Plant uptake potential of endosulfan from soil by carrot and spinach (다소비 채소작물인 시금치와 당근의 토양 중 엔도설판 흡수이행능)

  • Residual endosulfan in an agricultural environment has been reported, although endosulfan was listed to persistent organic pollutants and banned. To produce the safe crop from endosulfan residue risk, the plant uptake potential of endosulfan from soil to crop should be studied. In here, the plant uptake potentials of endosulfan in various crops were surveyed and ranged from 0.002-4.460. And the bioconcentration factors (BCF) of total endosulfan in carrot and spinach were calculated from the pot experiment. The BCFs in carrot and spinach were 0.285 and 0.040-0.047 respectively. Endosulfan sulfate was contributed to over 42.8% of the crop residue as a major contributor among the three endosulfan congeners in both of carrot and spinach.

Preparation and Characteristics of Crosslinked SEBS/HIPS Cation Exchange Membrane Using Epoxidized Polybutadiene/Divinylbenzene (Epoxidized Polybutadiene/Divinylbenzene을 이용한 가교 SEBS/HIPS 양이온교환막의 제조 및 특성)

  • The cation-exchange membrane which was sulfonated styrene-ethylene/buthlene-styrene(SEBS) block copolymer containing the high impact polystyrene (HIPS) was prepared via post-sulfonation and casting method using the epoxidized polybutadiene and divinylbenzene as crosslinking agents. Post-sulfonation was carried out with sulfuric acid as sulfonating agent and silver sulfate as initiator in the nitrogen atmosphere. The basic properties of membranes, degree of sulfonation (DS), water uptake, ion-exchange capacity (IEC), electrical resistance, and modulus have been examined. DS of membrane increased with increasing the sulfonation time. The maximum DS of membrane containing 10 wt% HIPS was 83.6 %. The water uptake and IEC of membranes gradually increased as increasing the DS. The maximum water uptake and IEC of membranes were 43.8 % and 1.14 meq/g, respectively. The lowest electrical resistance of membrane containing the 20 wt% HIPS was $83\;\Omega{\cdot}cm^2$. The electrical conductivity of membrane containing 10 wt% HIPS was $1.22\times10^{-4}S/cm$. The modulus of membrane increased with increasing DS and these values were 153 and $204\;kgf/cm^2$ before and after sulfonation, respectively.

Establishment of Safe Management Guideline Based on Uptake Pattern of Pesticide Residue from Soil by Radish (토양잔류 농약의 무 흡수양상 및 토양 안전관리기준 설정)

  • BACKGROUND: Uptake patterns of ${\alpha}$-, ${\beta}$-isomers and sulfate metabolite of endosulfan (ED) by radishes grown in treated soils with ED concentrations of 2 and 10 mg/kg were investigated to establish soil management guidelines for ensuring the safety of radishes from ED residues. METHODS AND RESULTS: All samples of soils and radish plants separated into shoot and root parts were analyzed for ED residues using a gas-chromatography mass spectrophotometer, and the results were used to calculate the bioconcentration factor (BCF), indicating the ratio of ED concentrations between radishes and soils. During the experimental period, uptake and distribution rates of ED-sulfate in radishes were the highest, followed by ${\alpha}$- and ${\beta}$-ED. The BCF values to initial ED concentrations in soils were greater for root parts (0.0077 to 0.2345) than for shoot parts (0.0002 to 0.0429) and used to obtain regression equations by time. Long-term BCFs estimated by the obtained equations ($R^2$ of 0.86 to 1.00) were evaluated with the maximum residue limit (0.1 mg/kg) of ED for radishes, in order to suggest safe management guidelines of ED for radish-cultivating soils. CONCLUSION: Suggested guidelines showed the significant dependency on duration for radish cultivation and exposed concentration of ED in soil.

Trace element levels and selenium uptake in cereals grown in lower Austria

  • Wheat, barley, rye, and maize were grown in field and pot experiments at various non-contaminated soils in order to establish uptake rates for added selenate, and to find baseline concentrations for various soil types. Edible parts (grains) and stalks of the crops were analyzed separately for Se, as well as for Ca, Cu, Fe, Mn, P, S, and Zn. The addition of Na-selenate in admixture with the NPK 20:8:8 fertilizer had no influence on the composition of the other elements investigated. The proportions of added nitrate: selenate, and sulfate:selenate were kept constant. The Se- uptake rate differed among the cereals tested, it was highest for winter wheat. Utilization of added Se in the field ranged from $0,4-4,7\%$, and and in the pots from $3,3-5,4\%$, it was markedly lower in clay soil. Whereas P and Zn were preferably found in the grains, Ca-Fe-Mn-S got enriched in the stalks. For the fields, the location had some influence upon Fe, Mn, and Zn, whereas it was not important for P, S, Cu, and strikingly, Ca. Pot and field experiments on similar soils led to different results, except for P and S. Maize (whole grains) was significantly lower in Ca, Cu, and Mn, and might even cause trace element deficiencies, if exclusively fed. Few correlations between the trace elements investigated led to the conclusion that most element contents were governed by plant metabolism. Variations of mobile Fe in the soils were balanced by uptake into the stalks. The data are compared with data from other presumably non-contaminated sites.

  • The concern for dissolved phosphate in water/wastewater has been increasing because of the risk for eutrophication. A variety of conventional and advanced technologies were applied to meet the enforced new regulation of phosphate around the world. However, there still remained a lot of challenge because most introduced/developed method, for example, biological and physic-chemical treatment is not easy to satisfy the new regulation of phosphate in water. In order to meet the new regulation, the application of ion exchanger has been tried which showed that the removal efficiency for phosphate was strongly determined by in the presence of the competing ion, especially sulfate. As results, a new class of ion exchanger governed by ligand exchange was developed and investigated to increase the selectivity for phosphate. The current study using organic/inorganic anion exchanger developed with Lewis acid-base interaction confirms the selectivity for phosphate over sulfate. According to isotherm test and column test, the value of the maximum phosphate uptake (Q) showed 64 mg/g as $po{_4}^{3-}$ and the breakthrough for phosphate occurs after 1000 min and completely finishes at 2500 min, respectively.

Utilization of Bamboo Leaves as a New Resource of Natural Green Colorants

  • The objective of this study was to find an appropriate method and process for protecting the green color in bamboo leaves and subsequent extraction of the green colorants, chlorophyll. Various inorganic salts including cupric sulfate, ferric sulfate, and zinc chloride were employed as green color protectors. Accordingly, the effects of metal salts and treatment conditions on color protection were investigated to find appropriate protector and conditions. And also, the efficacy of bamboo colorants as a natural green dye was evaluated through dyeing and colorfastness tests. Antimicrobial activity of dyed fabrics was measured by shake flask method in terms of bacterial reduction rate using Staphylococcus aureus(ATCC 6538). On the basis of experimental results for stabilizing bamboo leaves colorants, it was confirmed that Cu was the most appropriate metal type considering dye uptake, photostability and light fastness, and its optimum concentration was 0.025%. After the stabilization, the colorants were extracted efficiently at NaOH aqueous solution of 1.00%. It was concluded that bamboo leave has a high potentiality as new resources to produce a natural green dye with antimicrobial functionality.

  • In order to evaluate the neutral red uptake assay as an alternative method for phototoxicity test, we compared the potential of phototoxicity in vitro in cultured human fibroblasts and 3T3 fibroblast cells derived from Balb/c mice. Both fibroblasts were exposed to various known phototoxic chemicals (promethazine, neutral red, chlorpromazine, chlortetracycline, amiodarone, bithionol, 8-methoxypsoralen) and non-phototoxic chemical (ammonium laureth sulfate) and irradiated with 5 J/cm$^2$ of UVA.(omitted)

Functional characterization of Clonorchis sinensis choline transporter

  • Clonorchis sinensis is commonly found in East Asian countries. Clonorchiasis is prevalent in these countries and can lead to various clinical symptoms. In this study, we used overlap extension polymerase chain reaction (PCR) and the Xenopus laevis oocyte expression system to isolate a cDNA encoding the choline transporter of C. sinensis (CsChT). We subsequently characterized recombinant CsChT. Expression of CsChT in X. laevis oocytes enabled efficient transport of radiolabeled choline, with no detectable uptake of arginine, α-ketoglutarate, p-aminohippurate, taurocholate, and estrone sulfate. Influx and efflux experiments showed that CsChT-mediated choline uptake was time- and sodium-dependent, with no exchange properties. Concentration-dependent analyses of revealed saturable kinetics consistent with the Michaelis-Menten equation, while nonlinear regression analyses revealed a Km value of 8.3 µM and a Vmax of 61.0 pmol/oocyte/h. These findings contribute to widen our understanding of CsChT transport properties and the cascade of choline metabolisms within C. sinensis.

Optimization of uranium biosorption in solutions by Sargassum boveanum using RSM method

  • The potential use of Sargassum boveanum algae for the removal of uranium from aqueous solution has been studied by varying three independent parameters (pH, initial uranium ion concentration, S. boveanum dosage) using a central composite design (CCD) under response surface methodology (RSM). Batch mode experiments were performed in 20 experimental runs to determine the maximum metal adsorption capacity. In CCD design, the quantitative relationship between different levels of these parameters and heavy metal uptake (q) were used to work out the optimized levels of these parameters. The analysis of variance (ANOVA) of the proposed quadratic model revealed that this model was highly significant (R2 = 0.9940). The best set required 2.81 as initial pH(on the base of design of experiments method), 1.01 g/L S. boveanum and 418.92 mg/L uranium ion concentration within 180 min of contact time to show an optimum uranium uptake of 255 mg/g biomass. The biosorption process was also evaluated by Langmuir, Freundlich, Temkin and Dubinin-Radushkevich isotherm models represented that the experimental data fitted to the Langmuir isotherm model of a suitable degree and showed the maximum uptake capacity of 500 mg/g. FTIR and scanning electron microscopy were used to characterize the biosorbent and implied that the functional groups (carboxyl, sulfate, carbonyl and amine) were responsible for the biosorption of uranium from aqueous solution. In conclusion, the present study showed that S. boveanum could be a promising biosorbent for the removal of uranium pollutants from aqueous solutions.

  • The biotechnology including genetic engineering is expected to be applied to various fields of wastewater treatments in order to promote biological reaction rate, to grade up the effluent quality and to advance the stability of microorganisms against temperature, pH and toxic substances. The current topics in Japan on application of biotechnology to wastewater treatment will be reviewed at the beginning of the presentation. Next, the research of Biochemical Engineering Laboratory is to be presented, especially focused on the following subjects ; (1) Application of genetic engineering to the investiation of heavy metal uptake by the resistant bacteria of Hg Cd or Zn. (2) Relationship between sulfate reducing bacteria and wastewater treatment, offensive odor and corrosion of sewer tranks.
