS medium supplemented with $1.0{\sim}3.0$ mg/L BA with up to 30 shoots. But abnormally swollen hard calli were formed from basal parts of shoots on MS media with TDZ and high concentration of BA(10.0 mg/L). The proliferated shoots on same media also showed necrosis by the lack of chlorophyll. The shoot growth and rooting were favorable on MS media containing $0.5{\sim}2.0$ mg/L IBA. The rooted plantlets were acclimatizated effectively in soil mixed with perlite 1:vermiculite 1 or vermiculite alone. Fifteen mL of liquid medium containing 10 g/L activated charcoal and 30 g/L sucrose were added in same vessels after small shoots were proliferated to stimulate shoot growth and rooting. After 8 weeks in culture, the shoots were dipped into high concentration of IBA solution. and planted in soil mexed with perlite 1:vermiculite 1. The shoot growth and rooting were favorable in dipping treatments of $500{\sim}2,000$ ppm IBA solutions for 10 sec.

  • In order to micropropagate uniform plantlets of Alocasia cadieri Chantrier in vitro, the shoot tips were cultured on media containing various concentrations of BA and thidiazuron (TDZ). Multiple shoot formation from shoot tips was very effective on medium containing 0.1mg/L TDZ. The formed shoots from shoot tips were separated into a shoot, and cultured on media with BA, TDZ, and NM combination for proliferation. The shoots were multiplied very vigorously on medium with 0.5mg/L TDZ and 0.5mg/L NAA. The rooting and growth of multiplied shoots were more effective on medium with 2.0g/L activated charcoal, rather than those with IBA and NAA. Rooted plantlets show high survival in soil mixed with perlite 1: vermiculite 1 or vermiculite alone.

  • The production of high quality seedlings is a very important phase in silvicultural systems for successful reforestation or restoration. The purpose of this study was to measure both growth performances and nutrient responses of Fraxinus rhynchophylla and Pinus densiflora seedlings, which are commercially planted in Korea, according to soil improvement treatments. We applied 8 types of soil improvements: control with no treatment, compost B and compost Y as organic materials, vermiculite, perlite, two level of zeolite, and mix of vermiculite, perlite, and zeolite as inorganic materials in a permanent national nursery. Only compost B treatment significantly increased soil pH, organic matter, total nitrogen, available phosphorus, exchangeable potassium and calcium at the 0-10 cm soil depth. The growth of F. rhynchophylla and P. densiflora was the highest at the compost B treatment and the lowest at the vermiculate treatment. Compost B treatment allocated more carbon to aboveground than belowground by 39%, especially to foliage. On the vector diagnosis, there was 'shortage' on compost B treatment because of all increases of N contents, N concentrations, and growth and 'over accumulation' on vermiculite treatment because of more N uptake compared with dry weight increase. This study suggested optimal use of soil improvements is very important to improve soil quality in a permanently used nursery.

  • This experiment was conducted to establish the mass propagation system of Calanthe discolor Lindley. When the Calanthe discolor seeds were sown in Murashige and Skoog medium, the percentage of germination was 65%. Seedlings grew more rapidly in the liquid medium than the solid medium. All regenerated plantlets were survived in acclimatized condition of 70% shade and more than 80% humidity. Also, we found out that the 88% of survival ratio could be achieved in containing soil mixture of vermiculite and perlite as same as amount.

  • In order to develop effective acclimatization methods for Aralia elata plantlets regenerated from somatic embryos, various acclimatizing conditions were compared regarding both survival rate and growth of the plantlets. The plantlets were transplanted into plastic boxes containing artificial soil in the presence of either several levels of MS liquid media, distilled water, 2% sucrose or 0.1% hyponex solution. They were then cultured by spraying of distilled water twice a week and maintained in the normal tissue culture room. Perlite was proved to be better than vermiculite on survival rate and growth of the plantlets. As the size of perlite (larger than 0.2 cm in diameter) increased, both the survival rate and growth of the plantlets improved. Among the various MS liquid media and different aqueous solutions tested, distilled water appeared to result in the best survival rate and growth. MS media were also effective in increasing survival rate and supporting growth when diluted to 1/4 and/or 1/8. The acclimatized plantlets could be transplanted directly onto the nursery bed and grown normally. The above results suggest that plantlets regenerated from somatic embryos of Aralia elata be effectively acclimatized using a plastic box containing perlite with distilled water treatment.

  • Experiments were conducted to measure rooting and growth of rose cuttings as affected by medium composition and volume. Rosa hybrida 'Red Sandra' and 'Little Marble' were used as test plants. For medium composition test, rockwool, phenolic foam, perlite, vermiculite, peatmoss, coir and a commercial medium (Tosilee medium) were applied in single or mixed forms on the propagation bed with fog (200 sec./5 min.) installed to control the humidification time. 'Red Sandra' and 'Little Marble' cuttings were planted in 16-cell plastic pots and 72-cell plug trays, respectively. For the medium volume test, 72-cell plug trays and 16-cell or 9-cell plastic pots were used. Cuttings with an uniform node with a five-leaflet leaf was soaked in a $500mg{\cdot}L^{-1}$ IBA solution for 30 sec. A fogging system installed on the propagation bed was used for (100 sec./5 min.). Rockwool 1 : peatmoss 1 (v/v) was the most effective on growth and rooting for both cultivars and high volume of Tosilee medium and rockwool were most effective media far both cultivars.

  • This paper investigates the flexural behavior of concrete beams reinforced longitudinally with either steel bars, molded glass-fiber reinforced polymer (GFRP) grating mesh or pultruded glass-fiber reinforced polymer (GFRP) grating mesh, under four-point bending. The variables included in this study were the type of concrete (normal weight concrete, perlite concrete and vermiculite concrete), type of the longitudinal reinforcement (steel bars, molded and pultruded GFRP grating mesh) and the longitudinal reinforcement ratio (between 0.007 and 0.035). The influences of these variables on the load-midspan deflection curves, bending stiffness, energy absorption and failure modes were investigated. A total of fifteen beams with a cross-sectional dimension of 160 mm × 210 mm and an overall length of 2400 mm were cast and divided into three groups. The first group was constructed with normal weight concrete and served as a reference concrete. The second and third groups were constructed with perlite concrete and vermiculite concrete, respectively. An innovative type of stirrup was used as shear reinforcement for all beams. The results showed that the ultimate load of the beams reinforced with pultruded GFRP grating mesh ranged between 19% and 38% higher than the ultimate load of the beams reinforced with steel bars. The bending stiffness of all beams was influenced by the longitudinal reinforcement ratio rather than the type of concrete. Failure occurred within the pure bending region which means that the innovative stirrups showed a significant resistance to shear failure. Good agreement between the experimental and the analytical ultimate load was obtained.

  • This experiment was carried out to develop the mass propagation system for producing plug plantlets using stem cuttings of virus-tree microtubers in potato. Cocopeat, vermiculite, perlite and peatmoss were combined and used as plug nursery media to find out the best combination suitable for the growth of seedlings derived from microtubers. Seedling growth was favored in high temperature (above 2$0^{\circ}C$) and a long-day photoperiod(above 16 hours) condition, while stolons and microtubers formed in outdoor condition. Shoot and root multiplication was not affected by NAA 10mg /1 or IAA 10mg /1 treatment. At the early growth stage of plug plantlets, the number of leaves and roots and the length of root increased significantly when nodes from the upper (near to apex) part of shoots rather than from basal part were taken. But after transplanting, these differences among these characters were not observed. At ninety days after transplanting the plug plantlets in spring time, plant was around 70 to 80cm in height, and the number of stolons and tubers were ten and seven, respectively.

  • This experiment was conducted to investigate physical and chemical characteristics by volume fractions of root media using peatmoss, perlite, and vermiculite, along with effects on the growth of pot chrysanthemums (Dendranthema ${\times}$ grandiflorum 'Vemini') in a C-channel mat irrigation system. To evaluate the physico-chemical properties of 20 root media, the bulk density, particle density, total pore space, pore space, ash content, organic matter, pH, and electrical conductivity were measured and data were analyzed using principal component analysis (PCA). PCA scores revealed that physico-chemical properties changed by the blending of peatmoss, perlite, and vermiculite. The 20 root media were divided into three main groups by hierarchical cluster analysis. At the end of the experiment, the pH and EC of the root media were measured from media divided into four layers. The pH of root media without plants showed a strong linear relationship and the pH of root media with plants increased exponentially. The change of EC in the root medium was indicated as a hyperbolic curve. Plant growth characteristics according to growth in the 20 root media were analyzed by PCA. It was found that the mixing ratios of the root media affected plant growth characteristics. Therefore, mixing ratio is an important factor for pot-plant production in a subirrigation system.
