Purpose : Leptin and adiponectin are two representative adipocytokines. Leptin increases, but adiponectin decreases, with obesity and insulin resistance. We aimed to study the relationship between the leptin/adiponectin ratio and insulin resistance in healthy children. Methods : Seventy-seven healthy children (36 boys and 41 girls) were enrolled in this study. Anthropometric measurements were performed, and the percentage of weight for height (%WFH) was calculated in each subject. Fasting plasma levels of glucose, insulin, leptin, adiponectin, testosterone, estradiol, and sex-hormone binding globulin (SHBG) were measured. The free androgen index (FAI) was used as a representative of testosterone bioactivity. The homeostasis model assessment was used to estimate the degree of insulin resistance (HOMA-IR). Results : In the boys, HOMA-IR was significantly correlated with age, pubertal stage, free androgen index (FAI), leptin, and the leptin/adiponectin ratio. HOMA-IR was also significantly related to age, percentage of weight for height (%WFH), pubertal stage, estradiol, leptin, and the leptin/adiponectin ratio in girls. The leptin/adiponectin ratio was independently related to HOMA-IR after adjusting for age, %WFH, and FAI in the boys (P<0.05). The leptin/adiponectin ratio was not independently related to HOMA-IR after adjusting for age, %WFH, and estradiol in girls. Conclusion : In non-obese healthy children, the leptin/adiponectin ratio was significantly correlated with insulin resistance. The leptin/adiponectin ratio was independently related to insulin resistance even after adjusting for age, degree of obesity, and androgen levels in healthy boys.

The present study was carried out to investigate the potential toxicity of highly-developed rice varieties, Oryza sativa cvs. Jukjinjubyeo, Heuginjubyeo and Heughyangbyeo by a single oral dose in ICR mice. The test article was orally administered once by gavage at dose levels of 0, 2.5, 5.0, and 10.0 g/kg body weight (n=10 for male and female mice for each dose). We examined the number of deaths, general clinical signs, body weights and biochemical analysis for sexes and doses of mice between control and experimental groups. As a result all of the groups were alive during experimental periods and none of the significant clinical symptom and body weight changes were shown in all of the groups. Plasma glucose levels were valued for both control and experimental groups and there were no significant differences between the groups. The activities of aminotransferase were not increased in all experimental groups compared to control groups. The results suggest that the toxicity of the aleurone layer of Oryza sativa cvs. Jukjinjubyeo, Heugjinjubyeo and Heughyangbyeo are low and its $LD_{50}$ is over 10.0 g/kg body weight in both male and female mice.
An experiment was conducted to determine the effects of two levels of rumen undegradable protein (RUP) without or with whole or extruded flaxseed on milk yield, milk component, milk fatty acids (FAs) profile and plasma metabolites in transition ewes. Three weeks before and after lambing, seventy-two Baluchi ewes were used in a completely randomized design with a 3 × 2 factorial arrangement of treatments. The treatments contained 1) no flaxseed + 20% RUP (no flaxseed, low RUP [NFLR]); 2) no flaxseed + 40% RUP (no flaxseed, high RUP [NFHR]); 3) 10% whole flaxseed + 20% RUP (whole flaxseed, low RUP [WFLR]); 4) 10% whole flaxseed + 40% RUP (whole flaxseed, high RUP [WFHR]); 5) 10% extruded flaxseed + 20% RUP (extruded flaxseed, low RUP [EFLR]), and 6) 10% extruded flaxseed + 40% RUP (extruded flaxseed, high RUP [EFHR]). Ewes fed 10% extruded flaxseed exhibited higher (p < 0.001) dry matter intake (DMI) and colostrum yield (p < 0.1) compared to other treatments. Two types of flaxseed and RUP levels had no significant effect on milk yield, but milk fat and protein contents decreased and increased in diets containing 40% RUP, respectively. Ewes fed extruded flaxseed produced milk with lower concentrations of saturated fatty acids (SFA) and higher α-linolenic and linoleic acids and also polyunsaturated fatty acids (PUFA) compared to other groups (p < 0.05). During post-lambing, the ewes fed diets containing flaxseed exhibited higher concentration of serum non-esterified FAs (NEFA) compared to diets without flaxseed (p < 0.01). The concentration of serum β-hydroxybutyric acid (BHBA) decreased in the diets containing flaxseed types at pre-lambing, but increased in diets containing extruded flaxseed at post-lambing (p < 0.01). The serum glucose concentration of ewes (pre and post-lambing) which consumed diets containing extruded flaxseed or 40% RUP increased, but blood urea concentration was elevated following supplementation of diet with whole flaxseed or 40% RUP (p < 0.001). In conclusion, utilization of 10% extruded flaxseed in the diets of transition ewes had positive effects on animal performance with favorable changes in milk FAs profile. However, there is no considerable advantage to supply more than 20% RUP level in the diet of transition dairy sheep.

The present study was aimed at investigating effects of chicory inulin and three kinds of oligosaccharides on lipid metabolism in rats fed a high-cholesterol diet. Nine Sprague-Dawley male rats weighing, about 190g were given one of five experimental diets, which were basal cholesterol diet (Control) isomaltooligosaccharide diet (IMO), Iructooligosaccharide diet (FO), chicory inulooligosaccharide diet (CIO) and chicory inulin diet (CI) for 5 weeks. In the oligosaccharide and inulin diets, 6% was added at the expense of sucrose. Rats were pair-fed to the intake of FO group which consumed the least amount, and their feces were collected during the last 4 days. Body weight gain was lower in Fo and CI groups compared with the Control group. Plasma glucose levels of FO and CIO groups were lower and plasma triglyceride concentrations of FO, CIO, and CI groups were lower than those of IMO group. Plasma cholesterol concentration did not differ among groups. Relative liver weight was lower in CIO group. Hepatic triglyceride and cholesterol did not differ among. groups. Fecal excretion of neutral steroid and bile acid were not different among groups, but fecal triglyceride excretion was significantly increased in FO and CI groups compared with the Control group. In conclusion, supplementation of oligosaccharides and chicory inulin at 6% of diets showed no significant hypolipidemic effect in rats fed a high cholesterol diet.

Comparison of blood properties in juvenile black seabream (Acanthopans schlegeli) between transfer from seawater to freshwater (sFW) and transfer from freshwater to seawater (fSW) were investigated for 60 days. Plasma cortisol levels in sFW were significantly increased from 34.2 $\pm$ 28.6 ng/mL at the beginning to 365.6 $\pm$ 136.0 ng/mL at 3 hours, and decreased to the beginning level at 24 hours. However, in fSW, no significant differences in the cortisol levels were recognized throughout experimental period. No significant difference was found in the glucose levels between sFW and fSW. The transfer from seawater to freshwater of juvenile black seabream resulted in reduced $Na^{+}\;and\; Cl^{-}$ concentrations for 24 hours (decreasing from 165.3 $\pm$ 2.5, 146.0$\pm$ 2.2 to 118.3 $\pm$ 12.3, 78.0$\pm$ 7.0 mEq/L, respectively), but these were completely recovered the beginning levels at 30 days. Total protein and AST showed no significant differences between the two rearing conditions, while ALT was markedly elevated at 3 hour in sFW and at 24 hours in fSW. Ht in sFW was increased from 18.5 $\pm$$0.6\%$ at the beginning to 25.3 $\pm$$4.0\%$ at 12 hours, and was decreased to the beginning level at 24 hours. Ht, RBC and Hb in fSW were also significantly higher at 12 hours, but recovered to their initial levels at 24 hours. All fish were dead until 50 days in sFW while survival rate in fSW was $85\%$ at the end of experiment.

This study was performed to investigate the anti-diabetic mechanism of crude polysaccharides isolated from the fruiting bodies of Grifola frondosa. We treated 3T3-L1 adipocyte cells to observe whether the crude polysaccharides isolated from Grifola frondosa would stimulate insulin sensitivity. Significant insulin sensitizing activity was observed in the 3T3-L1 adipocytes, and giving the crude polysaccharide of Grifola frondosa with 1 nM of insulin caused glucose uptake to increase to a similar level as giving 50 nM of insulin alone. To confirm the mechanism for the anti-diabetic effect of the crude polysaccharides, we performed further examinations within $KK-A^{y}$ mice, an animal model of type 2 diabetes. The crude polysaccharides reduced blood glucose levels in the $KK-A^{y}$ mice for 2 weeks after feeding, and also significantly lowered plasma insulin levels. These results suggest that the anti-diabetic mechanism of the crude polysaccharide of Grifola frondosa is related to the enhancement of insulin sensitivity.

This study was conducted to investigate the effects of Kluyveromyces fragilis, Candida utilis and brewer's yeast as an additive in formulated diets containing different fish meal levels ($55{\%} and 45{\%}$) on growth and body composition of juvenile Korean rockfish (Sebastes schlegeli). Juvenile rockfish (average weight, 2.6 g) were distributed into 24 tanks with 3 replication groups and fed eight isoproteic ($41{\%}$) and isolipidic ($11{\%}$) diets containing each level ($3{\%}$) of K. frayilis, C. utilis and brewer's yeast as an additive. Survival rate of fish was not affected by dietary fish meal level and yeast kind (P > 0.05). Weight gain of fish fed the control and diets containing C. utilis and brewer's yeast was not influenced by fish meal level (P > 0.05), Weight gain of fish fed the $55{\%}$ fish meal diet containing C. utilis and $45{\%}$ fish meal diets containing K. fragilis and C. utilis was lower than that of fish fed the control diet (P < 0.05). Feed efficiency and protein efficiency ratio of fish fed the control and diet containing brewer's yeast were not affected by fish meal level, but these value of fish fed the diets containing K, fragilis and C. utilis were lower in $45{\%}$ fish meal diets than in $55{\%}$ fish meal diets. Hepatosomatic index and liver lipid content of fish fed the diet containing C. utilis were decreased regardless of fish meal level (P < 0.05), Hemoglobin, hematocrit, red blood cell, plasma total protein and plasma total glucose were not affected by dietary fish meal levels and yeast supplements (P > 0.05). From the above results, it can be concluded that yeast supplement in this formulated diet for growth of juvenile rockfish is not necessary.
The current study was designed to define whether a blend of prunus mume extract(25%) containing lactic acid(75%) and grape seed extract(10ppm) could affect in vitro antimicrobial activity and growth performance, intestinal microflora, plasma biochemical profiles and digestive enzymes activities in broiler chickens. In paper disc agar diffusion test, we clearly observed antimicrobial activity against E. coli in response to prunus mume extract or a blend of prunus mume extract. For in vivo test, a total of ninety six 3-d-old male broiler chicks were assigned to basal diet(CON), basal diet supplemented with antibiotics (ANTI) and 0.5% a blend of prunus mume extract(PRNUS) until 35 days of age. Throughout the entire experimental period(3-35 days), there were no differences in BW and FCR between the birds fed the basal diet with antibiotics and the diet supplemented with a blend of prunus mume. However, ANTI group showed a significant increase in BW and total gain compared to CON group. The weights of digestive organs such as the pancreas and mucosal tissues were not affected by dietary treatments. There was no difference in plasma levels of glucose, cholesterol, AST and ALT activity. However, triglyceride in plasma increased(P<0.05) in the birds fed the diet supplemented with 0.5% a blend of prunus mume extract compared to those fed antibiotics supplemented diet. The activities of pancreatic trypsin and amylase, and intestinal hydrolase including disaccharidase were not affected by dietary treatment. The colony forming units(CFU) of lactobacillus in the lower ileal-cecum of the birds fed the diet supplemented with a blend of prunus mume extract was significantly(P<0.05) higher than that of birds fed antibiotic supplemented diet without affecting the CFU of E. coli. In conclusion, the birds fed the diet supplemented a blend of prunus mume as an alternative to antibiotics showed a similar growth performance and an significant increase in lactobacillus population compared with the birds fed basal and antibiotics supplemented diets.

Background: Measurement of HbA1c levels is widely used to diagnose diabetes mellitus and to evaluate and monitor plasma-glucose concentrations over 6-8 weeks. In this study, we evaluated the diagnostic performance of the newly developed latex immunoturbidimetric method by using Autolab HbA1c. Methods: We analyzed and compared the diagnostic performance of Autolab HbA1c with that of Toshiba 200FR between April 2009 and July 2009. According to guidelines (EP5-A2, EP6-P, EP9-A2) of the clinical and laboratory standards institute (CLSI), we compared linearity, precision and correlation of Autolab HbA1c with those of G7 (Tosoh Corp., Kyoto, Japan) by using high-performance liquid chromatography (HPLC) method. Results: Data obtained using Autolab HbA1c showed good linearity in mixtures of samples with low (3.1%) and high (15.1%) levels of HbA1c (r2=0.9997). In the analysis of within-run precision of the samples with HbA1c levels of 5.1% and 12.1%, the SDs were 0.04 and 0.06 and covariances of these samples were 0.8% and 0.5%, respectively. In the Deming regression model, the regression equation was as follows: Autolab HbA1c=1.0859×Tosoh HPLC-0.6957. Conclusions: In this study, Autolab HbA1c method showed better performance characteristics than Tosoh G7 did. In reference review, there was no interference of variant hemoglobin. The data acquisition time of Autolab HbA1c was lower than that of Tosoh G7. The advantages of Autolab HbA1c are that it can be used as an autoanlyzer in routine chemical analysis, it does not require pre-analytical treatment, and the samples are automatically treated with distilled water for hemolysis.

The effects of Bulnesia sarmienti water extract on lipid metabolism were investigated in type-2 diabetes rats. The rats were fed a basal diet and B. sarmienti water extract for 8 weeks. The rats were randomly assigned to each treatment group : normal, control, two kinds of treatment group (BW1: 0.9 mg/ml, BW2: 1.8 mg/ml). After 8 weeks of experimental diets consumption, the body weights and food efficiency ratio of B. sarmienti water extract fed groups were not different from those in the control group. The concentrations of total-cholesterol, LDL-cholesterol, and triglyceride in blood plasma were significantly decreased in the B. sarmienti water extract groups comparison to the control group. Although blood glucose control effects were not observed, the B. sarmienti water extract feeds were highly effective in lowering the levels of blood lipid substances such as triglycerides, total-cholesterol, and LDL-cholesterol.
