Selection and Cultivation of Microorganism Producing Iron Superoxide Dismutase(Fe-SOD) (Iron Superoxide Dismutase( Fe-SOD)를 생산하는 미생물의 선발 및 배양)

  • Pseudomonas plycolor was used to investigated the optimal culture condition to examine the various properties of superoxide dismutase (SOD). this SOD was inhibited by $H_2O_2$, azide ion, but not by cyanide ion. This result indicates that the enzyme might be a Fe-SOD. The composition of optimal culture medium for the enzyme production was 3% of glycerin, 1% of polypeptone, 0.5% of meat extract, 0.2% of KCI and the initial ph was 9.0 . The cultivation for the enzyme production was carried out in 500ml shaking flask containing 100ml of the optimal medium at $30^{\circ}C$ on a reciprocal shaker. The enzyme production reached maximum at 15hrs of cultivation and then declined sharply afterward.

Standardized Sod Production Using Box Seeding (상자파종에 의한 규격화된 잔디의 생산)

  • The objective of this research was to produce sod by box seeding for zoysiagrass or by vegetative propagation for zoysiagrass and manilagrass.1 Various ratio of peatmoss to sand(v /v) were prepared to find idea[ medium for fast and light weight sod production. Then, the days required for sod formation, the effect of growth regulators on the growth of turfgrass, and the various storage methods for winter keeping of sods were also investigated. 1.The mixed medium of sand and peatmoss(v /v, 1 : 2) showed more biomass production than that of sand. 2.In comparison of seeding rate of zoysiagrass, the amount of log /$m^2$ was most effective in the fast and dense sod formation. The amount of 20g /$m^2$ also showed fast sod formation. But, it resulted in weak plant and less tillering. During April to June, about 100 days were required to form sod with seeding rate of 5g /$m^2$ regardless of seeding time. Whereas 80 days were required to form sod in the rate log /$m^2$, which was 20 days shorter than that of 5g /$m^2$. 3.More than 85% of shoots in sod stored in field or plastic house during the winter time resumed the growth in good appearance after transplanting. The whole covering of ground with sod resulted in less weeds and faster formation of lawn. 4.Vegetative propagation of manilagrass showed about 7 to 15 days faster formation of sod than that of zoysiagrass. Application of GA increased shoot growth and BA increased the total number of tillering. However, the effects of the combined application of GA and BA were negligable.

  • This study was conducted to make the utmost use of the good traits and to improve the poor traits of zoysiagrass through the production technology for mixed lines sod in addition to breeding. Mixed lines sod was produced by mixing broad or middle leaf line with narrow leaf line by volume, a ratio of 1 to 2. The mixed lines sod of zoysiagrass was superior to the single ling sod, which is used commonly, for the establishment rate, coverage rate, shoot density, texture, leaf color, dormancy period cnd color, and visual quality.

Production of Superoxide Dismutase by Deinococcus radiophilus

  • The production of superoxide dismutase (SOD) varied in Deinococcus radiophilus, the UV resistant bacterium, depending upon different phases of growth, UV irradiation, and superoxide treatment. A gradual increase in total SOD activity occurred up to the stationary phases. The electrophoretic resolution of the SOD in cell extracts of D. radiophilus at each growth phase revealed the occurrence of MnSOD throughout the growth phases. The SOD profiles of D. radiophilus at the exponential phase received oxidative stress by the potassium superoxide treatment or UV irradiation also revealed the occurrence of a single SOD. However, these treatments caused an increase in SOD activity. The data strongly suggest that D. radiophilus has only one species of SOD as a constitutive enzyme, which seems to be a membrane-associated protein.

  • Background Diabetes is characterized by chronic hyperglycemia, which can increase reactive oxygen species (ROS) production by the mitochondrial electron transport chain. The formation of ROS induces oxidative stress and activates oxidative damage-inducing genes in cells. No research has been published on oxidative damage-related extracellular superoxide dismutase (EC-SOD) protein levels in human diabetic skin. We investigated the expression of EC-SOD in diabetic skin compared with normal skin tissue in vivo. Methods The expression of EC-SOD protein was evaluated by western blotting in 6 diabetic skin tissue samples and 6 normal skin samples. Immunohistochemical staining was also carried out to confirm the EC-SOD expression level in the 6 diabetic skin tissue samples. Results The western blotting showed significantly lower EC-SOD protein expression in the diabetic skin tissue than in the normal tissue. Immunohistochemical examination of EC-SOD protein expression supported the western blotting analysis. Conclusions Diabetic skin tissues express a relatively small amount of EC-SOD protein and may not be protected against oxidative stress. We believe that EC-SOD is related to the altered metabolic state in diabetic skin, which elevates ROS production.

Present Situation of Zoysiagrass (Zoysia spp.) Culture, Sod Production, and Bland by Prefecture in Japan (일본의 한국잔디의 재배, 생산 및 현별 브랜드화 현황)

  • This study was carried out to provide recent information on zoysiagrass (Zoysia spp.) sod production in Japan. Total zoysiagrass sod production acreage in Japan was 6,623 ha. The acreage by prefecture was 3,900 ha (58.9%) in Ibalaki, 779 ha (11.8%) in Tostoli, 609 ha (9.2%) in Kagosima, and 550 ha (8.3%) in Sijeumoka, respectively. Sod was harvested by 1.5 cm sod thickness. Mowing height of Goraisiba (Zoysia matrella) and Nosiba (Zoysia japonica) cultivars was cut by 10 mm and 25 mm height, respectively. Japan agricultural cooperative issued a warranty of sod production quality to zoysiagrass consumers. A quality evaluation for sod production was carried out continuously to maintain a reasonable degree of uniformity and hight quality of sod production three times a year. Also, zosiagrass sod production was rotated with Chinese cabbage (Brassica rapa var. glabra Regel) for improving fertility of soil every five to six years. Patented cultivars for promoting bland in Ibaraki prefecture of Japan were 'Tsukuba himae', 'Tsukuba kagayaki', 'Tsukuba Talou' and 'Tsukuba green'.

Sod Production in South Korea (한국의 잔디 생산 현황)

  • This study was carried out to provide data on current sod production in South Korea. Fifteen items were surveyed including production acreage, species and cultivars, experience on farming, production cost and the others during April to October in 2006. To estimate production acreage, 73 turfgrass growers were interviewed. Estimated acreage for turfgrass production was 2,947 ha. Acreages by province were 1,417 ha in Jeollanam-do(48%), 442 ha in Jeollabuk-do(14.9%), 344 ha in Gyeonggi-do(11.6%), 248 ha in Gyeongsangnam-do(8.4%), and 240 ha in Chungcheongnam-do(8.1%), respectively. The major sod producing regions were Jangseong-gun, Hampyeong-gun, Yeonggwang-gun, Gochang-gun, Sacheon city, Iksan city, Yeoju city, Yeoncheon-gun. Percentage of turf growers over 50 years old was 72.4 percent, which means production was carried out mainly by old farmers. Grower's career over 10 years was 84.8%. Cultivated species and varieties were 'Junggi'(55.5%) of fine-textured Korean lawngrass, common Korean lawngrass(37.4%), Kentucky bluegrass(3.8%), and new zoysiagrass cultivars(0.1%), respectively. Sod size were variable. Sod size of $18{\times}18cm $ was 43.5 percent. Sales route was mainly through sod distributor(40.5%). Sod producing cost was $2,160{\sim}2,730$ won per square meter at Jeolla-do areas.

Effect of 2,4,5-Trichlorobiphenyl (PCB-29) on Oxidative Stress and Activities of Antioxidant Enzymes in Tomato Seedlings

  • Leaves of two-week old seedlings of tomato (Lycopersicon esculentum) were treated with various concentrations (0, 0.2 and 0.4 $\mu$g/1) of 2,4,5-trichlorobiphenyl (PCB-29) and subsequent growth of seedlings, symptoms of oxidative stress and activities of antioxidant enzymes were investigated. Compared with the non-treated control, foliar application of PCB-29 decreased both biomass and superoxide ($O_2$) radical production but increased hydrogen peroxide production and lipid peroxidation such as malondialdehyde (MDA) formation with increased activities of superoxide dismutase (SOD), ascorbate peroxidase (APX) and guaiacol peroxidase (GPX). Further studies on the isozymes of SOD, peroxidase (POD) and APX showed that all three isozymes of SOD such as Mn-SOD, Fe-SOD and Cu/Zn-SOD, two among four isozymes of POD and all three isozymes of APX were selectively increased in response to PCB. Therefore, we suggest that a possible cause for the reduction of seedling growth by PCB exposure is the oxidative stress including over production of hydrogen peroxide and the selective expression of specific isozymes of some antioxidant enzymes.

Insect Pests in Turf Sod Production Areas in Korea (잔디 재배지 발생 해충 종류)

  • Turfgrass insect pests were investigated in different turf sod production areas of Korea. Twelve insect pest species of 7 families in 6 orders and one Eriophyidae mite, Aceria zoysiae were collected from turf sod production areas. The zoysiagrass mite was most frequently occurred zoysiagrass sod production areas. Damaged rate by zoysiagrass mite was increased from May to September. Noctuidae (Spodoptera depravata, Agrotis ipsilon and A. segetum) and Pyralidae (Crambus sp.) insect pests in Lepidoptera were attracted in A. ipsilon sex pheromone trap. Eggs and larva of A. ipsilon was the highest occurred August. Zoysiagrass mite and A. ipsilon were main insect pests in turf sod production areas in Korea.

Optimization of SOD Biosynthesis by Controlling Sucrose Concentration in the Culture of Carrot Hairy Root

  • In order to optimize the carrot hairy root culture for SOD production, a fed-batch culture of hairy roots was performed in a bioreactor. Maximum SOD activity was obtained when the hairy roots were transferred to the MS medium containing 110 g/1 concentration of sucrose. By controlling the sucrose concentration (70 g/1 sucrose for growth and 110 g/1 sucrose far production, respectively) In a two-stage fed-batch culture, 29 g/1 of the hairy roots was obtained based on the final dry mass. The volumetrically determined SOD activity and productivity in the fed-batch culture were about 6 times higher than those from the flask culture containing sucrose at 30 g/1 concentration.
