Activation of the Mating Pheromone Response Pathway of Lentinula edodes by Synthetic Pheromones

  • Pheromone (PHB)-receptor (RCB) interaction in the mating pheromone response pathway of Lentinula edodes was investigated using synthetic PHBs. Functionality of the C-terminally carboxymethylated synthetic PHBs was demonstrated by concentration-dependent induction of a mating-related gene (znf2) expression and by pseudoclamp formation in a monokaryotic strain S1-11 of L. edodes. Treatment with synthetic PHBs activated the expression of homeodomain genes (HDs) residing in the A mating type locus, and of A-regulated genes, including znf2, clp1, and priA, as well as genes in the B mating type locus, including pheromone (phb) and receptor (rcb) genes. The synthetic PHBs failed to discriminate self from non-self RCBs. PHBs of the B4 mating type (B4 PHBs) were able to activate the mating pheromone response pathway in both monokaryotic S1-11 and S1-13 strains, whose B mating types were B4 (self) and B12 (non-self), respectively. The same was true for B12 PHBs in the B4 (non-self) and B12 (self) mating types. The synthetic PHBs also promoted the mating of two monokaryotic strains carrying B4-common incompatible mating types ($A5B4{\times}A1B4$). However, the dikaryon generated by this process exhibited abnormally high content of hyphal branching and frequent clamp connections and, more importantly, was found to be genetically unstable due to overexpression of mating-related genes such as clp1. Although synthetic PHBs were unable to discriminate self from non-self RCBs, they showed a higher affinity for non-self RCBs, through which the mating pheromone response pathway in non-self cells may be preferentially activated.

Mechanisms Underlying Plk1 Polo-Box Domain-Mediated Biological Processes and Their Physiological Significance

  • Mammalian polo-like kinase 1 (Plk1) has been studied intensively as a key regulator of various cell cycle events that are critical for proper M-phase progression. The polobox domain (PBD) present in Plk1's C-terminal noncatalytic region has been shown to play a central role in targeting the N-terminal kinase domain of Plk1 to specific subcellular locations. Subsequent studies reveal that PBD binds to a phosphorylated motif generated by one of the two mechanisms - self-priming by Plk1 itself or non-self-priming by a Pro-directed kinase, such as Cdc2. Here, we comparatively review the differences in the biochemical steps of these mechanisms and discuss their physiological significance. Considering the diverse functions of Plk1 during the cell cycle, a better understanding of how the catalytic activity of Plk1 functions in concert with its cisacting PBD and how this coordinated process is intricately regulated to promote Plk1 functions will be important for providing new insights into different mechanisms underlying various Plk1-mediated biological events that occur at the multiple stages of the cell cycle.

  • The Experiment to Detect the Global EoR Signature (EDGES) has probed the status of the early Universe through the global 21cm observation. The claimed (brightness temperature) of ~ 500 mK absorption dip at z~17 against the continuum background cannot be explained in the standard LambdaCDM framework. In the meantime, the Planck 2018 Legacy Data, especially the E-mode polarization power spectrum, puts rather strong constraints on the high-redshift reionization process. We show how these two observational contraints can be accomodated in a series of reionization scenarios, with a special focus on the strongly self-regulated reionization by first stars.

Web-based E-learning System Supporting an Effective Self-directed Learning Environment (효과적인 자기주도적 학습 환경을 지원하는 웹 기반 이-러닝 시스템)

  • For success in E-learning, support from a learning environment that enables learners to perform self-directed learning more effectively is assumed. However, most existing e-learning systems do not maximize the improvement in learners' self-regulated learning ability because they only partially accommodate factors that can facilitate self-directed learning. In this paper, a web-based e-learning system is designed and proposed that enables support of an enhanced self-directed learning environment by providing various learning methods, evaluation methods, learning content levels, and strategies for learning motivation in various conditions, and synthetically reflecting them. To validate the effectiveness of the proposed system, it was applied to the subject of data structures in a university course, and an online survey was conducted with the students. The results indicated that the proposed system can support a learning environment in which students can perform more effective self-directed learning, enhancing their learning ability.

Development of Learning Strategy Scale for College Students (전문대학생을 위한 학습전략 진단 도구의 개발)

  • The purpose of this study was to develop of learning strategy scale for college students. This study further classified several sub-areas and defined each concepts of learning strategy. Based upon the classification of each sub-areas, tentative test items were developed through the verification of validity by three professionals. A pilot study of the developed scale was administered to 239 college students. And the research collected major data from 1,012 college students. Data were analyzed to obtain item quality, reliability, and validity analysis. The results of this study were as follows. The scale for learning strategy was defined by eight factors and they were 'self-management strategy', 'examination-readiness strategy', 'cognitive strategy', 'memorizing strategy', 'reporting strategy', 'resource-utilization strategy', 'self-regulated strategy', 'cooperative learning strategy'. The results of the confirmatory factor analysis proved the eight factors in the learning strategy. And criterion validity evidence was also obtained from a correlation analysis of the level of academic achievement.

Clustering Approaches to Identifying Gene Expression Patterns from DNA Microarray Data

  • The analysis of microarray data is essential for large amounts of gene expression data. In this review we focus on clustering techniques. The biological rationale for this approach is the fact that many co-expressed genes are co-regulated, and identifying co-expressed genes could aid in functional annotation of novel genes, de novo identification of transcription factor binding sites and elucidation of complex biological pathways. Co-expressed genes are usually identified in microarray experiments by clustering techniques. There are many such methods, and the results obtained even for the same datasets may vary considerably depending on the algorithms and metrics for dissimilarity measures used, as well as on user-selectable parameters such as desired number of clusters and initial values. Therefore, biologists who want to interpret microarray data should be aware of the weakness and strengths of the clustering methods used. In this review, we survey the basic principles of clustering of DNA microarray data from crisp clustering algorithms such as hierarchical clustering, K-means and self-organizing maps, to complex clustering algorithms like fuzzy clustering.

  • In Web-based learning systems, the techniques, as self-regulated learning, self-directed learning, are used to improve the effect of learner's study. These techniques are methods considering learner's study level but to consider the learner's study ability properly, the tailored course for learner should be applied. In this research, the learning system considering learner's study ability was proposed. To decide a learner's study ability, IRT(Item Response Theory) was applied and learning contents and question items were developed and applied by the degree of difficulty.

  • The complement system is part of the innate immune response and as such defends against invading pathogens, removes immune complexes and damaged self-cells, aids organ regeneration, confers neuroprotection, and engages with the adaptive immune response via T and B cells. Complement activation can either benefit or harm the host organism; thus, the complement system must maintain a balance between activation on foreign or modified self surfaces and inhibition on intact host cells. Complement regulators are essential for maintaining this balance and are classified as soluble regulators, such as factor H, and membrane-bound regulators. Defective complement regulators can damage the host cell and result in the accumulation of immunological debris. Moreover, defective regulators are associated with several autoimmune diseases such as atypical hemolytic uremic syndrome, dense deposit disease, age-related macular degeneration, and systemic lupus erythematosus. Therefore, understanding the molecular mechanisms by which the complement system is regulated is important for the development of novel therapies for complement-associated diseases.

  • This paper presents a 6-bit self-calibrated D/A converter designed with current cell matrix structure. This structure is based on the current-cell matrix configuration using a regulated gate cascode current cell with 3-way switch. using from CMOS process and 5V power supply, the simulated conversion rate is 45.78MHz and the average mismatching properties among current sources are reduced to 0.02% and 0.005%, respectively when 1% and 0.5% errors of current sources are considered.

  • The purpose of this study was to examine the effect of coaches' transformational transactional leadership on athletes' self-efficacy among high school students. A total of 400 high school athletes from S and D city were selected through the convenience sampling method. A total of 258 questionnaires were returned and 39 were discarded due to missing values. Therefore, a total of 219 questionnaires were analyzed using SPSS 20.0. This study found the following: First, the intellectual stimulation of leadership had the greatest effect on self-regulated efficacy of self-efficacy. Second, inspirational motivation of leadership had the greatest effect on confidence of self-efficacy. Third, leadership factors did not affect task difficulty preference of self-efficacy.
