Effect of Growth Regulators on the Organ Differentiation and the Growth from the Axillary Bud of Sweetpotatoes in Vitro Culture (고구마의 액아배양에서 생장조절물질이 기관분화 및 생장에 미치는 영향)

  • This study was conducted to determine the optimum concentrations of growth regulators and their responses on the clonal propagation in axillary bud culture. Cultivars, Hongmi and Shinmi, responded differently to the levels of growth regulators, proliferation rate and shoot growth. The shoot and root of Hongmi cultivar in axillary bud culture were conspicuously induced by combination of NAA(0.1mg/l) and Kinetin(1mg/l) while Shinmi cultivar were affected by the single concentration of Kinetin(1mg/l) and BA(0.1mg/l), and also by the combination of NAA(0.1mg/l) and Kinetin(1mg/l). Better shoot growth and root initiation were obtained in the combination of NAA(0.1mg/l) and Kinetin(1mg/l) regardless of cultivars used when 5mm axillary buds were cultured. The shoots regenerated at the high levels of BA(1-5mg/l) were abnormally thicker and narrower leaves than normal plants and short in shoot height. Frequencies of abnormal plants were higher than that of the low level (0.1mg/l) of BA.

In vitro mass propagation and acclimatization of Haworthia truncata (하월시아 옥선(Haworthia truncata)의 기내 대량 증식 및 순화 조건 구명)

  • The purpose of this study was to investigate suitable parts for callus induction and optimal concentrations of growth regulators, contained in the medium affecting shoot and rooting for in vitro mass production of Haworthia truncata. Leaves and flower bud showed 100% callus formation rate at NAA $1{\sim}2mgL^{-1}$ treatment, and NAA $1mgL^{-1}$ + TDZ $2mgL^{-1}$ treatment. The flower stalk showed 75% callus formation rate, at NAA $2mgL^{-1}$ + TDZ $2mgL^{-1}$ treatment in H. truncata. While the rate of callus formation was high in leaves and flower bud, leaves were the most efficient in obtaining most culture parts. Shoot induction rate from callus was highest, at NAA $0.1mgL^{-1}$ treatment in H. truncata. Additionally, the number of shoots formation was 66.3 shoots high, in NAA $1mgL^{-1}$ + BA $0.1mgL^{-1}$ treatment in H. truncata. In the case of acclimatization of regenerated plant, growth characteristics did not show significant difference (95%) shading with respect to the different ratio of substrate mixture, and it was determined that would be appropriate, considering plant height and appearance preference of H. truncata. It was established that optimization of culture condition, was responsible for mass propagation in vitro cultures of H. truncata.

  • Adventitious shoot buds, without an intervening callus phase, were induced from stem explants of Trichosanthes kirilowii on MS medium supplemented with 2 mg/L BA. The culture medium, carbon source as well as plant growth regulators were found to influence further shoot multiplication and eventual shoot growth. A maximum number of shoots was obtained on a MS medium containing 0.5 mg/L BA and 3% sucrose. Shoot produced in vitro were rooted on half-strength phytagelled 1/2MS medium prior to transfer to green house condition.

Efficient Procedures for Direct Shoots Regeneration from Leaf Explants of Rehmania glutinosa Lib. (지황 잎조직 절편으로부터 신초 형성)

  • Adventitious shoots were directly induced from leaf explants of R. glutinosa, an important medicinal plant. Proliferating shoot cultures were obtained by culturing leaf discs on Murashige and Skoog(MS) medium alone or combination with auxins and cytokinins. MS medium supplemented with 1 $mg/{\ell}\;BA\;and\;2\;mg/{\ell}$ IAA was the most effective, providing high shoot bud formation frequency without formation of intervening callus. The effect of leaf age on adventitious shoot formation was also investigated. The maximum shoot bud production (93.4%) was achived using 3rd leaf from apex of 6 weeks old plantlets after seed germination. Plantlet were rooted on an half-strength MS (1/2MS) medium containing 0.1 $mg/{\ell}\;IBA$. This prtocol is useful for clonal propagation and Agrobacterium-mediated transforamtion in R. glutinosa.

Asexual Propagation of Kalopanax pictus by Root Cutting (근삽에 의한 음나무의 무성번식)

  • This research was carried out to establish the large quantity propagation system for Kalopanax pictus using root cutting. The results are as follows. As there was no significant difference in the promotion of adventitious shoot development from the root cuttings of Kalopanax pictus among kinds (IBA, NAA and Kinetin) and concentrations (0 to 2000 mg/L) of growth regulators, no application is recommended, which showed more than 90% of shoot development. Horizontal and vertical placement of rooting showed more than 93% of shoot occurrence, but the latter showed more desirable growth traits in the number of shoot development, number of leaf, petiole length, and leaf length and leaf width Polarity was observed and thus normal vertical placement of root cutting is preferred. However, root cuttings placed upside down showed the shoot development at the distal end, which indicates further sophisticated test is necessary to investigate this phenomenon.

Mass Propagation by In Vitro Culture of Bupleurum latissimum Nakai (섬시호(Bupleurum latissimum Nakai)의 조직배양을 통한 대량생산)

  • This study was carried out to establish the micropropagation system of Bupleurum latissimum Nakai that is a Korean native endangered species. Callus were induced from the leaf, petiole and floral bud and the percentage of callus formation was highest in the floral bud on the MS medium containing 2.0 mg ${\cdot}$ $L^{-1}$ 2,4-D. Especially, callus induced from floral bud was formed 77.8% and the percentage of shoot formation was 42.6% on the MS medium containing 2.0 mg ${\cdot}$ $L^{-1}$ 2,4-D plus 1.0 mg ${\cdot}$ $L^{-1}$ TDZ. For simultaneously callus formation and shoot regeneration, 1/2 MS medium was more effective than MS medium. The percentage callus formation, shoot regeneration and rooting were 46.3%, 13.0%, 13.0% in 1/2 MS medium, respectively. Soot regeneration from callus was good in 1/2 MS medium supplemented with 2.0 mg ${\cdot}$ $L^{-1}$ 2,4-D plus 1.0 mg ${\cdot}$ $L^{-1}$ BA where percentage of shoot regeneration was 74.1 %, and the number of shoot per explant was 2.4. The percentage of rooting was lowest (57.8%) in control while it was highest (97.8%) in 1.5 mg ${\cdot}$ $L^{-1}$ NAA. In acclimatization of regenerated plantlets, the percentage of survived plantlets was highest (86.1%), and plant height, root length and fresh weight were good in the soil for horticulture.

Influence of Medium and Plant Growth Regulator on Micropropagation Efficiency in Blueberry (블루베리의 미세번식에서 배지와 식물생장조절제의 영향)

  • The aim of this study was to develop an effective production system of blueberry plants by using tissue culture technique. Murashige and skoog medium (MS) and woody plant medium (WPM) were compared for shoot formation of highbush blueberries. Also medium supplemented with zeatin/2-isopentenyl adenine (2iP)/benzyl aminopurine (BA) (1, 2/10, 15/4, $6mg{\cdot}L^{-1}$)and zeatin/2iP/BA (0.5/10, 15/$0.05mg{\cdot}L^{-1}$) as plant growth regulators to determine the effect of shoot formation and shoot proliferation, respectively. The shoot explants cultured on WPM showed higher shoot formation rates, more number of nodes, and longer root length than those on MS medium during the primary culture. Shoots were not formed when the explants were cultured on the medium without plant growth regulators or on only BA. The shoot explants cultured on the medium supplemented with 2iP showed low rates of shoot formation. On the other hand, zeatin was the most effective for shoot formation and growth of the explants. Also influence of different cytokinins (zeatin, 2iP) on the shoot proliferation of subcultured shoot explants was studied. There was no significant difference among the different concentrations of zeatin in the rate of shoot formation and number of shoots. However at higher concentration of zeatin, number of nodes was increased, and shoot length was shorted. The proper concentrations of zeatin for shoot propagation in subculture were found to be $0.5mg{\cdot}L^{-1}$ and $1mg{\cdot}L^{-1}$.

  • Since garlics (Allium sativum L.) are propagated through cloves, infection by virus or other pathogens may become severe problem if not using high quality seed bulbs every year resulting in the reduction of yield and bulb quality, In order to solve this problem, the establishment of virus-free bulb production and its supply system have been required because no chemicals were found to eliminate viruses from seed bulbs. This experiment was conducted to develop an effective production technique of high quality seed bulbs using shoot-tip culture. Over 90% of shoot-tips explanted on January L 1990 were survived at constant temperature of either 20, 24 or 28$^{\circ}C$, wheres 88% at alternate temperature (28/20$^{\circ}C$). The growth of shoot and root was most vigorous at constant 24$^{\circ}C$, and least at alternate temperature (28/20$^{\circ}C$) condition. When shoot-tips were explanted June 21 to August 1,1991, survival and growth of shoot-tips was most vigorous on MS medium supplymented with 0.1 mg/L NAA and 2 mg/L kinetin and least 1 mg/L Gh$_3$. The shoot-tips taken from the seed bulbs stored at 4$^{\circ}C$ for 15 to 60 days were placed on MS medium, shoot growth and in vitro bulblet formation increased slightly as affected by the increase of told treatment period at 4$^{\circ}C$.

  • We, Tong Yang Moolsan Co. Ltd. (TYM) set up the mass-production system for virus-free seed garlic via tissue culture technique. TYM's tissue culture technique is called as 'Multiple shoot propagation technique'. This technique can lead mass propagation of genetically homogeneous seed garlic in a short period because of its highly proliferation rate of in vitro shoots ($15^{10}$ /year). TYM researchers applied the technique to some selected garlic cultivars with superior characteristics and carried out field test of productivity in the inside and outside of the country for several years. According to the yearly results of field test with virus-free seed garlic, we ascertained that virus-free seed garlic can produce the highly yield increase (max. above $50\%$) and also can enhance the product quality. Consequently, we estimated that TYM's seed garlic will contribute to farmers with increase of income and can elevate the national position of garlic market in the world for its competitive power of technical and production cost.
