Screening of Inducible Resistance Genes to Macrolide-Lincosamide-Streptogramin B(MLS) Antibiotics (마크로라이드-린코사마이드-스트렙토그라민 B(MLS)계 항생물질에 대한 유도 내성)

  • Forty nine clinical isolates of S. aureus showing resistance to erythromycin(EM) were selected from 83 strains isolated recently in Korea. Fourteen strains of S. aureus showing inducible resistance to MLS antibiotics were selected by disc agar diffusion method. Colony hydridization was executed using two MLS inducible resistance genes, ermA and ermC, identified previously from S. aureus as probes. S. aureus 375 and S. aureus 507 whose genes were not homologous to those probes were finally selected. It was confirmed that the resistance genes of S. aureus 375 and S. aureus 507 had no homology with those probes in southern hybridization test using ermA, ermC and ermAM as probes. It was determined that S. aureus 375 had a plasmid whose size was about 35 kb. To know if the plasmid may have the genes related to inducible resistance to MLS antibiotics, it was attempted to transform Bacillus subtillis BR151 and S. aureus RN4220 with the plasmid isolated from S. aureus 375. It was shown that the gene related to inducible resistance to MLS antibiotics did not exist in this plasmid. These results indicate that two clinical isolates of S. aureus showing inducible resistance to MLS antibiotics have novel genes that have no homology with MLS resistance genes identified so far. It is assumed that these genes may exist in chromosomal DNA.

Selection of Environmental Friendly Organic Agricultural Materials for Controlling Ginseng Gray Mold (인삼 잿빛곰팡이병의 친환경방제를 위한 유기농업자재 선발)

  • Background : To control ginseng gray mold, farmers have mainly used inorganic chemical based fungicides. The recent emergence of fungicide resistance has reduced the effectiveness of such control methods. Such pesticides also carry additional problems, such as diffuse pollution. Methods and Results : Six treatments of organic agricultural materials were tested for control of ginseng gray mold, CAPW (Chrysophanic acid + Phytoncide + Wood vinegar), EmEWV (Emodin + Ethanol + Wood vinegar), CEWV (Curcumin + Eugenol + Wood vinegar), Bacillus subtilis, soybean oil and sulfur. The control effect for gray mold by a single application of the agrochemical fungicide industrial Fenhexamid wettable powder (WP) was 84.4%. The control effect by CAPW, EmEWV and CEWV varied between 52.7 - 64.9%. The control effect by B. subtilis, soybean oil, and sulfur were 32.9 - 59.2%. Conclusions : In the field tests, CAPW showed the highest control effects when used before, and at first stage of disease incidence, against ginseng gray mold.

Pre-immigration Screening for Tuberculosis in South Korea: A Comparison of Smear- and Culture-Based Protocols

  • Background: Tuberculosis (TB) is the most important disease screened for upon patient history review during preimmigration medical examinations as performed in South Korea in prospective immigrants to certain Western countries. In 2007, the U.S. Centers for Disease Control and Prevention (CDC) changed the TB screening protocol from a smear-based test to the complete Culture and Directly Observed Therapy Tuberculosis Technical Instructions (CDOT TB TI) for reducing the incidence of TB in foreign-born immigrants. Methods: This study evaluated the effect of the revised (as compared with the old) protocol in South Korea. Results: Of the 40,558 visa applicants, 365 exhibited chest radiographic results suggestive of active or inactive TB, and 351 underwent sputum tests (acid-fast bacilli smear and Mycobacterium tuberculosis culture). To this end, using the CDOT TB TI, 36 subjects (88.8 per $10^5$ of the population) were found to have TB, compared with only seven using the older U.S. CDC technical instruction (TI) (p<0.001). In addition, there were six drug-resistant cases which were identified (16.7 per $10^5$ of the population), two of whom had multidrug-resistance (5.6 per $10^5$ of the population). Conclusion: The culture-based 2007 TI identified a great deal of TB cases current to the individuals tested, as compared to older U.S. CDC TI.

Development of an Efficient Screening System for Resistance of Tomato Cultivars to Ralstonia solanacearum (토마토 풋마름병에 대한 효율적인 저항성 검정 방법 개발)

  • This study was conducted to establish an efficient screening system for resistant tomato to bacterial wilt caused by Ralstonia solanacearum. Under several conditions such as inoculation methods, growth stages of tomato seedlings, inoculum concentrations, and incubating temperatures after inoculation, development of bacterial wilt on nine resistant or susceptible cultivars of tomato was investigated. To inoculate by drenching the non-cut roots with the bacterial suspension was better to distinguish resistance and susceptibility of tomato cultivars than by drenching the cut roots using scalpel. And 'Hawaii7996' a resistant tomato to R. solanacearum showed high resistance at all the tested conditions including growth stages (3-, 6-, 8-, 10-leaf stages), inoculum concentrations ($OD_{600}=0.1-0.4$) and incubation temperatures (25, 30, $35^{\circ}C$). On the other hands, susceptible cultivars represented disease index of 3.7 and 3.9 at 6- and 8-leaf stages, respectively. At 3- and 10-leaf stages, the cultivars demonstrated lower disease severity of 2.1 and 0.5, respectively, than at 6- and 8-leaf stages. When the inoculated seedlings were incubated in growth chambers of 25, 30 and $35^{\circ}C$, disease severity of susceptible cultivars was significantly greater at 30 and $35^{\circ}C$ than at $25^{\circ}C$. In addition, the level of resistance of the tomato cultivars was not significantly affected by inoculum concentrations of $OD_{600}=0.1-0.4$. On the basis of the results, we suggest an efficient screening method to measure resistance level of tomato cultivars to bacterial wilt. The eight-leaf stage seedlings transplanted 7 days before inoculation, are inoculated with R. solanacearum by drenching the non-cut roots with a bacterial suspensions ($OD_{600}=0.4$) to give inoculum volume of 50 ml/soil l. The inoculated plants are incubated in a growth room at $30^{\circ}C$ for 12-13 days with 12-hour light a day.

Developing screening system for resistance to anthracnose in grapes by using culture filtrates from Elsinoe ampelina

  • It was investigated whether culture filtrates produced by X. fastiduosa could be used to determine varietal susceptibility in grape cultivars to anthracnose as a substitute for pathogen inoculation or field screening. Bioassay of grape leaves with culture filtrates showed that their phytotoxicities were active and host-selective. Ethyl acetate extracts from those also showed the toxicities and host selectivity among grape cultivars. The sensitive range of plants to culture filtrates and their ethyl acetate extracts was consistent with the host range to the pathogen. Susceptible cultivars were sensitive to even highly diluted culture filtrates but resistant cultivars were not affected even at original culture filtrates. Susceptible cultivars were sensitive to the undiluted culture filtrates than highly diluted culture filtrates and the younger leaves were the more sensitive to the culture filtrates and their ethyl acetate extracts in grapes.

Selection of Clinically Isolated Strains for Evaluation of the Newly Synthesized Antibiotics (새로운 $\beta$-lactam계 항생물질 개발을 위한 검정용 균주의 개발)

  • Clinically isolated bacterial strains resistant to almost of all the clinically superior .betha.-lactam antibiotics can be used to screen the promising ones among the newly synthesized $\beta$-lactam antibiotics. To select the resistant strains, the susceptibility of 389 strains of S. aureus, 144 strains of coagulase negative staphylococci, 509 strains of E. coli, 115 strains of E. cloacae and 187 strains of P. aeruginosa to methicillin, ampicillin, piperacillin and gentamicin was determined. The susceptibility of 19 bacterial strains selected through the first screening to cefixime, cefotiam, cefotaxime, flomoxef, cepfirome, cefdnir, SCE-2787, panipenem and imipenem was determined. Four strains of S. aureus finally selected have high degree of resistance to almost of all $\beta$-lactam antibiotics used and also produce $\beta$-lactamases. These 4 strains of S. aurues can be used to screen effectively the promising $\beta$-lactam antibiotics among the numerous numbers of the newly synthesized $\beta$-lactam antibiotics.

Antibody-based Screening of Porphyromonas gingivalis Proteins Specifically Produced in Patients with Chronic Periodontitis

  • Porphyromonas gingivalis is among the major etiological pathogens of chronic periodontitis. The virulence mechanisms of P. gingivalis is yet to be identified as its activity is largely unknown in actual disease process. The purpose of this study is to identify antigens of P. gingivalis expressed only in patients with chronic periodontitis using a unique immunoscreening technique. Change Mediated Antigen Technology (CMAT), an antibody-based screening technique, was used to identify virulence-associated proteins of P. gingivalis that are expressed only during infection stage in patients having chronic periodontitis. Out of 13,000 recombinant clones screened, 22 tested positive for reproducible reactivity with rabbit hyperimmune anti-sera prepared against dental plaque samples acquired from periodontitis patients. The DNA sequences of these 18 genes were determined. CMAT-identified protein antigens of P. gingivalis included proteins involved in energy metabolism and biosynthesis, heme and iron binding, drug resistance, specific enzyme activities, and unknown functions. Further analysis of these genes could result in a novel insight into the virulence mechanisms of P. gingivalis.

Screening of Rice Cultivars for Adult-Plant Resistance to Pyricularia oryzae (도열병에 대해 성체식물 저항성을 지닌 벼 품종의 선발)

  • Thirty two rice cultivars which have been cultivated or used as breeding materials in Korea were tested for screening rice cultivars resistant to leaf blast at adult-plant stages in the blast nursery hill plots. When compared on the basis of disease severities in individual leaves at different growth stages of rice plants under natural field infection, the 16 indica-japonica hybrids tested were highly resistant but the 16 japonica cultivars tested showed various degrees of resistance to leaf blast. With aging of rice plants, the quantitative levels of resistance to leaf blast increased in all the cultivars, although the levels of resistance to leaf blast varied according to rice genotypes. The leaf position of rice plants in which changed from susceptible to resistant reactions varied also with rice genotypes. The susceptible reactions of the rice cultivars to Pyricularia oryzae were distinctly changed to a resistant reaction on upper leaves of rice plants. The rice cultivars, in which the quantitative level of resistance to leaf blast was higher, were resistant on the lower leaves of rice plants. The cultivars Akibare, Palkeum, Jinheung, Olchal, Dobong and Ginga which drastically decreased blast infection at late growth stage were evaluated as adult-plant-resistant to leaf blast in the field. The adult-plant-resistant cultivars became resistant to leaf blast, both qualitatively and quantitatively, as rice plants matured.

Development of a bioassay for screening of resistance to Tomato spotted wilt virus isolate from Korea (국내 분리 토마토반점위조바이러스의 저항성 판별을 위한 생물검정법 개발)

  • Tomato spotted wilt virus (TSWV) is one of the most destructive viruses worldwide, which causes severe damage to economically important crops, such as pepper and tomato. In this study, we examined the molecular and biological characterization of a TSWV isolate (SW-TO2) infecting tomato and compared it to the recently reported isolates from boxthorn, butterbur, and angelica plants. The phylogenetic analysis based on the complete genome sequences confirmed that SW-TO2 was clustered with those of isolates from boxthorn and pepper in Korea with the maximum nucleotide identities ranging from 98% to 99%. We developed the bioassay method for screening TSWV resistance and tested some commercial pepper and tomato cultivars for resistance evaluation of four isolates of TSWV. TSWV resistance was evaluated as TSWV resistance when all the following three conditions were satisfied: first, when symptoms of necrotic spots or no symptoms were present in the inoculated leaves; second, when there were no symptoms in the upper leaves; and third, when the upper leaves were negative as a result of RT-PCR diagnosis.

Optimization of Evaporator for a Vapor Compression Cooling System for High Heat Flux CPU (고발열 CPU 냉각용 증기 압축식 냉각 시스템의 증발기 최적화)

  • This paper presents the optimization process of evaporator for a vapor compression cooling system for high heat flux CPU. The CPU thermal capacity was given by 300W. Evaporating temperature and mass flow rate were $18^{\circ}C$ and 0.00182kg/s respectively. R134a was used as a working fluid. Channel width(CW) and height(CH) were selected as design factors. And thermal resistance, surface temperature of CPU, degree of superheat, and pressure drop were taken as objective responses. Fractional factorial DOE was used in screening phase and RSM(Response Surface Method) was used in optimization phase. As a result, CW of 2.5mm, CH of 2.5mm, and CL of 484mm were taken as an optimum geometry. Surface temperature of CPU and thermal resistance were $33^{\circ}C\;and\;0.0502^{\circ}C/W$ respectively. Thermal resistance of evaporator designed in this study was significantly lower than that of other cooling systems such as water cooling system and thermosyphon system. It was found that the evaporator considered in this work can be a excellent candidate for a high heat flux CPU cooling system.
