Growth Responses of Lactic Acid Bacteria to Leguminous Seed Extracts (콩과식물 종실 추출물의 유산균에 대한 생육반응)

  • Methanol extracts from 25 seed samples belonging to the family Leguminosae were subjected to an in vitro screening for their growth-promoting and inhibitory activities towards Bifidobacterium adolescentis, B. longum, B. bifidum, and Lactobacillus casei, using spectrophotometric and paper disc agar diffusion methods under $O_2-free$ conditions, respectively. The responses varied with both bacterial strains and plant species. Among seed extracts, extracts from Glycine max (light-green color) and Arachis hypogaea (dark-brown) enhanced the growth of lactic acid bacteria in media with or without carbon sources, suggesting that bifidus factor(s) might be involved in the phenomenon. This growth-promoting effect was most pronounced with L. casei among lactic acid bacteria used. Additionally, all seed extracts did not adversely affect the growth of the lactic acid bacteria tested.

Levels of Serum Lipids, Copper, Zinc, Ceruloplasmin and Ferroxidase Activity in Smoking College Women (흡연 여대생의 혈청 지질, Cu, Zn, Ceruloplasmin 농도 및 Ferroxidase 활성도)

  • This study was done to compare serum levels of lipids, trace minerals, ceruloplasmin and ferroxidase activity between female college smokers and nonsmokers. Serum levels of lipids, Cu, Zn, ceruloplasmin and ferroxidase activity were determined in 33 smokers and 42 nonsmokers residing in the Seoul area. Serum levels of TG, total cholesterol and HDl-cholesterol were measured using test kits. Serum Cu Zn were determined by the AAS. Serum ceruloplasimin level and ferroxidase activity were determined by spectrophotometric methods. All data were statistically analyzed by the SAS PC package program. Mean and standard errors were calculated for each item, and the significance of the difference between two groups was evaluated by Student's t-test at p〈0.05. There was no significant difference in levels of total cholesterol, TG, VLDL-cholesterol, LDL-cholesterol, LDL/HDL and atherogenic index, but HDL-cholesterol level was significantly lower(p$\ll$0.05) in female college smokers than in nonsmokers. Serum zinc level showed no significant difference, whereas copper level was significantly higher in smokers, consequently yielding higher Cu/Zn ratio in smoking college women. Ceruloplasmin level was 32.6mg/dl in smoking college women, which was significantly higher(p$\ll$0.05) than that of 28.6mg/dl in nonsmokers. However, cerulopasmin ferroxidase activity, 0.34U/mg in nonsmokers was higher(p$\ll$0.05) than that of 0.29U/mg in smokers. From these results, it is confirmed that cigarette smoking itself has a very detrimental effect on health and mineral nutritional status although the smoking history is relatively short among college women(pack-years=1.7). Screening of the Natural Plant Extracts for the Antimicrobial Activity on Dental Pathogens (구강 질환 유발 미생물에 대한 항생작용을 갖는 천연물 추출물 검색)

  • Many of antimicrobial components used in the mouthwash have disadvantages such as coloring of periodontal tissue and homing sensation. To find the more biocompatible antimicrobial agent, we have screened the 2,125 kinds of natural plant extracts obtained from by the Plant Extract Bank (Daejeon, Korea). The antimicrobial activity was determined by spectrophotometric growth inhibition assay for the major dental pathogens. For the Porphyromonas gingivalis, 19 plant extracts had an antimicrobial activity, 17 plant extracts for the Prevotella intermedia, 45 plant extracts for the Haemophilus actinomycetemcomitans and 85 plant extracts for the Streptococcus mutans. Among these effective extracts, 8 kind of natural plant extracts had an antimicrobial activity for more than 3 species of dental pathogens. In our experiment, Cudrania tricuspidata, Morus bombycis and Mallotus japonicas have antibacterial effects on the all kinds of major dental pathogens. Therefore these plant extracts have a possibility to be a candidate for major antibacterial components in dental products.

Accuracy and Precision of Spectrophotometric Measurement of Clay Content in Soils (분광광도계를 이용한 점토함량 분석의 정확성 및 정밀성 평가)

  • Accuracy and precision of the spectrophotometeric analysis of clay content in soils were estimated by comparison with the conventional pipet method. Clay contents in 25 soil samples of various physico-chemical properties including texture were determined by spectrophotometry and pipet methods, and the two sets of data an clay content were compared by several statistical analyses. The correlation between clay contents determined by spectrophotometry and pipet methods was highly significant. The regression coefficient was $0.98^{**}$ and the slope of regression equation was close to 1.0. The standard deviation and CV of clay contents measured by spectorphotometry were smaller than those found in the data of clay contents obtained by pipet method. In conclusion, compared to the conventional pipet method, spectrophotometry was a rapid, convenient, accurate and precise method for the measurement of clay content in soils.

  • Photocatalytic degradation of Congo red was performed using various semiconductors as ZnO, CdS, rutile-$TiO_2$ or mixed rutile-$TiO_2$/ZnO. The change of degradation of the dye was investigated by UV-visible spectrophotometric method. The photocatalytic action of CdS was greater than ZnO and rutile-$TiO_2$ in account of low band gap energy of CdS. The rate of photocatalytic degradation reaction increased drastically in according to increasing ratio of ZnO on mixed rutile-$TiO_2$/ZnO. These photocatalytic effect of rutile-$TiO_2$ was suppressed by more stable rutile-$TiO_2$, doping the hydrolysis product with $Zn^{2+}$ prior to calcination onto the rutile-$TiO_2$ surface.

Structural and thermal analysis of electrochemically Li intercalated synthetic graphite (전기 화학적으로 리튬이 층간 삽입된 인조흑연의 구조와 열적 특성 분석)

  • The characteristics of the electrochemically Li intercalated synthetic graphite were determined from the studies with XRD method, DSC and solid $^7Li-NMR$ spectrophotometric analysis. From the results of X-ray diffraction method, it was found that the compounds in the stage 1 structure were predominantly formed. The enthalpy and entropy changes of the compounds can be obtained from the differential scanning calorimetric analysis results. From these results, it was found that exothermic and endothermic reactions of lithium intercalated into synthetic graphite are related to thermal stability of lithium ion between carbon graphene layers. From the $^7Li-NMR$ data, scientific observation found that bands are shift toward higher frequencies with increasing lithium concentration because non-occupied electron shells of Li increased in charge carrier density. Line widths of the Li intercalated synthetic graphite compounds decreased slowly because of non-homogeneous local magnetic order and the random electron spin direction for substituted Li.

  • The interaction of monochlorophenol isomers with the micellar system of TTAB (tetradecyltrimethylammonium bromide) was studied by the UV/Vis spectrophotometric method. The solubilization constants ($K_s$) of monochlorophenol isomers into this micellar system have been measured with the change of temperature. Various thermodynamic parameters (${\Delta}G^{\circ}_s$, ${\Delta}H^{\circ}_s$, and ${\Delta}S^{\circ}_s$) have been calculated and analyzed from the dependence of $K_s$ values on the temperature. The results show that the values of ${\Delta}G^{\circ}_s$ and ${\Delta}H^{\circ}_s$ are all negative but the values of ${\Delta}S^{\circ}_s$ are all positive for the solubilizations of all isomers within the measured temperature range. The effects of additives (n-butanol and NaCl) on the solubilization of monochlorophenol isomers by the same surfactant system have been also measured. There was a great change on the values of $K_s$ and CMC simultaneously with these additives. From these changes we can postulate the solubilization sites of each isomer in the micellar system of TTAB.

Characteristics of Cu(II) Extraction by β-ketohexanal (β-ketohexanal에 의한 Cu(II)의 추출 특성)

  • $\beta$-ketohexanal as a chelating extractant was synthesized from the reaction of ethyl formate and methyl propyl ketone in the presence of sodium amide, and the equilibrium characteristics in the extraction of copper by $\beta$-ketohexanal-chloroform were investigated. The equilibrium constants such as the dissociation constant and the partition coefficients of $\beta$-ketohexanal, the stability constant and the partition coefficient of copper chelating complex, and the overall equilibrium constant in the extraction of copper were determined by spectrophotometric measurements, and the mechanism of extraction was proposed. The percent of extracted copper by $\beta$-ketohexanal-chloroform was near 100%, and the selective extraction of copper from Cu-Zn-Cd mixture was possible. Copper was found to be extracted as $CuR_2$ and the equilibrium reaction was expressed as $Cu^{2+}+2{\overline{HR}}{\rightleftarrows}{\overline{CuR{_2}}}+2H^+$.

Screening of HMG-CoA Reductase Inhibitory Activity of Ethanol and Methanol Extracts from Cereals and Regumes (곡류 및 두류 추출물로 부터 HMG-CoA reductase 저해활성 검색)

  • A study was conducted to screen the inhibitory activity of 3-hydroxy-3-methylglutaryl coenzyme A(HMG-CoA) reductase, which is known to be rate-limiting enzyme in cholesterol bosynthesis, from the extracts of 80% methanol and 70% ethanol of cereals and regumes. The strongest inhibitory activity was shown in the ethanol extract of sorghum among the ethanol extracts. The inhibitory activity of HMG-CoA reductase of prosomillilet methanol extract was 73%, and highest among the methanol extracts. The inhibitory activity of 44.7% was observed in sorghum methanol extract. The methanol extracts of prosomillet and sorghum were further fractionated with hexane, chloroform, ethylacetate, butanol and water. HMG-CoA reductase inhibitory activity was shown in all fractions of prosomillet and sorghum methanol extracts. Hexan fraction of both prosomillet and sorghum had the strongest inhibitory activity among five fractions, and the inhibitory activity was increased compared to each crude extracts.

Conjugated linoleic acid producing potential of lactobacilli isolated from goat (AXB) rumen fluid samples

  • Objective: The present investigation was aimed to explore the potential of lactobacilli for conjugated linoleic acid (CLA) production, isolated from rumen fluid samples of lactating goats. Methods: A total of 64 isolates of lactobacilli were obtained using deMan-Rogosa-Sharpe (MRS) agar from rumen fluid of goats and further subjected to morphological and biochemical characterizations. Isolates found as gram-positive, catalase negative rods were presumptively identified as Lactobacillus species and further confirmed by genus specific polymerase chain reaction (PCR). The phylogenetic tree was constructed from the nucleotide sequences using MEGA6. Results: Out of the 64 isolates, 23 isolates were observed positive for CLA production by linoleate isomerase gene-based amplification and quantitatively by UV-spectrophotometric assay for the conversion of linoleic acid to CLA as well as gas chromatography-based assay. In all Lactobacillus species cis9, trans11 isomer was observed as the most predominant CLA isomer. These positive isolates were identified by 16S rRNA gene-based PCR sequencing and identified to be different species of L. ingluviei (2), L.salivarius (2), L. curvatus (15), and L. sakei (4). Conclusion: The findings of the present study concluded that lactic acid bacteria isolated from ruminal fluid samples of goat have the potential to produce bioactive CLA and may be applied as a direct fed microbial to enhance the nutraceutical value of animal food products.
