생쥐 정자세포의 분리와 체외 배양에 관한 연구 (In Vitro Culture Following Purfication of Mouse Spermatogenic Cells)

  • 본 연구는 생쥐의 고게정관내에 존재하는 분화단계의 정자세포를 발생단계별로 분리하여, 체외에서 단기간 배양체계를 확립하기 위하여 실시하였다. 8주령 이상된 생쥐의 정소로부터 정소막을 제거시킨 후, collagenase (1mg/ml), trypsin(2.5mg/ml)를 처리하여 곡세정관을 간질세포와 분리하여 배양액에 부유시켰다. 부유세포는 Celcep장치를 이용하여 세포크기와 밀도 차이에 의해 분화 단계별로 분리하였다. 회수된 세포의 균질성은 haematoxylin/eosin 또는 orcein으로 염색한 후, 광학현미경하에서 확인한 결과 약 85% pachytene spermatocyte와 round spermatid을 성공적으로 분리해냈다. 따라서 sedimentation velocity에 의해서 생쥐의 spermatogenic cell의 발달단계별 분리가 가능함을 알 수 있었다. 이러한 방법으로 분리된 pachytene spermatogenic cell들은 DMEM 배양액에서 6일 이상 배양한 결과 약 36%의 생존율을 보였다. 따라서, 분화단계별 정자 세포의 분리 및 배양체계의 확립은 웅성생식세포의 발생과정에 따른 생리 또는 분자생물학적 현상을 규명함은 물론 세포융합기술의 이용에 의한 형질전환동물 생산에의 응용을 통해 가축에 있어서의 형질전환 생산효율의 개선에 기여할 수 있으리라 사료된다.

Protein and hematological evaluations of infant formulated from cooking banana fruits(Musa spp, ABB genome) and fermented bambara groundnut(Vigna subterranean L. Verdc) seeds

  • Protein-energy malnutrition is regarded as one of the public health problems in developing countries as a result of poor feeding practices due to poverty. This study, therefore, aimed at evaluating nutritional quality of a potential weaning food formulated from locally available food materials. The cooking banana fruit(CB) and bambara groundnut seeds(BG) were purchased from local market in Akure, Ondo State, Nigeria. The CB and BG were processed into flours, mixed in ratios of 90:10, 80:20, 70:30 and 60:40 and subjected into proximate, sensory and biochemical analyses using standard procedures. Nutrend(a commercial formula) and ogi(corn gruel, a traditional weaning food) were used as control. The nutritient composition(g/100 g) of the food samples were ranged as follows: moisture 2.94-6.94, protein 7.02-16.0, ash 1.76-2.99, fat 0.76-8.45, fibre 1.52-3.75, carbohydrate 63.84-88.43 and energy 1569.8-1665.7 kcal. The biological value(BV), net protein retention(NPR), protein efficiency ratio(PER) and feed efficiency ratio(FER) of the experimental food samples were significantly(p<0.05) lower than nutrend, but higher than ogi. The haematological variables of rats fed with formulated food samples, commercial formula(nutrend) and traditional weaning food(ogi) were not significantly(p>0.05) influenced by the dietary treatment. However, the values obtained for red blood cell(RBC), white blood cell(WBC), pack cell volume(PCV) and erythrocyte sedimentation rate(ESR) were higher in the experimental food samples than the commercial food. The growth rate of animals fed with experimental food samples were lower than those fed with the nutrend, but higher than those fed with ogi. In conclusion, the nutritional quality of CB and fermented BG mix of 60:40 ratio was better than ogi; and comparable to the nutrend. This implies that it can be used to replace low quality traditional weaning food and the expensive commercial weaning formula.

  • 수중 부유입자의 효과적인 제거 방안을 모색하고자 $CaCO_3$로 구성된 부유물의 계면화학적 특성을 고찰하였다. pH에 따른 전기영동도 측정에서는 수중 $OH^-$의 작용으로 pH가 상승함에 따라 electrokinetic potential은 음의 방향으로 변환하는 것으로 나타났다. 계면활성제는 부유물의 안정성에 영향을 끼치는 바, 계면활성제의 농도 및 전하에 따라 영향을 미치는 양상이 다르게 나타났다. 무기응집제의 경우에 있어서도 그 전하가 및 농도에 따라 부유물의 계면화학적 거동이 달라졌으며 이는 DLVO 이론에 의거하여 작성된 potential energy의 변화추이와 관련하여 이론적으로 해석될 수 있었다. Non-specific adsorption은 전기이중층의 압축을 초래하여 electrokinetic potential의 절대값 감소를 야기시켰으며 specific adsorption은 부유입자의 IEP 및 PZC가 상호 반대방향으로 이동하는 결과를 나타내었다.

  • 본 연구에서는 격자볼츠만 방법을 기반으로 유체-입자 상호작용에 대한 수치계산을 수행하였다. 유체 유동은 격자볼츠만 방법을 이용하였으며, 유동장 내에서의 고체입자 운동은 계산점(node) 기반의 가상영역으로 간주하여 해석하였다. 유체-입자의 상호작용은 격자볼츠만 방법의 지배방정식에 국부적으로 운동량 교환량을 추가하여 해석하며, 가상영역 내에 위치한 고체입자의 병진 및 회전 운동은 뉴턴 운동 방정식과 오일러(Euler) 방정식을 이용한다. 구성된 상호작용 모델의 유효성을 검증하기 위하여 중립상태에서의 부유 입자운동 및 단 입자의 침강에 대한 수치계산을 수행하였으며, 기존 연구들과의 비교를 통하여 본 연구의 유체-입자 상호작용 모델이 갖는 신뢰성과 효용성을 평가하였다.

금강하구 및 인근해역에서 부유퇴적물의 계절적 변동에 관한 연구 (Seasonal Variations of suspended Matters in the Keum Estuary and its Adjacent coastal Area)

  • 한반도 서해중부에 위치하는 금강하구 및 인근 연안역에서 염분 및 부유물함량의 계절적 변화양상을 관측하였다. 금강하구에서 부유물함량은 계절적으로 크게 변하였으 며, 동계에 높고 하계에 낮게 나타났다. 특히 홍수기의 하계에는 담수의 유입이 증가 하여 금강하구에서 염분은 5$\textperthousand$ 미만이었고, 염분이 28$\textperthousand$ 미만의 기수가 인근 연안역으 로 확장하였다. 부유물함량은 금강하구에서 100 mg/l 이상으로 높았지만, 외해에서는 5mg/l 이하로 감소하였다. 이와 같이 홍수기인 하계에 부유물함량이 낮은 것은 금강을 통해 공급되는 육상기원 부유물질의 대부분이 강하구역에 집적되기 때문으로 해석된 다. 금강하구 및 인근해역에서 부유물함량 및 분포양상의 계절적 변동은 한반도 서해 연악역에서 현생 퇴적물의 주기적 퇴적작용으로 해석되었다. 즉, 하계동안 금강을 통 해 공급되는 세립물질은 강하구에 우세하게 집적되며, 동계에는 재부유되어 남쪽방향 으로 운반되는 반복작용이 주기적으로 나타나는 것으로 해석된다.

Vortex separator와 연속식 섬유사여과를 이용한 CSOs 처리연구 (A Study on Treatment of CSOs by Vortex Separator and Continuous Fiber-Filter System)

  • This study was conducted to confirm the CSOs characteristics, and to estimate treatment efficiency of CSO treatment process. Flowrate was average $53,500m^3$/d, maximum $58,100m^3$/d during dry season, but after rain-fall, the flowrate was increased more than twice that of the dry season. And, water pollution concentrations, such as $COD_{Cr}$, SS, $BOD_5$, TN and TP of after rain-fall, were also increased. Thus, for more efficient treatment of pollutants during rainy season, The vortex separator and continuous fiber filter devices were used. From the results on particle range, removal efficiency of particle was 99.7% at the particle size range of $40{\sim}100{\mu}m$ but decreased as 55-80% at the below $40{\mu}m$. The removal efficiencies of $COD_{Cr}$, SS, TN and TP were approx. 70, 60, 70 and 50, respectively during the dry season and approx. 50, 50, 8 and 18% during the rainy season. Also, when compared with the primary sediment basin, $COD_{Cr}$, SS, TN and TP removal efficiencies were high. especially, at the case of TN and TP, TN was more removed than TP because of higher conversion factor value. But we needed more study for the injection of a coagulants to get more stable treatment efficiency for soluble pollutants. Consequently, This process can be used for CSOs treatment as well as replace the primary sedimentation basin during the dry season.

소아의 하악에 발생한 Garre 골수염의 근관치료에 관한 증례보고 (GARRE'S OSTEOMYELITIS OF THE MANDIBLE RESOLVED BY ENDODONTIC TREATMENT IN CHILDREN: A CASE REPORT)

  • Garre's osteomyelitis is a unique form of osteomyelitis characterized rediographically by localized thickening of the periosteum and deposition of laminated subperiosteal bone. The most common inciting factor is a mandibular infection in permanent first molar with necrotic pulp. This disease occurs primarily in children and to date in all instances it has occured only in mandible. It usually results in hard swelling over the jaws, producing facial asymmetry with little or no pain. The overlying skin is normal but can occasionally be inflammed mostly when pain is present. Palpation reveals a usually smooth, bone-hard lesion which feel like an inherent part of the mandible. Unlike other forms of osteomyelitis, there is no marked increase in fever, white bloods cell count, sedimentation rate or alkaline phosphatase value. The treatment of Garre's osteomyelitis usually consist of elimination of the sourses of infection, i.e., either extration of an offending infected teeth or root canal therapy. This treatment almost always results in resolution of the Garre's osteomyelitis. Resistant cases have involved secondary surgery, i.e., decortication and sequestrectomy. This report presents three cases of Garre's osteomyelitis resolved by endodontic treatment. Cliniqtl examination revealed swelling on the face with no tenderness. Periapical radiograph showed deep caries lesion extending into pulp chamber and periapical radiolucency. Occlusal radiograph showed an enlargement of bone and stretching the periosteum. A clinical diagnosis of the Garre's osteomyelitis was made. Endodontic treatment was accomplished with conventional method and restored facial symmetry. Long-term check-ups are necessary to evaluate the results of endodontic treatment.

여수 해만을 통한 광양만의 부유퇴적물 수지균형 (Suspended Sediment Budget in Gwangyang Bay through the Yeosu Sound)

  • 여수해만 광양만 입구의 조류 및 부유 퇴적물 함량 분석을 통하여 광양만 내의 부유 퇴적물 수지 균형을 조사하였다. 만 입구의 부유 퇴적물 함량은 바닥이 평균 약 17.80mg/l로 표층의 평균 약 4.75mg/l에 비해 상당히 높으며, 조류도 표층 부근에서는 썰물이 강하나 저층에서는 오히려 밀물이 강하여 유속의 비대칭 현상을 보이고 있다. 이러한 저층의 높은 부유 퇴적물 함량과 밀물우세 조류로 인하여 이 지역의 부유 퇴적물 이동은 저층을 통한 이동이 큰 중요성을 갖으며, 따라서 광양만의 내부 특히 서쪽 부분은 전체적으로 세립 퇴적물의 순수 퇴적이 이루어지고 있다. 이러한 조류에 의한 세립 퇴적물의 순수 유입량은 약 $5.66\times10^8g/day$로 계산되며, 이 순수 유입량이 만의 서부에 퇴적된다면 그 퇴적율은 약 1.15m/1,000years 정도가 될 것이다.

Serum levels of interleukin-6 and interleukin-8 as diagnostic markers of acute pyelonephritis in children

  • Purpose: Early diagnosis and treatment of acute pyelonephritis in children is of special importance in order to prevent serious complications. This study was conducted to determine the diagnostic value of serum interleukin (IL)-6 and IL-8 in children with acute pyelonephritis. Methods: Eighty-seven patients between 1 month to 12 years old with urinary tract infection (UTI) were divided into 2 groups based on the result of 99m-technetium dimercapto-succinic acid renal scan: acute pyelonephritis (n=37) and lower UTI (n=50) groups. White blood cell (WBC) count, neutrophil (Neutl) count, erythrocyte sedimentation rate (ESR), C-reactive protein (CRP) concentration, platelet count, and serum IL-6 and IL-8 concentrations of both groups were measured and compared. Results: There was a significant difference between two groups regarding WBC count, Neutl count, ESR, and CRP concentration (P<0.05). In addition, the difference between the two groups regarding serum IL-6 and IL-8 concentrations was not significant (IL-6, 60 and 35.4 pg/mL and IL-8, 404 and 617 pg/mL, respectively). The sensitivity and specificity of serum IL-6 and IL-8 for diagnosis of acute pyelonephritis were 73%, 42% and 78%, 32%, respectively. Sensitivity, specificity, negative and positive predictive values of serum IL-6 and IL-8 were less than those of acute phase serum reactants such as CRP. Conclusion: This study showed that there was no significant difference between acute pyelonephritis and lower UTI groups regarding serum IL-6 and IL-8 levels. Therefore, despite confirming results of previous studies, it seems that IL-6 and IL-8 are not suitable markers for differentiating between acute pyelonephritis and lower UTI.

Characterization and Application of a Novel Thermostable Glucoamylase Cloned from a Hyperthermophilic Archaeon Sulfolobus tokodaii

  • A gene for a putative glucoamylase, stg, of a hyperthermophilic archae on Sulfolobus tokodaii was cloned and expressed in Escherichia coli. The recombinant glucoamylase (STGA) had an optimal temperature of $80^{\circ}C$ and was extremely thermostable with a D-value of 17 hr. The pH optimum of the enzyme was 4.5. Being different from fungal glucoamylases, STGA hydrolyzed maltotriose (G3) most efficiently. Gel permeation chromatography and sedimentation equilibrium analytical ultracentrifugation analysis showed that the enzyme existed as a dimer. STGA was stable enough to hydrolyze liquefied com starch to glucose in 4 hr at $90^{\circ}C$ with a yield of95%. Comparison of the $k_{cat}$ values for the hydrolysis and the reverse reaction at $75^{\circ}C$ and $90^{\circ}C$ indicated that glucose production by STGA was more efficient at $90^{\circ}C$ than $75^{\circ}C$. Therefore, STGA showed great potential for application to the industrial glucose production process due to its high thermostability.
