Plant parasitic nematodes, Meloidogyne and Pratylenchus spp., are mostly detected in imported bulbs and tubers including a ginger, Zingiber officinale in Korea by quarantine inspection. However, there is little information on hot water immersion treatment (HWIT) for control of exotic nematodes, which induce economic loss by discard or send back to exporter, in imported gingers. In here, we determined that mortality of two plant parasitic nematodes and thermal stability of ginger. Meloidogyne and Pratylenchus spp. were completely killed at $48^{\circ}C$ and $49^{\circ}C$ for 30 sec by HWIT. Thermal conduction of Z. officinale to reach a target temperature as $50^{\circ}C$ take 10~32 min and 6~16 min for core and inner 5 mm region from surface, respectively. When ginger exposed at $51^{\circ}C$ for 30 min, growth of Z. officinale was not affected by heat treatment compared with control. Based on these results, HWIT at $51^{\circ}C$ for 30 min completely killed artificially infected juveniles of Meloidogyne spp. in Z. officinale. Therefore, this condition for HWIT will be used as fundamental information on phytosanitory to kill two plant parasitic nematodes without damage on ginger.
The white soybean cyst nematode Heterodera sojae, isolated from the roots of soybean in Korea, is widespread in most provinces of the country and has the potential to be as harmful to soybean as H. glycines. Determining the virulence phenotypes of H. sojae is essential to devising management strategies that use resistant cultivars. Consequently, virulence phenotypes of 15 H. sojae populations from Korea were determined on seven soybean lines and one susceptible check variety. Two different HS types were found to be present in Korea; the more common HS type 2.5.7, comprising 73.3% of the H. sojae populations and the less common HS type 0, constituting only 26.7% of the tested populations. Considering the high frequency of H. sojae adaptation to soybean indicator lines, the PI 88788 group may not be a possible source of resistance while PI 548402, PI 90763, PI 437654, and PI 89772 can be used as resistance sources for soybean breeding programs aimed at developing H. sojae-resistant soybean cultivars in Korea.
The distribution and form of pine wood nematode (PWN) were investigated in Jinju-si and Sacheon-si areas, where infested with pine wilt disease (PWD). The average PWN population per g of the tubulation part in the wilted Japanese balck pine were 381.2 and 341 in Jinju-si and Sacheon-si, respectively. The PWN population per g of affected Japanese black pine with wilt rate below 60% were 556 and 518 in Jinju-si and Sacheon-si, respectively. The ratios of PWN and other plant-parasitic nematode in the wilted Japanse black pines were 48% vs. 52% in Jinju-si and 53% vs. 47% in Sacheon-si after exposure to PWD for 1 year. The survivorship of pine wood nematode in the sampled trees after 1 year was 0-10% in the Jinju-si area and 5-20% detected in the Sacheon-si area. In the forest areas affected by PWN, the highest survival rates were 37.2% and 39.8% at 25 cm diameter at breast height (DBH) in Jinju-si and Sacehon-si, respectively, while the highest wilt rates were 30.5% and 28.3% at 30 cm DBH in Jinju-si and Sacehon-si, respectively.
Fifty-two nematode predatory fungi were isolated from 37 soil samples collected from eight provinces in Korea. Isolated fungi were tested their predacity against Rhabditis sp. and Meloidogyne incognita in petri dish, and against M. incognita in greenhouse pot experiments. Fifty isolates had trapping organ of adhesive networks and two isolates had adhesive column or adhesive knob. In petri dish experiments, 5 1 isolates against Rhubditis sp. and 26 isolates against M. incognita showed over 91 % of predacity; in greenhouse experiments, however, only three isolates showed over 81% of predacity. These results imply that the results from the laboratory experiments are not consistent with those from the greenhouse experiments. Therefore, to select a promising biocontrol predatory fungi for plant-parasitic nematodes, the screening experiment should be conducted in conditions close to nature.
A survey on the geographical distribution of two soybean cyst nematodes, Heterodera glycines and Heterodera sojae, was conducted in soybean cultivation areas in the Gangwon-do and Chungcheongbuk-do provinces. In 67 fields of 10 counties or cities in Gangwon-do province, H. glycines was detected in 13 fields, H. sojae was detected in five fields, and both species were found in two fields. In 84 fields of nine counties or cities in Chungcheongbuk-do province, H. glycines was detected in 27 fields, H. sojae was detected in four fields, and both species were found in five fields. The diversity and dominance of the plant parasitic nematodes in the Gangwon and Chungbuk provinces were not significantly different among the soybean cultivars. The diversity and dominance index of the plant parasitic nematodes according to the presence or absence of soybean cyst nematodes did not show any significant difference between the soybean cultivation areas of both provinces. The relationship between the soybean cyst nematodes and soil chemical properties showed a low coefficient of determination, and the density of soybean cyst nematodes tended to increase as the pH and available phosphate content in soil samples increased. The soil texture of cyst nematode-detected fields was mostly sandy soils, which amounted to about 60% of the total. As the clay and silt content increased, the density of soybean cyst nematodes tended to decrease, and as the sand content increased, the density of cyst nematodes tended to increase.
Clover cyst nematode, Heterodera trifolii, is one of the most important plant-parasitic nematode on Kimchi-cabbage in highland in Korea. Recently, a demand of soil disinfection machine for fumigants is increasing. This study was conducted to assess the control effects of a newly developed prototype soil disinfection machine to the cyst nematode. Two fumigants, dimethyl disulfide (DMDS) and metam sodium (MNa), were treated using the machine in a field, which was infected with the cyst nematode. After 4 weeks, control effects of fumigants were assessed as eggs hatching rates inside of cysts extracted from the soil, and as a number of females reproduced in roots of Kimchi-cabbage. DMDS (39 l/10 a) suppressed over 99.0% of the eggs hatching rate and the number of females reproduced. On the other hand, MNa (29 l/10 a) controlled the egg hatching rates from minimum 78.3% to maximum 99.4%, and the number of females from 34.7% to 49.3%. The control effects of two fumigants to clover cyst nematode by treated depth were no significant differences statistically. These results showed that DMDS treatment using the soil disinfection machine was expected to have the control effects for the clover cyst nematode.
Sweetpotato (Ipomoea batatas [L.]) is a globally important root crop cultivated for food and industrial processes. The crop is susceptible to the root-knot nematode (RKN) Meloidogyne incognita, a major plant-parasitic RKN that reduces the yield and quality of sweetpotato. Previous transcriptomic and proteomic analyses identified several genes that displayed differential expression patterns in susceptible and resistant cultivars in response to M. incognita infection. Among these, several sporamin genes were identified for RKN resilience. Sporamin is a storage protein primarily found in sweetpotato and morning glory (Ipomoea nil). In this study, transcriptional analysis was employed to investigate the role of sporamin genes in the defense response of sweetpotato against RKN infection in three susceptible and three resistant cultivars. Twenty-three sporamin genes were identified in sweetpotato and classified as group A or group B sporamin genes based on comparisons with characterized sweetpotato and Japanese morning glory sporamins. Two group A sporamin genes showed significantly elevated levels of expression in resistant but not in susceptible cultivars. These results suggest that the elevated expression of specific sporamin genes may play a crucial role in protecting sweetpotato roots from RKN infection.
The study was conducted to investigate the infestation and distribution of plant-parasitic nematodes on Chinese yam (Dioscorea batatas) in Andong, Korea. Root-knot nematodes, Meloidogyne incognita was the most prevalent nematode species which is found from 43 yam fields (81.1%) with high population densities (average of 450 juveniles/$300cm^3$). Other nematodes, root-lesion nematode (Pratylenchus spp.) and pin nematode (Paratylenchus spp.) were less problematic. Density of juveniles (J2) of M. incognita was $10\;J2/300cm^3$ by August, then increased to 274 in September and 624 in October. The highest J2 density was found at the soil depth between 40 and 50 cm ($1,840\;J2/300cm^3$). M. incognita was able to infest yam tuber down to a depth of 70 cm and developed galls outside and brown spots inside. The highest number of females were found at 40-50 cm (79 females/10 cm piece) tuber from the top.
This study was conducted to investigate the plant-parasitic nematodes of ornamental foliage plants, citrus orchards, and tea plantations from July to December 2022. As a result of the investigation of plant-parasitic nematodes in 415 foliage plants, root-lesion nematodes were detected most frequently, followed by root-knot nematodes, pin nematodes, and other nematodes. In tea plantations, spiral nematodes, cyst nematodes, and root-knot nematodes were detected. Citrus nematodes, ring nematodes, and root-knot nematodes were discovered in citrus orchards. In foliage plants, tea plantations, and citrus orchards, the detection rate of plant-parasitic nematodes was not high, but root-lesion nematodes were detected. Therefore, it is necessary to apply appropriate control methods to manage root-lesion nematodes during the cultivation of foliage, tea, and citrus plants.

Serine proteases form one of the most important families of enzymes and perform significant functions in a broad range of biological processes, such as intra- and extracellular protein metabolism, digestion, blood coagulation, regulation of development, and fertilization. A number of serine proteases have been identified in parasitic helminths that have putative roles in parasite development and nutrition, host tissues and cell invasion, anticoagulation, and immune evasion. In this review, we described the serine proteases that have been identified in parasitic helminths, including nematodes (Trichinella spiralis, T. pseudospiralis, Trichuris muris, Anisakis simplex, Ascaris suum, Onchocerca volvulus, O. lienalis, Brugia malayi, Ancylostoma caninum, and Steinernema carpocapsae), cestodes (Spirometra mansoni, Echinococcus granulosus, and Schistocephalus solidus), and trematodes (Fasciola hepatica, F. gigantica, and Schistosoma mansoni). Moreover, the possible biological functions of these serine proteases in the endogenous biological phenomena of these parasites and in the host-parasite interaction were also discussed.
