A cyber-physical system is a technology that connects the physical systems of a manufacturing environment with a cyber space to enable simulation. One of the major challenges in this technology is the seamless communication between these two environments. In complex manufacturing processes, it is crucial to adapt to various protocols of manufacturing equipment and ensure the transmission and reception of a large volume of data without delays or errors. In this study, we propose a method for constructing agent models for real-time simulation-capable cyberphysical systems. To achieve this, we design data collection units as independent agent models and effectively integrate them with existing simulation tools to develop the overall system architecture. To validate the proposed structure and ensure reliability, we conducted empirical testing by integrating various equipment from a real-world smart microfactory system to assess the data collection capabilities. The experiments involved testing data delay and data gaps related to data collection cycles. As a result, the proposed approach demonstrates flexibility by enabling the application of various internal data collection methods and accommodating different data formats and communication protocols for various equipment with relatively low communication delays. Consequently, it is expected that this approach will promote innovation in the manufacturing industry, enhance production line efficiency, and contribute to cost savings in maintenance.

The CRISPR-Cas9 system has significantly advanced regenerative medicine research by enabling genome editing in stem cells. Due to their desirable properties, mesenchymal stem cells (MSCs) have recently emerged as highly promising therapeutic agents, which properties include differentiation ability and cytokine production. While CRISPR-Cas9 technology is applied to develop MSC-based therapeutics, MSCs exhibit inefficient genome editing, and susceptibility to plasmid DNA. In this study, we compared and optimized plasmid DNA and RNP approaches for efficient genome engineering in MSCs. The RNP-mediated approach enabled genome editing with high indel frequency and low cytotoxicity in MSCs. By utilizing Cas9 RNPs, we successfully generated B2M-knockout MSCs, which reduced T-cell differentiation, and improved MSC survival. Furthermore, this approach enhanced the immunomodulatory effect of IFN-r priming. These findings indicate that the RNP-mediated engineering of MSC genomes can achieve high efficiency, and engineered MSCs offer potential as a promising therapeutic strategy.
Objective: The goal of the current study was to investigate the impact of various concentrations of xylanase in energy-deficient corn-based diets on the growth performance, carcass characteristics, nutrient digestibility, and digesta viscosity in broilers from 7 to 35 days of age. Methods: A total of 280 seven-day-old Ross 308 broilers were randomly allocated to one of the five dietary treatments following a completely randomized design with 8 replicates and 7 birds per cage. The treatments were: i) positive control (PC, without xylanase); ii) NC-1 (80 kcal/kg ME reduced from PC); iii) NC-2 (100 kcal/kg ME reduced from PC); iv) NCX-1 (NC-1 + 2,000 U/kg xylanase); and v) NCX-2 (NC-2 + 3,000 U/kg xylanase). Body weight, weight gain, feed intake, and feed conversion ratio were determined weekly to evaluate growth performance. One bird per pen was sacrificed for ileal digesta collection to determine the viscosity and digestibility of energy, dry matter, crude protein on days 24 and 35, however breast and leg meat samples were obtained for proximate analysis (moisture, crude protein, fat, and ash) on day 35. Results: Birds fed diets supplemented with xylanase regardless of the amount had higher (p<0.05) body weights, daily gains, and improved feed efficiency compared to NC diets all throughout the experimental period. Feed intake was not affected (p>0.05) by the addition of xylanase. Moreover, lowered (p<0.05) viscosity of the ileal digesta were observed upon xylanase inclusion in the diets compared to the birds fed NC diets on day 24. Ileal nutrient digestibility and meat proximate composition were not affected (p>0.05) by xylanase. Conclusion: The present study indicated that the xylanase at 2,000 U/kg and 3,000 U/kg levels compensates for the 80 kcal/kg and 100 kcal/kg dietary energy levels, respectively, without having adverse effects on the growth performance, carcass characteristics, nutrient digestibility, and digesta viscosity of broilers.

Since the cement industry generates more than 60 % of CO2 during the clinker production process, supplementary cementitious materials are used worldwide to reduce CO2 efficiently. Mainly used supplementary cementitious materials such as blast furnace slag and fly ash, which are used in various industries including the cement industry, concrete admixtures, and ground solidification materials. However, since their availability is expected to decrease in the future according to the carbon neutrality strategy of each industry, new supplementary cementitious materials should be used to achieve the cement industry's goal for increasing the additive content of Portland cement. Limestone is a material that already has a large amount in the cement industry and has the advantage of high grinding efficiency, so overseas developed countries established Portland limestone cement standards and succeeded in commercialization. This study was an experimental study conducted to evaluate the possibility of utilizing domestic PLC, the effect of fineness and replacement ratio on the physical properties of cement was investigated, and the environmental impact of cement was evaluated by analyzing CO2 emissions.

A representative problem in domestic chestnut industry is the high loss of flesh due to excessive knife peeling in order to increase the peeling rate, resulting in a decrease in production efficiency. In this study, a prediction model for weight loss rate of chestnut by stage of knife peeling process was developed as undergarment study to optimize conditions of the machine. 51 control conditions of the two-stage blade peeler used in the experiment were derived and repeated three times to obtain a total of 153 data. Machine learning(ML) models including artificial neural network (ANN) and random forest (RF) were implemented to predict the weight loss rate by chestnut peel stage (after 1st peeling, 2nd peeling, and after final discharge). The performance of the models were evaluated by calculating the values of coefficient of determination (R), normalized root mean square error (nRMSE), and mean absolute error (MAE). After all peeling stages, RF model have better prediction accuracy with higher R values and low prediction error with lower nRMSE and MAE values, compared to ANN model. The final selected RF prediction model showed excellent performance with insignificant error between the experimental and predicted values. As a result, the proposed model can be useful to set optimum condition of knife peeling for the purpose of minimizing the weight loss of domestic chestnut flesh with maximizing peeling rate.

In recent years, the government has strongly promoted multi-purpose utilization of paddy field. However, poor drainage causes waterlogging stress in upland crops, requiring subsurface drainage technology, resulting in high installation and management costs. To address this issue, a low-cost and high-efficiency technique was developed that utilizes wasted coir substrates which have characteristics of high porosity and good drainage, for upland crop cultivation in paddy fields. Soybeans were grown in both paddy soil and wasted coir slab with two planting densities (80×20 cm and 60×20 cm). The results showed that the coir substrates had better performance than the paddy soil in terms of soil physical and chemical properties and the growth and yield of upland crops are improved. The treatments using wasted coir substrate showed a 41.4% increase in yield and a 21.3% increase in protein content compared to PS treatment. Our findings demonstrate that recycling waste coir substrates to grow upland crops is a positive cultivation strategy to solve some drainage problems in paddy fields. This approach offers a sustainable solution for upland crop production while also addressing the issue of waste management in agriculture.
A proper level of UV-A light treatment in terms of intensity, spectrum, and exposure duration is known to have a positive impact on plant growth, photosynthesis, and the biosynthesis of secondary metabolites. However, there are few studies investigating the physiological responses of spinach (Spinacia oleracea L.) to UV radiation. Hence, this study aimed to assess the effects of short-term UV-A radiation on the growth and bioactive compounds of spinach. Spinach seedlings were cultivated in a vertical farm module under the following environmental conditions: photosynthetic photon flux density 200 µmol·m-2·s-1, white LED, 12 h on/off, 20℃ air temperature, 70% relative humidity, and 500 µmol·mol-1 CO2 concentration. After 5 weeks of sowing, the seedlings were subjected to continuous UV-A (peak wavelength; 385 nm) irradiation at two different energy levels: 20 W·m-2 and 40 W·m-2 for 7 days. As a result, the UV-A20W treatment increased the shoot fresh and dry weights of spinach. However, there were no significant differences observed in photosynthetic parameters between the UV-A treatments and the control. The maximum quantum efficiency of photosystem II (Fv/Fm) consistently decreased across all UV-A treatments for 7 days in UV-A treatments. Additionally, the total phenolic content and antioxidant capacity increased in the UV-A20W treatment at 7 days of treatment as well as the total flavonoid content significantly increased at 5 and 7 days of treatment. These findings suggest that supplemental UV-A LED radiation can enhance the growth and quality of spinach cultivated in closed type plant production systems such as vertical farms.

Recently, active research in Korea and worldwide has begun to focus on gene function and cultivar development using gene editing tools. This research, in addition to studies on edible mushroom, aims to enhance the physical and biochemical characteristics of mushrooms for applications in materials and substance production. For these studies, efficient isolation of protoplasts from the target mushroom is critical. However, several commercial cell wall-lysing enzyme cocktails, including Novozyme234, Glucanex, and Lysing enzymes, have recently been discontinued. In this study, we aimed to identify alternative enzyme systems to replace the discontinued cell wall-lysing enzymes for stable isolation of protoplasts from Ganoderma lucidum. To select an optimal osmotic buffer, enzyme function in 0.6 and 1.2 M Sorbitol, Sucrose, Mannitol, and KCl was assessed. The effect of reaction time was also evaluated. Protoplast isolation efficiency of each alternative enzyme was tested using lysing enzymes from Trichoderma harzianum, Chimax-N, and Yatalase, either individually or in combination. This matrix of studies identified enzymes and optimal conditions that could replace the discontinued lysing enzymes.

In process plant construction, the implementation of design automation technologies is pivotal in reducing the timeframes associated with the design phase and in enabling the generation and evaluation of a variety of design alternatives, thereby facilitating the identification of optimal solutions. These technologies can play a crucial role in ensuring the successful delivery of projects. Previous research in the domain of design automation has primarily focused on parametric design in architectural contexts and on the automation of equipment layout and pipe routing within plant engineering, predominantly employing rule-based algorithms. Nevertheless, these studies are constrained by the limited flexibility of their models, which narrows the scope for generating alternative solutions and complicates the process of exploring comprehensive solutions using nonlinear optimization techniques as the number of design and engineering parameters increases. This research introduces a framework for automating plant design through the use of generative neural network models to overcome these challenges. The framework is applicable to the layout problems of process plants, covering the equipment necessary for production processes and the facilities for essential resources and their interconnections. The development of the proposed Neural-network (NN) based Generative Design Model unfolds in four stages: (a) Rule-based Model Development: This initial phase involves the development of rule-based models for layout generation and evaluation, where the generation model produces layouts based on predefined parameters, and the evaluation model assesses these layouts using various performance metrics. (b) Neural Network Model Development: This phase transitions towards neural network models, establishing a NN-based layout generation model utilizing Generative Adversarial Network (GAN)-based methods and a NN-based layout evaluation model. (c) Model Optimization: The third phase is dedicated to optimizing the models through Bayesian Optimization, aiming to extend the exploration space beyond the limitations of rule-based models. (d) Inverse Design Model Development: The concluding phase employs an inverse design method to merge the generative and evaluative networks, resulting in a model that outputs layout designs to meet specific performance objectives. This study aims to augment the efficiency and effectiveness of the design process in process plant construction, transcending the limitations of conventional rule-based approaches and contributing to the achievement of successful project outcomes.
Two experiments were conducted to compare the effects of the natural and synthetic commercial pigments on the laying Performance, Pigmentation and fatty acid contents in e99 yolk of laying hens. The experimental diets were formulated to have isocalories and isonitrogen. In experiment I, the diet does not contain the com in which the xanthophylls are free, and in experiment II, the diet contained 54% of corn in which the level of xanthophylls are 19.34 ppm/kg (calculated levels). In the experiment 1, 480 ISA Brown laying hens were divided into eight groups. Each group has 60 birds fed the 0% of corn with 8 types of pigment levels for 4 weeks. in the experiment 2, 600 ISA Brown laying hens were divided into ten groups. Each group has 60 birds fed the 54% of com diet with 10 types of pigment levels for 4 weeks. Feed intake, e99 Production, egg weight and feed efficiency did not have significant difference in experiments I and II. Albumen height and haugh unit did not have significant difference in both experiments. In order to approach the yolk pigmentation to 12∼13 of Roche color fan, addition level of natural red pigment was 25∼30 ppm. In the case of synthetic red pigment, the level was 15∼20 ppm. In this experimental condition, the pigmenting effect of the synthetic pigment was better than that of the natural pigment. In the experiment 2, the pigmenting effect of mixing pigments were investigated between TM2 mixed with natural red pigment, and TM6 mixed with synthesis red pigment. The pigmenting effect of synthetic red pigment was greater than that of the natural red pigment. However, the pigmenting effect of natural pigment was greater than that of synthetic pigments when the levels of synthetic and natural red pigments in diet are higher in TM3 and TM8. The fatty acid content in yolk was not affected by pigment addition.
