Differences of Phytoalexin-like Activities in Compatible and Incompatible Combinations of Rice Cultivars and Races of Pyricularia oryzae, and Decrease of those Activities by Heavy Application of Nitrogen (수도품종과 도열병균레이스와의 친화 및 비친화조합에 있어서 Phytoalexin 활성차이와 질소과용에 의한 활성 저하)

  • Thytealexin activities were investigated in compatible and incompatible combinations between rice cultivars and races of Pyricularia oryzae by using the spore-germination test in the diffusates from surface of leaf-blade treated with the spore suspensions. In compatible and incompatible combinations between nine cultivars of rice and three races of the fungus, phytoalexin activities were recognized in all combinations, and were much higher in incompatible combinations than in compatible combinations with significant differences. Activities of phytoalexin were increased with the time, and maximal peaks of them were at 36 hours after inoculation, and thereafter decreased. The activities were generally decreased in all combination by heavy applications of nitrogen fertilizer, especially in incompatible combinations. On the other hand, fresh juices from leaves of rice cultivars which were not inoculated with the fungus showed no phytoalexin activities.

Histological Detection of Phytoalexin Scoparone from Heat-Treated and UV-Illuminated Lemon Fruits After Inoculation with Penicillium digitatum

  • Phytoalexin scoparone (6,7-dimethoxycoumarin) was induced in flavedo tissue of lemon fruit inoculated with Penicillium digitatum during heat treatment for 3 days at $36^{\circ}$. The compound was also induced in the flavedo tissue after UV illuminatiion. Induction of scoparone was deteected in the flavedo tissue by histological analysis. This fluorescent scoparone accumulated only on the 4-5 layers of cells adjacent to the inoculation site. Preinoculation with P. digitatum and subsequent heat-treatment induced resistance in the lemon fruit tissues after challenge-inoculation at the site of the first infection. the data obtained in the study suggest that lemon fruit acquired resistance against P. digitatum parallel with the scoparone production at the infection site.

  • Extracellular capsidiol, sesquiterpenoid phytoalexin, in the medium of pepper (Capsicum annuum L.) suspension cells was not identified from control cells, but highly accumulated in the elicitor-induced cells within 6 hours after the addition of 0.05$\mu\textrm{g}$/$m\ell$ cellulase. Capsidiol production in elicitor-induced cells was markedly suppressed by cytochrome P450 inhibitors, such as ancymidol and ketoconazole demonstrating that biosynthesis of capsidiol is catalyzed by at least on hydroxylation enzyme in the biochemical pathway. Based on protein electrophoresis, two bands, 23.0kDa and 27.5kDa, were identified as newly synthesized polypeptides in the elicitor-induced suspension cells, suggesting that pepper cells which were subjected to elicitor treatment activate specific gene(s) for capsidiol biosynthesis in cultured cells.

  • To induce the phytoalexins in plant cell culture systems, we surveyed the antimicrobial activity following the feeding of five naphthoquinones in cell cultures of petunia. Among naphthoquinones treated, 2,5,7-trihydroxy-3-(5'-hydroxyhexyl)-1,4-naphthoquinone (3-OH NQ ) was most efficiently absorbed into the cells within 48 hr. The crude extracts of cells treated with 3-OH NQ showed a strong inhibition activity on spore germination of Aspergillus candidus $(MIC:\;32\;{\mu}g/ml)$, whereas the untreated cells showed no activity. The two active compounds, 4,2',4',${\beta}$-tetrahydroxychalcone and 4',7-dihydroxyflavone, were isolated from petunia cells treated with 3-OH NQ. The major phytoalexin, 4,2',4',${\beta}$-tetrahydroxychalcone, inhibited strongly the spore germination of A. candidus $(MIC:\;16\;{\mu}g/ml)$.

  • The extracellular sesquiterpenoids were accumulated in cell and hairy root cultures of Hyoscyamus muticus by elicitation using extracts of Rhizoctonia solani. The vetispiradiene synthase (VS) which is the first committed step in biosynthetic pathway leading to formation of solavetivone, lubimin, and rishitin from isoprenoid intermediate farnesyl pyrophosphate was induced upon elicitation, whereas no sesquiterpenoids and VS activity were detected in both control cell and hairy root cultures. VS activity increased rapidly and reached its maximum 12 h in both cell and hairy root cultures upon elicitor treatment. VS activities were paralleled with the absolute levels of VS polypeptide(s). Interestingly, the profiles of sesquiterpenoid accumulation in hairy root cultures were different from those in cell cultures. The hairy root culture seemed to fail to metabolize solavetivone further to lubimin.

  • BACKGROUND: Polychlorinated biphenyls (PCBs) are one of the most common environmental contaminants. Because of their recalcitrant properties and long-term toxicity, numerous studies have been performed. The toxicological concerns are focused on endocrinological effects of animal. Several different metabolites have been reported, including hydroxy PCBs, PCB quinones, and methylsulfonyl PCBs from animal tissues. However, details in plants have never been studied. It is well-known that plants can produce phytoalexin in response to chemical, physical, or pathological stress.METHODS AND RESULTS: In this study, the several PCBs and hydroxy derivatives were prepared by chemical syntheses. Their effects on secondary metabolite biosynthesis were determined in carrot roots. The levels of 6-methoxymellein were determined in several different treatments, using gas chromatography-mass spectrometry. In general, the concentration of 6-methoxymellein reached a maximum at 2 days and gradually decreased to trace level at 5 days in control experiments. However, the effects of PCBs or hydroxy derivatives were highly dependent on compounds. For example, the maximum concentrations of 6-methoxymellein were observed at 3 days for 2-hydroxy/4-hydroxybiphenyl, while 3,3',4,4',5-pentachlorobiphenyl and 3,5-dichloro-2-hydroxybiphenyl showed a rapid accumulation within 1 day, followed by rapid dissipation to undetectable levels.CONCLUSION: Biphenyl derivatives were effective elicitor of 6-methoxymellein accumulation. In general, hydroxybiphenyls (phenols) more efficiently induced phytoalexin biosynthesis than those without hydroxy groups. It can be concluded that PCBs or their possible metabolites could change the plant secondary metabolism.

Enzyme Activities and Compounds Related to Self-Defense in UV-Challenged Leaves of Rice

  • The induction of enzymes and the accumulation of their end products associated with self-defense mechanism in rice were investigated. When rice leaves were irradiated with UV light, activities of diterpene cyclase, phenylalanine ammonia-lyase (PAL), and cinnamic acid 4-hydroxylase (CA4H) were induced and rice phytoalexin, momilactone A was accumulated. The content of p-coumaric acid in rice leaves was closely correlated with self-defense or allelopathic potential against barnyardgrass. UV-challenged rice leaves gave rise to the inhibition of barnyardgrass growth.

  • The last enzyme of the sesquiterpen phytoalexin capsidiol synthesis in tobacco cell, 5-epi-aristolochene hydro-xylase which convert 5-epi-aristolochene (EAS) to capsidiol, was cloned by a reverse transcription polymerase chain reaction strategy and cDNA library screening. Cloned CYP-B3 contained high probability amino acid matches to known plant cytochrome P450 sequences and open reading frame with the conserved FxxGxRxCxG heme-binding region. Transcripts of CYP-B3 were not detected in control cells, but induced in elicitor-treated cells. Furthermore, CYP-B3 transcripts were induced by fungal extracts and cellulase but not by other stimuli(chilling, heat shock and 2,4-D). Induction of CYP-B3 transcripts by elicitor treatment was not affected by ancymidol and ketoconazole treat-ments suggesting that an inhibition of hydroxylase activity by Cyt P450 inhibitors resulting from post translational processing event.
