Effects of White Radish (Raphanus sativus) Enzyme Extract on Hepatotoxicity

  • Raphanus sativus (Cruciferaceae), commonly known as radish is widely available throughout the world. From antiquity it has been used in folk medicine as a natural drug against many toxicants. The present study was designed to evaluate the hepatoprotective activity of radish (Raphanus sativus) enzyme extract (REE) in vitro and in vivo test. The $IC_{50}$ values of REE in human liver derived HepG2 cells was over 5,000 ${\mu}g/ml$ in tested maximum concentration. The effect of REE to protect tacrine-induced cytotoxicity in HepG2 cells was evaluated by MTT assay. REE showed their hepatoprotective activities on tacrine-induced cytotoxicity and the $EC_{50}$ value was 1,250 ${\mu}g/ml$. Silymarin, an antihepatotoxic agent used as a positive control exhibited 59.7% hepatoprotective activity at 100 ${\mu}g/ml$. Moreover, we tested the effect of REE on carbon tetrachloride ($CCl_4$)-induced liver toxicity in rats. REE at dose of 50 and 100 mg/kg and silymarin at dose of 50 mg/kg were orally administered to $CCl_4$-treated rats. The results showed that REE and silymarin significantly reduced the elevated levels of serum enzyme markers induced by $CCl_4$. The biochemical data were supported by evaluation with liver histopathology. These findings suggest that REE, can significantly diminish hepatic damage by toxic agent such as tacrine or $CCl_4$.

청피홍심무우(Raphanus sativus cv. Chungpihongsim)의 모상근 배양에 의한 안토시아닌 생성 (Production of Anthocyanin by Culture of Hairy Roots of Raphanus sativus cv. Chungpihongsim)

  • A. rhizogenes A4 균주를 접종하여 청피홍심무우(Raphanus sativus cv. Chungpihongsim)의 모상근 배양을 확립하였다. 형질전환된 뿌리는 MS 배지의 기본염을 1/2로 희석하고, pH는 5.2, sucrose는 3%로 조정한 배양조건에서 최적성장을 보였다. 형질전환된 조직에서 합성되는 물질인 opine, 즉 agropine과 mannopine이 모상근의 추출액에서 검출되었다. 배지에 2, 4-D와 Kinetin이 첨가될 경우 모상근의 탈분화와 더불어 세포내에 안토시아닌의 합성이 유도되었으며, 그 중 2, 4-D $0.45\;\mu\textrm{M}$과 kinetin $2.3\;\mu\textrm{M}$의 첨가에서 최대의 색소 축적능을 보였다. 모상근의 탈분화와 함께 유도되는 안토시아닌의 paper chromatography 전개양상은 경작뿌리에서 추출한 색소에 비하여 다소 차이를 보였지만 모든 안토시아닌의 aglycone은 pelargonidin으로 확인되었다. 이러한 시료의 총 안토시아닌 함량은 0.49 mg/g(f.w.)로 계산되었다.

Antifungal and Plant Growth Promotion Activities of Recombinant Defensin Proteins from the Seed of Korean Radish (Raphanus sativus L.)

  • In the present study, we analyzed the defensin protein deduced from Korean radish (Raphanus sativus L.) seeds.To express the genes in E. coli, we constructed a recombinant expression vector with a defensin gene, named rKRs-AFP gene isolated from Korean radish seeds. Over expressed rKRs-AFP proteins was separated by SDS-PAGE to determine the purity, and protein concentration was determined by the Bradford method. Antifungal activity was assessed by disk assay method against the tested fungi. As a result, when 500 mL of cell culture were disrupted by sonicator, 32.5 mg total proteins were obtained. The purified protein showed a single band on SDS-PAGE with estimated molecular weight about 6 KDa, consistent with the molecular mass calculated from the deduced amino acid sequence. The purified rKRs-AFP protein showed remarkable antifungal activities against several fungi including Aspergillus niger, Botrytis cinerea causing the gray mold disease, and Candida albicans. In field tests using the purified rKRs-AFP protein, the protein showed the reducing activity of disease spot and the mitigating effect of spreading of disease like agrichemicals. The immuno-assay of rKRs-AFP protein showed that the purified protein entirely accumulated at B. cinerea cytoplasm through the hyphal septa shown by fluorescence imaging. There was no fluorescence inside the cell, when the hypha was incubated without the protein. These all results indicate that the recombinant rKRs-AFP proteins can be utilized as a potential antifungal drug to control harmful plant fungal pathogens.

  • Toxic effert of water extract from leaves of Pinus rigida and some phenolic compounds on the seeds germination of Raphanus sativus var. hortensis for. acanthiformis Makino has been studied. There was little difference of germination percentage among the pH value of leaf extract (pH3~9). The germination percentage drastically with increased concentration level of leaf extract at about 60 percent. Seeds gemination of Raphanus sativus var. hortensis for. acanthiformis Makino inhibited severely by caffeic acid, ferulic acid and p-coumaric acid at M, but the germination percentage was higher than that of the control group in vanillic acid. In electrophoresis, there was no differences at earlier seedling stage of protein band between allelochemical treated and non-treated group, but in late stage, two protein band near 58kd and 27kd did not appeared in the toxic affected group. In case of caffeic acid treatment, two protein band near 58kd and 27kd did not found at late stage too.

무 유식물에서의 니켈내성과 히스티딘의 작용 (Nickel Tolerance and the Complexing Role of Histidine in Raphanus sativus)

  • The effect of nickel (Ni) on growth and some tolerance strategies with regard to heavy metal tolerance mechanism was investigated in radish (Raphanus sativus) seedlings. The protective effect of histidine on nickel stress conditions was also monitored. The seedling growth decreased with an increase in metal concentrations. The inhibitory effect was more pronounced in the root elongation than in the shoot elongation. Increasing Ni supply showed a progressive increase of Ni concentrations in the roots and shoots. Ni content was higher in the shoots than in the roots. In the presence of nickel, radish exhibited an antioxidative defense mechanism, as evidenced by the elevated malondialdehyde(MDA), showing that nickel is an efficient inducer of lipid peroxidation. Exposure of radish to elevated concentrations of nickel was accompanied by an increase in the proline content. Supplemental histidine in the presence of Ni ameliorated metal-induced growth inhibition and lipid peroxidation. Combinations of Ni and histidine resulted in a significant decline in proline content compared with Ni stress alone, indicating that histidine may provide protection against the adverse effect of Ni stress. From the results it is suggested that histidine is an efficient chelator by complexing metal ion within the plant and may playa role in nickel tolerance implicated in metal detoxification.

Leaves of Raphanus sativus L. Shows Anti-Inflammatory Activity in LPS-Stimulated Macrophages via Suppression of COX-2 and iNOS Expression.

  • Raphanus sativus L. (RS) is a cruciferous vegetable that is widely consumed in Korea. The anticancer activity of leaves of RS (RSL) extract has been investigated; however, no studies focused on its anti-inflammatory effects. Therefore, the aim of the current study was to evaluate the anti-inflammatory effects of RSL extract. In brief, RSL powder was fractionated into n-hexane, chloroform, ethyl acetate, n-butanol, and water-soluble fractions. Lipopolysaccharide (LPS)-stimulated RAW264.7 cells were treated with each fraction for initial screening. It was found that the chloroform fraction significantly inhibited nitric oxide release in LPS-stimulated RAW264.7 cells with a half maximal inhibitory concentration value of $196{\mu}g/mL$. In addition, the mRNA and protein expression levels of inducible nitric oxide synthase, measured using reverse transcriptase-polymerase chain reaction and western blotting, respectively, were reduced in a concentration-dependent manner. Moreover, the inflammatory cyclooxygenase-2 enzyme expression decreased. Furthermore, the expression of nuclear factor-kappa B ($NF-{\kappa}B$), the key regulator of the transcriptional activation of the inflammatory cytokine genes, was reduced by the RSL chloroform fraction. Therefore, the results of our study suggest that RSL exhibits anti-inflammatory effects in LPS-stimulated macrophages via $NF-{\kappa}B$ inactivation.

Genetic Variation in Korean Populations of Wild Radish, Raphanus sativus var.hortensis f. raphanistroides (Brassicaceae)

  • Raphanus sativus L. var. hortensis f. raphanistroides (wild radish: Brassicaceae), a herbaceous perennial, occurs only on beaches in East Asia. Genetic diversity and population structure of seven Korean populations were investigated using starch gel electrophoresis. Although the Korean populatins are small, isolated with patchy distribution, the population maintain a moderate level of genetic diversity; the mean percentage fo polymorphic loci was 51.4%, mean number of alleles per locus was 1.84, and mean expected heterozygosity was 0.116. A combination of animal-outcrossing breeding system, wide geographical distribution, restricted ecological distribution, and a propensity for high fecundity may in part be explanatory factors contributing the moderate level of genetic diversity within populations. An overall excess of homozygotes relative to Hardy-Weinberg expetations (mean FISa=0.116) indicates that consanguineous mating occur within wild radish populations, leading to a family structure within a circumscribed area. Although population of wild radish experience a limited gene flow, only 5% of the total genetic variation found in Korean wild radish populations examined is due to differences among populations (mean GST=0.052). This value is considerably lower than the mean values of species with similar life history and ecological characteristics. However, significant differences were found in allele frequencies between populations for all polymorphic loci (P<0.01). It is supposed that directional selection toward genetic uniformity (similar gene frequencies) in a relatively homogenous habitat is thought to be operated among Korean wild radish populations.

몇가지 식물에 있어서 어린순의 식품학적 가치개발을 위한 기초연구 (Basic Research of Development for Food of Seedling in Some Plants)

  • 최근 소비자들의 신선한 채소 요구에 따른 수요증가로 볼 때 어린순의 소비는 크게 늘어 날 것으로 보나 국내에서는 무순외에는 거의 개발이 안된 실정이다. 이에 본 연구에서는 약용식물중에서 어린순을 채소로 개발할 수 있는 것을 선발하고자 기초연구로써 수행한 결과는 다음과 같았다. 1. 실험에 사용된 12종의 식물중에서 가걱은 들깨, 메밀, 무등이 낮은 값을 보였다. 2. 발아율은 전체적으로 낮았으나 그중 무, 소엽, 메밀, 항금 및 들깨가 상대적으로 높은 발아율을 보였다. 3. 발아세는 발아율보다 낮으며 그중 소엽, 무, 메밀 및 황금이 상대적으로 높은 발아세를 보였다. 4. 식미특성중 외관상으로 무가 가장 높은 점수를 보였다. 5. 맛은 황금이 가장 높은 점수를 보였다. 6. 조직감은 메밀이 가장 높은 점수를 보였다. 7. 기호도는 소엽이 가장 높은 점수를 보였다.

Plant Growth-Promoting Rhizobacteria Improved Salinity Tolerance of Lactuca sativa and Raphanus sativus

  • Salinity stress is an important environmental problem that adversely affects crop production by reducing plant growth. The impacts of rhizobacterial strains to alleviate salinity stress on the germination of Lactuca sativa and Raphanus sativus seeds were assessed using different concentrations of NaCl. Plant growth-promoting rhizobacteria (PGPR) strains were also examined to improve the early germination of Chinese cabbage seeds under normal conditions. Lactobacillus sp. and P. putida inoculation showed higher radicle lengths compared with non-inoculated radish (Raphanus sativus) seeds. LAP mix inoculation increased the radicle length of lettuce (Lactuca sativa) seedlings by 2.0 and 0.5 cm at salinity stress of 50 and 100 mM NaCl concentration, respectively. Inoculation by Azotobacter chroococcum significantly increased the plumule and radicle lengths of germinated seeds compared with non-inoculated control. A. chroococcum increased the radicle length relative to the uninoculated seeds by 4.0, 1.0, and 1.5 cm at 50, 100, and 150 mM NaCl concentration, respectively. LAP mix inoculation significantly improved the radicle length in germinated radish seeds by 7.5, 1.3, 1.2, and 0.6 cm under salinity stress of 50, 100, 150, and 200 mM NaCl concentration, respectively. These results of this study showed that PGPR could be helpful to mitigate the salinity stress of different plants at the time of germination.

무(Raphanus sativus L.) 첨가 속성발효 멸치(Engraulis japonicus) 액젓의 제조 및 품질 (Processing and Quality Characteristics of Rapidly Fermented Anchovy Engraulis japonicus Sauce with Radish Raphanus sativus L.)

  • To develop a value-added anchovy Engraulis japonicus sauce, the processing conditions and quality characteristics of a rapidly fermented and high-purity anchovy sauce (RRAS) were examined by adding 5-10% (w/w) radish Raphanus sativus L. RRAS exhibited higher yield, total nitrogen content, and amino nitrogen and calcium contents as well as lower salinity than those of conventional anchovy sauce (control). The salinity, yield, and total amino acid contents of RRAS and control were 17.2-17.7% and 19.6%, 81.2-88.7% and 61.0%, and 13,117.8-14,174.9 mg/100 g and 10,041.1 mg/100 g, respectively. The major amino acids recorded were aspartic acid, glutamic acid, alanine, valine, isoleucine, leucine, phenylalanine, ornithine, lysine, and histidine. The histamine contents of RRAS and control were 8.3-8.6 mg/100 g and 19.2 mg/100 g, respectively, while the protease activity levels were 0.901-0.958 unit/mg and 0.695 unit/mg, respectively. Overall, this study establishes that RRAS can not only significantly shorten the salt fermentation period, but can also serve as an anchovy sauce with superior nutritional quality and higher levels of amino acid and calcium.
