연초의 근권환경이 뿌리와 지상부의 생육에 미치는 영향 (The effect of root zone environment on the growth of shoot and root of tobacco plant)

  • This experiment was conducted to investigate the effect of growth medium in pots composed of upland soil(S), rice straw manure(M), carbonated rice hull(CRH), and their mixture on growth of tobacco cv. NC 82. The growth of shoot and root was vigorous in order of medium S+M+CRH>M>CRH>S. In M+S medium, root growth in the part of manure was superior to upland soil. But root growth of upland soil part in M+S plot was more vigorous than that in upland soil only. It is possibly due to be influenced by manure in M+S plot. Total length and weight of root, number of roots, and especially for development of adventitious root were closely related to shoot growth. Roots grown in upland soil part was brownish gray in color, while the roots in manure part was milky white. The milky white colored roots had longer life than others. It was concluded that root zone environments derived from several media in pots closely related to shoot growth and disease tolerance of tobacco plant.

수입 오렌지로부터 갈색썩음병균(Phytophthora citrophthora)의 분리 및 동정 (Isolation and Identification of Phytophthora citrophthora from Imported Orange Fruits)

  • Occurrence of brown rot was observed on imported orange fruits, and irregular brown spots were appeared as an intial symptom. White colored mycelia were developed rapidly on the surface of the fruits under the humid condition and resulted in rotting of the fruits. The causal organism of brown rot of orange was identified as Phytophthora citrophthora. Most sporangia were nondeciduous, papillate, ovoid, and measured 25.5~51.5$\times$17.9~34.7 ${\mu}{\textrm}{m}$ in size. P. citrophtora was pathogenic on Satsuma mandarin as well as orange. Symptoms were produced 3~7 days after artificial inoculation of sporangia and mycelia mixture on the fruits of orange and Satsuma mandarin, which were similar to those on the naturally infected fruits.

수수꽃다리 잎중의 다가 알코올과 유리 환원당의 소장에 관한 연구 (Studies on the Seasonal Variation of the Polyalcohols and the Free Reducing Sugars in the Leaves of Syringa dilatata $N_{AKAI}$)

  • One of the indigeneous plants to this country, Syringa dilatata $N_{AKAI}$ (Oleaceae) is known in commerce as 'Ya-Jung-Hyang' (野丁香) and has been known to be of effect as bitter stomachics. Seasonal variations in the content of polyalcohols and free reducing sugars in leaves of this plant which contains syringin glycoside, mannitol and free reducing sugars etc. were studied. Application of chromotropic acid to formaldehyde which was obtained from polyalcohols and reducing sugars by treatment with periodic acid results in reddish violet coloration and the solution has absorption maximum at wave length $570m\;{\mu}$. By the use of ionic exchange resin chromatography, poyalcohols were separated from the above mixture. The content of polyalcohols of this plant was plentiful in the growing season while that of free reducing sugars was decreasing vice versa.

  • Containment Filtered Vent Systems (CFVSs) have been mainly equipped in nuclear power plants in Europe and Canada for the controlled depressurization of the containment atmosphere under severe accident conditions. This is to keep the containment integrity against overpressure during the course of a severe accident, in which the radioactive gas-steam mixture from the containment is discharged into a system designed to remove the radionuclides. In Korea, a CFVS was first introduced in the Wolsong unit-1 nuclear power plant as a mitigation measure to deal with the threat of over pressurization, following post-Fukushima action items. In this paper, the overall features of a CFVS installation such as risk assessments, an evaluation of the performance requirements, and a determination of the optimal operating strategies are analyzed for the Wolsong unit 1 nuclear power plant using a severe accident analysis computer code, ISAAC.

  • Antileukemic tropoloisoquinoline alkaloids, pareirubine A (1) and gandirubrine (2), at first have been isolated from.Abuta concolor (Menisper-maceae). Methylation of 1 and 2, existing in solution as a mixture of tautomersgave the corresponding four methyl derivatives (3 -6). Thioimerubrine (7) andthioisoimerubrine (8) were prepared by the nucleophilic substitutions of themethoxyl groups at C-11 and -10, respectively. Acetylation of 1 and 2produced the corresponding mono-acetyl tautomers (9 and 10). Antileukemicactivity of these derived tropoloisoquinoline alkaloids is also reported.

담배(Nicotiana tabacum L.) 현탁배양 세포에서 DNA 합성에 미치는 Polyamine의 효과 (Effects of Polyamines on DNA Synthesis in Nicotiana tabacum L. Suspension Cultured Cells)

  • Effects of polyamines on DNA synthesis were studied in synchronized culture of Nicotiana tabacum L. When DFMO and DFMA, inhibitors of ornithine decarboxylase and arginine decarboxylase, respectively were initially applied to the cells, the polyamine contents were rapidly dropped and [methyl-3H] thymidine incorporation into DNA was markedly reduced during the early stage of culture period. Inhibition of DNA synthesis, however, was partially reversed when these inhibitors were applied simultaneously with putrescine. In addition, exogenous administration of putrescine also increased the DNA synthesis during the all over the culture period. In vitro activity of DNA polymerase from Nicotiana tabacum L. was promoted by increasing concentrations of polyamines in the reaction mixture. Maximal activity was shown at 5 mM putrscine, 0.5 mM spermidine and spermine, respectively. Lack of Mg2+ ion in the reaction buffer resulted in an inhibition of the enzyme activity by about 30%. The inhibition could not be completely reversed by application of polyamines at optimal concentrations. These results suggest that polyamines promote the DNA synthesis in vivo and in vitro by stabilizing the DNA-helix upon binding to negatively charged groups on DNA or increasing the activity of DNA polymerase in Nicotiana tabacum L.

새우난초(Calanthe discolor Lindley)의 대량증식 시스템 확립 및 이용 (Establishment and Using of mass propagation system for Calanthe discolor Lindle)

  • 새우난초의 대량 증식 시스템을 확립하기 위하여 새우난초 종자 무균파종 및 순화 방법에 따른 생육특성 을 조사하였다. 발아율은 MS배지 에서 65% 이상이었고 유묘의 대량증식이 가능하였다. 계대배양한 유묘의 증식은 고체배지 보다는 액체배지에서 생육상태 가 더 양호한 경향을 보였다. 순화실험 결과, 70%차광조건에서 플라스틱 멀칭으로 습도를 80% 이상 유지하였을 때 100% 순화 생존하였다. 기내 육성된 배양묘는 백태로 뿌리를 감싸 심었을 때 발근이 가장 잘 이루어졌으나 순화과정 중에서 백태 대신에 질석과 펄라이트를 1:1 부피비로 이용한 경우에도 88%의 비교적 높은 생존율을 보였다.

동전해도금공장 Sludge로부터 동의 회수 (Recovery of Copper from Sludge of Copper Electro-Plating Plant)

  • The metallic copper was recovered from sludge of the copper electro-plating plant by pyrometallurgical process. The reducing agent was Pyrolysized from waste tires and the flux was a mixture $Na_2CO_3$, $NaB_4O_7$, and glass. The green sludge contained 87.5% moisture and 12.5% solid with 56.5% Cu and 1.59% Fe. The sludge dried at $100^{\circ}C$ was analyized to be $Cu_4SO_4(OH)_6{\cdot}2H_2O$ and CuO by XRD analysis. The former was 84% and the latter 16%, However, the calcined sludge at $500^{\circ}C$ was 49% $Cu_2O(SO_4)$ and 51% CuO. The sludge could by smelted at $1100^{\circ}C$ for two hours with 6 to 8 moles carbon with respect to copper to produce metallic copper (>90%) with recovery of 9% above.

중부지역 초지에서 혼파조합 및 유기질 비료 시용이 목초 수량, 식생 구성 및 사료가치에 미치는 영향 (Effect of Seed Mixture and Organic Fertilizer Application on Productivity, Botanical Composition and Forage Quality in Middle Mountainous Pasture)

  • 본 시험은 우리나라 중부지역(충남 금산, 해발 250m)에서 유기물 시용과 및 혼파조합에 따른 신규조성 초지의 생산성 변화를 평가하기 위하여 수행하였다. 총 6개의 처리 (오차드 그라스 위주, 티머시 위주 그리고 톨페스큐 위주의 혼파 초지에 유기질 비료의 시용 유무)를 두고 2014년 가을에 조성하여 2015년부터 2016년까지 조사하였다. 초장은 오차드그라스 위주 혼파조합에서 길었으며 2차 수확시는 오차드그라스 위주 및 톨페스큐 위주의 혼파조합에서 길었다. 건물함량은 대체적으로 처리간에 유의적인 차이가 없었다(P<0.05). 식생은 1차 수확시 오차드그라스 위주 혼파조합에서 잡초의 비율이 높았으며 3차 수확시에는 모든 처리구에서 두과목초가 사라졌다. 생초 및 건물수량은 1년차보다는 2년차에서 더 높게 나타났다. 또한 1년차에 유기물을 시용한 오차드그라스 위주 혼파조합에서 유의적으로 높았으며(P<0.05), 2년차에서는 유기물을 시용한 톨페스큐 위주 혼파조합에서 가장 높게 나타났다. 조단백질 함량은 톨페스큐 혼파조합에서 높게 나타났으며 티머시 위주 혼파조합에서 ADF 및 NDF 함량이 낮았고 소화율은 높았다. 조성 2년차에서는 2회 및 4회 예취시 목초의 사료가치가 높게 나타났으며 평균 RFV값도 4차에서 가장 높았다. 이상의 결과를 종합하여 볼 때 중부지역 구릉지 초지 조성시 생산성 면에서는 유기물을 시용한 오차드그라스와 톨페스큐 위주 혼파조합이 우수하였으며 사료가치적 측면에서는 티머시 위주 혼파조합이 우수한 것으로 나타났다.

An Efficient in vitro Micropropagation for Production of Disease-free Bulbs in Korean Native Lilium

  • Cryopreservation is one of the ideal and suitable methods for long-term storage of plant germplasm. The plant contaminated with diseases and pathogens are decreased the multiplication rate, survival rate and high quality of plants after cryopreservation. The aim of this work was to improve a micropropagation method for lily in Korea, which is indigenous plant. In the last process of rinsing scales after surface-sterilization, we tried to control the diseases and pathogens lived within the tissue by rinsing in 0.03% sodium hypochlorite (NaClO) instead of sterile distilled water. Bulb scales of Lilium were cultured in vitro on MS medium supplemented with Plant Preservative Mixture (PPM). The results showed that L. tsingtauense accessions were observed ranged from 53.9 to 100% with a mean value of 76.8% and L. hansonii accessions were checked from 84.5 to 85.5% with a mean of 85% survival rate. The newly small bulb formed from bulb-scales was transferred to MS medium. We checked the presence of microorganisms and survival rate after 3 weeks in culture after examination of bacterial incidences. The results indicated that the non-contamination rate were shown ranged from 75.0 to 94.1% with mean value of 83.2% in L. tsingtauense species, and that L. hansonii were observed 85.1 to 91.7% with mean value of 88.4%. This study will provide a valuable basis for establishment of effective axenic cultures for in vitro micropropagation of Korean native lily species.
