In the course of in vitro and in vivo screening for bioherbicidal agents, a hyphomycete fungus, Myrothecium sp. F0252 was selected as a candidate for the biocontrol of weeds. The isolate was identified as Myrothecium roridum Tode ex. Fries based on the morphological characteristics and 18S ribosomal DNA sequence analysis and registered as Myrothecium roridum F0252. In order to evaluate the in vitro effect of M. roridum F0252 on germination of ladino clover and white clover (Trifolium repens L.) seeds, spore solution of the fungus was employed in two concentrations, $6.5{\times}10^6$ and $2.5{\times}10^7$ spores per mL and then inoculated to the seeds. The fungal spores inhibited the seed germination, infected the seedlings, and caused an abnormal withering and inhibition of seedling growth. In addition, when the herbicidal activity of crude ethyl acetate extract from the liquid culture was assessed on a mini-plant, duck-weed (Lemna paucicostata (L.) Hegelm.), the extract showed high inhibitory effect at the level of $12.5{\mu}g$ per mL. On the other hand, in vivo herbicidal activity of M. roridum F0252 was evaluated by a whole plant spray method. M. roridum F0252 exhibited strong and broad-spectrum herbicidal activity. The herbicidal values ranged from 95-100% against 7 weeds, including Abutilon avicennae and Xanthium strumarium, and 70-80% against Digitaria sanguinalis and Sagittaria pygmaea. When the nutritional utilization (95 carbon sources) pattern of M. roridum F0252 was investigated, it varied with water activity ($a_w$) and temperature conditions, supplying good, basic information in regard to nutritional utilization for proper cultivation and formulation. Our results showed that M. roridum F0252 might be used as a potential biocontrol agent against weedy plants.
A moderately halophilic, Gram-positive, spore-forming bacterium was isolated from the roots of Blutaparon portulacoides, a plant found in sandy soil parallel to the beach line in Restinga de Jurubatiba, Rio de Janeiro, Brazil. The strain, designated $M9^T$, was motile and strictly aerobic with rod-shaped cells. It grew in the absence of NaCl and up to 20% NaCl, and was able to hydrolyze casein and starch. Strain $M9^T$ had a cell-wall peptidoglycan based on L-Orn-D-Asp, the predominant menaquinone present was menaquinone-7 (MK-7), diaminopimelic acid was not found, and anteiso-$C_{15:0}$ and iso-$C_{15:0}$ were the major fatty acids. A phylogenetic analysis based on 16S rRNA gene sequences showed that strain $M9^T$ belonged to the genus Halobacillus and exhibited 16S rRNA gene similarity levels of 97.8-99.4% with the type strains of the other nine Halobacillus species. The DNA-DNA relatedness of strain $M9^T$ with H. trueperi, the closest relative as regards 16S rRNA gene similarity, and H. locisalis was 21% and 18%, respectively. Therefore, on the basis of phenotypic, genotypic, and phylogenetic data, strain $M9^T$ (=ATCC BAA-$1217^T$, =CIP $108771^T$, =KCTC $3980^T$) should be placed in the genus Halobacillus as a member of a novel species, for which the name Halobacillus blutaparonensis sp. nov. is proposed.

This study attempted to determine important meteorological parameters related to airborne pollen concentrations in urban areas. Hourly pollen measurement data were prepared from a regular sampling with a volumetric Burkard spore trap at a site in the Ulsan city, during the spring season (March~May) of 2011. Results showed that the daily mean and maximum concentrations for total pollen counts during the spring season were statistically significantly correlated with both air temperature and wind speed; daily mean pollen concentration was the most highly related to daily maximum temperature (r=0.567, p<0.001). It was also identified that pollen concentration has a stronger relationship with wind speed at the rural site than at the urban one, which confirms that strong wind conditions over the pollen sources area can be favorable for pollen dispersal, resulting in increases in airborne pollen concentrations downwind. From the results of an oak-pollen episode analysis, it was found that there was a significant relationship between hourly variation of oak pollen concentrations and dynamic meteorological factors, such as wind and mixing height (representing the boundary layer depth); especially, a strong southwestern wind and elevated mixing height was associated with high nocturnal concentrations of oak pollen. This study suggests that temperature, wind, and mixing height can be important considerations in explaining the pollen concentration variations. Additional examination of complex interactions of multiple meteorological parameters affecting pollen behavior should be carried out in order to better understand and predict the temporal and spatial pollen distribution in urban areas.
Pseudomonas fluorescens GL20 is a plant growth-promoting rhizobacterium that produces a large amount of hydroxamate siderophore under iron-limited conditions. The strain GL20 considerably inhibited the spore germination and hyphal growth of a plant pathogenic fungus, Fusarium solani, when iron was limited, significantly suppressed the root-rot disease on beans caused by F. solani, and enhanced the plant growth. The mechanism for the beneficial effect of strain GL20 on the disease suppression was due to the siderophore production, evidenced by mutant strains derived from the strain. Analysis of the outer membrane protein profile revealed that the growth of strain GL20 induced the synthesis of specific iron-regulated outer membrane proteins with molecular masses of 85- and 90 kDa as the high-affinity receptors for the ferric siderophore. In addition, a cross-feeding assay revealed the presence of multiple inducible receptors for heterologous siderophores in the strain. In order to induce increased efficacy and potential in biological control of plant disease, a siderophore-overproducing mutant, GL20-S207, was prepared by NTG mutagenesis. The mutant GL20-S207 produced nearly 2.3 times more siderophore than the parent strain. In pot trials of beans with F. solani, the mutant increased plant growth up to 1.5 times compared with that of the parent strain. These results suggest that the plant growth-promoting P. fluorescens GL20 and the genetically bred P. fluorescens GL20-S207 can play an important role in the biological control of soil-borne plant diseases in the rhizosphere.

A method for the simple, rapid, specific, and accurate detection of Bacillus anthracis spores was developed by employing specific capture peptides conjugated with fluorescent quantum dots (QDs). It was possible to distinguish B. anthracis spores from the spores of B. thuringiensis and B. cereus using these peptide-QD conjugates by flow cytometric and confocal laser scanning microscopic analyses. For more convenient high-throughput detection of B. anthracis spores, spectrofluorometric analysis of spore-peptide-QD conjugates was performed. B. anthracis spores could be detected in less than 1 h using this method. In order to avoid any minor yet false-positive signal caused by the presence of B. thuringiensis spores, the B-Negative peptide, which can only bind to B. thuringiensis, conjugated with another type of QD that fluoresces at different wavelength was also developed. In the presence of mixed B. anthracis and B. thuringiensis spores, the BABA peptide conjugated with QD525 and the B-Negative peptide conjugated with QD585 were able to bind to the former and the latter, specifically and respectively, thus allowing the clear detection of B. anthracis spores against B. thuringiensis spores by using two QD-labeling systems. This capture peptide-conjugated QD system should be useful for the detection of B. anthracis spores.
Keratinases are of particular interest because of their action on insoluble keratins and generally on a broad range of protein substrates. Alkalophilic and neutrophilic actinomycete strains isolated from different soil samples, rich in keratinaceous substances were screened for keratinolytic activity. An alkalophilic isolate, TBG-S13A5, was found to possess good keratinolytic activity and was able to utilize feather as the sole carbon and nitrogen source. TBG-S13A5 exhibited an off-white aerial mass color with a rectus-flexibilis type of spore chain. The morphological, microscopical and biochemical characters were comparable with that of Streptomyces albidoflavus. Fatty acid methyl ester profiling (FAME) and 16S rDNA sequence analysis confirmed its identity as a strain of S. albidoflavus. Under submerged fermentation conditions, maximum protease production was recorded on the $5^{th}$ day of incubation at $30^{\circ}C$, using basal broth of pH 9.0 with 0.25% (w/v) white chicken feather. This strain could affect feather degradation when the initial pH was 8 and above and maximum protease production was recorded when the initial pH was around 10.5. The effectiveness of the crude enzyme in destaining and leather dehairing were also demonstrated.
Soil samples were screened for actinomycete strains capable of producing phospholipase D, and a strain, Streptomyces sp. YU100, showing a high transphosphatidylation activity was isolated. This strain secreted phospholipase D in a culture broth after 12 h of cultivation, and its productivity continued to increase for 36 h of fermentation. In addition, its transphosphatidylation rate of phosphatidylcholine to phosphatidylserine was almost $68\%$ within 1 h. The morphological and chemotaxonomical characteristics showed that this strain could be classified as a number of the Streptomycetaceae family, particularly due to the spiral form of its spore chain consisting of 60-70 smooth spores $(0.75{\times}1.0{\mu}m$) on an aerial mycelium, FA-2c type of fatty acid profile in the cell wall, and LL-DAP component in the cell wall peptidoglycan. A phylogenetic analysis of the 16S rDNA provided a clue that the strain YU100 was actually a member of the genus Streptomyces, because the determined sequence exhibited a higher homology with Streptomyes sp. ASB27, S. peucetius JCM9920, and S. griseus ATCC10137. A dendrogram based on the 16S rDNA sequences also showed a phylogenetic relationship between the strain YU100 and these strains. However, the strain YU100 has not yet been assigned to a particular species, because of absence of any other classified species with a high matching score.

This study investigated the dif usion characteristics of Ecklonia cava spores around marine forest reefs. For this purpose, a numerical analysis was conducted using field observations in the target area and an EFDC model. Based on the results of field observations and the EFDC model, Flow-3D was performed on three types of marine forest reefs, mamely triangular pyramid reef, double-dome reef, and ribbed reef, to monitor the movement direction and maximum movement distance for E. cava spores. As a result, the equilaterally triangular pyramid reef and double-domed reef were found to have a maximum settlement of spores of 10 m in the northwestern direction and 6 m in the western direction. The ribbed reef had a maximum settlement of spores at 4m. These results show that consideration of the diffusion characteristics of E. cava spores when the seaweeds are installed can increase the effectiveness of the algae as substrate of adhesion.

In this study, we isolated two novel chitinase producing bacterial strains from yellow loess samples collected from Jullanamdo province. The chitinase producing bacteria were isolated based on the zone size of clearance in the chitin agar plates. Both of them were gram positive, rod ($2{\sim}3{\times}0.3{\sim}0.4{\mu}m$), spore-forming, and motility positive. They were facultative anaerobic, catalase positive and hydrolyzed starch, gelatin, and casein. From the 16s rRNA gene sequence analysis, the isolates were labeled as Bacillus licheniformis KYLS-CU01 and B. licheniformis KYLS-CU02. The isolates showed higher extracellular chitinase activities than B. licheniformis ATCC 14580 as a control. The optimum temperature and pH for chitinase production were $40^{\circ}C$ and pH 7.0, respectively. Response Surface Methodology (RSM) was used to optimize the culture medium for efficient production of the chitinase. Under this optimal condition, 1.5 times higher chitinase activity of B. licheniformis KYLS-CU02 was obtained. Extracellular chitinases of the two isolates were purified through ammonium sulfate precipitation and anion-exchange DEAE-cellulose column chromatography. The specific activities of purified chitinase from B. licheniformis KYLS-CU01 and B. licheniformis KYLS-CU02 were 7.65 and 5.21 U/mg protein, respectively. The molecular weights of the two purified chitinases were 59 kDa. Further, the purified chitinase of B. licheniformis KYLS-CU01 showed high antifungal activity against Fusarium sp.. In conclusion, these two bacterial isolates can be used as a biopesticide to control pathogenic fungi.

Some species of the Trichoderma genus are reported as the major problem in oak wood mushroom production in Korea. In spite of economic loss by the fungi, scientific information on airborne Trichoderma species is not much available. To generate information for disease management development we analyzed airborne Trichoderma. A total of 1,063 fungal isolates were purely obtained from indoor air sampling of cultivation houses used for oak wood mushroom using sawdust media. Among the obtained isolates, 248 isolates were identified as Trichoderma fungi including T. harzianum, T. atroviride, T. citrinoviride, and T. pseudokoningii, by morphological and molecular analysis. T. harzianum was dominant among the four identified species. All the four Trichoderma species grew fast on solid nutrient media tested (potato dextrose agar [PDA], malt extract agar [MEA], Czapek's Dox + yeast extract agar [CYA] and cornmeal dextrose agar). Compact mycelia growth and mass spore production were better on PDA and CYA. In addition, T. harzianum and T. citrinoviride formed greenish and yellowish mycelium and spores on PDA and CYA. Greenish and yellowish pigment was saturated into PDA only by T. pseudokoningii. These four Trichoderma species could produce extracellular enzymes of sawdust substrate degradation such as β-glucosidase, avicelase, CM-cellulase, amylase, pectinase, xylanase, and protease. Their mycelia inhibited the growth of oak wood mushroom mycelia of two tested cultivars on dual culture assay. Among of eleven antifungal agents tested, benomyl was the best to inhibit the growth of the four Trichoderma species. Our results demonstrate that the airborne Trichoderma fungi need to be properly managed in the cultivation houses for safe mushroom production.
