HIF-1-Dependent Induction of Jumonji Domain-Containing Protein (JMJD) 3 under Hypoxic Conditions

  • Jumonji domain-containing proteins (JMJD) catalyze the oxidative demethylation of a methylated lysine residue of histones by using $O_2$, ${\alpha}$-ketoglutarate, vitamin C, and Fe(II). Several JMJDs are induced by hypoxic stress to compensate their presumed reduction in catalytic activity under hypoxia. In this study, we showed that an H3K27me3 specific histone demethylase, JMJD3 was induced by hypoxia-inducible factor (HIF)-$1{\alpha}/{\beta}$ under hypoxia and that treatment with Clioquinol, a HIF-$1{\alpha}$ activator, increased JMJD3 expression even under normoxia. Chromatin immunoprecipitation (ChIP) analyses showed that both HIF-$1{\alpha}$ and its dimerization partner HIF-$1{\beta}$/Arnt occupied the first intron region of the mouse JMJD3 gene, whereas the HIF-$1{\alpha}/{\beta}$ heterodimer bound to the upstream region of the human JMJD3, indicating that human and mouse JMJD3 have hypoxia-responsive regulatory regions in different locations. This study shows that both mouse and human JMJD3 are induced by HIF-1.

NMR Hydrogen Exchange Study of DNA Duplex Containing the Consensus Binding Site for Human MEIS1

  • Transcription factors are proteins that bind specific sites or elements in regulatory regions of DNA, known as promoters or enhancers, where they control the transcription or expression of target genes. MEIS1 protein is a DNA-binding domain present in human transcription factors and plays important roles in various biological functions. The hydrogen exchange rate constants of the imino protons were determined for the wild-type containing the consensus DNA-binding site for the MEIS1 and those of the mutant DNA duplexes using NMR spectroscopy. The G2A-, A3G- and C4T-mutant DNA duplexes lead to clear changes in thermal stabilities of these four consensus base pairs. These unique dynamic features of the four base pairs in the consensus 5'-TGAC-3' sequence might play crucial roles in the effective DNA binding of the MEIS1 protein.

  • Purpose - The purpose of this study was to expound the packaging framework system(PFS) under international trade rules. Design/methodology/approach - This study analyzed packaging framework system and packaging regulations and technical standards in international regulations and major countries and classified these in some standards Findings - First, the study provides packaging provisions associated with packaging regulations and technical standards (PRTS). Then, the basis of PFS in international trade rules was proposed; it is believed that the PFS was composed of packaging legal system in WTO institutional framework (WTO-PLS), international organization packaging standard system (IO-PSS) and major countries or regions packaging regulatory system (MCR-PRS). Secondly, this paper expounded relations and legal characteristics of the three packaging systems. Finally, This study investigated PRTS coverage under technical barriers to trade (TBT) and Sanitary and Phytosanitary measures (SPS). Research implications or Originality - The regulations associated with PRTS have played a key role in challenges to merchandise trade in international trade. This study has significance in classifying packaging related reticulations into several criteria unlike previous studies. Therefore it is hope that this study can provide a supplementary document for the existing research of PRTS.

  • In eukaryotic cells, a key RNA processing step to generate mature mRNA is the coupled reaction for cleavage and polyadenylation (CPA) at the 3' end of individual transcripts. Many transcripts are alternatively polyadenylated (APA) to produce mRNAs with different 3' ends that may either alter protein coding sequence (CDS-APA) or create different lengths of 3'UTR (tandem-APA). As the CPA reaction is intimately associated with transcriptional termination, it has been widely assumed that APA is regulated cotranscriptionally. Isoforms terminated at different regions may have distinct RNA stability under different conditions, thus altering the ratio of APA isoforms. Such differential impacts on different isoforms have been considered as post-transcriptional APA, but strictly speaking, this can only be considered "apparent" APA, as the choice is not made during the CPA reaction. Interestingly, a recent study reveals sequential APA as a new mechanism for post-transcriptional APA. This minireview will focus on this new mechanism to provide insights into various documented regulatory paradigms.

Revolutionizing Nepal's Transportation: The Potential of Advanced Air Mobility (AAM) in Overcoming Geographical Challenges

  • This paper examines the unique transportation challenges posed by Nepal's diverse and rugged terrain, which significantly hampers socio-economic development due to its negative impact on infrastructure, trade, and accessibility. Despite ongoing efforts to enhance road and traditional air transport systems, Nepal's geographic and environmental conditions continue to obstruct efficient connectivity, particularly in rural and remote areas. This study proposes Advanced Air Mobility (AAM) as a transformative solution, leveraging recent technological advancements in unmanned aerial vehicles (UAVs) and electric vertical takeoff and landing (eVTOL) aircraft. By conducting a comprehensive analysis of Nepal's current transportation infrastructure and the feasibility of AAM implementation, the paper highlights the potential benefits of AAM, including improved accessibility, economic growth, and environmental sustainability. Furthermore, it addresses the anticipated challenges and regulatory considerations necessary for integrating AAM into Nepal's transportation network. Through a multidisciplinary approach, this research aims to contribute to the discourse on overcoming transportation barriers in mountainous regions, offering policy recommendations and identifying areas for future study to facilitate the adoption of AAM in Nepal and similar contexts worldwide.

참산부추(Allium sacculiferum Max.) 메탄올 추출물의 지방세포 내 ROS 생성 및 지질 축적 억제 효능 (Inhibitory Effects of Allium sacculiferum Max. Methanol Extracts on ROS Production and Lipid Accumulation during Differentiation of 3T3-L1 Cells)

  • 본 연구에서는 3T3-L1 지방전구세포를 이용하여 Allium 속 식물의 하나인 참산부추(ASM) 메탄올 추출물의 ROS 생성 저해 및 지질 축적 억제 효과를 확인하고자 하였다. 먼저 ASM 메탄올 추출물 $100{\sim}2,000{\mu}g/mL$의 모든 농도에서 유의적인 DPPH 라디칼 소거능을 나타내었으며, 지방전구세포에 ASM 메탄올 추출물 $10{\sim}100{\mu}g/mL$를 처리하였을 때 세포 독성을 나타내지 않았다. 지방세포 내 ROS 관련 효소와 분화 관련 전사인자의 조절로 인한 중성지방 축적 저해 활성을 확인하기 위하여 지방전구세포를 분화 유도하면서 추출물을 농도별(10, 50, $100{\mu}g/mL$)로 처리하였다. 그 결과 ASM 메탄올 추출물은 대조군에 비해 ROS 생성량과 ROS 관련 효소인 G6PDH mRNA 발현을 농도 의존적으로 감소시켰다. 또한 ASM 메탄올 추출물에 의하여 지방세포 내 중성지방 축적량이 유의적으로 감소하였으며, 지방세포 분화에 관련된 전사인자인 SREBP1c, $PPAR{\gamma}$$C/EBP{\alpha}$ mRNA 발현도 유의적으로 감소하였다. 이상의 결과들로 볼 때 ASM 메탄올 추출물로 인한 ROS 생성 저해와 지질 축적 억제는 ROS 생성 및 ROS 관련 유전자의 발현감소로 인한 지방 생성 주요 전사인자의 유전자 발현 억제로 인한 것으로 보이며, ASM이 항비만 효과가 있는 천연물 소재로 개발 가능성이 있을 것으로 사료된다.

세균 게놈 유래성 PyrR Orthologue의 기능 분석 (Characterization and Functional Study of PyrR Orthologues from Genome Sequences of Bacteria)

  • 그람 양성세균에서 PyrR단백질에 의하여 피리미딘의 생합성이 조절된다는 발견을 바탕으로 하여, Synechocystis sp.PCC6803과 Haemophilus influenzae의 PyrR orthologue 유전자를 Bacillus subtilis에서 형질전환 시켜 피리미딘 생합성의 조절 유무를 조사하였다. Synechocystis sp.PCC6803과 H. influenzae의 PyrR orthologue유전자를 pUC19과 T-vector에 클로닝 한후 pKH1, pKH2, pHPSK1, pHPSK2으로 각각 명명하였다. 이것을 다시 Escherichia. coli와 B. subtiius용 shuttle vector인 pHPS9에 클로닝 하여 pKH3, pKH4, pHPSK3, pHPSK4로 각각 명명하였다. B. subtilis DB104Δ PyrR에 pKH3, pKH4, pHPSK3, pHPSK4을 형질전환후 ATCase 활성을 측정결과 pHPSK3을 가진 균주만 피리미딘에 의한 조절작용이 일어난다는 사실을 통하여, H. influenzae의 PyrR orthologue 유전자의 선도 부분에 조절에 관여하는 미지의 부분이 있음을 예측할 수 있었다. 서로 다른 유래의 PyrR orthologue단백질을 정제하기 위하여 pET14b에 클로닝후 pKH5, pHPSK5으로 각각 명명하였다. SDS-PAGE로 분석한 결과 각각 약 18 kDa과 21 kDa의 분자량을 나타내었다. 정제된 PyrR orthologue 단백질의 UPRTase 활성을 측정한 결과 H. infuenzae의 PyrR orthologue 단백질은 UPRTase 활성을 나타내었으며 다양한 pH에서 측정한 결과 pH 5에서 가장 높은 활성을 나타내었다. 반면에, Synechocystis sp. PCC6803의 PyrR orhologue 단백질은 UPRTase 활성을 나타내지 않았다. 여러 가지 균주의 PyrR 아미노산 서열을 비교한 계통수 분석은 PyrR 단백질의 조절기작과 어느 정도 연관됨을 시사해 주었다.

  • Biosynthesis and secretion of anterior pituitary hormones are under the control of specific hypothalamic stimulatory and inhibitory factors. Among them, Growth Hormone Releasing Hormone (GHRH) is the major stimulator of pituitary somatotrophs activating GH gene expression and secretion. Human GHRH is a polypeptide of 44 amino acids initially isolated from pancreatic tumors, and the gene for the hypothalamic form of GHRH is organized into 5 exons spanning over 10 kilobases (kb) on genomic DNA and encodes a messenger RNA of 700-750 nucleotides. Several neuropeptides classically associated with the hypothalamus have been found in the extrahypothalamic regions, suggesting the existence of novel sources, targets and functions. GHRH-like immunoreactivity has been found in several peripheral sites, including placenta, testis, and ovary, indicating that GHRH may also have regulatory roles in peripheral reproductive organs. Furthermore, higher molecular weight forms of the GHRH transcripts were identified from these organs (1.75 kb in testis; 1.75 and >3 kb in ovary). These tissue-specific expression of GHRH gene suggest the existence of unique regulatory mechanism of GHRH expression and function in these organs. In fact, placenta-specific and testis-specific promoters for GHRH transcripts which are located in about 10 kb upstream region of hypothalamic promoter were reported. The use of unique promoters in extrahypothalamic sites could be refered in a different control of GHRH gene and different functions of the translated products in these tissues. Somatotrophs and lactotrophs have been thought to be derived from a common bipotential progenitor, the somatolactotrophs, which give origins to either phenotypes. Although the precise mechanism responsible for the lactotroph differentiation in the anterior pituitary gland has not been yet clalified, there are several candidators for the generation of lactotrophs. In human, the presence of GHRH peptides with different size from authentic hypothalamic form in the normal anterior pituitary and several types of adenoma were demonstrated. Recently our group found the existence of immunoreactive GHRH and its transcript from the normal rat anterior pituitary (gonadotroph> somatotroph> lactotroph), and the GHRH treatment evoked the increased proliferation rate of anterior pituitary cells in vitro. The transgenic mouse models clearly shown that GHRH or NGF overexpression by anterior pituitary cells induced development of pituitary hyperplasia and adenomas particularly GH-oma and prolactinoma. Taken together, we hypothesize that the pituitary GHRH could serve not only as a modulator of hormone secretion but as a paracrine or autocrine regulator of anterior pituitary cell proliferation and differentiation. Interestingly enough, the expression of Pit-1 homeobox gene (the POU class transcription factor) was confined to somatotrophs, lactotrophs and somatolactotrophs in which GHRH receptors are expressed commonly. Concerning the mechanism of somatolactotroph and lactotroph differentiation in the anterior pituitary, we have focused following two possibilities; (1) changes in the relative levels or interactions of both hypothalamic and intrapituitary factors such as dopamine, VIP, somatostatin, NGF and GHRH; (2) alterations of GHRH-GHRH receptor signaling and Pit-1 activity may be the cause of lactotroph differentiation or pituitary hyperplasia and adenoma formation. Extensive further studies will be necessary to solve these complicated questions.

Overlapping Region of p53/Wrap53 Transcripts: Mutational Analysis and Sequence Similarity with microRNA-4732-5p

  • Background: Although the majority of investigations concerned with TP53 and its protein have focused on coding regions, recently a set of studies highlighted significant roles of regulatory elements located in p53 mRNA, especially 5'UTR. The wrap53${\alpha}$ transcript is one of those that acts as a natural antisense agent, forming RNA-RNA hybrids with p53 mRNA and protecting it from degradation. Materials and Methods: In this study, we focused on the mutation status of exon $1{\alpha}$ of the WRAP53 gene (according to exon 1 of p53) in 160 breast tumor tissue samples and conducted a bioinformatics search for probable miRNA binding site in the p53/wrap53 overlapping region. Mutations were detected, using single stranded conformation polymorphism (SSCP) and sequencing. We applied the miRBase database for prediction of miRNAs which target overlapping region of p53/wrap53 transcripts. Results: Our results showed all samples to have wild type alleles in exon 1 of TP53 gene. We could detect a novel and unreported intronic mutation (IVS1+56, G>C) outside overlapping regions of p53/wrap53 genes in breast cancer tissues and also predict the presence of a binding site for miR-4732-5p in the 5'UTR of Wrap53 mRNA. Conclusions: From our findings we propose designing further studies focused on overexpression of miRNA-4732-5p and introducing different mutations in the overlapping region of wrap53 and p53 genes in order to study their effects on p53 and its ${\Delta}N$ isoform (${\Delta}$40p53) expression. The results may provide new pieces in the p53 targeting puzzle for cancer therapy.

Resveratrol Induces Glioma Cell Apoptosis through Activation of Tristetraprolin

  • Tristetraprolin (TTP) is an AU-rich elements (AREs)-binding protein, which regulates the decay of ARE-scontaining mRNAs such as proto-oncogenes, anti-apoptotic genes and immune regulatory genes. Despite the low expression of TTP in various human cancers, the mechanism involving suppressed expression of TTP is not fully understood. Here, we demonstrate that Resveratrol (3,5,4'-trihydroxystilbene, Res), a naturally occurring compound, induces glioma cell apoptosis through activation of tristetraprolin (TTP). Res increased TTP expression in U87MG human glioma cells. Res-induced TTP destabilized the urokinase plasminogen activator and urokinase plasminogen activator receptor mRNAs by binding to the ARE regions containing the 3' untranslated regions of their mRNAs. Furthermore, TTP induced by Res suppressed cell growth and induced apoptosis in the human glioma cells. Because of its regulation of TTP expression, these findings suggest that the bioactive dietary compound Res can be used as a novel anti-cancer agent for the treatment of human malignant gliomas.
