Isolation and Characterization of an Acyclic Isoprenoid from Semecarpus anacardium Linn. and its Antibacterial Potential in vitro - Antimicrobial Activity of Semecarpus anacardium Linn. Seeds -

  • Objectives: Semecarpus anacardium Linn. is a plant well-known for its antimicrobial, antidiabetic and anti-arthritic properties in the Ayurvedic and Siddha system of medicine. This has prompted the screening of this plant for antibacterial activity. The main aims of this study were to isolate compounds from the plant's seeds and to evaluate their antibacterial effects on clinical bacterial test strains. Methods: The n-butanolic concentrate of the seed extract was subjected to thin layer chromatography (TLC) and repeated silica gel column chromatography followed by elution with various solvents. The compound was identified based on observed spectral (IR, $^1H$ NMR, $^{13}C$ NMR and high-resolution mass spectrometry) data. The well diffusion method was employed to evaluate the antibacterial activities of the isolated acyclic isoprenoid compound (final concentration: $5-15{\mu}g/mL$) on four test bacterial strains, namely, Staphylococcus aureus (MTCC 96), Bacillus cereus (MTCC 430), Escherichia coli (MTCC 1689) and Acinetobacter baumannii (MTCC 9829). Results: Extensive spectroscopic studies showed the structure of the isolated compound to be an acyclic isoprenoid ($C_{21}H_{32}O$). Moreover, the isoprenoid showed a remarkable inhibition of bacterial growth at a concentration of $15{\mu}g/mL$ compared to the two other doses tested (5 and $10{\mu}g/mL$) and to tetracycline, a commercially available antibiotic that was used as a reference drug. Conclusion: The isolation of an antimicrobial compound from Semecarpus anacardium seeds validates the use of this plant in the treatment of infections. The isolated compound found to be active in this study could be useful for the development of new antimicrobial drugs.

Fitness cost and competitive ability of transgenic herbicide-tolerant rice expressing a protoporphyrinogen oxidase gene

  • The expression of transgenic traits in genetically modified crops is sometimes associated with decreases in crop performance or fitness. These decreases in performance or fitness of transgenic plants in unfavourable conditions may provide valuable information about the ecological consequences of transgene escape. In a glasshouse trial, we tested the cost associated with resistance to herbicides by comparing the growth, yield, and competitive ability of transgenic rice with its parental non-transgenic line. This new line was developed for constitutive overexpression of protoporphyrinogen oxidase (PPO) to increase resistance to herbicides. We evaluated nine agronomic traits of transgenic and non-transgenic rice grown in a replacement series design over four densities. Competitive ability was also assessed between transgenic and non-transgenic plants by analyzing their relative yields based on biomass and seed weight data. Our results indicated that non-transgenic plants showed greater performance than did the transgenic plants when those genotypes were grown in mixtures. The non-transgenic rice plants exhibited superior competitive ability at certain combinations of planting densities and genotype proportions. These results suggest that PPO-herbicide resistance incurs some costs in plant performance and competitive ability.

Estimation of Carcass Cut Traits in Live Pigs (돼지 생체에서 부분육 형질의 추정)

  • Means measuring carcass cut traits in live pigs is needed to genetically improve the production of favorite cuts in swine. The data of body measurements as well as carcass traits were collected from 432 heads of 4 crossbred lines. Weights of most parts and percentages of belly and boston in carcass were significantly influenced by days to slaughter. Most of off test body measurements show higher correlations with carcass cut traits than body measurements of on test and market do. The multiple regression equations for estimation of carcass cut traits by off test body measurements have higher accuracy than by body measurements of on test and market. The coefficients of determination in estimation of polynomial regression with off test body measurements after adjustments of carcass cut traits for days to market were ranged 0.59 to 0.68 and 0.33 to 0.43 in weights and percentages of carcass cuts, respectively. Develop- ment of an excellent Korean type seed stock for favorite cuts can be expected, if the estimation of carcass cut traits for live animals is implemented in swine genetic improvement program.

  • In coagulant control of water treatment plants, rule extraction, one of datamining categories, was performed for coagulant control of a water treatment plant. Clustering methods were applied to extract control rules from data. These control rules can be used for fully automation of water treatment plants instead of operator's knowledge for plant control. To perform fuzzy clustering, there are some coefficients to be determined and these kinds of studies have been performed over decades such as clustering indices. In this study, statistical indices were taken to calculate the number of clusters. Simultaneously, seed points were found out based on hierarchical clustering. These statistical approaches give information about features of clusters, so it can reduce computing cost and increase accuracy of clustering. The proposed algorithm can play an important role in datamining and knowledge discovery.

Variation of the Germination Responses to Temperature of Plantago asiatica Seed Population along Altitude in Mt. Chiri (지리산에서 고도에 따른 질경이(Plantago asiatica) 개체군의 온도에 대한 발아습성의 변이)

  • This study was focused on the effect of the altitude on the geographical variations of germination characteristics in the populations of Plantago asiatica L. distribute in Mt. Chiri. There was a significant difference among the 14 groups in the phenological pattern in relation to altitudes. When the altitude becomes higher, the thermal time which was required for 10 to 80% germination rate showed higher and wider distribution. On the other hand, the germination response of increasing temperature (IT) and secreasing temperature (DT) regime was classified into 3 group. The first group was the spering germination type. This group showed that the IT regime hadhigher germination rate than that of the DT regime, and was distributed in Macheon(300m) and Packmudong (500m). The second was the spring-fall germination type which was distributed in Hadong (900m) and Saemt대 (1100m). This group also showed higher germination rate in the IT regime, but the difference of the germination rate between IT and DT regime was less than that in the first group (the spring type). The third group was the early fall germination type which was found in the Nogodan (1507m), Changetomok (1750m) and Cheonwangbong (1915m). The germination rate of this group showed almost 100% similarity between IT and DT regime. These data suggested that the geographical varations of germination characteristics within species was an important ecological strategy for the survival from severe environmental conditions.

Immersion rate of Protothaca jedoensis spat on Different Grain Sizes (살조개, Protothaca jedoensis 치패의 저질입도에 따른 잠입율)

  • In order to obtain the basic biological data for effective seed production of Protothaca jedoensis, the influence of sediment condition on its immersion rate was investigated. Immersion rate of P. jedoensis spats was investigated in the different substrate groups; mud, fine sand, medium sand and coarse. In 72 hours, immersion rate and survival rate of spat was 90.0 and 90.0, 76.7 and 98.3, 61.7 and 86.7, and 85.0 and 93.3%, respectively. And also, survival rates of the groups were 90.0%(mud), 98.3%(fine sand), 86.7% (medium sand) and 93.3% (coarse sand) respectively. The immersion time and rate of spat was increased with an increase in shell length.

The Upper Ascending Reticular Activating System between Intralaminar Thalamic Nuclei and Cerebral Cortex in the Human Brain

  • Purpose: The ascending reticular activating system (ARAS) is responsible for regulation of consciousness. In this study, using diffusion tensor imaging (DTI), we attempted to reconstruct the thalamocortical projections between the intralaminar thalamic nuclei and the frontoparietal cortex in normal subjects. Methods: DTI data were acquired in 24 healthy subjects and eight kinds of thalamocortical projections were reconstructed: the seed region of interest (ROI) - the intralaminar thalamic nuclei and the eight target ROIs - the medial prefrontal cortex, dorsolateral prefrontal cortex, ventrolateral prefrontal cortex, orbitofrontal cortex, premotor cortex, primary motor cortex, primary somatosensory cortex, and posterior parietal cortex. Results: The eight thalamocortical projections were reconstructed in each hemisphere and the pathways were visualized: projections to the prefrontal cortex ascended through the anterior limb and genu of the internal capsule and anterior corona radiata. Projections to the premotor cortex passed through the genu and posterior limb of the internal capsule and middle corona radiata; in contrast, projections to the primary motor cortex, primary somatosensory cortex, and posterior parietal cortex ascended through the posterior limb of the internal capsule. No significant difference in fractional anisotropy, mean diffusivity, and fiber volume of all reconstructed thalamocortical projections was observed between the right and left hemispheres (p>0.05). Conclusion: We reconstructed the thalamocortical projections between the intralaminar thalamic nuclei and the frontoparietal cortex in normal subjects. We believe that our findings would be useful to clinicians involved in the care of patients with impaired consciousness and for researchers in studies of the ARAS.

Suspended Time Dependent Meat Weight Increase of Oysters, Crassostrea gigas, in Pukman Bay, Korea (북만의 양식 참굴, Crassostrea gigas의 수하시기에 따른 육중량 변화)

  • From September 1994 to April 1996, we observed the suspending time dependant meat increase (dry weight) for oysters at Pukman Bay in Korea. The oysters which suspended in September increased to 5 g in meat weight (dry weight) by January. Ones in June increased rapidly but lessened during summer and this stagnation of meat increase extended to autumn. It took long time to compensate for the loss of meat weight owing to spawning. Oysters suspended in July or August showed low growth or stagnation due to the high water temperature and spawning activities after suspending. However, meat weight showed sudden increasement after December and achieved 5 g at the late farm period. The meat increase of oysters suspended in October or November was very slow during entire growing period. Most of them did not achieved 5 g during farm period. The negative correlation was observed between meat growth and water temperature in September. Also, there was a high correlation between meat growth and chlorophyll-a. It was believed that deficiency of food organisms in the water column lead slow growth in terms of meat growth, particularly right after transplanting seed oysters to the farm. Our study suggests that proper time for transplanting oysters is between August and mid-September. However, the data indicate that good growth of oysters is depending upon amount of food available in the water column.

Investigation on the Inhabitation Environments and Growth Conditions of Machilus thunbergii Community in Pyonsanbando (변산반도내 후박나무군락의 서식환경 및 생육실태에 관한 조사연구)

  • The inhabitation environments and growth conditions of Machilus thunbergii community in the Pyonsanbando located at southwestern area in Korea were examined and analyzed to provide some practical data to be used to establish measures for long term succession monitoring, protection and sustainable management. The Machilus thunbergii community are located at Kyokpo-ri, Pyonsan-myon, Puan-gun(35$^{\circ}$35′24"N~35$^{\circ}$ 42′30"N, 126$^{\circ}$28′18"E~126$^{\circ}$40′40"E). Within the Pyonsanbando area the mean temperature is 12.4$^{\circ}C$, the warmth index 101.5$^{\circ}C$.month, the coldness index -12.3$^{\circ}C$ month and the annual precipitation 1,016mm. The soil within the community is silt loam or clay loam and so fertile. 30 species and 1 varieties of 21 families inhabited at the community. There were 18 grown trees and 131 saplings of Machilus thunbergii within the community. The mean basal diameter, mean height, and crown width of grown trees was measured to be 57.4cm, 7.4m and 9.0~9.8m respectively. And 9 trees were full flowering, 2 trees small flowering here and there, and 7 trees non flowering in 1998. The mean branch growth length was 8.3cm, mean leaf area 18.3$\textrm{cm}^2$, mean inflorecence length 9.3cm, mean fertilization 14.5% and mean seed diameter 7.6mm. Further rich management measure and investigation were recommended such as sapling protection, signboard construction, soil erosion controlling and regular monitoring within the community.

Comparison of antioxidant activities in Korean black soybean landraces

  • Black soybean with the anthocyanin in the seed coat is known to have higher antioxidant activity than the yellow soybean. This study was carried out to evaluate antioxidant activity of 231 Korean black soybean landraces which conserved at RDA gene bank. Antioxidant activities were measured using DPPH, TPC, TFC, ABTS, and FRAP assay. DPPH showed wide variations, ranging from 16.4 to 200.4($IC_{50}$). TPC, TFC, ABTS, and FRAP were ranged from 0.8 to 13.2 mg gallic acid equivalent/g (mg GAE/g), 0.15 to 0.82 mg quercetin equivalent/g (mg QE/g), 2.0 to 8.3 mg ascorbic acid/g (mg ASC/g) and 0.2 to 3.1 mg ASC/g, respectively. Among 231 Korean black soybean landraces, IT177715 showed the highest antioxidant activity in DPPH assay. In TPC, TFC, ABTS, and FRAP assays, IT274975, IT274551, IT167725, and IT178047 showed the highest antioxidant activity, respectively. In Relative Antioxidant Capacity Index (RACI), IT178047 showed the highest antioxidant activity, while IT177197 the lowest. This study will be able to provide useful data to select black soybean landraces with high antioxidant activity.
