  • The pine needle gall midge, Therodiplosis japonensis Uchida et Inouye, is the most important insect pest. It requires two different habitats for the development; on trees and under the ground. The habitat specific mortality rates ere $30\~40\%,\;and\;50\~60\%$ for the respective habitats. The key developmental stage is the prepupa, and the key mortality factor is the moisture contents of the soil and its variability. Since the insect is an exotic, the population status is the periphery and in the source of infestation are considerably different. Such a difference in habitats and the population status of the insect should be considered in relation to suppression of the insect. The control strategies should be directional and rational based on the reality of the pest status. There have been substantial information on the control methods of the pine needle gall midge, and each control method has an important place, but none has always provided a satisfactory solution to the many problems associated by this insect. These methods should be applied to a system based on the ecology of the insect. There should be continued support for directed effort on the development of operational management systems for the insect: specifically, estmation of the critical economic injury level, and of the absolute density of the insect.

년생 및 재배조건이 인삼의 엽형에 미치는 영향 (Effects of Plant Age and Cultural Conditions on Leaf Shape of Korean Ginseng)

  • This study was conducted to know the influence of plant age and cultural conditions such as plant population density and light intensity under the shading on the leaf shape of panax ginseng. The result obtained were as follows; 1) Leaf length(L)/maximum width(W) was no difference with different age of over 3-over 3year old plant, but that of 1 or 2-yearold was smaller than those of over 3-year old. The values of L/W showed in the order of 2 or 4,3, 1 or 5 leaflet. 2) Ratios of leaflet area to leaf area were 32.0% in leaflet 3, from 209.% to 27.9% in leaflet 2 or 4, and from 6.5% to 7.1% in leaflet 1 or 5. 3) The coefficients of variability for L/W and ratio of leaflet area to leaf area of leaflet 3 were smallest among leaflets. 4) There were significant differences between largest and smallest leaflet 3, leaf areas and ratio of leaflet 3 area to leaf area in same plant. 5) LW and ratio of leaflet 3 area to leaf area were not affected by plant population density. 6) It showed a tendency that the L/W was increased with increasing the light transmittance rate (LTR). The ratio of leaflet 3 area to leaf area of ginseng grown under 20% LTR was not different comparing to that of plant grown under 5% LTR, but it was significantly increased in plant grown at 100% LTR.

Genetic structure of Larimichthys polyactis (Pisces: Sciaenidae) in the Yellow and East China Seas inferred from microsatellite and mitochondrial DNA analyses

  • Genetic variation was surveyed at four microsatellite loci and 1416 base pairs (bp) of the mitochondrial DNA (mtDNA) cytochrome c oxidase I gene (COI) to clarify the genetic structure of the small yellow croaker, Larimichthys polyactis, in the Yellow and East China Seas, especially regarding four provisional populations, (one Korean and three Chinese populations). Based on microsatellite DNA variations, the estimated expected heterozygosity ($H_E$) in each population ranged from 0.776 to 0.947. The microsatellite pairwise $F_{ST}$ estimates showed no significant genetic differentiation between the populations. MtDNA variations also indicated no genetic structure in L. polyactis, but very high variability. The absence of genetic differentiation among and within populations of L. polyactis may either result from the random migration of the adult or the passive dispersal of the eggs and larvae.

대두 양적형질의 유전적 변이와 선발(I) (Genetic Variability of Important Quantitative Characters and Selection for Yield in Soybean (I))

  • 본 실험은 대두육종을 위한 유전자원으로서 우리나라 재래종 825계통을 대상으로 2년간 재배하면서 그 집단내의 유전적 변이성과 특성을 구명하고 이들을 이용한 다수확 품종의 육성에 필요한 정보를 획득하는데 목적이 있으며 그 결과는 다음과 같다. 1. 단위면적당 평균 수량은 1,004.1kg/ha이고 대부분의 계통이 795kg에서 1,245kg의 수량성을 보였으며 백립중은 8.6gr∼44.4gr의 범위에서 평균 23.0gr이였다. 성숙기는 평균 144.7일로써 대부분의 계통이 만숙성이였고 초장 및 주당협수의 변이폭은 비교적 컸다. 2. 단백질함량은 평균 42.83%이고 40.7%∼44.2%, 지방함량은 평균 17.46%로서 전체계통의 70%가 16.5%∼19.0% 범위에 속해 있어 저지방성임을 나타내었다. 3. 유전분산은 수량과 초장 및 주당협수가 컸고 백립중 및 성숙기도 비교적 컸으나 단백질과 지방함량은 적은 유전분산을 보였다. 4. 백립중, 초장 및 주당협수의 유전력과 선발에 대한 기대치는 비교적 높았으며 성숙기의 유전력은 높았으나 선발효율은 낮았는데 그 원인은 재래종 집단의 표현형분산이 적은데 있다고 할 수 있다.

Evaluation of clinical usefulness of reference interval of some selected hematological parameters in canine blood

  • To estimate the source of variance components for some hematological parameters and assess the utility of the conventional population-based reference interval, this study computed index of individuality for blood samples, which were from 13 dogs drawn once weekly for 4 consecutive weeks. Results were subjected to nested analysis of variance. For all parameters measured between-dog variations were greater than within-dog variation. Except for the parameters RBC and MCHC the index of individuality was <1.4. The low reliability coefficient and high index of individuality of ${\leq}0.8$ were found for the majority of hematological parameters. In practical term, the present study indicated that use of hemogram profiles alone in the evaluating clinical state of a single patient should be avoided because of their physiological or natural random variations, and that comparing a single measurement on the blood analytes from an individual dog to the conventional population-based reference range may be too insensitive to detect any significant changes in the blood components of that particular dog. A single measurement may not characterize an individual's average concentration of the parameters even shortterm period.

한 자동차공장의 1주연속 12시간주야맞교대근무 노동자들의 심박동수변이 (Circadian Disruptions of Heart rate Variability among Weekly Consecutive-12-hour 2 Shift Workers in the Automobile Factory in Korea)

  • Objectives : The objective of this study is to compare the circadian patterns of heart rate variability assessed by 24-hour ambulatory electrocardiographic (ECG) recordings during day shift and night shift among the workers in the 5 days-concecutive-12-hour shift in an automobile factory in Korea. Methods : The study population consisted 300 workers, who were randomly selected among the 8700 total workers in one car factory. To analyse circadian variation, the 24-hour ECG recordings (Marquette) were measured during day shift (08:00-20:00 h) and night shift (20:00-08:00 h). Analysis was performed for all time and frequency domain measures of HRV. 233 workers completed taking 24-hour ECG recordings. Results : This study shows that the 24 hourcircadian variation mainly follows work/sleep cycle rather than day/night cycle among shift workers. This study also shows that among the night shift, the circadian variation between work and sleep cycle decreased compared to the work/sleep cycle among day shift workers. All time and frequency domain parameters (except LF/HF ratio) show significantly different between work and sleep in the day shift and night shift. Conclusion : These changes in heart rate variability circadian rhythms reflect significant reductions in cardiac parasympathetic activity with the most marked reduction in normal vagal activity among the shift workers. Especially, it suggests the circadian rhytm has blunted among the night workers. The quantification of the circadian variation in HRV can be a surrogates of workers' potential health risk, as well as suggests possible mechanisms through which the shift works compromise workers' health.

Experimental Infection of Different Tomato Genotypes with Tomato mosaic virus Led to a Low Viral Population Heterogeneity in the Capsid Protein Encoding Region

  • The complete genome sequence of a Slovak SL-1 isolate of Tomato mosaic virus (ToMV) was determined from the next generation sequencing (NGS) data, further confirming a limited sequence divergence in this tobamovirus species. Tomato genotypes Monalbo, Mobaci and Moperou, respectively carrying the susceptible tm-2 allele or the Tm-1 and Tm-2 resistant alleles, were tested for their susceptibility to ToMV SL-1. Although the three tomato genotypes accumulated ToMV SL-1 to similar amounts as judged by semiquantitative DAS-ELISA, they showed variations in the rate of infection and symptomatology. Possible differences in the intra-isolate variability and polymorphism between viral populations propagating in these tomato genotypes were evaluated by analysis of the capsid protein (CP) encoding region. Irrespective of genotype infected, the intra-isolate haplotype structure showed the presence of the same highly dominant CP sequence and the low level of population diversity (0.08-0.19%). Our results suggest that ToMV CP encoding sequence is relatively stable in the viral population during its replication in vivo and provides further demonstration that RNA viruses may show high sequence stability, probably as a result of purifying selection.

Intron sequence diversity of the asian cavity-nesting honey bee, Apis cerana (Hymenoptera: Apidae)

  • The Asian cavity-nesting honeybee, Apis cerana (Hymenoptera: Apidae), has been extensively studied for its biogeography and genetic diversity, but the molecules utilized in past studies were mainly ~90 bp long mitochondrial non-coding sequences, located between $tRNA^{Leu}$ and COII. Thus, additional molecular markers may enrich our understanding of the biogeography and genetic diversity of this valuable bee species. In this study, we reviewed the public genome database to find introns of cDNA sequences, with the assumption that these introns may have less evolutionary constraints. The six introns selected were subjected to preliminary tests. Thereafter, two introns, titled White gene and MRJP9 gene, were selected. Sequencing of 552 clones from 184 individual bees showed a total of 222 and 141 sequence types in the White gene and MRJP9 gene introns, respectively. The sequence divergence ranged from 0.6% to 7.9% and from 0.26% to 17.6% in the White gene and the MRJP9 introns, respectively, indicating higher sequence divergence in both introns. Analysis of population genetic diversity for 16 populations originating from Korea, China, Vietnam, and Thailand shows that nucleotide diversity (π) ranges from 0.003117 to 0.025837 and from 0.016541 to 0.052468 in the White gene and MRJP9 introns, respectively. The highest π was found in a Vietnamese population for both intron sequences, whereas the nine Korean populations showed moderate to low sequence divergence. Considering the variability and diversity, these intron sequences can be useful as non-mitochondrial DNA-based molecular markers for future studies of population genetics.

Molecular Characterization of Hallikar Breed of Cattle Using Microsatellite Markers

  • Molecular characterization of Hallikar, the native cattle breed of Karnataka, was undertaken using 19 cattle specific, highly polymorphic microsatellite markers recommended by FAO. The genomic DNA was subjected to PCR amplification and alleles were resolved through six per cent denaturing PAGE with a 10 bp DNA ladder followed by silver staining. Genotyping of animals was done based on allele size. The number of alleles ranged from three to nine with allele sizes ranging from 102 bp to 294 bp. These alleles were distributed in the frequency range between 0.0306 and 0.8673 in the population. The mean observed number of alleles was $6.368{\pm}1.4225$. The mean observed and expected heterozygosities were $0.7515{\pm}0.1734$ and $0.7850{\pm}0.1381$, respectively. The high heterozygosity observed implies presence of higher genetic variability within Hallikar breed. The PIC (Polymorphism Information Content) values ranged from 0.2322 (ETH152) to 0.8654 (ETH225). The percentage of polymorphic loci obtained was 100 as all the 19 microsatellite markers were found to be polymorphic. Except for ETH152, all the other loci had high PIC values, indicating that these markers are highly informative for characterization of Hallikar breed. The population was tested for Hardy-Weinberg equilibrium at 19 microsatellite loci, and at 74 per cent of the loci the population was found to be in disequilibrium.

RAPD에 의한 지리산 내 산거울 집단의 공간적 상관관계 분석 (Spatial Autocorrelation Analysis of Carex humilis on Mt. Giri by RAPD)

  • RAPD에 의한 지리산 내 산거울 집단의 유전자 빈도와 지리적 거리에 따른 공간적 상관관계를 분석하였다. 전체 102 DNA 분절(밴드)이 107 개체에서 탐지되었다. 102 밴드 중 48(47.1%)개 밴드는 다형성을 나타내었다. 분집단간 다형성의 비교에서 거리 구간 I와 V가 가장 낮은 변이(16.7%)를 나타내었고, 거리 구간 VIII이 가장 높은 변이를 나타내었다(22.6%). 전체 다양도는 0.076이었다. 구간 VIII이 가장 높은 다양도(0.093)를 나타내었고, 구간 I가 가장 낮았다(0.063). 구간 사이의 유전적 유사도는 60 m 거리까지는 유사하였다. 지리산 집단에서 산거울은 강한 공간구조를 나타내고 있음이 RAPD 마커로 알 수 있었다. 이는 지리산 집단에서 낮은 이주자수와 개체들이 덩어리 모양의 분포를 나타내기 때문으로 판단된다. 본 연구에서 RAPD 마커로 산거울의 공간구조와 유전적 구조를 파악하는데 유용하게 이용될 수 있음을 입증하였다.
