Effects of Flos Sophora japonica Extract on the Changes of Cerebral Hemodynamics

  • The purpose of this study was to measure the changes of regional cerebral blood flow(rCBF) and blood pressure(BP) in rats, following the intravenous injection of Flos Sophora japonica L.(FSJ) water extract. The measurement was continually monitored by Laser-Doppler Flowmeter and pressure transducer in anaesthetized adult Sprague-Dawley rats for 2 hours to 2 hours and a half through the data acquisition system. FSJ increased the changes of rCBF in rat significantly. The rCBF of FSJ did not change by pretreated propranolol, atropine, L-NNA($N^G$-nitro-L-argininine) and indomethacin. But the rCBF of Flos Sophora japonica L. was increased by pretreated methylene blue. FSJ decreased the changes of BP, significantly. The BP of FSJ did not change by pretreated propranolol, atropine, L-NNA and indomethacin. But the BP FSJ was decreased by pretreated methylene blue. There results indicated that FSJ can increase the rCBF and decrease the BP, that is related to guanylyl cyclase activity.

The Study of Preservative System in Cosmetics using Botanical Antimicrobial (식물유래 천연항균제를 이용한 화장품에서의 방부시스템에 관한 연구)

  • Anti-microbial activities of Scutellariae Radix Extracts and Sophora flavecens Extracts was tested by formulation, such as skinsoftener, emulsion for 4 weeks. Control contained no preservative, test group contained 1.00% Scutellariae Radix extracts and 1.00% Sophora flavecens extracts and positive control contained parabens and imidazolidinyl urea. To determine the anti-microbial activity of these extract, the 4 germs such as Escherichia coil, Pseudomonas aeruginosa, Staphylococcus aures, Canida albicans were used. The test groups showed significant anti-microbial activities against the 4 germs at 2 and 3 weeks as compared with control. Anti-microbial activities of these extracts were similar to positive control. Considering that the Scutellariae Radix Extracts and the Sophora flavecens Extracts have a significant anti-microbial activities against 4 germs, it is possible as natural preservative in cosmetics.

  • Natural extracts were isolated from 20 kinds of medicinal plants that have been traditionally widely used in Korea to develop treatments for dental caries. Medicinal plants showing antibacterial activity against a cariogenic bacterium, Streptococcus sobrinus KCOM 1157 were investigated. The natural extracts of Glycyrrhiza uralensis, Sophora flavescens, Coptis Chinensis, Mentha arvensis, Pinus densiflora, Spirodela polyrhiza showed antimicrobial activities against S. sobrinus KCOM 1157. The extracts of Pinus densiflora, Sophora flavescens, Coptis Chinensis among these medical herbs showed significant antimicrobial activity. These results suggested that the natural products of Pinus densiflora, Sophora flavescens, Coptis Chinensis could be the potential source of anticariogenic agent against dental pathogens including S. sobrinus.

Screening of $5{\alpha}-Reductase$ Inhibition and Comedolytic Effects from Natural Products (천연물로부터 $5{\alpha}-Reductase$ 활성억제 및 면포용해효과의 검색)

  • The antibacterial activity against Propionibacterium acnes (P. acnes), $5{\alpha}-reductase$ inhibition and comedolytic effects are the important pharmacological target sites of antiacne drughs. We previously reported on the antibacterial activities against P. acnes by natural products. In the present study the screening of $5{\alpha}-reductase$ inhibition and comedolytic effects from natural products were performed. Seven natural products such as Angelica koreana, Sophora flavescens, Prunus persica, Bombyx mori, Areca catechu, Galla rhois and Gleditschia koraiensis perfectly inhibited the activity of $5{\alpha}-reductase$ at the concentration of 0.01% (w/v). Sixteen natural products which were shown to have the potent antibacterial activities against P.acnes or $5{\alpha}-reductase$ inhibition activities were assayed for the comedolytic test. In the results of comedolytic effects on experimentally-induced comedones (EIC), Sophora flavescens showed the strongest comedolytic effect on EIC, and Polygonum cuspidatum and Angelica koreana showed stronger comedolytic effects on EIC than azelaic acid used for a positive control at the concentration of 3% (w/v). These results suggest that several natural products including Sophora flavescens can be developed as noble antiacne agents.

  • Rhizoma of Dryopteris crassirhizoma, Root of Glycyrrhiza uralensis, Root of Sophora flavescens and Pericarp of Citrus unshiu were extracted with ethanol. The ethanol extracts of 4 medicinal plant were tested for the antimicrobial activity against Streptococcus mutans. The extracts of Rhizoma of Dryopteris crassirhizoma, Root of Sophora flavescens, Root of Glycyrrhiza uralensis showed antimicrobial activity against Streptococcus mutans. At the 300ug/disk concentration of the ethanol extract from Dryopteris crassirhizoma and Sophora flavescens showed significant antimicrobial activity against Streptcoccus mutans. These results suggested that the extracts from Dryopteris crassirhizoma could be the potential source of antimicrobial agents against S. mutans.

  • This study was carried out to evaluate cytotoxicity of the extracts from Sophora flavescens Ait. against L1210 (lymphocytic leukemia) and $P388D_1$ (lymphoid neoplasms) cells in vitro. We have determined cytotoxicity by MTT {3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazo-liumbromide} assay. The order of cytotoxicity of Sophora flavescens Ait. extracts against L1210 and $P388D_1$ cells in vitro is as follows: AM> EASF > CFSF > MTSF > WSF > HXSF and AM> EASF> CFSF> MTSF> HXSF> WSF. These results suggest that the ethyl acetate soluble extract of Sophora flavescens Ait may be a valuable choice for the development of antitumor agents.

  • This study was carried out to evaluate cytotoxic effects of the roots of Sophora flavescens Ait. extracts on murine leukemia tumor cells lines $(P388D_1\;and\;L1210)$. Disruptions in cell organelles were determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay. The comparison of $IC_{50}$ values of the ethyl acetate of Sophora flavescens Ait. extract in leukemia cell lines showed that their susceptibility to these extracts decreased in the following order : Adriamycin>Fr.4>Fr.5>Fr.3>Fr.1>Fr.2 by the MTT assay. These results suggest that the fraction 4 of the ethyl acetate soluble extract of Sophora flavescens Ait. may be a valuable choice for the studies on the treatment of murine leukemia cell lines.

  • A yeast, Torulopsis bombicola, was cultured in the media fortified with soybean oil as a additional carbon source for 7 days with reciprocal shaking. From the culture broth, sophorose-lipid was isolated and treated with alkali to afford sophorose. The sophorose contained in the medium was acetylated and isolated through silica gel column chromatography. The aceylated sophorose was hydrolyzed with 5% KOH at room temperature to give rise to sophorose. Meanwhile, the MeOH extracts obtained from the pod of Sophora japonica was solvent-fractionated with n-BuOH and $H_2O$, and butanolic layer was chromatographed on silica gel column to afford a flavonoide-glycoside. The glycoside was hydrolyzed with 0.02 N $H_2SO_4$ to yield sophorose.

  • A high performance liquid chromatographic method was designed for the quantitative analysis of oxymatrine in Sophora Radix. The separation of oxymatrine was performed by reversed-phase chromatography with a $C_{18}$ column and a buffered aqueous solution containing acetonitrile, and monitored by UV absorption at 215 nm. Extraction of oxymatrine in Sophora Radix was carried out using various solvents and extraction methods. The optimum extraction efficiency for the crushed Sophora Radix was achieved by reflux at $80^{\circ}C$ in 50% ethanol for five hours. Most extraction methods used to complicate pretreatments. In this study, sublimation was employed for a extraction method without going through complicate pretreatments. Sublimation was carried out under high vacuum ($1{\times}10^{-3}$ torr) and at high temperature ($200^{\circ}C$). Extraction efficiency using Sublimation was found to be inferior to other extraction methods.

A Study on the Characteristics of Design and Acculturation of Planting of 'Guǐ(槐)' in Chirinjeong Wonlim of Pohang (포항 칠인정원림(七印亭園林)의 조영특성과 '괴목(槐木)' 식재(植栽)의 문화변용(文化變容))

  • The purpose of this study is to illuminate historical sensitivity and design in the days of the Joseon era and examines the phenomenon of acculturation as revealed in the planting of Sophora japonica and Zelkova serrata in the garden grove by looking into the purpose underlying the construction, formative intention and the process of change in Pohang's Chirinjeong wonlim. Chirinjeong, also called Sanggaejeong(雙槐亭) after the fact that two Zelkova serrata trees were planted there, is a hermit pavilion in Sa-il village, Chogok-ri, built in the 9th year of King Taejong(1409) by Jang, Pyo(張彪, 1349~?) of an officer of bureaucratic origin toward the end of the Goryeo Dynasty, the Chirinjeong garden grove is an element essential to Chogok, the 3rd Gok of Sanggaegoogok(雙溪九曲). The wonlim of Chirinjeong is divided into the pavilion section composed of the entrance area, Chirinjeong, composed of Zelkova serrata and Sophora japonica, and the pond section composed of a quadrangular pond and island(方池方島), and Lagerstroemia indica. In view of the intent of the name Chirinjeong or of the fact that the pavilion was named based on that the royal seal string was hung on the Sanggwaesoo(雙槐樹) or two Zelkova serrata trees, it is reasonable to view it as Zelkova serrata trees having been planted early in the days of the Joseon Dynasty and those additionally planted later on that symbolizes the position of the three nobles and the Sophora japonica. In addition, in spite of the fact that the type of trees supplementarily planted in the 21st year of King Yeongjo(1745) is known as Sophora japonica, it is impossible to rule out the possibility of the tree planted outside of Chirinjeong being Zelkova serrata. In short, the three Zelkova serrata trees planted along with Sophora japonica must certainly be evidence that Zelkova serrata planted along with Sophora japonica in Chirinjeong wonlim indicates that the off spring of the Indong Jang family could tell the difference between Sophora japonica and Zelkova serrata. In the recognition process of Zelkova serrata trees finding their way into Korea, it was known as Zelkova serrata on the one hand and as Sophora japonica on the other, and the former, which enjoys a comparative advantage over in terms of the easiness with which to purchase, growth speed and possibility of growing into a long-lived tree was interchanged with Zelkova serrata, a case of acculturation as manifested in the planting of Sophora japonica in the Chirinjeong garden grove.
