Purification and Physical Proerties of Biosurfactant Produced from Rhodotorula muciloginosa (Rhodotorula muciloginosa G-1에서 생산되는 biosurfactant의 정제 및 물리적 성질)

  • The surface tension-decreasing biosurfactant was purified from Rhodotorula muciloginosa G-1. The purification procedure was the solvent extraction of culture broth. To ensure complete extraction, the sample was extracted twice with equal volume of ethylacetate. The crude solution was washed with n-hexane to remove unconsumed soybean oil. The crude sample of biosurfactant was applied to Silica gel column chromatography equilibrated with chloroform, and eluted with chloroform : methanol gradient. Serveral solvent system was used to developed the thin layer chromatography (TLC). The purified biosurfactant sample gave one spot (Rf 0.78). It was estimated that biosurfactant was glycolipid about having M.W.1,500 with standard of polyethyleneglycol by Sephadex LH-20 column chromatography.

The Metabolism of (4-$^{14}C$) Cholesterol on Photoperiodism in Solanum andigena

  • (4-$^{14}$ C) Cholesterol was administered to the leaves of Solanum andigena during photoperiodic induction. Radioactive products converted from cholesterol were studied by thin-layer chromatography and gas-chromatography. The major products from cholesterol were shown to be esterified cholesterol in lipid and solanine in the aqueous ethanolic phase in SD-and LD-leaves. The radioactive solanidine was isolated by chromatography and crystallized to constant specific activity. Short-day condition did not stimulate the conversion of cholesterol into solanine in the leaves, but both groups of leaves converted cholesterol into solanine at the about same rate. Incorporation of radioactivity into aqueous ethanolic phase of tubers and stolons, containing storied glycoalkaloid, was very much higher than that into lipid phase, contrary to those in the leaves and the stems.

  • Component having antioxidant activity on lard during heating was separated from hexane extract of spinach, and its characteristic chemical structure was speculated through nuclear magnetic resonance spectroscopy, liquid chromatography-mass spectrometry, and Fourier transform infrared spectrophotometry. Lard was heated with hexane-, ethyl ether-, ethyl acetate-, or ethanol extract of spinach at $180^{\circ}C$ for 20 hr. Hexane extract of spinach, having highest antioxidant activity on lard during heating, was fractionated by silicic acid column chromatography (SACC), and SACC fractions having higher antioxidant activity on lard during heating were further separated by thin layer chromatography (TLC). Isolated compound from SACC fractions of hexane extract of spinach by TLC had sugar moieties and benzene ring along with hydroxy, carbonyl, and alkyl groups in the structure.

  • Antibiotic and physico-chemical properties of an active compound from actinomycetes isolate G-37, of which the culture filtrate had an inhibitory effect against tobacco mosaic virus(W) infection, were examined. The active compound, which was purified by ethylacetate extraction, silica gel column chromatography, preparative thin layer chromatography, and high performance liquid chromatography, showed strong antibacterial activities especially against Gram-positive bacteria including Bacillus subtillis, Sarcina lutea and Staphylococcus aureus. From the IH-NMR, FAB/RfS, UV spectral data, and physicochemical properties, the active compound of G-37 appears to belong to a peptide antibiotic group. Among the known peptide antibiotics in the antibiotic group, No. 280, A-30912, and Taitomycin showed molecular weights and ultra violet spectrum similar to those of the active compound from G-37, but was not identical to the compound, which suggests that it may be a new peptide antibiotics.

  • Evaluation method of crude drug preparations was studied in Saengmaek-san. Zig-zag TLC scanning profiles and high performance liquid chromatograms were obtained from Saengmaek-san and its each crude drug. A method using TLC densitometry and high performance liquid chromatography was established for the precise determination of ginsenoside $Rb_1$ in Saengmaek-san containing Ginseng Radix. Consequently, ginsenosicle $Rb_1$ content was 0.45-0.48 mg per g of Saengmaek-san. This method was found to be useful for the quality evaluation of oriental medicinal preparations.

  • Red ginseng powder was administered at a dose of 2.7 g per day for 3 months to 21 diabetic patients who were under the treatment with insulin. It was found that the ginseng powder was effective to 12 patients and ineffective to 9 patients. Based on these clinical results, experiments were carried out to elucidate factors which concerned with improvement of pathological conditions of diabetes mellitus. In the previous symposium, we reported that red ginseng powder contained an anti-lipolytic peptide, or an insulin-like peptide. In the course of purification of the insulin-like peptide in the ginseng, we found another fraction which possessed anti-lipolytic activity. The anti-lipolytic factor of the fraction was purified by gel filtration on Bio Gel P-2 column and Dowex $50W{\times}4$ column chromatography. The character of the finally purified material was examined by thin-layer chromatography, high-speed liquid chromatography and mass spectrometry. With these examinations, the active principle was indentified to be adenosine. Pharmacological significance of these insulin-like substances, the peptide and adenosine, was discussed.

Studies of Antipyretic Component of the Earthworm (지렁이(지룡)의 해열성분에 관한 연구)

  • In order to confirm the exact antipyretic component in the earthworm, etherial extract of American earthworm(Red Worm) was fractionated into five fractions by using silica gel column chromatography and thin layer chromatography. The fraction including free fatty acids was found to possess artipyretic response and standard arachidonic acid showed marked antipyretic response on typhoid vaccinated rabbits. Arachidonic acid was identified from the free fatty acid fraction of the earthworm by using gas liquid chromatography. Thus it was considered that the antipyretic activity of the free fatty acid may be due to the presence of arachidonic acid. Lipid-free earthworm powder was extracted with phosphate buffer (pH, 8.0, 0.1M) and all the proteins was salted out by ammonium sulfate. The crude precipitate was dialyzed and the impure proteins were eliminated at pH 5.4 and 4.6. The remaining protein solution was fractionated with various concentrations of acetone. The acetone fractions were identified by using S.D.S. polyacrylamide gel electrophoresis and disc gel electrophoresis. The precipitate at 85% acetone concentration and the remaining proteins in the supernatant did not exhibit the antipyretic activity.

  • Three sterol fractions; 4-desmethyl-, 4-monomethyl-, and 4,4-dimethylsterol, separated by thin layer chromatography from the unsaponifiables of five Cucurbitaceae (cucumber, watermelon, sponge cucumber, gourd and snake gourd) seed oils were analyzed by gas liquid chromatography and combined gas liquid chromatography-mass spectrometry. ${\alpha}$-Spinasterol, ${\Delta}^{7,22,25}$-stigmastatrienol and ${\Delta}^{7,25}$-stigmastadienol isolated from the 4-desmethylsterol fraction were identified by IR, NMR and mass spectrometry.

Studies on the Sterol Components of Torreya Nut of Korea (한국산 비자중의 스테롤 성분에 관한 연구)

  • "Bija"(Torreya nut), a tree belonging to the Torreyaceae family, is cultivated in Jae Joo Do and Jun Ra Do, Korea. The seed of Torreya nucifera, which has been widely used as folkmedicine in the treatment of tapeworm infestation, was examined on the sterol composition determined by gas liquid chromatography and thin layer chromatography on the preparative plates. Sterols were obtained from the nonsaponifiable matters of ether extract of the seed. It was noted that .betha.-sitosterol was the major sterol in the nut. The results showed that contents of sterols were campesterol 3.15-3.75%, stigmasterol 5.38-5.67%, $\beta$-sitosterol 85.61-86.28% and ${\delta}^{7}$-sterol 4.91-5.19% by gas liquid chromatography.

  • A method for isolation of some phenolic acids from Korean ginseng(Panax ginseng C.A.Meyer)was studied using silicic acid column chromatography. preparative thin layer chromatography and high performance liquid chromatography. Two phenolic compounds were isolated and identified as ferulic acid, mp $156-157^{\circ}C$ and vanillic acid. mp $154-156^{\circ}C$ by spectral data of Mass and NMR spectroscopy.
