경남 지역 학교 급식소의 미생물학적 오염도 평가: COVID-19 이후의 위생관리 현황 조사 (Microbiological Contamination Assessment of School Cafeterias in the Gyeongnam Area: Investigation of the Status of Hygiene Management since COVID-19)

  • School meals are prepared based using the HACCP system, which is designed to ensure the physical and mental health of students. However, operational recommendations in school cafeterias have changed due to COVID-19 and include delays in serving time to restrict the number of meals and the installation of screens to prevent droplet transmission. Unfortunately, these changes may have detrimentally affected hygiene practices and generated new food poisoning sources. This study aimed to determine the hygiene state of school cafeterias in the Gyeongnam area from December 2020 to September 2021 based on the monitoring of total aerobic and coliform bacteria. Kitchen floors were the most contaminated areas with an average number of total aerobic bacteria of 4.3 log CFU/100 cm2, whereas counts in dining areas were relatively low at 0.1~3.5 log CFU/100 cm2. Newly installed partitioned areas had the lowest contamination level of 0.1~2.8 log CFU/100 cm2. Escherichia coli was not detected on any surface, while coliform was detected on workbenches and floors. In conclusion, hygienic practices appear to have been adequately managed in school cafeterias despite COVID-19 driven changes. Nonetheless, continuous monitoring is recommended to ensure prompt response to changing environments.

Variation in adhesion of Streptococcus mutans and Porphyromonas gingivalis in saliva-derived biofilms on raw materials of orthodontic brackets

  • Objective: To evaluate differences in the adhesion levels of the most common oral pathogens, Streptococcus mutans and Porphyromonas gingivalis, in human saliva-derived microcosm biofilms with respect to time and raw materials of orthodontic brackets. Methods: The samples were classified into three groups of bracket materials: 1) monocrystalline alumina ceramic (CR), 2) stainless steel metal (SS), and 3) polycarbonate plastic (PL), and a hydroxyapatite (HA) group was used to mimic the enamel surface. Saliva was collected from a healthy donor, and saliva-derived biofilms were grown on each sample. A real-time polymerase chain reaction was performed to quantitatively evaluate differences in the attachment levels of total bacteria, S. mutans and P. gingivalis at days 1 and 4. Results: Adhesion of S. mutans and P. gingivalis to CR and HA was higher than the other bracket materials (SS = PL < CR = HA). Total bacteria demonstrated higher adhesion to HA than to bracket materials, but no significant differences in adhesion were observed among the bracket materials (CR = SS = PL < HA). From days 1 to 4, the adhesion of P. gingivalis decreased, while that of S. mutans and total bacteria increased, regardless of material type. Conclusions: The higher adhesion of oral pathogens, such as S. mutans and P. gingivalis to CR suggests that the use of CR brackets possibly facilitates gingival inflammation and enamel decalcification during orthodontic treatment.

Rotifer, Brachionus plicatilis, 성장을 위한 광합성세균의 첨가 효과와 넙치, Paralichthys oliraceus, 자어에 대한 먹이효율 (Effect of Photosynthetic Bacterial Addition to Chlorella or ${\omega}-Yeast$ on Growth of Rotifer, Brachionus plicatilis, and its Dietary Value for Flounder, Paralichthys olivaceus, Larvae)

  • Rotifer의 대량배양에는 해산 chlorella가 가장 적합하나 비용이 높고, 유지효모는 경제적이 긴 하나 먹이효율이 낮은 문제점이 있다. 이와 같은 문제점을 개선하기 위하여 본 연구에서는 광합성세균 (Rhodopseudomonas capsulata)의 첨가 효과를 조사 하였다. Rotifer 한 개체당 1일 먹이량으로 10,000, 50,000, 100,000, 200,000, 300,000세포의 Chlorella ellipsoidea를 공급하고 각 chlorella 수의 10, 20, 30배의 광합성세균을 첨가한 결과 200,000세포에 광합성 세균 20배 ($4{times}10^6 cell$)를 공급한 것이 가장 성장이 높았으나 경제적인 측면에서는 chlorella 100,000세포에 광합성세균 30배를 공급하는 것이 더 효과적이었다. 또, rotifer 1개체당 1일 먹이량으로 유지효모를 100,000, 200,000, 300,000세포를 공급하고 각 유지효모 수의 10, 20, 30배의 광합성세균을 첨가한 결과 200,000세포에 광합성세균 20배를 첨가한 실험구에서 rotifer의 성장이 가장 높았다. Chlorella와 유지효모를 각각 200,000 세포씩 그리고 chlorella와 유지효모에 광합성세균을 20배의 농도로 첨가하여 rotifer를 배양하여 넙치 자어를 사육한 결과 chlorella에 광합성세균을 첨가한 실험구의 생존율이 $96.8{\%}$로 가장 높았으며 성장의 경우도 광합성세균을 첨가한 실험구에서 높은 경향을 보였다. 또 광합성세균을 첨가한 실험구에서의 넙치 자어는 총지질과 EPA와 DHA의 함량이 광합성세균을 첨가하지 쟈은 실험구에서보다 높게 나타났다. Artemia nauplius를 6시간동안 광합성세균으로 영양강화하여 넙치 치어를 20일간 사육한 결과 영양강화한 것은 하지 않은 것보다 생존율과 성장이 높았으며 광할성세균의 최적영양걍화농도는 $ml 당 2{times}10^7 cells$로 나타났다. Chlorella와 유지 효모에 광합성 세균의 첨가는 rotifer의 성장과 자치어의 생존율과 성장에 효과적이었다. 그러나 과다한 양의 광합성세균은 오히려 negative effect가 있는 것으로 나타났다.

만경.동진강 염하구에서 종속영양성 및 혼합영양성 미소편모류의 수도(數度)와 박테 리아 섭식 (Abundances and Bacterivory of Heterotrophic and Mixotrophic Nanoflagellates in and Estuarine System of the Mankyung and Dongjin Rivers, Korea)

  • 만경.동진강 염하구에서 종속영양성 및 홉합영양성 미소편모류(Nanoflagellates) 의 분포와 이들에 의한 박테리아섭식의 생태적 중요성을 추정하기 위해 1993년 10부터 1995년 3월 까지 총 4차에 걸쳐 풍속영양성 및 자가영양성 미소편모류의 수도와 FLB(fluorescently labeled bacteria)를 이용하여 박테리아 섭식률을 측정하였다. 연 구기간 동안 종속영양성 미소편모류와 자가영양성 미소편모류의 수도는 각각 438-4,149 개체 ml/SUP -1/(평균 2,145 개체 ml/SUP -1/, n=20), 971-4,935 개체 ml/SUP -1/(평균 2,226 개체 ml/SUP -1/,n=20)의 변화폭을 보였으며, 이들 편모류는 전반적으로 서로 유사한 수도를 나타냈다. 박테리아의 주요 섭식자로 알려진 종속영양 성 미소편모류와 엽록체가 있는 자가영양성 미소편모류의 수도는 보고 테리아의 수도 와 모두 유의성 있는 양의 상관관계(각각 $r^2$=0.51)과 $r^2$=0.47, p<0.05)를 보였다. 혼합영양성 미소편모류 군집의 크기는 적어도 자가영양성 미소편모류 군집의 4.23%에 달하는 것으로 추정되었다. 개체별 박테리아 섭식률(individual predation rate)은 종 속영양성 미소편모류가 2.2-14.2 박테리아 미소편모류/SUP -1/h/SUP -1/(평균 3.7 박 테리아 미소편모류/SUP -1/h/SUP -1/,n=16), 혼합영양성 미소편모류가 1.6-9.7 박테리 아 미소편모류에 의한 박테리아 섭식에서 30-69%를 차지하여, 박테리아 섭식자료서의 역할이 중요함을 알 수 있었다. 박테리아 이차생산력에 대한 총 미오편모류의 박테리 아 섭식률의 비는 0.06-1.23으로 넓은 범위를 보였으나, 미소편모류의 수도가 매우 낮 은 1994년 6월의 경우(8$\pm$1%,써 =4)를 제외하면 총 미소편모류의 섭식률은 박테리아 이차 생산량의 상당 부분(66$\pm$37%,n=9)에 해당하였다. 따라서, 만경.동진강 염하구에 서 종속영양성 및 혼합영양성 미소편모류는 박테리아의 주요 섭식자로서 박테리아 이 차생산량을 상위 영양단계의 동물플랑크톤에 전달하는 연결자 역할을 할 것으로 보여 진다.

Comparison of Saponin Content and Antioxidant Effect depending on the Processing Method of Codonopsis lanceolata

  • Codonopsis lanceolata, called deodeok in Korean, has been verified for various effects, including anti-aging and anti-inflammatory effects and insomnia improvement, and is one of the healthy foods that Koreans enjoy eating. In this study, the saponin content, lansemaside A content, and total saponin content of deodeok were analyzed using high-pressure sterilization and solid fermentation by mixed lactic acid bacteria. The antioxidant effect was compared to determine improved processing methods of deodeok. The lansemaside A content of deodeok samples depending on the preprocessing methods was analyzed: 2,594.10 mg/kg for dried deodeok, 2,100.93 mg/kg for steamed deodeok, and 1,151.31mg/kg for fermented deodeok. The total saponin content was found to be 7,209 mg/kg for dried deodeok and 8,605 mg/kg for steamed deodeok, showing a high saponin content. The total polyphenol content was highest for dried deodeok, steamed deodeok, and fermented deodeok. As for the antioxidant effect, it was analyzed that the effect of dried deodeok was the highest, followed by steamed deodeok and fermented deodeok; Dried deodeok had the highest value in total polyphenol content, not in total saponin content, which is considered to have a positive influence on its antioxidant effect. The content of lansemaside A was the highest for dried deodeok. When fermented deodeok is consumed, more beneficial effects on health can be expected by ingesting it with lactic acid bacteria cultured using saponins and polyphenols. Therefore, this study suggests the possibility of manufacturing products suitable for the needs of consumers, such as the flavor of deodeok, according to the processing methods.

비배양식 총세포수 평가를 통한 병물 위생관리 연구 (Study on Hygiene Management of Bottled Water through Non-culture-based Total Cell Count)

  • The demand for bottled water in South Korea is steadily increasing, but there are challenges regarding water sources and violations of water quality standards. Consumers struggle to identify products that do not meet these standards, highlighting the need for improved water management. This study aims to investigate the use of flow cytometry to identify microbial behavior in bottled water. Twelve different bottled water brands were selected for this study. A novel non-culture-based analysis method called total cell count via flow cytometry was utilized, which is not commonly used to assess drinking water quality. This method was compared to conventional culture-based methods for heterotrophic plate count and E. coli experiments, in order to introduce new indicators for hygiene management. Adenosine triphosphate analysis was also conducted to assess cell activity, and total organic carbon was measured to determine the presence of organic matter. The total cell counts varied among the different bottled water brands. The adenosine triphosphate levels ranged from 37.1ng/L to 221.7ng/L, while the total organic carbon ranged from 0.4 to 0.6 mg/L. Furthermore, E. coli was not detected in any of the bottled waters, and with the exception of two cases, the levels of heterotrophic bacteria did not exceed the drinking water standard of 100 CFU/mL. This study demonstrated a correlation between total cell count and heterotrophic plate count, suggesting that non-culture-based analysis could be valuable in promptly assessing microbial contamination, in contrast to the conventional methods that require approximately 48 hours for incubation.

우포늪, 순천만, 서해 갯벌에서부터 분리한 황산염/황-환원 세균의 특성 분석 (Isolation and Characterization of Sulfate- and Sulfur-reducing Bacteria from Woopo Wetland, Sunchun Bay, and Tidal Flat of Yellow Sea)

  • 황화합물은 혐기성환경에서 혐기성호흡을 위한 매우 중요한 전자수용체이다. 본 연구를 통하여 한국의 다양한 습지에서 배양을 통한 황산염/황-환원세균의 특성연구를 실시하였다. 이를 분리하기 위하여 혐기성 roll tube법을 통해 총 11개의 순수 배양체를 확보하였다. 16S rDNA를 이용한 계통분석 및 상동성 분석을 실시하여 Desulfovibrio 속의 세균 8종, Sulfurospirillum 속의 세균 2종, Desulfitobacterium 속의 세균 1종을 얻을 수 있었다. 이들 황산염/황-환원세균은 모두 lactate와 pyruvate를 전자공여체로 이용하였으며, sulfite and thiosulfate를 전자수용체로 이용할 수 있었다. 앞으로, 다양한 전자공여체와 배양조건을 통하여 유용한 절대혐기성 황산염/황-환원세균의 생물자원 확보에 기여할 것으로 기대된다.

시판 식물성 식품의 오염지표세균 분포 및 저장온도, 기간별 오염지표세균의 변화 (Distribution of Indicator Organisms and Influence of Storage Temperature and Period in Commercial Plant Food)

  • There were few data for the distribution of the indicator organisms in the commercial plant foods, and for the normal flora and for the foodborne agents within the country. First of all it must be investigated the distribution of the indicator organisms. And also it is very important to prepare the sanitation criteria for the plant foods through the microbiological examination and the investigation of tendency to change of the indicator organisms according to the storage temperature and period. The average number of total viable counts for grains was 2.9$\times$105/g, psychrophilic bacteria 2.9$\times$105/g, heterotrophic bacteria 3.1$\times$105/g, heat-resistant bacteria 2.1$\times$103/g, Pseudomonas aeruginosa 23/g. That for beans was 6.3$\times$102/g, psychrophile 34/g, heterotroph 1.7$\times$102/g. That for sesames was 1.4$\times$105/g, coliform 350/g, psychrophile 7.4$\times$104/g, heterotroph 5.8$\times$104/g, Pseud. aeruginosa 2.3$\times$103/g. heat-resistant bacteria 150/g. That for potatoes was 2.0$\times$107/g, coliform 5.0$\times$104/g, psychrophile 1.8$\times$107, heterotroph 1.4$\times$107/g, heat-resistant bacteria 3.3$\times$104/, Staphylococcus 2.7$\times$105/g, fecal streptococcus 4.5$\times$103/g, Pseud. aeruginosa 7.0$\times$103/g. That for mushrooms was 1.2$\times$108/g, psychrophile 9.4$\times$107/g, heterotroph 1.0$\times$109/g, heat-resistant bacteria 1.6$\times$105/g, Pseud. aeruginosa 1.3$\times$103/g. That for vegetables was 5.9$\times$1011/g, coliform 1.8$\times$106g/, Staphylococcus 1.1$\times$1012/g, heterotroph 8.4$\times$1011/g, heat-resistant bacteria 7.6$\times$106/g, Staphylococcus 1.1$\times$107/g, fecal streptococcus 1.1$\times$104/g, Pseud. aerugniosa 5.2$\times$104/g. That for nuts 3.9$\times$104/g, coliform 3.9$\times$103/g, psychrophile 4.0$\times$104/g, heterotroph 3.2$\times$104/g, heat-resistant bacteria 400/g. In commercial grains and beans, SPC, psychrophile, heterotroph and heat-resistant bacteria stored at 1$0^{\circ}C$, 2$0^{\circ}C$, 3$0^{\circ}C$ were constant. Staphylococcus, coliform, Pseud. aeruginosa were decreased a little n grains, but were not detected in beans. In mushrooms, all indicator organisms were increased as time goes on and were increased rapidly at 2$0^{\circ}C$. In sesames, coliform was not detected at all temperature. psychrophile was increased for 7 days, the others were constant. In potatoes, SPC, psychrophile, heat-resistant bacteria, heterotroph had a tendency to increase and the others were constant. In vegetables, indicator organisms were had a tendency to increase, psychrophile, heterotroph were rapidly increased after 7 days. In nuts, SPC, coliform, psychrophile heterotroph, heat-resistant bacteria, Pseud. aeruginosa were constant, staphylococcus and fecal streptococcus were not detected.

해빙기 바이칼호에서 부유세균과 Aggregates에 부착한 세균의 군집구조 (Bacterial Community of Free-living and Aggregated Bacteria at Thawing Period in Lake Baikal)

  • 러시아 바이칼호에서 해빙기에 부유세균과 aggregates에 부착한 세균의 군집구조를 FISH (fluorescent in situ hybridization)방법으로 0 m부터 250 m수심에서 비교 분석하였다. 조사대상은 Eubacteria에 속하는 세균과 class Proteobacteria에 속하는 세균 중 $\alpha$-, $\beta$-, $\gamma$ -group과 Cytophaga-Flavobacterium group,그리고 Planctomycetales였다. 부유세균의 수는 $0.2{\times}10^6\cells{\cdot}ml^-1 to 3.2{\times}10^6 \cells{\cdot}ml^-1$범위였으며, 수심이 깊어질수록 감소하였다. Aggregate에 부착한 세균은 부유세균과 반대로 수심이 깊어질수록 증가하였고, 개체수의 범위는 0.4~$3.3{\times}10^4$ $cells{\cdot}ml^-1$ 이였다. 총세균수에 대한 Eubacteria 수의 비율은 부유세균의 경우 52.3~74.1%, aggregates에 부착한 세균은 39.6~66.7%로 부유세균보다 부착세균에서 그 비율이 낮았으며, 세균의 군집구조 분석 결과에서도 부유세균과 aggregates애 부착한 세균의 군집구조가 다른 양상으로 나타났다. 특히 두 세균의 군집구조는 식물플랑크톤이 밀집해 있는 25 m 수심에서 급격히 변화하여 식물플랑크톤이 부유세균과 부착세균의 군집의 변화에 밀접한 관련이 있는 것으로 확인되었다, Aggregates에 부착한 세균 군집은 수심에 따라 매우 특이한 변화 양상을 나타내었다. $\beta$-proteobacteria group은 수심이 깊어지면서 그 비율이 높아져, 100m에서는 $\beta$-group이 총세균수는 51.8%를 차지하였으나, 250m에서는 $\gamma$-group이 43.8%를 차지하여, 급격하게 우점 group이 변화하였다. 그러나, 부유세균에서는 전혀 다른 군집 구조를 이루고 있었다. 이러한 결과에서 aggregates에 부착한 세균은 부유세균과는 다른 다양성을 이루고, 다른 천이과정을 거치는 것으로 확인되었다.

Effect of Bamboo (Pseudosasa japonica Makino) Leaves on the Quality and Sensory Characteristics of Dongchimi

  • Effect of covering various levels(0, 1, 3, 5 and 7% per Chinese radish weight) of bamboo(Pseudosasa japonica Makino) leaves on the surface of Dongchimi was studied in the course of fermenting at 1$0^{\circ}C$ up to 75 days. Physicochemical, microbiogical, and sensory characteristics of Dongchimi were analyzed at regular intervals of 5 to 7 days during fermentation. Both the degree of pH drop from the initial 6.47~6.54and increase total acid content from the initial 0.004%, with the accumulation of organic acids tended to be more gradual depending upon the amounts of bamboo leaves covered. The increases of total solid content and turbidity were also suppressed in similar patterns, notably after 13 days of fermentation. The growth of total bacteria(1.5$\times$10$^4$cfu/ml, initial)was partly inhibited while that of lactic acid bacteria(1.8$\times$10$^4$cfu/ml, initial)was favorably encouraged by the presence of bamboo leaves. As a result of sensory evaluation, Dong-chimi covered with 1 and 3% bamboo leaves on showed the higher scores significantly(p<0.05, p<0.01, p<0.001) in overall acceptability till 39 days. But after 61 days, those covered with 3 and 5% bamboo leaves were rather preferable than any other one.
