Plant growth promoting rhizobacteria (PGPR) are known to confer disease resistance to plants. Bacillus sp. JS demonstrated antifungal activities against five fungal pathogens in in vitro assays. To verify whether the volatiles of Bacillus sp. JS confer disease resistance, tobacco leaves pre-treated with the volatiles were damaged by the fungal pathogen, Rhizoctonia solani and oomycete Phytophthora nicotianae. Pre-treated tobacco leaves had smaller lesion than the control plant leaves. In pathogenesis-related (PR) gene expression analysis, volatiles of Bacillus sp. JS caused the up-regulation of PR-2 encoding ${\beta}$-1,3-glucanase and acidic PR-3 encoding chitinase. Expression of acidic PR-4 encoding chitinase and acidic PR-9 encoding peroxidase increased gradually after exposure of the volatiles to Bacillus sp. JS. Basic PR-14 encoding lipid transfer protein was also increased. However, PR-1 genes, as markers of salicylic acid (SA) induced resistance, were not expressed. These results suggested that the volatiles of Bacillus sp. JS confer disease resistance against fungal and oomycete pathogens through PR genes expression.
Leading soybean cultivars such as Kwanggyo, Yugu No.3, Dongbugtae, Gangrim, and Eundaedu were heavily diseased by a virus in Korea. The disease was most severe in the northern provinces where soybean mosaic virus also occurrs, but the disease has also been observed in other provinces where soybean diseases are less prevalent. The disease symptoms were similar to bud blight caused by tobacco ringspot virus; but this was not confirmed in inoculation tests on indicator plants and serological experiments. There were some differences in varietal susceptibility to the disease, with symptom variation depending on the soybean cultivar and source of inoculm. Disease symptoms on infected soybean plants were mottling and necrosis. The present results, therefore, indicate some strains of SMV or a mixture of legume viruses may or may not be responsible for the disease.
This study was carried out to identify the protein patterns and to estimate molecular weights of the ginseng and other plants. Polyacrylamide gradient 2-30% gels for identification of protein patterns and 11% SDS polyacrylamide gels for molecular weight estimation of various plants were used. Proteins extracted with distilled water and phosphate buffer were most clearly separated. Although minor bands were a little different according to kinds of buffer, major bands showed similar pattern. Major protein bands were two bands of 45,000, 66,000 in Panax ginseng, 3 bands of 32,000-39,000 in Codonopsis lanceolata and one double band around 39,000 kd in Platycodon grandiflorum.
Experiment was conducted to get the information about physico-chemical properties of flue-cured tobacco on the degree of maturity cultivated in paddy-upland rotated field, and compared to upland ones. For the samples of this experiment, 3~4 leaves at each stalk position were harvested from the bottom of plants. Physico-chemical properties of cured leaves were determined from the samples collected at weekly intervals, and of obtained from 4 stalk positions. For the degree of maturity, harvested leaves were separated with visual characters into four classes such as immature, mature, ripe, and mellow. Regardless of stalk position, the order of shrinkage rate with length and width of leaves was mellow> immature> ripe> mature, and ripe leaves from paddy field showed higher shrinkage rate than those of upland. Nicotine and total nitrogen contents were decreased with the degree of maturity while reducing sugar content were showed a reverse tendency. Ripe leaves from paddy field had lower reducing sugar contents, comparing with upland tobacco. Filling capacity of cured leaves from paddy field was decreased with degree of maturity, but there was no difference between upland and paddy tobacco. Shatter index was increased in the oeder of immature > mellow > mature > ripe. Chemical components of cigarette smoke from paddy field tobacco were little higher in $CO_2$ total particulate matter and tar contents, while combustibility was little lower than that of upland tobacco. It was also evaluated that paddy field tobacco was unfavorable for the non-volatile organic and higher fatty acids contents comparing with upland tobacco.
Ginseng fields abolished by potato rot nematode (Ditylenchus destructor) (heavily infested with the nematode) were treated with selected nematicides after preplanting treatment with fumigants or non fumigant chemicals in the fall of 1982. Suppression of the nematode populations was noted in the fields fumigated by cylon (Cy) and treated with ethoprop (E) or triazophos (T). Also in these treatments, high percentages of shoot sprouting and low nematode damages were observed. In the fields fumigated by Cy, subsequent treatments with nonfumigant nematicides such as E, aldicarb (A), phenamiphos (Ph), oxamyl (O) and carbofuran (C) during cultivation had no effect on the significant decrease of the nematode, compared with Cy alone. No differences in shoot sprouting and symptom appearance were observed; however, survivalities of plants and root weights of 4-year old ginseng were high in A and E or E following preplanting treatment with E. Correlation coefficients of the nematode densities to host responses such as sprouting of shoots, symptom appearance, survivality of plants and weight of 4-year old ginseng roots were significant at and mostly before and after the time periods of survey.

Antioxidative responses of transgenic tobacco plants expressing both superoxide dismutase (SOD) and ascorbate peroxidase (APX) in chloroplasts was investigated with several herbicides. In greenhouse test, tolerance of SOD/APX-overexpressed tobacco (CA) to photosystem (PS) I inhibitor paraquat was increased by about 40%. However, any response differences between CA and wild type (WT) tobacco was not observed in a treatment with PS II inhibitors (bromoxynil, diuron and bromacil), chlorophyll biosynthesis inhibitor(oxyfluorfen), carotenoid biosynthesis inhibitor (fluridone) and 5-enolpyruvylshikimate-3-phosphate (EPSP) synthase inhibitor (glyphosate). This tendency was also similar in the growth chamber test of low light intensity, using paraquat and diuron. That is, increased antioxidant activity of CA was shown only in paraquat treatment. When paraquat was foliar-treated to 6 to 9-leaf stage plant, the third to fourth placed leaf from shoot tip showed relatively higher antioxidant activity. Ascorbate supplemented to paraquat solution alleviated the phytotoxicity with a similar range in both CA and WT. In conclusion, CA specifically responded to oxidative stress induced by paraquat among tested herbicides in a whole plant assay.

Recombinant plasmid, pBKH4, containing NPT II and P35S-BcHSP17.6 was constructed by ligation of Bum H I -digested pBKSl-l and BcHSP 17.6 (thermotolerance gene) 6om pBLH4. The tobacco leaf disc was cocultivated with transformed Agmbacterium tumefaciens bearing pBKH4 for 24 hours and transformed shoots were selected on MS-n/B medium containing $100\;{\mu\textrm{g}}/ml$ of kanamycin. Heat-killing temperature of Nicotima tabacum was $50^{\circ}$ for >15min, and transformed tobacco plants with BcHSP17.6 cDNA exhibited thermotolerance at the heat-killing temperature. The transgenic plants were analyzed by Southern blot hybridization with the probe of ${\alpha}^{_32}P$ labelled BcHSP17.6 cDNA. Transcription and expression level of BcHSP17.6 cDNA were also continued by Northern blot analysis and Ouchterlony double immunodiffusion assay. In this study, we suggest that the BcHSP17.6 cDNA introduced to tobacco plant is related to thenuoto-lerance and 17.6-kD LMW HSP acts as a protector from heat damage in plants.

A mutant of Potato vims Y (PVY) was occurred in PVY resistance flue-cured tobacco breeding line KF0402 $(TC1146{\times}KF117)$ showing vein necrosis at Suwon in Korea. This isolate, PVY-SWM, was differentiated from other PVY based on biological properties and nucleotide sequence analyses of coat protein gene. PVY-SWM caused typical symptoms on 21 indicator plants as compared to the PVY-TOJC37. Remarkably, the PVY-SWM induced distinctly different symptom of systemic vein necrosis on tobacco cultivars V.SCR, PBD6, TN86, TN90, Virgin A Mutant (VAM), Wislica, NC744, KB108 and KB111, which were reported to have the recessive potyvirus resistance gene va. In RT-PCR assays with specific primers for detection of PVY, a single band of about 800bp in length was produced. The amplified DNA was cloned and the nucleotide sequence was determined. The coat protein gene of PVY-SWM showed 88.4%-99.0% and 92.5%-98.5% identities to the 12 different PVY isolates of Genbank Database at the nucleotide and amino acidi respectively. Multiple alignments as well as cluster dendrograms of PVY-SWM isolate revealed close phylogenetic relationship to the $PVY^{NTN}$ subgroup.
Ground-based remote sensing can be used as one of the non-destructive, fast, and real-time diagnostic tools for predicting yield, biomass, and nitrogen stress during growing season. The objectives of this study were: 1) to assess biomass and nitrogen (N) status of tobacco (Nicotiana tabacum L.) plants under N stress using ground-based remote sensors; and 2) to evaluate the feasibility of spectral reflectance indices for estimating an application rate of N and predicting yield of tobacco. Dry weight (DW), N content, and N uptake at the 40th and 50th day after transplanting (DAT) were positively correlated with chlorophyll content and normalized difference vegetation indexes (NDVIs) from all sensors (P<0.01). Especially, Green NDVI (GNDVI) by spectroradiometer and Crop Circle-passive sensors were highly correlated with DW, N content and N uptake. The yield of tobacco was positively correlated with canopy reflectance indices measured at each growth stage (P<0.01). The regression of GNDVI by spectroradiometer on yield showed positively quadratic curve and explained about 90% for the variability of measured yield. The sufficiency index (SI) calculated from data/maximum value of GNDVI at the $40^{th}$ DAT ranged from 0.72 to 1.0 and showed the same positively quadratic regression with N application rate explaining 84% for the variability of N rate. These results suggest that use of reflectance indices measured with ground-based remote sensors may assist in determining application rate of fertilizer N at the critical season and estimating yield in mid-season.
This experiment was conducted to study the nature of gene effects for the leaf characters in flue-cured tobacco. The genetic populations were derived from crosses between NC 2326 and Mc Nair 373, and NC 628 and DG-72. The generation means experiment Included the Pl, P2. Fl, F2, Bl and B2, which were frown at Taegu Experiment Station, Korea Ginseng & Tobacco Research Institute in 1984. Seedlings were transplanted to the field in a randomized block design with 3 replications. In each block, parental and Fl Plots contained 15 plants in a single row, F2, Bl and B2 plots being composed of 75 plant, in 5 rows. Leaf characters were measured of largest (middle leaf) and 5th leaf(top leaf) from the top after topping. Measurements of the length and width of leaf were obtained from the fresh the middle and top leaves, and weight of leaf, weight and width of midrib were from the satrap leaf after curing. Estimates of additive and dominance genetic variance were analyzed according to Gamble's biometrical model. The results obtained are as follows: 1. The additive gene effects were significant and larger than the dominance gene effects for all leaf and midrib characters in both stalk positions. 2. The dominance gene effects were significant for the length and width of leaf, and weight of midrib in the middle leaves. 3. The digenic epistatic effects were significant for the length and width of leaf in both stalk positions. 4. The additive gene effects were larger in the top than in the middle leaves and midrib characters.
