Seasonal Fluctuations of Marine Environment and Phytoplankton Community in the Southern Part of Yeosu, Southern Sea of Korea (여수 남부 협수로 해역의 해양환경과 식물플랑크톤 군집의 계절변동 특성)

  • Field observations on the seasonal variations of environmental factors and phytoplankton community were carried out four times at 30 stations in the narrow strait between Yeosu and Dolsan Island of the Korean South Sea from September 2005 to May 2006. The ranges of water temperature, salinity and extinction coefficient in the surface waters were 5.6~26.3, 25.36~33.92 psu and 0.13~2.13, respectively. The water temperature measured higher at Gamak Bay in summer and spring. It measured higher at Yeosuhae Bay in autumn and winter. Salinity showed uniformity of distributions in almost all areas, except for an area near a sewage disposal outlet. Extinction coefficient indicated that the turbidity of Gamak Bay and the area near the sewage disposal outlet were higher than that of the Yeosuhae Bay. In the phytoplankton community were identified a total of 99 species belonging to 51 genera. The species composition showed itself to be various in summer and autumn, but poor in winter and spring with a high ratio of centric diatoms all the year round. Seasonal succession of dominant species were Skeletonema costatum and Chaetoceros curvisetus in summer, Eucampia zodiacus in autumn and winter, and Chaetoceros affinis and Thalassionema nitzschioides inspring. Standing crops of phytoplankton and Chlorophyll $\alpha$ concentration were greatly higher at Gamak Bay in summer with ranges of $0.2{\times}10^4\;cells\;L^{-1}$ to $296{\times}10^4\;L^{-1}$, and $1.94\;L^{-1}$ to $22.12\;L^{-1}$, respectively. From the results of principal component analysis (PCA), the northern part of Dolsan Island was divided into two or three regions from the characteristics of marine environment and phytoplankton community.

Effect of Night-break Timing on Growth, Bolting and Anthesis of Orostachys japonicus (암기중단 처리시기에 따른 바위솔의 생장, 추대 및 개화)

  • Orostachys japonicus, Wasong as herb medicine, has been artificially cultivated as an anti-tumor medicinal. The experiment was carried out to examine the effect of natural daylength as control or night-break treatment (NB) imposed at June 20, July 18 or Aug. 15 on its growth, dry weights of leaf and bract, stem, floret and root, and morphological characters including bolting and floret flowering. After a plant was grown in a 15cm plastic pot containing a 2 : 1 soil : peat moss mixture on May 23, three treatments with above differing night-break had been imposed around midnight up to Nov. 7. The plants were sampled 3 times at the same day forced to night-break and then done 6 times by 2-week interval after the final NB. Plant height and inflorescence length of all the NB increased with delayed NB but declined in comparison with the natural daylength. No. of leaves including bracts showed similar response to plant height although NB given before July 18 showed less leaves and bracts. Stem diameters of NB were continuously increased to middle Sept. to middle Oct. while that of natural daylength decreased after middle Oct. Natural daylength or NB given on Aug. 15 had greater fraction, shoot and total dry weights resulting from increment of leaf and bract up to Aug. or of floret, stem and root after Sept. The earlier NB, the later formation of florets and the less number of flowering florets whereas in natural daylength florets on inflorescence begun to be formed from middle Sept. were sharply increased up to middle Oct. when all the plants were flowered. Bolting was not formed in the plant of the earliest NB of June 20, and thereby no anthesis of florets up to early Nov. It was concluded that year-round cultivation of Orostachys japonicus plants was possible through controlling the NB timing because its bolting and flowering of florets separately occurred.

Effect of $CO_2$ Concentration, NAEH and Light Intensity on the Photoautotrophic Growth of Campanula punctata 'Rubriflora' Plantlets In Vitro (자주초롱꽃의 기내 자가영양배양시 $CO_2$농도, 환기횟수 및 광도가 생장에 미치는 영향)

  • Growth of Campanula punctata 'Rubriflora' plantlets, as affected by three levels of photosynthetic photon flux (PPF), 70, 110, and $220{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, two levels of $CO_2$ concentration, 500 and $1,500{\mu}mol{\cdot}m^{-1}$, and two levels of number of air exchanges per hour (NAEH), 0.1 and $2.8 h^{-l}$, was studied. Explants were obtained from photomixotrophically-micropropagated plantlets. Four explants were planted in each $3.7{\times}10^{-4}m^3$ polycarbonate box containing MS basal medium and no added sucrose. Explants were cultured under cool-white fluorescent lamps for $16h{\cdot}d^{-1},\;at\;25\pm1^{\circ}C$ temperature, and $70\~80\%$ relative humidity In treatments of $2.8h^{-1}$ NAEH, a 10mm round hole made on the vessel cap was sealed with a microporous filter. For higher $CO_2$ concentrations in the culture room, $CO_2$ gas was provided from a tank of liquefied $CO_2$. Fresh and dry weights, length of the longest root, and number of leaves significantly increased with increasing PPF and especially $CO_2$ concentration. Length of the longest root, number of leaves, fresh and dry weights, and chlorophyll concentration were enhanced with increased NAEH. However, leaf area was the smallest in the $220{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}\;PPF\;2.8h^{-1}$ NAEH and especially, $1,500{\mu}mol{\cdot}mol^{-1}\;CO_2$ concentration treatment. Treatment effect became more produced with time. Overall, treatment with $220{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}\;PPF\;and\;1,500{\mu}mol{\cdot}mol^{-1}\;CO_2$ gave the most vigorous growth.

Comparative Study on the Nucleus accumbens septi and the Nucleus fundus striati I. Classification of Neuronal Cell Types (중격측좌핵과 선조체 기저핵의 비교연구 I. 세포형의 구분)

  • There's been arguments on the different morphological status between the nucleus accumbens septi and nucleus fundus striati of ventral striatum. Authors carried out the comparative study on the neuronal cell types of these nuclei, in the chick and the rat. Results are summarized as follows: In the nucleus accumbens septi of the chick, there found 3 main cell types. Type I cells are oval or spindle-shaped. They are the most abundant cell types, comprising more than 80% of neurons. The pale nucleus is usually indented. The cytoplasm is also pale and contains small amount of mitochondria, rough r-ER and Golgi complexes. This cell has a few symmetric synapses on the cell membrane. Type II cells are pale large cells. They are polygonal or irregularly-shaped. They contain pale spherical nucleus, and the pale cytoplasm with relatively large amount of mitochondria, free ribosomes and well-developed Golgi complexes. Some axo-somatic synapes are found on the cell. Type III cells are oval or spherical-shaped. The nucleus is relatively pale and large, In the dense cytoplasm, well developed. r-ER formed typical Nissl's body, and there found many mitochondria, ribosomes and lysosomes. In the chick fundus striati nucleus, there also found 3 main cell types. Type I cells are small and spindle-shaped. This type is the most abundant one and constitutes more the 80% of the neurons. Morphological features other than it's shape, is generally similar with that of Type I cell in the nucleus accumbens. Type II cells are irregularly shaped large cells. Dense cytoplasm contains large amount of cell organelles. Some axo-somatic synapses are found. Type III cells are small dense cells. This oval cell contains the oval nucleus, and the plentiful cytoplasm with well developed r-ER, ribosomes and mitochondria. In the nucleus accumbens septi of the rat, there found 4 main cell types. Type I cells are small, oval or spherical cells, comprising more than 90% of all the neurons. Spherical nucleus shows typical chromatin rim along the nuclear membrane. Dense cytoplasm contains many ribosomes and mitochondria. Type II cells are large oval cells. The eccentric nucleus is deeply invaginated. Pale cytoplasm contains large amount of ribosomes, Golgi complexes, mitochondria, and dense bodies. Type III cells are pale, large, oval cells. They contain moderate amount of ribosomes and mitochondria, and some scattered stacks of r-ER. Type IV cells are small pale cells. Small oval nucleus is indented and shows chromatin rim. Only small amount of ribosomes and mitochondria can be found. In the nucleus fundus striati of the rat, there also found 4 main cell types. Type I cells are spherical or oval cells, comprising more than 90% of the neurons. The chromatin rim of the spherical nucleus is not so prominent as compared to the rim of type I cell in the nucleus accumbens septi. The cytoplasm contains moderate amount of mitochondria, ribosomes and some scattered r-ER. A few axo-somatic synapses were found. Type II cells are small round or polygonal cells. Golgi complexes are especially well-developed in this cell type. The cytoplasm also contains moderate amount of mitochondria, ribosomes, and dense bodies. Type III cells are small cells. The large nucleus shows prominent chromatin rim. The cytoplasm contains many ribosomes and mitochondria. Type IV cells are large, spheircal or oval cells. The nucleus is deeply indented. The plentiful cytoplasm contains large amount of ribosomes, mitochondria, Golgi complexes, neurotubules, but not r-ER. In the present study, it is clear that the nucleus accumbens septi and the nucleus fundus striati are independant cell groups, according to their cytoarchitectonics and the ultrastructural features of their cell types.

Immunohistochemical and Ultrastructural Characterization of the Choline Acetyltransferase-immunoreactive Nerve Cells in the Diagonal Band of Broca of the Rat Basal Forebrains (흰쥐의 전뇌 기저부 대각 Broca대에서 Choline Acetyltransferase 면역반응 신경세포에 대한 면역조직화학 및 미세구조)

  • This study was performed to investigate the immunohistochemical and ultrastructural characterization of the choline acetyltransferase (ChAT)-immunoreactive nerve cells in the diagonal band of Broca of the rat basal forebrains, utilizing techniques of immunohistochemical and immunocytochemical microscopy. The ChAT-immunoreactivities were shown within neuronal cell bodies and processes by the light micoscope. According to cell shape and ratio of long axis vs short axis of cell body, the ChAT-immunoreaclive nerve cells in both vertical and horizontal limbs of the diagonal band of Broca were classified into 6 types. at the light microscopic level; round, oval, elongated, fusiform, triangular and polygonal types. As a result of the electron microscopic observation, the ChAT-immunoreactivated products appeared on the outer nuclear envelope, membranes of rough endoplasmic reticula (rER), free ribosomes and polysomes. Each cell type was subdivided into subtype I and II according to the several criteria such as volume of cell body, nuclear size relative to the cytoplasm, kinds and distribution of cell organelles and numbers and sorts of synapses. The subtype I of immnunoreactive nerve cells had large cell body and a small nucleus showing shallow indentations of nuclear evelope. In this subtype I with abundant cytoplasm, rER were well differentiated. Their long cisternae were parallelly ditributed and lamellated. One or two lamellar bodies and nematosomes were observed. The subtype II cell had small cell body and a large nucleus with deep indentations of nuclear envelope. In this subtype II with small cytoplasm, the rER were irregularly distributed and the lamellar body and nematosome were not found. A few axosomatic synapses in the subtype I and II were shown to be symmetric or asymmetric. The ratios of the symmetric synapse to the asymmetric one were investigated to be 1 : 2 and 1 : 4 in the subtype I and II, respectively. The axodendritic ones were almost asymmetric. But, the fusiform and triangular immunoreactive nerve cells were shown only to be subtype I. According to observations in this study, it is considered that the ultrastructural characterization in the 2 subtypes of each cell type may reflect the differences of the metabolic activities and projecting distances to the target cells.

Analysis of Environment Factors in Pleurotus eryngii Cultivation House of Permanent Frame Type Structure (영구형 큰느타리버섯 재배사의 환경요인 분석)

  • Pleurotus eryngii is one of the most promising mushrooms produced on the domestic farms. The quality as well as quantity of Eryngii is sensitively affected by micro climate factors such as temperature, relative humidity, $CO_2$ concentration, and light intensity. To safely produce high-quality Eryngii all the yew round, it is required that the environmental factors be carefully controlled by well designed structures equipped with various facilities and control systems. At the commercial mushroom cultivation houses of permanent frame type (A, B), this study was carried out to find out reasonable range of each environmental factor and yield together with economic and safe structures influencing on the optimal productivity of Eryngii. This experiment was conducted for about two-year ken Nov. 2003 to Dec. 2005 in cultivation house. Ambient temperature during the experiment period was not predominantly different from that of a normal year. The capacity of the hot water boiler and the piping systems were not enough. Because the capacity of electric heater and air circulation were not enough, air temperatures in cultivation house before improvement of system were maintained somewhat lower than setting temperature, and maximum air temperature difference between the upper and lower growth stage during a heating time period was about 5.1. But the air temperatures after system improvement were maintained within the limits range of setting temperature without happening stagnant of air. Air temperature distribution was generally distributed uniform. Relative humidity in cultivation house before , improvement was widely ranged about $44{\sim}100%$. But as the relative humidity after improvement was ranged approximately $80{\sim}100%$, it was maintained within the range of relative humidity recommended. And $CO_2$ concentration was maintained about $400{\sim}3,300mg{\cdot}L^{-1}$ range. The illuminance in cultivation house was widely distributed in accordance with position, and it was maintained lower than the recommended illuminance range $100{\sim}200lx$. The acidity of midium was some lower range than the recommend acidity range of pH $5.5{\sim}6.5$. The yield was relatively ununiform. In case of bottle capacity of 1,300cc, the mushroom of the lowest grade was less than 3%. The consumption electric energy was quite different according to the cultivation season. The electric energy consumed during heating season was much more than that of cooling season.

Hedgehog Oral Viral Papilloma (고슴도치 구강유두종)

  • Here we describe the histological lesion of a viral fibro-papilloma in a hedgehog. After surgical removal from maxilla, the solitary swollen mass was round to oval, yellowish with rough surface and measuring $6{\times}3{\times}2mm$ approximately. The tumor mass was submitted to the laboratory of pathology, college of veterinary medicine, Kyungpook National University for pathological diagnosis. Pathological examination of the tumor was established, the tumor was described grossly and sample was trimmed, sectioned and routinely prepared for histopathological evaluation. The tumor mass was diagnosed as viral fibro-papilloma, as the histological picture showed characteristic features of warts caused by papillomaviruses. The tumor characterized by thickening of the stratum spinosum (acanthosis) and basale, koilocytosis, intra-nuclear inclusion bodies in keratinocytes and fibrosis of submucosa. Further, viral inclusion bodies were demonstrated by Machiavello stain giving red color to the nuclei. No lymphocytes that responsible for regression of the wart could be detected, suggesting the poor possibility of spontaneous regression of the tumor. Papillomatosis is a disease of young animals, but in our case the infected Hedgehog was 5 years old, that maybe due to an impaired immune system, which is also shown by absence of lymphocytes. To the best knowledge of the author, this case presents the first report of viral fibropapillomatosis in Hedgehog.

A STUDY ON THE CHILD PATIENT'S PREFERENCE TOWARD DENTIST'S ATTIRE (소아환자의 치과의사 복장에 대한 선호도 연구)

  • The purpose of this study was to find a method for improving the children's behavior during dental treatment in relation with dentist's attire. For this study 150 children, visiting a dental clinic, were asked to participate in a survey on the attire of the dentist. The results of the survey were as follows: 1. In the survey for the preference between gown and ordinary clothes according to sex(P<0.05), age(P<0.05), residence (P<0.05) and number of visits(P<0.05), there was more preference for gown(65.3%). 2. The preference for the type of gown and attire under the gown was a long gown and white shirt (30.7%), followed by a suit-like gown (18.7%) and white shirt, and long gown and polo shirt(17.3%). 3. The preference for the color of gown was white(34.7%), followed by pink (18.7%), green(18.0%), blue(15.3%) and yellow(13.3%). 4. The preference for the pattern on the gown was cartoon characters(49.3%), followed by hospital logo(28.7%) and no pattern(14.7%). 5. The preference for ordinary clothes of female dentists was polo shirt(37.3%), followed by striped shirt(28.7%), round T-shirt(18.7%), and everyday Korean traditional dress(15.3%). 6. The preference for ordinary clothes of male dentists was pattern necktie and no pattern Y-shirt(28.0%), followed by no pattern necktie and Y-shirt(21.3%) or pattern shirt with no necktie, and no pattern Y-shirt with no necktie(14.7%). 7. The preference on the protective equipment worn was mask and glove(28.7%), followed by no protective equipment(26.7%), mask, glove and protective glasses(22.7%) at)d only mask(22.0%). Based on the above results, it seems that parting with the traditional white gown and wearing multi-color and pattern attire will provide psychological stability and help improve children's behavior during treatment.

The Change of the Relationship between Korean 4th Graders' Career Aspirations in Science and Science Identities (초등 4학년 학생들의 과학 관련 진로 포부와 과학 정체성 관계의 변화)

  • This study sought to explore longitudinal changes in Korean fourth graders' career aspirations in science. The sample included those who had high interest, but low career aspirations in science, and the analysis focused on the concept of science identity. To achieve this study's objectives, we selected 14 participants and then conducted two in-depth interviews with them. Compared with the first interviews (4th grade), changes were observed in eight students' career aspirations in science in the second round (5th grade). While six students maintained "negative" career aspiration, six revealed "medium" and two revealed "positive" career aspiration in science. To explore the relationship between science identities and career aspirations in science, three students were selected from the sample for further study. According to the results, the aspects of science identity that students valued regarding potential careers varied individually. Depending on these aspects, each student showed different patterns in the interactions between different aspects of science identity, and between aspects of science identity and science-related career aspirations. Over time, participants' experiences and interactions with people around them produced changes in their science identity, which led to changes in their career aspirations resulting from the interactions between aspects of their science identity. Their career aspirations in science were also significantly influenced by each participant's perceptions of relationships with peers (i.e., recognition by peers and of peers) and of science-related careers. Therefore, the results suggest that students need to be provided with appropriate learning environments and guidance relating to science careers.

Morphological Study on the Dorsal Lingual Papillae of Myotis macrodactylus (큰발윗수염박쥐(Myotis macrodactylus)의 혀 유두의 형태학적 연구)

  • The dorsal lingual papillae of Myotis macrodactylus were investigated morphologically using scanning electron microscopy. Three types of lingual papillae from Myotis macrodactylus were recognized. The filiform(Fi) papillae were distributed over the entire dorsal surface of the tongue, and they could be classified into 5 types of papillae by the shape, size and number of the protrusion. Type I was distributed on dorsal surface of the apex in the anterior region of the tongue, has a crow-like shape with ten to fifteen formed projections. Type II was located in the medial portion of the anterior region of the tongue, has an eight to ten spin-like protrusion. Type III was distributed on the medial portion of the tongue, has a scale-like papille with five to seven protrusion. Type IV was distributed on the both lateral portion of the posterior region of the tongue, has a small conical papillae, does not have needle projection. Type V have not needle projection, a large of conical papillae, on the both lateral portion of the posterior region of the tongue. Most Fu(Fungiform) papillae were distributed the both lateral region or medial portion of the posterior region of the tongue, has a round or oval shape with $40{\sim}45$ papillae. The small $(65{\mu}m)$, meddle $(75{\mu}m)$ and large $(120{\mu}m)$ fungiform papillae were distributed on dorsal surface of the apex in the anterior, the both lateral portions and medial portion of the posterior region of the tongue, respectively. Two large oval vallate (V) papillae are located in the medial portion of the posterior region of the tongue. In conclusion, the morphology of lingual papillae is useful to determine species relationship among Myotis species. It suggests that the difference of types and number of lingual papillae caused by the difference of food habit.
