Eggs and Egg Capsules of Three Stenoglossan Gastropod Species, Ceratostoma rorifluum, Ocinebrellus inornatum & Neptunnea constricta, (Mollusca: Neogatropoda) in the Korean Waters

  • The morphology of molluscan egg capsule is generally regarded as being species specific (Turner, 1992), and form and structure of the egg capsule reflect the degree of anatomical complexity of the reproductive organs (Fretter & Hian, 1984). Ceratostoma rorifluum and Ocinebrellus inornatum are common and widespread muricid species found throughout the Korean rocky and boulder shores, and Neptunnea constricta is found frequently from continental shelf of the eastern sea of Korea. In spite of their predominance along the Korean coasts, morphology of their eggs and egg capsules is unknown so far. The purpose of the present study, therefore, is to provide descriptions and illustrations for identifying egg capsules of the foregoing 3 stenoglossan gastropod species. (omitted)

  • Pig organ is thought to be the most suitable nonhuman organ for xenotransplanstation. However, one of the major constraints to using pig organs for xenotransplantation is human natural antibody-mediated hyperacute rejection (HAR). Elimination of a(1,3) galactosyltransferase (GGTA1) from the pig is expected to be a solution to the problem of hyperacute rejection. ry1any efforts have made characterization of GGTA1 in structure and function. improvement in the technique of DNA transfection of somatic cells and advancement of the pig NT, a specific modification has been made to one copy of the GGTA1 gene by Missouri group in 2002. To date because homozygousity of the genetic modification has been achieved in this gene, the role of gala(1,3) gal specific natural antibody in HAR and the efficacy of xenotransplantation in a nonhuman primate model will be addressed. If other genes are found to be involved in rejection of pig donors by primates, the technology will be available to modify those genes so that rejection can be overcome.

한국산 홍조식물 보라색 우무속(Symphyocladia)직물의 분류 (Taxonomy of the Genus Symphyocladia (Rhodophyta))

  • Four species of Symphyocladia growing along the coast of Korea were investigated in taxonomic point of view. Characters available for identification of the species were; difference in gross morphology, degree of cortication in vegetative axis, presence or absence of vegetative trichoblasts, and developmental pattern of stichidia. Vegetative structure and reproductive organs in developmental anatomy were similar among species. Branching pattern and occurrence of vegetative trichoblasts adopted for systematic criteria between Pterosiphonieae and Polysiphonieae, and between Symphyocladia and Pterosiphonia were no more confidential. The degree of congenital fusion of laterals with the parent axes was considered as only main character to distinguish Symphyocladia from Pterosiphonia.

Oxidative Modification of Cytochrome c by Tetrahydropapaveroline, an Isoquinoline-Derived Neurotoxin

  • Tetrahyropapaveroline (THP) is compound derived from dopamine metabolism and is capable of causing dopaminergic neurodegenerative disorder, such as Parkinson's disease (PD). The aim of this study was to evaluate the potential of THP to cause oxidative damage on the structure of cytochrome c (cyt c). Our data showed that THP led to protein aggregation and the formation of carbonyl compound in protein aggregates. THP also induced the release of iron from cyt c. Reactive oxygen species (ROS) scavengers and iron specific chelator inhibited the THP-mediated cyt c modification and carbonyl compound formation. The results of this study show that ROS may play a critical role in THP-induced cyt c modification and iron releasing of cyt c. When cyt c that has been exposed to THP was subsequently analyzed by amino acid analysis, lysine, histidine and methionine residues were particularly sensitive. It is suggested that oxidative damage of cyt c by THP might induce the increase of iron content in cells and subsequently led to the deleterious condition. This mechanism is associated with the deterioration of organs under neurodegenerative disorder such as PD.

Oxidative Modification of Human Ceruloplasmin by Methylglyoxal: An in vitro study

  • Methylglyoxal (MG) is an endogenous physiological metabolite which is present in increased concentrations in diabetics. MG reacts with the amino acids of proteins to form advanced glycation end products. In this in vitro study, we investigated the effect of MG on the structure and function of ceruloplasmin (CP) a serum oxidase carrier of copper ions in the human. When CP was incubated with MG, the protein showed increased electrophoretic mobility which represented the aggregates at a high concentration of MG (100 mM). MG-mediated CP aggregation led to the loss of enzymatic activity and the release of copper ions from the protein. Radical scavengers and copper ion chelators significantly prevented CP aggregation. CP is an important protein that circulates in plasma as a major copper transport protein. It is suggested that oxidative damage of CP by MG may induce perturbations of the copper transport system and subsequently lead to harmful intracellular condition. The proposed mechanism, in part, may provide an explanation for the deterioration of organs in the diabetic patient.

  • Vestibular hair cells, the sensory receptors of vestibular organs, selectively amplify miniscule stimuli to attain high sensitivity. Such selective amplification results in compressive nonlinear sensitivity, which plays an important role in expanding dynamic range while ensuring robustness of the system. In this study, negative stiffness mechanism, a mechanism responsible for the selective amplification by vestibular hair cells, is applied to a simple mechanical system consisting of an array of inverted pendulums. The structure and working principle of the system have been inspired by gating spring hypothesis proposing that opening and closing of transduction channels contributes to the global stiffness of vestibular hair bundle. Parameter study was carried out to analyze the effect of each parameter on the compressive nonlinearity of suggested model.

  • In tissue engineering, scaffolds play an important role in the growth of cells to 3-D organs or tissues. For the success of tissue engineering, they should be mimicked to meet the requirements of natural extracellular matrix (ECM) in the body, such as mechanical properties, adhesiveness, porosity, biodegradability, and growth factor release, etc. Contrary to other materials, polymeric materials are adequate to engineer scaffolds for tissue engineering because controlling the structure and the ratio of components and designing various shapes and size are possible. In this review, the importance, major characteristics, processes, and recent examples of polymeric scaffolds for tissue engineering applications are discussed.

  • The antiinflammatory mechanism of NSAIDs is attributed to the reduction of prostaglandin synthesis by the direct inhibition of cyclooxygenase. Inhibition of prostaglandin production in organs such as stomach and kidney can result in gastric lesions, nephrotoxicity and increased bleeding. In this study, newly designed COX-2 inhibitors, synthesized 1,2-benzothiazine derivatives, were screened in vitro for selectivity of COX-1 and COX-2 inhibition properties. Lead compounds in the structure-activity relationship were studied to synthesize new highly selective COX-2 inhibitors.13 determine inhibitory effect of COX-2, synthesized 1,2-benzothiazine derivatives were screened with accumulation of prostaglandin by lipopolysaccharide (LPS) in aspirin-treated macrophages and murine macropharge cell. Some of synthesized 1,2-benzothiazine derivatives were shown to be effective as selective COX-2 inhibitory activity. Others exhibited a preferential inhibition of COX-2, although some COX-1 inhibitory activity was still present. As a conclusion, simple monomer derivatives were more active than dimer derivatives. Substitution of halogen (Br, C1) on the benzothiazine nucleus slightly enhanced inhibition activity.

Application of Photobiomodulation in Hearing Research: Animal Study

  • Hearing organs have unique characteristics and have a role in processing external sensory signals. Sensory hair cells and nerve fibers in the organ of Corti can be damaged by various causes and they do not regenerate themselves. Medication used for clinical treatment for the inner ear is limited due to the anatomical structure of the inner ear. Photobiomodulation (PBM) is a therapeutic approach that uses various sources of light and the success of PBM therapy is highly reliant on the parameters of the light sources. The positive effects of PBM have been reported in various clinical fields. This paper summarizes the previously reported research on PBM for the treatment of hearing damage in animal models.

Mass Spectrometry in the Determination of Glycosylation Site and N-Glycan Structures of Human Placental Alkaline Phosphatase

  • Alkaline phosphatase (AP) is a membrane-bound glycoprotein that is widely distributed in the plasma membrane of cells of various organs and also found in many organisms from bacteria to humans. The complete amino acid sequence and three-dimensional structure of human placental alkaline phosphatase have been reported. Based on the literature data, AP consists of two presumptive glycosylation sites, at Asn-144 and Asn-271. However, it only contains a single occupied N-linked glycosylation site and no occupied O-linked glycosylation sites. Hydrophilic interaction chromatography (HILIC) has been primarily employed for the characterization of the glycan structures derived from glycoproteins. N-glycan structures from human placental alkaline phosphatase (PLAP) were investigated using HILIC-Orbitrap MS, and subsequent data processing and glycan assignment software. 16 structures including 10 sialylated N-glycans were identified from PLAP.
