RAD2 and PUF4 Regulate Nucleotide Metabolism Related Genes, HPT1 and URA3

  • Yeast RAD2, a yeast homolog of human XPG gene, is an essential element of nucleotide excision repair (NER), and its deletion confers UV sensitivity and NER deficiency. 6-Azauracil (6AU) sensitivity of certain rad2 mutants revealed that RAD2 has transcription elongation function. However, the fundamental mechanism by which the rad2 mutations confer 6AU sensitivity was not clearly elucidated yet. Using an insertional mutagenesis, PUF4 gene encoding a yeast pumilio protein was identified as a deletion suppressor of rad2${\Delta}$ 6AU sensitivity. Microarray analysis followed by confirmatory RT-qPCR disclosed that RAD2 and PUF4 regulated expression of HPT1 and URA3. Overexpression of HPT1 and URA3 rescued the 6AU sensitivity of rad2${\Delta}$ and puf4${\Delta}$ mutants. These results indicate that 6AU sensitivity of rad2 mutants is in part ascribed to impaired expression regulation of genes in the nucleotide metabolism. Based on the results, the possible connection between impaired transcription elongation function of RAD2/XPG and Cockayne syndrome via PUF4 is discussed.

경계요소법을 이용한 사출성형금형 냉각시스템의 최적설계 (Optimum design of injection molding cooling system via boundary element method)

  • The cooling stage is the very critical and most time consuming stage of the injection molding process, thus it cleary affects both the productivity and the part quality. Even through there are several commercialized package programs available in the injection molding industry to analyze the cooling performance of the injection molding coling stage, optimization of the cooling system has npt yet been accomplished in the literature due to the difficulty in the sensitivity analysis. However, it would be greatly desirable for the mold cooling system designers to have a computer aided design system for the cooling stage. With this in mind, the present study has successfully developed an interated computer aided design system for the injection molding cooling system. The CAD system utilizes the sensitivity analysis via a Boundary Element Method, which we recently developed, and the well-known CONMIN alforuthm as an optimization technique to minimize a weighted combination (objective function) of the temperature non-uniformity over the part surface and the cooling time related to the productivity with side constranits for the design reality. In the proposed objective function , the weighting parameter between the temperature non-uniiformity abd the cooling time can be adjusted according to user's interest. In this cooling system optimization, various design variable are considered as follows : (i) (design variables related to processing conditions) inlet coolant bulk temperature and volumetric flow rate of each cooling channel, and (ii) (design variables related to mold cooling system design) radius and location of each cooling channel. For this optimum design problem, three different radius and location of each cooling channel. For this optimum design problem, three different strategies are suffested based upon the nature of design variables. Three sample problems were successfully solved to demonstrated the efficiency and the usefulness of the CAD system.

축방향으로 압축을 받는 GFRP 원통형 판넬의 유한요소 모델링 (Finite Element Modelling of Axially Compressed GFRP Cylindrical Panels)

  • 복합재료를 효과적으로 사용하기 위하여 복합재료 구조물의 설계기준 개발을 위한 연구가 진행되고 있다. 금속 셀의 좌굴에 대한 설계시에는 초기 결함의 영향과 탄성임계좌굴응력을 근거로 한 녹다운계수(Knock-down factor)를 정의하는 것이 중요한 과정이나 복합재료 쉘의 좌굴에 대한 설계시에는 초기 결함에 대한 민감도가 거의 연구되어 있지 않은 실정이다. 복합재료 쉘의 좌굴거동에 영향을 주는 설계변수는 많기 때문에, 쉘의 설계시 이 변수들로 인한 초기 결함 민감도를 분석하기 위하여 많은 실험을 필요로 하고 있으며 실험 이외의 다른 방법으로서는 이미 검증된 수치모델을 사용하는 것이다. 본 논문에서는 복합재료 쉘요소를 개발하는데 사용된 이론을 요약, 정리 하였으며 수치예제를 통하여 본 연구에서 제안한 쉘요소의 정확성을 검증하였다. 그리고 축방향으로 압축을 받는 GFRP 곡선형 판넬의 설계시 고려해야 하는 각 변수들을 다양하게 변화시키면서 좌굴거동에 미치는 영향을 유한요소 모델링에 의해 고찰하였다. 방법으로서 초기 결합 및 두께의 진폭을 고려한 비선형 해석과 고유치 해석을 수행하였으며 이 결과를 이용하여 녹다운 계수를 산출하였다.

양방향 진화적 구조최적화를 이용한 신뢰성기반 위상최적화 (Reliability-Based Topology Optimization Based on Bidirectional Evolutionary Structural Optimization)

  • This paper presents a reliability-based topology optimization (RBTO) based on bidirectional evolutionary structural optimization (BESO). In design of a structure, uncertain conditions such as material property, operational load and dimensional variation should be considered. Deterministic topology optimization (DTO) is performed without considering the uncertainties related to the design variables. However, the RBTO can consider the uncertainty variables because it can deal with the probabilistic constraints. The reliability index approach (RIA) and the performance measure approach (PMA) are adopted to evaluate the probabilistic constraints in this study. In order to apply the BESO to the RBTO, sensitivity number for each element is defined as the change in the reliability index of the structure due to removal of each element. Smoothing scheme is also used to eliminate checkerboard patterns in topology optimization. The limit state indicates the margin of safety between the resistance (constraints) and the load of structures. The limit State function expresses to evaluate reliability index from finite element analysis. Numerical examples are presented to compare each optimal topology obtained from RBTO and DTO each other. It is verified that the RBTO based on BESO can be effectively performed from the results.

Seismic damage detection of a reinforced concrete structure by finite element model updating

  • Finite element (FE) model updating is a useful tool for global damage detection technique, which identifies the damage of the structure using measured vibration data. This paper presents the application of a finite element model updating method to detect the damage of a small-scale reinforced concrete building structure using measured acceleration data from shaking table tests. An iterative FE model updating strategy using the least-squares solution based on sensitivity of frequency response functions and natural frequencies was provided. In addition, a side constraint to mitigate numerical difficulties associated with ill-conditioning was described. The test structure was subjected to six El Centro 1942 ground motion histories with different Peak Ground Accelerations (PGA) ranging from 0.06 g to 0.5 g, and analytical models corresponding to each stage of the shaking were obtained using the model updating method. Flexural stiffness values of the structural members were chosen as the updating parameters. In model updating at each stage of shaking, the initial values of the parameter were set to those obtained from the previous stage. Severity of damage at each stage of shaking was determined from the change of the updated stiffness values. Results indicated that larger reductions in stiffness values occurred at the slab members than at the wall members, and this was consistent with the observed damage pattern of the test structure.

Finite element impact analysis for the design of structurally dissipating rock-shed

  • This paper presents finite element impact analysis for the design of Structurally Dissipating Rock-shed (SDR), an innovative design of reinforced concrete rock-shed. By using an appropriate finite element impact algorithm, the SDR structure is modelled in a simplified but efficient way. The numerical results are firstly verified through comparisons with the results of the experiments recently realized by ESIGEC and TONELLO I.C. It is shown that, using this impact algorithm, it is possible to correctly predict the SDR structural behaviour under different rock-fall impact conditions. Moreover, the numerical results show that the slab centre is the critical impact location for reinforced concrete slab design. The impact analyses have thus been focused on the impacts at the slab centre for the SDR structural optimization. Several series of parametric studies have been carried out with respect to load cases and engineering parameters choices. These numerical results support the robustness of the new SDR concept, and serve to optimize SDR structure and improve its conventional engineering design, especially for ensuring the slab punching shear resistance.

  • 철근콘크리트 구조물을 해석하는 경우, 하중조건 및 이들 하중의 증가에 따라 나타나게 되는 상이한 거동 특성들 즉, 인장균열, 압축파괴, 다축상태일 때의 강도의 증가, 전단파괴 시의 취성적 파괴특성 등을 반드시 고려해야만 한다. 유한요소법에 의한 해석적 평가 시, 최대하중 이후에는 요소크기에 따라 상이한 구조응답을 나타내는 요소의존적인 응답결과를 보인다. 본 연구에서는 이러한 요소의존성을 극복하기 위한 연구결과들 중 하나인 적분형 비국소화 모델을 이용하여 전력구 모형 실험체의 파괴거동을 해석하였다. 실험결과와 해석결과를 비교하여 적분형 비국소 모델의 신뢰성과 타당성을 검토하는 것을 목적으로 한다.

터빈로터 수명예측 프로그램의 개발 및 민감도 분석 (Development and Sensitivity Analysis of Life Estimation Program for Turbine Rotors)

  • Steam turbine rotors are the most critical and highly stressed components of a steam power plant; therefore, the life expectancy of the turbine rotor is an important consideration for the safety of a steam power plant. The objective of this paper is to develop a life estimation program for turbine rotors for all possible operating conditions. For this purpose, finite element analysis was carried out for four normal operating modes (cold, warm, hot and very hot starts) using ABAQUS codes. The results are made into databases to evaluate the life expenditure for an actual operating condition. For any other possible abnormal operating condition, the operating data are transmitted to the server (workstation) through a network to carry out finite element analysis. Damage estimation is carried out by transmitting the finite element analysis results to the personal computer, and then the life expectancy is calculated.

유한요소법을 이용한 자기센서용 자속집속기의 해석 (Analysis of Magnetic Concentrator of Magnetic Sensor by Using Finite Element Method)

  • 본 연구에서는 홀센서의 감도를 높이기 위해 사용되는 자속집속기의 두께, 홀소자와의 상대적 위치, 모서리 형상에 따르는 발생 자계를 유한요소법을 이용하여 계산하였다. 자속집속기의 두께가 얇을수록 발생 자계가 커지는 것을 알 수 있었고, 이 경향은 반자계를 고려한 겉보기 상대투자율의 두께의존성과 유사하다는 것을 알 수 있었다. 홀소자와의 상대적 위치에 따라 최대의 자계를 발생시키는 자속집속기의 최적의 두께가 변화하는 것을 알 수 있었다. 자속집속기와 홀소자의 간격이 가까울 경우, 자속집속기의 모서리 형상에 경사가 없는 것이 유리하지만, 자속집속기와 홀소자의 간격이 멀어질수록 자속집속기의 모서리를 경사지게 하는 것이 유리하다는 것을 알 수 있었다.

Mach-Zehnder 광섬유 간섭계를 이용한 압전형 진동발생기의 동특성 조사 (The Measurements of Vibration Displacement of the Piezoelectric Exciter Using Mach-Zehnder Optical Fiber Interferometer)

  • The vibration exciter with the accurate calibration requires a low distortion along a single axis over a wide range of frequency. The fabricated piezoelectric exciter was composed of a base, piezoelectric element(Venitron PZT 5A), electrode and seismic mass. Its performance characteristics is evaluated the Mach-Zehnder optical fiber interferometer. The phase of the optical wave passing through the optical fiber around the piezoelectric element was related the vibrational amplitude with a change of the applied sinusoidal voltage on the piezoelectric element. The dynamic characteristics of vibration exciter can be obtained by measuring the vibrational amplitude with a sinusoidal applied voltage on the piezoelectric element. The sensitivity of the fabricated piezoelectric exciter had a 0.4 nm/V which was uniform up to 20 kHz.
