Isolation of Defense-Related Genes from Nicotiana glutinosa Infected by Tobacco Mosaic Virus Using a Modified Differential Screening

  • Many of plant defense responses are consequence of transcriptional activation of related genes. We have developed a modified differential screening procedure to isolate tobacco genes that are involved in the defense responses against TMV infection. A cDNA library was constructed from Nicotiana glutinosa leaves infected by TMV under temperature shift conditions. Each of plasmid DNA in the library was hybridized on a set of slot blots to a pool of cDNA probes prepared from either TMV-infected or mock-treated tobacco leaves. Among 900 plasmid DNAs, 81 clones exhibiting significantly enhanced or reduced level of hybridization to either probe were selected for nucleotide sequencing. The clones were listed into 61 genes considering redundancy between the sequences. The genes were identified to be defense-related genes including PR-genes and genes involved in primary or secondary metabolisms. This results supports the implication that plant defense process entails a major shift in total cellular metabolisms rather than activation of a limited number of defense-related genes. Expression patterns of a number of defense-related genes. Expression patterns of a number of selected genes were examined in northern blot analyses. It is notable that the clone 630 of unknown function exhibits expression pattern similar to those of previously known PR-genes. Experiments to elucidate the roles in defense mechanism of a couple of genes newly identified in this study are in progress.

Characterization Of Rainrate Fields Using A Multi-Dimensional Precipitation Model

  • In this study, we characterized the seasonal variation of rainrate fields in the Han river basin using the WGR multi-dimensional precipitation model (Waymire, Gupta, and Rodriguez-Iturbe, 1984) by estimating and comparing the parameters derived for each month and for the plain area, the mountain area and overall basin, respectively. The first-and second-order statistics derived from observed point gauge data were used to estimate the model parameters based on the Davidon-Fletcher-Powell algorithm of optimization. As a result of the study, we can find that the higher rainfall amount during summer is mainly due to the arrival rate of rain bands, mean number of cells per cluster potential center, and raincell intensity. However, other parameters controlling the mean number of rain cells per cluster, the cellular birth rate, and the mean cell age are found invariant to the rainfall amounts. In the application to the downstream plain area and upstream mountain area of the Han river basin, we found that the number of storms in the mountain area was estimated a little higher than that in the plain area, but the cell intensity in the mountain area a little lower than that in the plain area. Thus, in the mountain area more frequent but less intense storms can be expected due to the orographic effect, but the total amount of rainfall in a given period seems to remain the same.

Pharmacognostical Studies on the Folk Medicine 'JoRitDae' (민간약 조릿대의 생약학적 연구)

  • Korean folk medicine 'JoRitDae' has been used mainly to cure vomiting and cancer. With regard to the botanical origin of 'JoRitDae', it has been considered to designate the rhizome of Sasa species (Gramineae), but there was no pharmacognostical confirmation on it. To clarify the botanical origin of 'JoRitDae', the anatomical characteristics of the rhizome of Sasa species growing wild in Korea and Japan, Sasa borealis, S, kurilensis, S. quelpaertensis, S. charatacea and S. senanensis were studied. As a result, it was found the morphological criteria for the five Sasa species that could discriminate them by the number of cell layers in cortex, the ratio of thickness of cortex to rhizome radius, the number of cell layers in the vascular bundle sheath, the total number of vascular bundle and thickness of vascular bundle sheath. According to these criteria, it was clarified that the commercial folk medicine 'JoRitDae' was the rhizome of Sasa borealis.

Anti-inflammatory Activity of Chihyo-san to Protect Respiratory Tissues from Asthmatic Damage

  • The present study was carried out to investigate the effect of Chihyo-san (CHS) administration on asthma induced by Alum/OVA treatment in the mice. In CHS-treated animal group, lung weight, which was increased after asthma induction, was significantly decreased, and total number of cells in the lung, peripheral lymph node (PLN) and spleen tissue was significantly decreased in CHS-treated group compared to the asthma control group. The number of immune cells including natural killer (NK) cells in asthmatic animals was largely regulated by CHS treatment, showing a similar pattern as that of CsA-treated positive control group. Levels of mRNAs encoding inflammatory cytokines IL-5, IL-13, $TNF-{\alpha}$, and eotaxin were determined by RT-PCR in the lung tissue and showed decreases in CHS-treated group to the similar levels of CsA-treated control group, Histamine level in the serum was significantly lower in CHS-treated group than asthma-induced control group. Both haematoxylin and eosin staining and Masson's trichrome staining results showed decreased number of inflammatory cells, reduced immune cell infiltration, and normalized epithelial cell layering in the bronchial tissue of CHS-treated mouse group. Thus, the present findings suggest that CHS may be useful for protecting bronchial tissues from consistent inflammatory damages that occur in asthma patients.

Radiation Effect on Mouse Jejunal Crypt Cells by Single and Split Irradiation (단일조사와 분할조사시 마우스 공장 소낭선세포의 방사선효과에 관한 실험적 연구)

  • To determine the dose·survival and repair characteristics of the jejunal crypt cells, experimental study was carried out using total 70 mice. Single or split irradiations of 1,100 to 2,200 rad were delivered to whole bodies of $C_{57}$ BL mice, using a cesium 137 animal irradiator and those mice were sacrificed after 90 hours. The number of regenerating crypts per jejunal circumference was counted by a jejunal crypt cell assay technique and dose·response curve was measured. The results were as follows : 1. The average number of jejunal crypts per circumference in control group was 140. In a single irradiation group, the number of regenerated jejunal crypts was, 125, 56, 2 in each subgroup of 1,100 rad, 1,400 rad and 1,800 rad respectively. In split irraiation group, it was 105,44,2 in each subgroup of 1,400rad 1,800rad and 2,200rad respectively. 2. Mean lethal dose of mouse jejunal crypt cell was 167 and 169 rad respectively in a single and split irradiation. 3. Repair dose of sublethal damage was 280 rad. 4. Sublethal damage was completely repaired within 4 hours between the split dose of irradiation.

A Study on the Somatic Cells in Half Milk Samples of Dairy Goats (젖염소 분방유즙에서 체세포에 대한 연구)

  • In the present study, a total of 547 half milk samples were collected from 274 dairy goats to perform somatic cell counts (SCC) and California mastitis test (CMT). Milk smear was stained with Pyronin Y-Methyl Green stain were classified into either epithelial or blood cells, etc. Of the 547 halves the percentage of CMT negative milk samples were 86%. Among these, 58.2% were CMT negative with SCC<500,000/ml, while 27.8% were CMT negative with SCC>500,000 ml. As expected, CMT score increased with the increase of SCC. The number of epithelial cells decreased with the increasing number of somatic cells, while the opposite was observed with the number of blood cells. These results indicate that the critical point in milk quality & CMT should be considered on the false (pseudo-SC) SCC in dairy goat.

  • This study aimed at constructing a draft genome of the adult female worm Toxocara canis using next-generation sequencing (NGS) and de novo assembly, as well as to find new genes after annotation using functional genomics tools. Using an NGS machine, we produced DNA read data of T. canis. The de novo assembly of the read data was performed using SOAPdenovo. RNA read data were assembled using Trinity. Structural annotation, homology search, functional annotation, classification of protein domains, and KEGG pathway analysis were carried out. Besides them, recently developed tools such as MAKER, PASA, Evidence Modeler, and Blast2GO were used. The scaffold DNA was obtained, the N50 was 108,950 bp, and the overall length was 341,776,187 bp. The N50 of the transcriptome was 940 bp, and its length was 53,046,952 bp. The GC content of the entire genome was 39.3%. The total number of genes was 20,178, and the total number of protein sequences was 22,358. Of the 22,358 protein sequences, 4,992 were newly observed in T. canis. Following proteins previously unknown were found: E3 ubiquitin-protein ligase cbl-b and antigen T-cell receptor, zeta chain for T-cell and B-cell regulation; endoprotease bli-4 for cuticle metabolism; mucin 12Ea and polymorphic mucin variant C6/1/40r2.1 for mucin production; tropomodulin-family protein and ryanodine receptor calcium release channels for muscle movement. We were able to find new hypothetical polypeptides sequences unique to T. canis, and the findings of this study are capable of serving as a basis for extending our biological understanding of T. canis.

Study on Effect of Human Hair by Deer Antler Fermented Keratin Peptide (녹용 발효 케라틴 펩타이드에 의한 인체 모발 연구)

  • In this study, keratin peptides were produced through high-temperature anaerobic fermentation of keratin, a protein contained in deer antler, with Fervidobacterium islandicum AW-1, and factors related to human hair, confirming the possibility of keratin peptides as cosmetic ingredients. As a result of the cytotoxicity and proliferation of deer antler fermented keratin peptide according to the concentration in the dermal papilla cell line, cytotoxicity was not observed and the cell proliferation effect was shown. For human dermal papilla cells, statistically significant increasing in growth factors according to the deer antler fermented keratin peptide was determined possiblity of effects on hair growth. Cosmetic products containing deer antler fermented keratin peptides were manufactured and skin safety and anti hair loss efficacy clinical tests were conducted. As a result, after 12 weeks of use, the total number of hairs statistically significant increased compared to before using the product and the difference in total number of hairs compared to the control group was found. In conclusion, we suggest that the possibility of fermented deer antler keratin peptide as a cosmeceutical ingredient as well as a health functional food material was confirmed.

A Study on the Somatic Cells in Quarter Milk Samples of Holstein Cows with Suspected Mastitis (유방염으로 의심된 유우 분방유즙의 체세포 연구)

  • This study was conducted to survey the relationship between the somatic cell count (SCC), and California mastitis test (CMT) & mastitis. A total of 328 quarter milk samples were collected from 211 cows suspected to have mastitis; Both SCC and CMT were performed on the samples. Milk smear was stained with Broadhurst and Paley stain and the cells were classified into either epithelial or blood cells. Bacterial isolation was made and antimicrobial susceptibility was tested. Of the 328 quarters, 78 ($23.8{\%}$) were CMT negative with SCC <750,000/ml. As expected, CMT score increased with the increase of SCC. The number of epithelial cells decreased with the increasing number of somatic cells, while the opposite was the case with the number of blood cells. The critical point was when the SCC reached 1,000,000/ml. Up to 1,000,000 cells/ml, the number of epithelial cells was greater than that of blood cells. The results indicate that when epithelia:blood cell ratio is 58.1:41.9, the milking line should be checked and bacterial isolation be performed on the samples in order to identity mastitis.

The Effect of Guibitang(歸脾湯) on the Ovarian Functions and Differential Gene Expression of Caspase-3, MAPK and MPG in Female Mice (귀비탕(歸脾湯)이 자성생쥐의 생식능력과 Caspase-3, MAPK, MPG유전자 발현에 미치는 영향)

  • Purpose: To evaluate the effect of administration of Guibitang on ovarian functions and differential gene expressions related cell viabilities caspase-3, MAPK and MPG in female mice. Methods: We administered the Guibitang to 6-week-old female ICR mice for 4, 8, or 12 days. Then, the female mice were injected PMSG and hCG for ovarian hyperstimulation. The mice divided into 3 different groups for each experiment. To compare the differences, we set a control group treated with plain water at the same volume by the same way. Results: administration of Guibitang, the mean number of total ovulated oocytes and the number of morphologically normal oocytes increased significantly compared to control group. And the rates of blastocyst formation from 2-cell stages alsa increased significantly compared to control group. Capase-3 gene expression which is known to maker gene for cell apoptosis were significantly lower than that of control group. And MAPK and MPG gene expressions for cell viability and DNA repair were same that of control group. Conclusion: From our results suggested that the medication of Guibitang has beneficial effect on reproductive functions of female mice via prevention of cell apoptosis and DNA damaging and promotion of cell proliferation.
