Effect of Organic Gemanium, Oligosaccharide and Starters on Fermentation of Fresh Kimchi Juice (김치즙액의 발효에 미치는 유기게르마늄, 올리고당 및 Starter의 영향)

  • Changes in pH, titratable acidity, CO$_2$production, reducing sugar, and lactic acid bacteria of fresh kimchi juice supplemented with combinations of 3 lactic acid bacteria and germanium(Ge)-132 or fructooligosaccharide(FO) during fermentation at 30$^{\circ}C$ were investigated to assess the potential for extending the shelf life and enhancing the functional properties in kimchi. In kimchi juice containing Ge-132, sample(I) (inoculated with a mixture of bacteriocin-producing SNF-13 strain and E. faecium) exhibited that the amounts of organic acid and evolved CO$_2$gas were lower than those of the other starter samples(II-IV). The growth of lactic acid bacteria naturally present in kimchi juice, particularly Lb. plantarum and Leu. mesentroides, may be inhibited due to competition of the isolated SNF-13 strain and E. faecium by Ge-132. During fermentation of kimchi juice containing FO sugar, the contents of organic acid and evolved CO$_2$gas On juice broth with 4 starters were predominantly higher than those of control and Ge-132 groups, and then the growth of lactic acid bacteria originated from kimchi ingredients was thought to be markedly accelerated. Our results indicated that functional properties like the extension of shelf life and increase of biological activity in kimchi were enhanced by adding Ge-132 and bacterium-producing lactic acid bacterium, which were resistant to organic acid and stimulated by Ge-132.

Fermentation of onion extract by lactic acid bacteria enhances its physicochemical properties (유산균에 의한 양파 착즙액의 발효효과와 이화학적 특성)

  • This study aimed to improve the physiological activity of onion juice via lactic acid bacterial fermentation. Seven types of lactic acid bacteria were used for the fermentation of onion juice. The pH and sugar content of the onion juice decreased, while its titratable acidity increased after lactic acid bacteria fermentation, and the cell count of lactic acid bacteria was 7.31-10.40 log CFU/mL. The total free sugar content decreased, while the total organic acid content increased in the fermented onion juice. Quercetin content of the fermented juice was 0.13-0.53 mg/kg. The total polyphenol and flavonoid contents increased after fermentation. Additionally, the 2,2-diphenyl-1-picryl-hydrazyl-hydrate free radical and 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonate) radical cation scavenging activities were increased by fermentation. Overall, lactic acid bacteria fermentation of onion juice enhanced its physiological activity. Based on these findings, Bifidobacterium breve KCTC 3441 was selected as the onion juice fermentation strain.

The Effect of Mugwort Extract on the Characteristics of Curd Yogurt (쑥 추출물의 첨가가 요구르트 특성에 미치는 영향)

  • Changes in titratable acidity, viable cells and sensory properties of curd yogurt during lactic fermentation in milk added with mugwort extract have been studied. Milk added with 5%, 10%, 15% or 20% of mugwort extract was fermented with single or mixed culture of Lactobacillus bulgaricus and Streptococcus thermophilus for 24 hours. Addition of mugwort extract markedly stimulated the acid production and propagation of lactic acid bacteria. Among the organism tested, the mixed culture of Lactobacillus bulgaricus and Streptococcus thermophilus produced the highest amount of acid, and Lactobacillus bulgaricus showed the highest number of viable cell counts. Sensory property of curd yogurt added with mugwort extract was evaluated as better than reference sample(milk yogurt). The sample prepared by fermenting milk containing mugwort extract at 5% level with Lactobacillus bulgaricus showed the most favorite characteristics.

Effects of Coagulants and Soaking Solutions of Tofu (Soybean Curd) on Extending its Shelf Life (두부의 저장에 미치는 응고제와 침지액의 효과)

  • In order to improve the shelf life of Tofu, the effect of calcium chloride or acetic acid as coagulants were investigated for microbial and physicochemical changes during the storage in different kinds of soaking solution. The soaking water of Tofu prepared from calcium chloride was found to be spoiled after 15-17 hours of storage at $30^{\circ}C$, by reaching the bacteria count to 10million per ml. The shelf life of the Tofu prepared from acetic acid was better than those prepared by calcium chloride. For the effect of soaking solution on storage life, soaking Tofu in 0.1% acetic acid was found to be more effective than soaking in other solutions of 3% NaCl or 0.1% K-sorbate. The optical density of soaking solution of Tofu increased in proportion to spoilage, and in case of soaked Tofu in 3% sodium chloride and 0.1% K-sorbate solution, it greatly increased. Titratable acidity, and amino nitrogen contents in soaking solution increased as spoilage of Tofu progressed, but pH decreased the early period, one or two days, of stored Tofu and then increased.

Assessment of Characteristics and Functional Properties of Lactobacillus Species Isolated from Kimchi for Dairy Use

  • The objective of this study was to identify lactic acid bacteria (LAB) isolated from kimchi and to evaluate its characteristics and functional properties for application in fermented dairy products as a probiotic or commercial starter culture. Eight stains isolated from kimchi were selected through an investigation of phenotypic characteristics. Two strains (DK211 and DK303) were identified as Lactobacillus plantarum, another two (DK207 and DK215) as Lactobacillus paracasei, and one (DK301) as Lactobacillus sakei. The remaining three strains were identified as species of Weissella. All selected Lactobacillus strains had acid and bile tolerance, even though there was wide variation in the ability of each strain. DK303 showed a remarkably higher proteolytic activity. There were no significant differences in β-galactosidase activity among the tested strains, except that DK301 showed no activity. Auto-aggregation varied between 82.1 and 90.0%, and hydrophobicity values ranged from 0.5 to 51.6%.The strongest auto-aggregation and hydrophobicity were observed in DK211. All selected strains showed better 1,1-diphenyl-2-picrylhydrzyl (DPPH) scavenging activity than commercial strains. DK211, DK215, DK301, and DK303 had effective inhibitory activity against all pathogens tested except E. coli. When selected strains were used for yogurt preparation as a single starter culture, the time required to reach target titratable acidity (0.9) was 11-12 h. The yogurt fermented with DK211 had favorable panelists ratings for most sensory attributes, which were comparable with yogurt fermented with a commercial strain. The results suggest that strains isolated from kimchi could be potential probiotic and starter cultures for use in yogurt manufacturing.

Effects of Coagulants on Storage of Packed Tofu (충진 두부의 저장성에 미치는 응고제의 효과)

  • Several coagulants was investigated for their effects on shelf-life of packed Tofu. The titratable acidity, amino nitrogen content in Tofu and the optical density of Tofu suspension were increased as the spoilage developed during storage. The number of microorganisms was increased to 13 million cells per gram after 1 day of storage at $30^{\circ}C$ in the Tofu prepared with $MgSO_{4}\;and\;CaCl_{2}$. But, the Tofu prepared from acetic acid was found to be more stable for storage. The quality of Tofu was maintained up to 12 days or 18 days at $30^{\circ}C\;or\;15^{\circ}C$, respectively. These results suggested that acetic acid was desirable as a coagulant of packed Tofu to extend its shelf life.

  • Meat is a perishable product since it offers favorable conditions for microbial contamination and multiplication. Besides, undercooked and raw meat has been linked to outbreaks of hemorrhagic diarrhea due to the presence of microbial pathogens. Therefore, this study was conducted to find out the effect of the use of gamma-irradiation in order to improve preservation and eliminate this microorganisms. The Initial level of microbial contamination in pork was 1.1$\times$10$^3$ CFU/g (aerobic bacteria), 5.3$\times$10$^3$CFU/g (psychrophile), 3.1$\times$10$^3$ CFU/g (coliforms) and 3.2$\times$10$^3$ CFU/g(Listeria), All kinds of contaminated microorgnisms were sterilized by gamma irradiation at 3 kGy. The proximate composition of pork was not significantly changed by irradiation dose and storage period. The pH was slightly increased during storage period and titratable acidity decreased. However, no significant changes in pH and acidity were observed by gamma irradiation. There was no difference in total amino acid content regardless of irradiation dose and storage period. Glutamic acid was detected at high levels throughout the storage period. The major mineral compositions of pork were potassium, phosphorus, sodium, magnesium. No significant difference in the components of minerals were observed by gamma irradiation.

Influence of Exogenous Abscisic Acid Concentration on the Coloration of 'Kyoho' Grapes. (Abscisic acid의 처리농도가 포도 '거봉'의 착색에 미치는 영향)

  • This experiment was carried to investigate the optimum concentration of abscisic acid (ABA) treatment for enhancing fruit coloration of 'Kyoho' grapes. Cluster and berry weights showed a tendency that increased in proportion to concentration of ABA treatment, but were not significant in all treatments. Also, soluble solids and titratable acidity were not significant during fruit development in all treatments. L-phenylalanine ammonia-lyase (PAL) activity showed a tendency that decreased after rapidly increased in all treatments, and was the highest in 1000 mg/l ABA treatment at final harvest. Anthocyanin and total phenolics contents were high in proportion to ABA treatment concentration, and anthocyanin content in 1000 mg/l ABA treatment was 2.5 folds of that in control. Fructose and glucose as soluble sugars were detected, but sucrose was not detected. Both fructose and glucose contents increased during fruit development, but showed little difference in all treatments.

Polyphasic Microbial Analysis of Traditional Korean Jeung-Pyun Sourdough Fermented with Makgeolli

  • Jeung-pyun, a fermented rice cake, is prepared by fermenting rice sourdough using makgeolli, a traditional Korean rice wine, in the presence of yeast and lactic acid bacteria (LAB). The goal of this study was to conduct biochemical and microbial analyses of five different rice sourdoughs, each fermented with a different commercial makgeolli, using culture-dependent and culture-independent approaches. All sourdough samples fermented with different makgeolli for 6.5 h showed different profiles in pH, total titratable acidity, organic acid concentration, and microbial growth. LAB belonging to different genera were identified based on colony morphology on modified MRS and sourdough bacteria agar medium. PCR-denaturing gradient gel electrophoresis analyses of the five sourdoughs showed different bands corresponding to LAB and yeast. 16S/26S rRNA gene sequence analyses of the samples confirmed that the predominant LAB in the five fermented rice doughs was Lactobacillus plantarum, Lb. pentosus, and Lb. brevis. Various other Lactobacillus spp. and Saccharomyces cerevisiae were common in all five fermented samples. This study provides comprehensive and comparative information on the microflora involved in fermentation of rice sourdough and signifies the need to develop effective starters to enrich the quality of jeung-pyun.

Antimicrobial Effects of Ethanol Extracts of Pinus densiflora Sieb. et Zucc. on Lactic Acid Bacteria (김치 발효 미생물에 대한 소나무(Pinus densiflora Sieb. et Zucc.) 에탄올 추출물의 항균효과)

  • This study was carried out to investigate the effect of ethanol extract of Pinus densiflora (PD) on the growth of lactic acid bacteria (LAB), A-1, B-9, K-7, M-7 isolated from kimchi. The growth of isolated LAB was inhibited significantly in the modified MRS broth containing 40 mg/mL PD ethanol extract. Ethyl acetate fraction showed the strongest antimicrobial activities against LAB strains compared to other fractions. The addition of PD ethanol extract to kimchi did not change the pH of kimchi greatly compare with the control during the fermentation for 25 days. Change of titratable acidity in control was more higher than in the PD ethanol extract added kimchi during fermentation. The growth of total bacteria and LAB was inhibited about 1 to 2 log cycle by the addition of PD ethanol extract during the kimchi fermentation for 25 days at 1$0^{\circ}C$. Sensory quality of PD ethanol extract added kimchi was lower than that of control.
