무발정우에서 Progesterone Implant와 Follicular Rupture에 따른 발정유도 및 임신율 (Effect of Progesterone Implant and Follicular Rupture on Estrus Induction and Fertility in Anestrus Cows)

  • 무발정 증상을 보이는 젖소 65두를 대상으로 몇 가지 호르몬 처리기법을 사용하여 소의 번식효율 향상시키고자 본 실험을 수행하였다. Group 1. Ovsynch program (GnRH-PGF$_2$ a /PGF$_2$ a/GnRH), Group 2. Two plus Two program (GnRH-PGF$_2$ a /PGF$_2$ a/GnRH), Group 3. progesterone implant (CIDR)-GnRH/PGF$_2$ a/PGF$_2$ a/GnRH과 Group 4. (Follicular rupture-progesterone implant-GnRH/ PGF$_2$ a/PGF$_2$ a/GnRH)로 구분하고 최종 GnRH 처지 후 발정 유도율, 발정유도시간, 임신율, 유산율, 인공수정 후 60일까지의 임신율을 조사하여 다음과 같은 결론을 얻었다. 1. 발정 유도율은 Group 1, 2, 3에서는 각기67%, 75%, 70%로서 비슷하게 나타났으나, Group 4에서는 86%로서 유의적(P<0.05)으로 높은 발정율을 나타내었다. 2. 발정 유도 시간은 progesterone implant를 사용한 Group 3과 Group 4는 각각 24, 23시간으로 발정유도 시간을 단축시키는데 효과가 있었다. 3. 각 호르몬 투여 임신율은 progesterone implant를 사용한 Group 3과 Group 4가 75%, 85%로서 사용하지 않은 Group 1과 Group 2보다 유의적(P<0.05)으로 높은 임신율을 나타내었다. 4. 임신 49일까지의 유산율은 Group 4가 5.9%로 낮은 유산율을 보였다. 이상의 실험결과를 종합하면 무발정우에서 각종 호르몬을 투여하기 전에 난소를 확인하여 폐쇄 난포가 있는 것은 폐쇄난포를 제거하고 progesterone implant 제제인 CIDR를 질내에 장착하는 것이 무발정우의 발정유기 및 수태율 향상에 효과적이라고 사료된다.

흰쥐 착상시기에 자궁내 난소 홀몬 수용체와 Prostaglandin 및 cAMP 농도변화에 관한 연구 (Studies on the Concentrations of Receptors for Ovarian Steroids, Prostaglandins and cAMP in Uterine Tissue during the Period of Implantation in Rats)

  • In the present study, hormonal changes in uterine tissue and circulation were evaluated during the implantation period in rats in order to understand the mechanism by which implantation takes place. The results obtained were as follows. 1. Concentrations of serum estradiol and progesterone were significantly increased on days 4 and 5. 2. Concentration of estrogen receptor reached maximum on day 5 when implantation normally occurred in rats. On the other hand, progesterone receptor was gradually decreased, reaching the lowest on day 5. 3. Uterine PGs and cAMP concentrations were significantly increased on day 5. 4. Uterine PGs and cAMP concentrations in implant sites were significantly greater than those in non-implant sites. It is, therefore, concluded that prostaglandins and cAMP in uterine tissue as well as circulating ovarian steroid hormones were increased during the period of implantation, suggesting that these hormones might be actively involved in the process of implantation in rats.

Prophylactic Mastectomy and Implant-Based Breast Reconstruction of BRCA1/2 Mutation-Positive Patients in Korea

  • Purpose: Mastectomy is performed as a surgical treatment for patients with breast cancer who have the BRCA 1/2 mutation. In this study, we have reported the trends in Korea for both immediate breast reconstruction and prophylactic mastectomy. Methods: This retrospective study was conducted from 2019 to 2021. Both skin-sparing mastectomy and immediate implant-based breast reconstruction with prepectoral and/or subpectoral techniques were performed in five patients with BRCA 1/2 mutations. Data on age; body mass index; cancer stage; BRCA 1/2 mutation; estrogen receptor, progesterone receptor, and human epidermal growth factor receptor 2 expression; diagnosis; and complications were collected. Results: The average (±standard deviation [SD]) age was 44.0±6.48 years old; BMI 24.5±2.25 kg/m2; and breast volumes were 365.8±70.34 and 382.4±96.33 cc for right and left ones, respectively. The BRCA 1 and 2 were diagnosed in four and one patients, respectively. The estrogen and progesterone receptors and human epidermal growth factor receptor 2 were detected in one (20%), one (20%), and three (60%) patients, respectively. The applied implant-based breast reconstruction techniques for ten breasts were subpectoral technique (n=7, 70%) and prepectoral technique (n=3, 30%). For the cancer stage, those with I, II, and III stages were one (20%), two (40%), and one (20%), respectively. There were no major complications such as Infection, seroma. Conclusion: When mastectomy is performed as surgical treatment in BRCA 1/2 mutation positive breast cancer patients, it is possible to obtain a better outcome with both implant-based breast reconstruction and different circumstances between breast cancer and contralateral breast.

여성 호르몬의 변화가 치은 섬유아세포와 치주인대세포의 교원질 분해 효소의 활성에 미치는 영향 (In Vitro Effects of Female Sex Hormones on Collagenase Activity of Gingival Fibroblast and Periodontal Ligament Fibroblast)

  • Many factors may affect periodontal changes during the physiologic conditions of woman(e.g. puberty, menstrual cycle, pregnancy, menopause). Recently many research has focused on the immunological changes of host, but the exact mechanism is not clear. Collagen is a major constituent of periodontium, and collagenase specifically digests the collagen and plays a role in destruction of periodontal tissue. So, I suppose that it participates with the cytokines in the inflammation of gingiva and vascular response during the changes of female sex hormones. Because there are some evidences of the existence of the receptors of estrogen and progesterone in the gingiva, it may be a target tissue of female sex hormones. In this experiment, gingival fibroblast and periodontal ligament cell were cultured in the presence of various concentrations of estrogen or progesterone corresponding to the menstrual cycle and pregnancy. Collagenase activity of the supernatant of culture media was determined by Spectrophotometric collagenase assay. The enzyme activity was calculated by the % decrease of the coated collagen. 1. The estrogen at both concentrations had no effect on the activity of collagenase of the gingival fibroblast. 2. The progesterone had some effect on the collagenase activity of the gingival fibroblast at low and high concentration of menstrual cycle, and elevated the enzyme activity at all range of pregnancy concentrations. 3. In periodontal ligament cells, estrogen elevated the enzyme activity at the early pregnancy concentration and progesterone elevated at the concentration just before menstruation. In this experiment, pregesterone elevated the collagenase activity of gingival fibroblast and periodontal ligament cells. But the mechanism of the up-regulation of the enzyme activity was not confirmed. The more experiments of direct effect of progesterone on gingival at the molecular level(e.g. northern blot analysis) can reveal the exact mechanism.

흰쥐 수정란 착상시기에 있어서의 호르몬 농도의 변화와 자궁내막의 구조에 관한 연구 (Studies on the concentrations of hormones and the structure of uterine endometrium during the implantation period in rats)

  • The mechanism by which blastocysts implant to uterine endometrium has not been clearly understood. In the present study, the following question was investigated: how are hormonal levels changed and how is uterine endometrium morphologically changed? Results obtained are as follows: Concentrations of serum estradiol and progesterone were significantly increased on day 4 and 5 of pregnancy. Uterine concentrations of PGE and $PGE_{2a}$ were sharply increased on day 1 and maintained similar concentrations thereafter, reaching the maximum on day 5. Both prostaglandins (PGs) concentrations were gradually decreased thereafter. Furthermore, concentrations of PGs in implant sites were greater than those in non-implant sites. PBR (pontamine blue reaction) in uterine endometrium was positive on day 6 of pregnancy, indicating that vascular permeability was increased. Edema and changes in structure of cell components were pronounced in stroma where PBR was developed. Moreover, these were more prominent in implant sites than non-implant sites. These results suggest that uterine PGs as well as steroid hormones increase during implantation in rats and these hormones might be involved in the process of implantation by modulating vascular permeability and the fine structures of uterine endometrial cells.

난소제거된 Shiba 염소에서 다른 농도의 Progesterone 처치에 의한 FSH Surge 및 Pulse 분비에 미치는 영향 (Patterns of Pulsatile and Surge Modes of Follicle-Stimulating Hormone Treated with Different Progesterone Levels in Ovariectomized Goats)

  • 본 연구의 목적은 FSH 분비 조절에 있어서 각각 다른 progesterone (P) 농도가 Pulse 또는 Surge 형태의 분비 패턴에 어떤 영향을 미치는가를 탐구한다. 선행된 연구에서 LH surge는 난포기 (follicular phase)에서 보이는 P 농도에서만 발현되었고 황체기(luteal phase) 또는 아황체기(subluteal phase) P 농도에서는 완전히 억제되었다. 또한 LH pulse 분비는 황체기 P농도에서만 시간 의존적으로 감소되었으나 난포기 또는 아황체기 농도에서는 높은 빈도로 유지되었다. 즉 아황체기 P농도 (스트레스, 황체형성부전에서 발생)는 LH surge는 억제시키나 LH pulse 조절에는 아무런 영향을 마치지 않았다. 이 결과를 바탕으로 FSH surge 및 pulse 분비패턴을 알아보기 위해 난소가 제거된 Shiba 염소를 이용하여 인위적으로 난포기, 아황체기, 기능성황체기의 P 농도 및 estradiol(E) 농도를 유도하였다. 스테로이드 호르몬 농도의 변화를 알아보기 위해 매일 혈액을 채취하였으며, LH pulse 용 혈액은 10분 간격으로 8시간 동안 Day 0, Day 3, Day 7 (Day 0: P packet 이식 직전)에 연속채혈을 실시하고, LH surge 분비패턴은 Day 8에 16시간 estradiol를 3 ${\mu}g/h$ 농도로 주사하고, 52시간 동안 2시간 간격으로 채혈을 실시했다. 그 결과 유도된 P 농도는 염소의 정상적 발정주기에서 볼 수 있는 난포기(<0.1 ng/ml), 아황체기($1.1{\pm}0.1$ ng/ml), 황체기 ($3.3{\pm0.1}$ ng/ml) 수준의 농도로 유도되었다. FSH pulse 분비는 3그룹에서 모두 높은 빈도로 유지되었고, FSH surge는 LH surge와 같은 시간대에서 난포기 그룹에서만 확인되었다. 본 연구의 결과는 FSH pulse는 LH pulse에서 보이는 progesterone에 의한 억제 효과와 다르게 나타났으며, 이러한 결과는 FSH 분비 조절에 있어서 LH 분비와 다른 기전 및 요소들이 관여하는 것으로 판단되었다.

Successful Birth of Pups Produced by GnRH-induced Estrus and Natural Mating in Captive Red Fox (Vulpes vulpes)

  • On January 6, 2010, two months earlier than normal breeding season, a red fox vixen was implanted with synthetic GnRH analogue, Deslorelin. Blood was sampled every 2~3 days from the day of implant to identifying spermatozoa on stains of epithelial cells. Estradiol and progesterone were examined. Even though the vixen was in non-breeding season, she was mated by a male fox. Pregnancy was confirmed by canine pregnancy detection kit that detect relaxin released from placenta. Four healthy pups were born on March 9, 2010. This is the first report showing synthetic GnRH can activate ovarian function and lead to fertile estrus of red fox in non-breeding season.

Changes of Sex Hormones and Cervical Mucus in Estrus-synchronized Himalayan Tahrs (Hemitragus jemlahicus) in Non-breeding Season

  • Eight female Himalayan tahrs (Hemitragus jemlahicus) were estrus-synchronized, and transcervically inseminated with frozen-thawed semen in September, 2009, about 2 to 3 months earlier than their natural breeding season. Intravaginal progesterone-releasing devices were inserted into vaginas of six Himalayan tahrs on September 7, and the other two on September 8 to suppress luteal function of ovaries. The devices had been placed deep inside the vagina prior to withdrawal on September 23. A day before CIDR removal, a combination of PMSG 400 IU and hCG 200 IU was intramuscularly injected. Forty hours later, frozen-thawed semen was transcervically inseminated. Pregnancy diagnosis was performed 39 days later by analyzing progesterone level of serum. Every treatment was done under anesthesia inducted by xylazine injection. In conclusion, vaginal discharge of cervical mucus, hormonal changes induced by implant-typed or muscularly injectable hormones and widening of cervix enough to insert an insemination gun into uterine body were achieved in non-breeding season. Moreover, the first inseminated Himalayan tahr, 36 hours after CIDR removal was assumed to be pregnant but the fetus may have been lost due to the use of anesthetic drug.
