Separation of Glabridin from Licorice by RP-HPLC (RP-HPLC를 이용한 감초에서 Glabridin의 분리)

  • By reversed-phase high-performance liquid chromatography, the extraction and separation of glabridin by from licoricce root was performed in this work. The column efficiencies and resolutions of glabridin were investigated with mobile phase composition on the reversed-phase chromatographic system. The glabridin collected from licorice root was identified by LC/MS. The mobile phase used to extract glabridin were composed of ethanol, methanol, acetone, and ethyl acetate. For one-hour ultrasonic extraction with solvent of ethyl acetate, the favorable content of glabridin was obtained as 1.26g/kg. The glabridin was well separated in the mobile phase composition of 50/50 vol. % (acetonitrile/water).

  • The retention mechanism and separation of various aromatic sulfonic and carboxylic acids on reversed-phase liquid chromatographic column were studied in the mobile phase containing dodecyltrimethylammonium bromide (DTAB). The retention mechanism was found to be followed the ion-interaction model where the DTAB occupies a primary layer at the stationary phase while the sample anions and other co-anions in the system compete for forming the secondary layer. The capacity factors of samples were influenced by the several factors such as pH, concentration of various organic solvents, co-anions in the mobile phase and functional groups in sample molecules. Some mixtures of organic samples were attempted to separate under optimum condition.

Effect of Concentration of Ionic Liquids on Resolution of Nucleotides in Reversed-phase Liquid Chromatography

  • The chromatographic behaviors of nucleotides (inosine 5'-monophosphate, uridine 5'-monophosphate, guanosine 5'-monophosphate, and thymine monophosphate disodium salts) on a C18 column were studied with different types of ionic liquids (ILs) as additives for the mobile phase in reversed-phase liquid chromatography (RPLC). Three ILs, 1-butyl-3-methylimidazolium tetrafluoroborate ([BMIm][BF4]), 1-ethyl-3-methylimidazolium tetrafluoroborate ([EMIm][BF4]), and 1-ethyl-3-methylimidazolium methylsulfate ([EMIm][MS]), were used. Eluents were composed of water and methanol (90/10%, vol) with the addition of 0.5-13.0 mM of ILs. The effects of the concentration of ILs on retention and separation were investigated and discussed. The results showed that the addition of ILs affects the retention and resolution of the tested compounds. Use of 13.0 mM of [BMIm][BF4] as the eluent modifier resulted in a baseline separation of nucleotides without requiring gradient elution. This study demonstrates that ILs can be potentially applied as a mobile phase modifier in RPLC.

  • This research was performed to investigate the effect of ageing on grain refinement of 82wt%Cu-13.5wt%Al-4.5wt%Ni-0.1wt% misch metal alloy with ageing temperature and time. The results obtained in this study are as follows; The variation of transformation temperature with ageing temperature had very influence on ageing time at $300^{\circ}C$. But it had not influence on ageing time at $100^{\circ}C$. The variation of transformation temperature after second reversed transformation cycle was not occured in martensitic phase ageing, but $M_s$ point was appeared equally with third reversed transformation cycle in parent phase ageing. The variation of $M_s$ point was not nearly with ageing time at $100^{\circ}C$ ageing temperature. But it was decreasing with ageing time at $300^{\circ}C$ ageing temperature and $A_s$ point was increasing with ageing time. Structure of parent phase ageing was being M18R martensitic and N2H martensitic phase. It was found that ${\alpha}$ and ${\gamma}_2$ phase were created by ageing of long time at $300^{\circ}C$ ageing temperature.

Optical Resolution of Dabsyl Amino Acids in Reversed-Phase Liquid Chromatography

  • The dabsylation of amino acids has been applied to resolve their optical isomers with the use of chiral mobile phase in high performance liquid chromatography. The dabsyl amino acids were successfully separated on reversed phase column($C_{18}$) by adding a chiral L-benzylproline-Cu(II) chelate to the mobile phase. The separation selectivity of the dabsyl amino acid enantiomers was not less than that of dansyl amino acids. The retention order of the dabsyl amino acid enantiomers was as those of the dansyl amino acid enantiomers except dabsyl threonine. The optical selectivity of the dabsyl amino acids increase with pH of the mobile phase and concentration of the chelate, but slightly decreases with concentration of buffer and organic solvent composition. However serine, methionine, valine, and leucine showed a slight decrease in the optical selectivity with increase in pH. The retention times of the dabsyl amino acids decreases with increasing pH and acetonitrile concentration but increases with the concentration of the chiral chelate added. The mechanism of the optical resolution is based on a stereospecific interaction including a intramolecular hydrophobic effect and SN-2 reactivity of the ligand exchange chromatography.It is advantageous to detect absorption at 436 nm, which is less interferent them the other detection systems. The derivatized dabsyl amino acids are stable for a month.

The Preparation of Reversed Micelle Containing Water Soluble Collagen Solution and Their Application on Lip Make up Products (콜라겐 수용액을 함유하는 역미셀의 제조 및 립 제품에 응용)

  • Water-soluble collagen (30 wt%) was entrapped within oil phase of lipstick using reversed micelle method to improve the moisture capacity of the lipstick. Reversed micelles containing collagen were prepared using caprylic/capric triglyceride as external phase and polyoxyethylene (10) octylphenyl ether (Triton X-100) and 1-dodecanol as surfactant and co-surfactant, respectively. The formation of reversed micelle encapsulating collagen was confirmed by measuring electric conductivity and UV-vis spectrum using methylene blue (MB). The stability and moisture capacity of the lipstick containing 20 wt% collagen encapsulated reversed micelles were observed by measuring rheology property, moisture content and amino acid content. The molecular ratio (W, water-pool) of water to surfactant (Triton X-100) in the most stable reversed micelle was ${\leq}$ 10. The hardness of the lipstick had no difference with that of the lipstick without reversed micelle, and the moisture content was increased to 59% and the amino acid content was 92.7%.

Micro-Determination of D-Amino Acids in Milk by using Column Switching System (Column-Switching System을 이용한 우유속의 D-아미노산의 미량정량)

  • Free amino acids were isolated from milk and their absolute amounts were determined by reversed phase high performance liquid chromatography after derivatization with dansyl chloride. The determination of D- and L-amino acids was based on achiral separation on a C18 column. It was found that milk contained totally 41.00 mg DL-amino acids in 100 mL milk. The level of D-amino acids to L-amino acids was determined by a column-switching system combining an achiral reversed phase separation and chiral chelate additive. The chiral separation was carried out with addition of the chiral Cu(N-benzyl-L-proline)2 chelate to the mobile phase in reversed phase liquid chromatography. It was found that the determination of 16 different amino acids is feasible in the milk sample with a C18 column separation and 12 D-amino acids out of the 16 amino acids can be determined via the column-switching system with chiral separation. 2.05% of D-glutamic acid and 2.93% of D-alanine were found in milk.

  • Effects of austenite on the pitting corrosion in 202 stainless steel with two phase of austenite and martensite were investigated through the electrochemical polarization test. Two phases structures of martensite and austenite were obtained by reversed annealing treatment at the range of $500^{\circ}C-700^{\circ}C$ for 10min. in 70% cold-rolled 202 stainless steel. Volume fraction of reversed austenite has increased rapidly with an increase of annealing temperature. Pitting corrosion has arisen mainly on martensite phase in 202 stainless steel with two phases of austenite and martensite. Pitting current density has decreased with an increase of volume fraction of austenite. Consequently, pitting corrosion at martensite has occurred largely with an increase of volume fraction of austenite. Pitting corrosion was affected by volume fraction of austenite.

  • 1-(2-Pyridylazo)-2-Naphthol (PAN) has been widely used as a spectrophotometric reagent and metallochromic indicator for many metal ions. In this work, the chelate reagent of PAN was used as mobile phase additive for the separation of metal ions by reversed phase chromatography. Metal ions could be detected by monitoring the effluent at 570 nm with spectrophotometric detector. In order to investigate retention behaviors of the metal ions, the chromatograms and capacity factors were obtained as the variation of pH, ionic strength and composition of organic modifier in mobile phase. Under the obtained optimum conditions, the mixtures of Fe(III), Ni(II), Cu(II), Zn(II) and Co(II) could be separated successfully and the calibration curves under the recommended conditions showed an excellent linearity. The detection limits(S/N) were feasible at the nanogram level.

  • Dodecyltrimethylammonium bromide (DTAB) was examined as a counter-ion in the investigation of capacity factor and separation of aromatic carboxylic acids on alkyl-modified silica (ODS) column as a stationary phase by ion-pair chromatography on reversed-phase system. The capacity factor of samples was influenced by the several factors such as a concentration of counter-ion as well as kinds and concentration of electrolyte, concentration of methanol in mobile phase and kinds and position of functional group in sample molecule. Some mixtures of samples were able to be separated under optimum condition.
