Toxicological Evaluation of Saposhnikoviae Radix Water Extract and its Antihyperuricemic Potential

  • Although the dried root of Saposhnikovia divaricata (Turcz.) Schischk. (Umbelliferae) is a popular medicinal plant in East Asia, there has been no systemic toxicological evaluation of a water extract of Saposhnikoviae Radix (SRE). In this experiment, an oral acute and 13-week subchronic toxicological evaluations of SRE (500-5,000 mg/kg body weight) were performed in both sexes of Crl:CD(SD) rats. Based on the results from mortality, clinical signs, effects on body weight and organ weight, clinical biochemistry, hematology, urinalysis, and histopathology, significant acute, 4-week repeated dose range finding (DRF) and 13-week subchronic toxicity of SRE was not observed in either sex of rats; thus, the no observed adverse effect level (NOAEL) was 5,000 mg (kg/day). To identify anti-hyperuricemia potential of SRE, the suppressive effect of SRE was determined in mice challenged with potassium oxonate (PO; 250 mg/kg) via intraperitoneal injection for 8 days (each group; n = 7). SRE supplementation suppressed the uric acid level in urine through significant xanthine oxidase (XO) inhibitory activity. Kidney dysfunctions were observed in PO-challenged mice as evidenced by an increase in serum creatinine level. Whereas, SRE supplementation suppressed it in a dose-dependent manner. Collectively, SRE was safe up to 5,000 mg (kg/day) based on NOAEL found from acute and 13-week subchronic toxicological evaluations. SRE had anti-hyperuricemia effect and lowered the excessive level of uric acid, a potential factor for gout and kidney failure.

Microstructures and Hardness of CO2 Laser Welds in 409L Ferritic Stainless Steel (409L 페라이트계 스테인리스강 CO2레이저 용접부의 미세조직과 경도)

  • The microstructure and hardness of $CO_2$ laser welds were investigated in the Ti-stabilized ferritic stainless steel 409L. The observed specimen was welded in a fully penetrated condition in which the power was 5 kW and the welding speed 5 m/min. The grain structure near the bond line of the laser welds was produced by epitaxial growth. The grain size was the largest in the fusion zone, and HAZ showed nearly the same grain size as that of the base metal. The HAZ microstructure consisted of subgrains and precipitates that were less than 100 nm in size and that were located along the subgrain boundaries. On the other hand, the hardness was the highest in the fusion zone due to the large amount of small precipitates present. These were composed of TiN, Ti(C,N) and $TiO_2$+Ti(C,N). The hardness decreased continuously from the fusion zone of the base metal. The HAZ hardness was slightly greater than that of the base metal due to the existence of subgrains and precipitates in the subgrain boundary.

Kv1.3 voltage-gated K+ channel subunit as a potential diagnostic marker and therapeutic target for breast cancer

  • Voltage-gated $K^+$ (Kv) channels are widely expressed in the plasma membranes of numerous cells such as epithelial cells. Recently, it has been demonstrated that Kv channels are associated with the proliferation of several types of cancer cells. Specifically, Kv1.3 seems to be involved in cancer cell proliferation and apoptosis. In the present study, we examined the expression of Kv1.3 in immortalized and tumorigenic human mammary epithelial cells. We also evaluated the expression level of Kv1.3 in each stage of breast cancer using mRNA isolated from breast cancer patients. In addition, treatment with tetraethylammonium, a Kv channel blocker, suppressed tumorigenic human mammary epithelial cell proliferation. Therefore, Kv1.3 may serve as a novel molecular target for breast cancer therapy while its stage-specific expression pattern may provide a potential diagnostic marker for breast cancer development.

Airborne Asbestos Concentrations of Dental Laboratories in One Metropolitan City (치과기공소의 공기 중 석면농도 분포에 관한 연구 -일개 광역시를 중심으로-)

  • This study was performed to investigate the problems of using asbestos for dental technicians. Samples for analysing were sampled by personal sampler in 40 sampling points of 10 dental laboratories in Daegu city from December 2010 to January 2011 and counted with phase contrast microscopy. The results were as follows: 1. Asbestos concentration(geometric mean) of 40 sampling points in 10 dental laboratories was 0.0061 f/cc and it was under recommended industrial and indoor air standard, but the concentration of asbestos in only two dental laboratories was over indoor recommended standard. 2. The concentration of asbestos in processing room and burnout room was 0.0099 f/cc and 0.0037 f/cc, respectively. 3. The mean concentration(GM) of asbestos by kinds of casting ring liners was 0.0159 f/cc in dental laboratories using industrial asbestos, 0.0104 f/cc in them using asbestos containing ring liners, and 0.0026 f/cc in them using non-asbestos ring liners. Casting ring liners using in dental laboratories should be substituted with non-asbestos liners, because most of dental laboratories were using asbestos containing ring liners and a few of them showed higher concentration than recommended standard.

  • We electroplated copper-cobalt thin films on a silicon substrate, which had 150 nm thick copper seed layer. The adhesion between the two metallic layers could be increased by utilizing a proper organic additive, pulse plating technique, and high temperature annealing. The thin films exhibited columnar growth of the deposits and enhanced adhesion. This is attributed to the grain growth mechanism introduced by the additive and annealing.

  • The general pharmacological effects of a new anthracycline anticancer agent, DA-125 $[7-0-(2,\;6-dideoxy-2-fluoro-{\alpha}-L-talopyranosyl)-adriamycinone-14-{\beta}-alaninate{\cdot}HCI]$ were investigated in mice, rats, guinea pigs, rabbits and dogs. Intravenous administration of DA-125 presented no significant effects on the central and peripheral nervous systems of ICR mice except a decrease in the numbers of acetic acid-induced writhing response at a dose of 10 mg/kg. In anesthetized rats and dogs, DA-125 produced a transient depression of blood pressure and an increase in heart rate, but did not affect the peripheral blood flow in the isolated ear vessels of rabbits and the mechanical functions of the isolated hearts of guinea pigs. No significant effects were observed on the gastrointestinal functions and the contractilities of smooth muscle preparations obtained from guinea pig trachea, rabbit ileum, pregnant and non-pregnant uterus and vas deferens of rats. DA-125 Increased the contractility of the isolated ileum of guinea pigs in a dose range of $10^{-6}{\sim}10^{-9}g/ml$, and also increased, but weaker than adriamycin, the vascular permeability in rat skin. DA-125 had no effect on the kallikrein-induced increase in permeability and the permeability of the visceral organs. DA-125 did not adversely affect the liver function and the blood coagulation system, and did not induce hemolysis in vitro. It is concluded from the results that the general pharmachological effects of DA-125 are similar to or weaker than those of adriamycin, and that little adverse effects are anticipated with a therapeutic dose range.

  • From water extract of red ginseng, two new-type glucosides, $A:\;C_{12}H_{16}O_8\;and\;B:\;C_9H_{16}\;O_7,$ were isolated by fractionation with highly porous polymer ollowed by chromatography on silica gel in yields of 0.04 and $0.16\%,$ respectivley. The structures of A and B were elucidated by means of NMR and MS. Neither glucosides were detected in the extract of white ginseng and the mechanism of formation of A and B during the process of steaming will be discussed. The isolation and identification of saponins of corms (head of the root) of red ginseng are also reported.

Comparison of authorized feed analysis laboratories in Korea: looking at feed chemical analysis

  • In Korea, chemical analysis of animal feed is done through authorized feed analysis laboratories (AFALs). Analysis results among the AFALs need to be similar or within acceptable variations; however, there is no experimental evidence of their comparability. We aimed to determine the level of variation of feed chemical analysis results from different AFALs. For this, we requested analysis of four kinds of feed (corn, soybean meal, corn gluten feed, and ryegrass) to eight AFALs and the Cumberland Valley Analytical Services (CVAS) which is an internationally well-recognized feed analysis laboratory. The AFALs spent more time on analysis than did CVAS. Fiber analysis results varied significantly among laboratories. However, moisture, CP, and ash content values showed almost no variation. At least one AFAL obtained results with significant differences from CVAS for all tested values. These differences can be explained by the followings: 1) the standard methods for feed analysis (SMFA) established for AFALs are not detailed enough to control the analytical variations among different laboratories and 2) guidelines are insufficient for the quality control of analysis results in Korea. Failure to accurately identify the nutritional components of the feed could mean failure to provide adequate nutrients to the animals. Therefore, efforts to reduce the differences among AFALs, such as revising SMFA and publishing guidelines on quality control of feed analysis results, are important.

Protective Effect of Taurine on TNBS-induced Inflammatory Bowel Disease in Rats

  • We had previously reported that the protective effect of taurine against indomethacin-induced gastric mucosal injury was due to its antioxidant effects, which inhibited lipid peroxidation and neutrophil activation. In this study, we examined the effect of taurine on reducing the inflammatory parameters of trintrobenzene sulfonic acid (TNBS)-induced inflammatory bowel disease (IBD) in rats. In order to induce IBD, ethanolic TNBS was given to rats intracolonically. Then they received 500 mg/kg.day of taurine orally and were sacrificed one week after IBD induction. While ulceration and inflammation of distal colon with formation of granuloma in the vehicle-treated IBD rats two days after administration of TNBS were observed, treatment with taurine ameliorated colonic damage and decreased the incidence of diarrhea and adhesion. also, colon weight as an index of tissue edema, which was mardedly increased in the IBD rats, became significantly lower after administration of TNBS were observed, treatment with taurine ameliorated colonic damage and decreased the incidence of diarrhea and adhesion. Also colon weitht as an index of tissue edema, which was markedly increased in the IBD rats, became significantly lower after taruine treatment. Myeloperoxidase (MPO) activity in the vehicle-treated IBD rats was substantially increased, compared with that of normal control. the taurine-treated animals significantly reduced MPO activity (35% lower) when compared with that of the vehicle-treated animals. Taurine treatment decreased both basal and formyl-methionyl leucyl phenylalanine-stimulated reactive oxygen generation from colonic tissue in the IBD rats. These results suggest that the administration of taurine reduce the inflammatory parameters in this IBD rat model by increasing defending capacity against oxidative damage.
