Purpose: Vascular endothelial growth factor (VEGF) is one of the most important growth factors for metastatic tumors. To clarify the role of VEGF-A and C in patients with peptic ulcer disease (PUD) or gastric cancer (GC), we evaluated the expression levels of these two molecules. We also analyzed the effect of Helicobacter pylori infection on VEGF-A and C expression levels. Materials and Methods: Patients with dyspepsia who needed diagnostic endoscopy were selected and divided into three groups: nonulcer dyspepsia (NUD), PUD, and GC, according to their endoscopic and histopathological results. Fifty-two patients with NUD, 50 with PUD, and 38 with GC were enrolled in this study. H. pylori infection was diagnosed by the rapid urease test. After RNA extraction and synthesis of cDNA, the expression levels of VEGF-A and C were determined by quantitative reverse transcriptase polymerase chain reaction. Results: The VEGF-C expression level in the PUD and GC groups was significantly higher than that in the NUD group. Moreover, the VEGF-A expression level in the PUD and GC groups was higher than in the NUD group, although the differences were not statistically significant. Significant positive correlations were also observed between the expression levels of these two molecules in the PUD and GC groups. In addition, the expression levels of these two molecules were higher in H. pylori positive patients with PUD or GC than in H. pylori negative patients of the same groups; however, these differences did not reach statistical significance. Conclusions: Up-regulation of VEGF-C expression during gastric mucosal inflammation may play a role in the development of peptic ulcers or GC.
Objectives: To obtain permeability surface (PS) values using multi-slice helical CT perfusion imaging and to evaluate the spatial distribution correlation between PS values and vascular endothelial growth factor (VEGF) expression in solitary brain metastases. Methods: Imaging was performed on 21 patients, PS values being calculated from the central, border and peripheral parts of tumours. VEGF expression was determined by immunohistochemical staining. Results: Rim enhancement was found in 16 cases, the border of the tumour featuring PS elevation with high VEGF expression in 13 cases. In the 5 cases with nodular enhancement, the border and the central part had high permeability and VEGF expression was high in all cases, the correlation being significant (P<0.01). Conclusion: VEGF expression in brain metastases positively correlates with PS values from CT perfusion imaging, so that the latter can be used in the surveillance of angiogenic activity in brain metastases.
Purpose : This study evaluated the effect of VEGF in the arterial anastomosis by using light and electron microscopy. Marerials and method : Rats underwent femoral arterial end-to-end anastomosis after transection and topical VEGF treatment. The proximal and distal segments of the femoral arteries was drenched with 1 drop of VEGF $(100ng/100{\mu}l/bottle)$. and when half of the repair was finished, the other 1 drop was drenched and then the repair was continued to complete the anastomosis. Gross and histologic characteristics of arterial wall were assessed after 3 days, 1, 3 and 5 weeks. In the control group, normal saline solution instead of VEGF was dropped with the same method in the anastomosis. Results : The histologic findings of the arterial wall were the vascular remodeling with the infiltration of inflammatory cells at early stages and the tissue fibrosis at lately stages in the anastomotic sites of the control and the VEGF-treated groups. The scanning electron microscopic results were; (1) the anastomotic sites were covered by many irregular cells with long cytoplasmic processes at the early stages. (2) After 1 week, endothelial cells started to cover the anastomotic sites. (3) After 3 weeks, the anastomotic sites were partially covered by endothelial cells in the control group. (4) After 5 weeks, the anastomotic sites were completely covered by endothelial cells in the control and VEGF-treated groups. (5) In the VEGF-treated group, the anastomotic site was completely covered by endothelial cells which directed parallel to longitudinal axis of arteries after 3 weeks. Conclusion : Topical VEGF maintained luminal integrity by decreasing fibrosis and increasing re-endothelialization. These findings suggest that topical VEGF may be a promising new strategy to enhance healing and improve the outcome of vascular anastomosis.
청국장은 대두발효식품으로 대두 단백질이 발효 중 분해되면서 다양한 생리활성물질이 만들어진다. 혈관내피세포는 혈관의 기능은 물론 신생혈관 생성을 주도하는 세포이다. 뇌졸중이나 심근경색, 뇌경색 등의 혈관관련 질병들은 신생혈관 생성을 촉진하는 치료법이 필요하다. Vascular Endothelial Growth Factor (VEGF)는 새로운 혈관 형성을 촉진하는 역할을 한다. 본 연구에서는 청국장 에탄올추출물(CEE)이 HUVEC (혈관 내피세포) 증식에 미치는 영향을 조사하여 보았다. 청국장 추출물(100, $1000{\mu}g/ml$)을 HUVEC에 처리했을 때, VEGF (10 ng/ml)를 처리한 대조군과 같은 정도로 세포를 증식시켰다. 열처리한 청국장추출물을 혈관 내피세포에 처리해도 세포 증식효과는 마찬가지였다. 청국장이 세포 증식뿐 아니라 HUVEC이동에도 영향을 주는지 sprout 분석법으로 확인하였다. 청국장 추출물($100{\mu}g/ml$)을 처리했을 때, VEGF (10 ng/ml)와 비슷할 정도로 HUVEC 이동이 일어났다. 청국장 추출물에서 HUVEC 증식과 이동에 영향을 미치는 특정 peptide의 분리가 필요할 것이다.
Tumor angiogenesis is a critical step f3r the growth and metastasis of solid tumors. Vascular endothelial growth factor (VEGF) is the most important angiogenic molecule associated with tumor-induced neovascularization. VEGF exerts its activity through binding to its receptor tyrosine kinase, KDR/Flk-1, expressed on the surface of endothelial cells. This study was carried out to investigate inhibitory effect of extracts from root cortex of Paeonia suffruticosa Andrews on VEGF binding to VEGF receptor. The MeOH extract from P. suffrutiocosa Andr. inhibited the binding of KDR/Flk-1-Fc to immobilized VEGF$_{165}$ more than 45% at the concentration of 100 ${\mu}$g/mL. The MeOH extract was further fractionated into n-hexane, ethyl acetate, n-BuOH, and aqueous fractions. Among the four fractions, the ethyl acetate fraction from the root cortex of P. suffruticosa Andr. exhibited highly effective inhibition (${\approx}$ 79% inhibition) and then n-BuOH fraction (${\approx}$ 45% inhibition) on the binding of KDR/Flk-1-Fc to immobilized VEGF$_{165}$ at the concentration of 100 ${\mu}$g/mL. The ethyl acetate fraction from the root cortex of P. suffruticosa Andr. more efficiently blocked VEGF-induced human umbilical vein endothelial cell proliferation, than the growth of HT1080 human fibrosarcoma. Our results suggest that P. suffruticosa Andr. may be used as a candidate fur developing anti-angiogenic agent.
본 연구는 중년여성 18명을 대상으로 저항성 운동군 A그룹 6명(1RM 40-60%), 유산소 운동군 B그룹 6명(VO2max 60-70%), 유연성 운동군 C그룹(10~60 sec/sets) 6명으로 무선 배정하여 12주간 주 3회(월, 수, 금) 1일 50~60분의 운동 형태에 따른 신체구성과 염증인자, 혈관내피 성장인자에 미치는 영향을 알아보기 위하여 비교·분석한 결과, 첫째, 염증인자의 변화에서는 CRP 및 IL-6의 시기의 주효과에서는 유의한 차이(p<.05)가 나타났다. 둘째, 혈관내피 성장인자의 변화에서는 VEGF의 시기의 주효과 및 시기×그룹의 상호작용에서는 유의한 차이(p<.05)가 나타났다. 본 연구의 결과를 토대로 운동 형태에 따른 염증인자와 혈관내피 성장인자에 관한 다양한 각도의 연구와 개선을 위한 기초 자료로 활용되기를 기대한다.
Adenoid cystic carcinoma (ACC) of salivary glands has a protracted clinical course with perineural invasion, delayed onset of hematogenous metastasis, and poor responses to classical cytotoxic chemotherapic agents. Most deaths from salivary ACC are caused by lung metastases that are resistant to conventional therapy. Therefore, knowledge of cellular properties and tumor-host interactions that influence the dissemination of metastatic cells is important for the design of more effective therapy of salivary cancer. I determined in vitro expression of epidermal growth factor receptor (EGFR) and its downstream effectors and vascular endothelial growth factor receptor (VEGFR)-2 on a human salivary ACC cell line (ACC2). I also evaluated the expression of EGF and VEGF signaling molecules and metastasis-related proteins on human salivary ACC cells orthotopically growing in nude mice. In Western blot and immunohistochemical analyses, EGFR and VEGFR-2 were presented and phosphorylated in ACC2 cells. In human parotid cancer xenografts in nude mice, EGF and VEGF signaling molecules, IL-8, and MMP-9 were expressed at markedly higher levels than in normal parotid tissues. Moreover, tumor-associated endothelial cells of this orthotopic parotid tumor expressed phosphorylated VEGFR-2 and phosphorylated Akt, which is a cell-survival protein. These data show that those biomarkers can be molecular targets for therapy of salivary ACC, which has a propensity for delayed lung metastasis.
목 적 : 가와사끼병은 전신성 혈관염으로 소아 후천성 심질환의 가장 흔한 원인이다. 혈관내피세포 생성인자는 미세혈관 투과성 및 혈관생성능력을 갖추고 있으며 가와사끼병에서 심장 합병증에 중요한 역할을 하는 것으로 알려져 있다. 가와사끼병에서 임상 양상, 심장 합병증, 감마글로불린 치료에의 반응 여부, 검사실 소견과 VEGF의 상관관계에 대하여 연구하였다. 방 법 : 가와사끼군은 49명을 대상으로 하였고, 대조군은 15명을 대상으로 하였다. 급성기, 아급성기, 회복기로 나누어 측정하였으며, 심장초음파 이상소견 및 감마글로불린 반응군 및 불응군을 정의하였다. 결 과 : 1) 가와사끼군과 대조군은 연령 및 체중에서 통계학적으로 유의한 차이는 없었다. 2) 급성 발열기 가와사끼군에서 혈청 VEGF가 대조군에 비해 유의하게 높았다. 3) 정형적 가와사끼군과 비정형 가와사끼군의 급성기 혈청 VEGF는 유의한 차이가 없었다. 4) 관상동맥 병변이 있는 군과 없는 군 간의 혈청 VEGF는 급성기와 아급성기 및 회복기에서 모두 유의한 차이가 없었다. 5) 심염이 있는 군과 없는 군 간의 혈청 VEGF는 급성기와 아급성기 모두에서 유의한 차이가 없었다. 6) 감마글로불린 반응군이 비반응군에 비해 급성기, 회복기 혈청 VEGF가 높았으나 유의한 차이는 없었고, 아급성기 혈청 VEGF는 불응군이 반응군에 비해 의미있게 높았다. 또한 감마글로불린 불응군 중에서 아급성기 혈청 VEGF가 급성기, 회복기에 비해 유의하게 높았다. 7) 회복기 관상동맥 변화 지속군과 비지속군의 급성기, 아급성기 혈청 VEGF의 차이는 의미 있었으나, 회복기 혈청 VEGF는 유의한 차이가 없었다. 8) 총 발열기간과 급성기 혈청 VEGF 간의 상관관계는 없었으나, 아급성기 혈청 VEGF는 뚜렷한 양의 상관관계를 보였다. 9) 급성기 혈청 VEGF는 적혈구침강속도, C-반응성단백과 양의 상관관계를 보였고 백혈구는 모든 시기의 혈청 VEGF와 뚜렷한 양의 상관관계를 보였다. 10) 급성기, 아급성기 혈청 VEGF는 혈색소와 뚜렷한 음의 상관관계를 보였으며 알부민과도 뚜렷한 음의 상관관계를 보였다. 결 론 : 혈청 VEGF는 가와사끼병의 중증도 판정에 도움을 줄 수 있으며 특히 아급성기 값이 심합병증의 예측인자로 사용 가능할 것으로 보인다. 검사실 소견과의 관계를 통해 염증반응지표와 밀접한 연관이 있는 것으로 생각되며 향후 더 엄격한 진단기준과 더 많은 대상 환아를 기초로 한 지속적인 연구가 필요할 것으로 사료된다.
Periodontitis is characterized by the loss of periodontal tissues, especially alveolar bone. Common therapies cannot satisfactorily recover lost alveolar bone. Periodontal ligament stem cells (PDLSCs) possess the capacity of self-renewal and multilineage differentiation and are likely to recover lost alveolar bone. In addition, periodontitis is accompanied by hypoxia, and hypoxia-inducible $factor-1{\alpha}$ ($HIF-1{\alpha}$) is a master transcription factor in the response to hypoxia. Thus, we aimed to ascertain how hypoxia affects runt-related transcription factor 2 (RUNX2), a key osteogenic marker, in the osteogenesis of PDLSCs. In this study, we found that hypoxia enhanced the protein expression of $HIF-1{\alpha}$, vascular endothelial growth factor (VEGF), and RUNX2 ex vivo and in situ. VEGF is a target gene of $HIF-1{\alpha}$, and the increased expression of VEGF and RUNX2 proteins was enhanced by cobalt chloride ($CoCl_2$, $100{\mu}mol/L$), an agonist of $HIF-1{\alpha}$, and suppressed by 3-(5'-hydroxymethyl-2'-furyl)-1-benzyl indazole (YC-1, $10{\mu}mol/L$), an antagonist of $HIF-1{\alpha}$. In addition, VEGF could regulate the expression of RUNX2, as RUNX2 expression was enhanced by human VEGF ($hVEGF_{165}$) and suppressed by VEGF siRNA. In addition, knocking down VEGF could decrease the expression of osteogenesis-related genes, i.e., RUNX2, alkaline phosphatase (ALP), and type I collagen (COL1), and hypoxia could enhance the expression of ALP, COL1, and osteocalcin (OCN) in the early stage of osteogenesis of PDLSCs. Taken together, our results showed that hypoxia could mediate the expression of RUNX2 in PDLSCs via $HIF-1{\alpha}$-induced VEGF and play a positive role in the early stage of osteogenesis of PDLSCs.
목 적 : 가와사키병은 어린 소아에서 전신성 혈관염을 일으키는 가장 흔한 원인이다. 가와사키병이 발병한 경우 여러 혈관 성장인자들의 분비가 촉진되어 내막층의 증식과 신생 혈관 생성작용이 진행되면서 관상동맥이 재구성된다. 수년간 가와사키병의 치료제로 IVIG와 코르티코스테로이드가 사용되었으나 그 치료기전이 명확히 밝혀진 바는 없다. 이런 IVIG와 코르티코스테로이드가 가와사키병의 관상동맥합병증을 감소시키는데 어떠한 기전으로 작용하는지 알아보고자 본 연구에서는 Matrigel을 이용하여 in vitro에서 VEGF에 의해 유도된 내막세포 분화에 대한 IVIG와 methylprednisolone의 효과를 보고자 하였다. 방 법 : Matrigel을 이용하여 in vitro에서 VEGF에 의해 유도된 내막세포 분화에 대한 IVIG와 methylprednisolone의 효과를 보고자 VEGF, VEGF와 IVIG, VEGF와 VEGF antibody, VEGF와 methylprednisolone, VEGF, IVIG와 methylprednisolone을 각각 넣어주고, HUVECs를 18시간 동안 배양 후 튜브의 총 길이를 측정하였다. 결 과 : 1) IVIG의 농도를 25 mg/mL, 그리고 40 mg/mL로 증량하였을 경우 IVIG를 투여하지 않은 대조군에 비하여 통계학적으로 각각 유의한 세포수(HUVECs) 감소 효과를 관찰할 수 있었으며(P<0.001), 40 mg/mL를 처리한 경우가 25 mg/mL를 처리한 경우보다 세포수 감소 효과가 더 뚜렷하였다(P<0.05). 세포배양 시간에 따른 억제 효과는 25 mg/mL과 40 mg/mL에서 배양 후 24시간과 48시간 모두에서 대조군과 비교하여 통계학적으로 유의한 효과를 보였다(P<0.001). 2) VEGF 20 ng/mL를 처리한 군에 IVIG 40 mg/mL 또는 여러 농도의 methylprednisolone($10^{-12}M$, $10^{-9}M$ or $10^{-6}M$)을 처치해 주었을 때 튜브 형성이 유의하게 억제되었으며(P<0.05), methylprednisolone의 경우 그 억제 정도가 농도에 비례하여 일어났으나 IVIG 투여시 보다는 억제 효과가 약하였다. 한편, IVIG와 methylprednisolone와 병합투여한 경우는 methylprednisolone 농도에 상관없이 모두 튜브 형성을 잘 억제하였으며(P<0.001), IVIG만 단독 투여한 것과 통계적 차이는 없었다. 결 론 : 본 연구에서는 in vitro에서 VEGF가 HUVECs의 분화를 유도하고 분화된 내막세포의 맥관 형성이 IVIG와 methylprednisolone에 의해 저해된다는 것을 보여주었다. 이는 가와사키병의 관상동맥합병증이 생기는 기전 중 VEGF와 내막세포의 분화로 인해 일어나는 관상동맥의 재구성을 IVIG와 methylprednisolone이 억제한다는 것을 시사하며 치료 기전 중 하나로 제시될 수 있다.

① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는

- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
