Evaluation of Allergenic Potency of an Inactivated Combination Vaccine against Hantaan and Puumala Viruses Using Mice and Guinea Pigs

  • Hantaan (HTN) and Puumala (PUU) viruses are major etiological agents of hemorrhagic fever with renal syndrome (HFRS), an important public health problem in Korea after the Korean War. The objective of present study was to determine allergenic potency of an inactivated combination vaccine against HTN and PUU viruses inflection. As a series of allergenicity assessment, a homologous active systemic anaphylaxis (ASA) and homologous/heterologous passive cutaneous anaphylaxis (PCA) tests using the mice and guinea pigs were carried out. In the ASA test, no anaphylactic symptoms were observed in the guinea pigs sensitized with the vaccine alone as well as the vaccine emulsified with an adjuvant. By homologous PCA test, the vatscine did not induced the potential IgE antibody production in the sera obtained from the sensitized guinea pigs. In addition, IgE against the vaccine was not significantly enhanced from the mice inoculated with the vaccine, which was judged by the heterologous PCA test in rats. On the other hand, the inoculation of ovalbumin appeared to allergenic reactions both in the ASA and PCA tests. The results suggest that a combination vaccine against HW and PUU viruses have no allergenic potential in mice or guinea pigs.

A Study on Antigenicity of G009, a Polysaccharide Isolated from Gandoderma lucidum IY009 in Mice and Guinea pigs (영지의 단백다당체 G009의 마우스와 기니픽에 있어서의 항원성에 관한 연구)

  • In the present study, the antigenic potential of G009, a polysaccharide isolated from Ganoderma lucidum IY009, was determined in BALB/C mice and Hartley guinea pigs. Antigenicity tests, including passive cutaneous anaphylaxis (PCA), active systemic anaphylaxis (ASA) and indirect hemagglutination test (IHA) were performed according to the established guidelines of National Institute of Safety Research. The results were as follows: 1. Mice showed no production of antibodies against G009 sensitized with an adjuvant, aluminum hydroxide gel (alum), when judged by the heterologous PCA test in rats. Meanwhile, antibodies against ovalbumin (OVA) sensitized with alum were clearly detected. 2. In the studies with guinea pigs, both the sensitization of G009 alone and of G009 with complete Freund's adjutant (CFA) did not produce positive reactions in homologous PCA. In the case of ASA, however, G009 alone and G009 with CFA produced positive reactions. 3. No G009 specific reaction was observed in an IHA assay using sera isolated from G009 sensitized mice. These findings suggest that G009 have no antigenicity potential in mice but may have weak antigenicity in guinea pjgs.

A Study on the Allergenicity of Egg Protein (달걀 단백질의 Allergenicity에 관한 연구)

  • Egg is an important foods containing many good proteins. But it is well known that egg protein has a lot of allergenicity. The purpose of this study is to develop the methods to reduce the allergenicity of egg. I tried various experimental methods ; For example, heat treatment, irradiation with ultraviolet and microwaves, treatment with polyphosphate, enzyme hydrolysis and PCA inhibition test using guinea pigs and degrees of hydrolysis. The results obtained were as follows ; 1. Heat treatment reduced allergenicity of egg protein. The longer the heat time, the better the effect. 2. Irradiating with ultraviolet and microwave increased both the degree of protein hydrolysis and PCA inhibition reduced the allergenicity. Ultraviolet was more effective than microwaves on egg protein. Fertilized eggs did not reduce allergenicity. 3. Enzyme treatment increased the degree of hydrolysis and PCA inhibition, and reduced allergenicity considerably. Alcalase was more effective than neutrase. 4. Adding polyphosphate did not induced protein hydrolysis, but increased PCA inhibition and reduced allergenicity. 5. The picture of various treatments of egg gel by SEM showed a light surface which indicated that protein was desolved. Neutrase was lighter than alcalase, and the longer the heating time, the lighter the surface became. 6. Measurements of the hardness of egg gel by Instron showed that the longer the reaction time with enzyme, the softer it became.

Effect of Kamihyungbangjihwang-tang on Immediate-type Allergic Reaction (가미형방지황탕이 즉시형 알러지반응에 미치는 영향)

  • The purpose of this research was to investigate the effects of Kamihyungbangjihwang-tang (KHT) on immediate-type allergic reaction. KHT was composed of hyungbangjihwangtang (HJT) and Rubus coreanus Miquel (RC) and Aspalathus linear is (AL). KHT and HJT (500 mg/kg) inhibited the systemic anaphylaxis induced by compound 48/80 and inhibited the passive cutaneous anaphylaxis (PCA) induced by anti-dinitrophenyl (DNP)-IgE and DNP-human serum albumin (HSA) in vivo. In addition, KHT, HJT, RC and AL inhibited the release of histamine and increased the release of cAMP from rat peritoneal mast cells. The anti-allergic action of KHT was more potent than those of HJT. These results indicate that KHT may be useful for the prevention and treatment of type I allergy related disease via inhibition of histamine release from mast cells.

  • The purpose of this research was to investigate the effects of Kwakhyangjeonggisan (KJS) on the anti-allergic action. In the present study, we examined the effect of KJS on type I and type IV allergic reaction. KJS inhibited the systemic anaphylaxis induced by compound 48/80 and platelet activating factor (PAF), and inhibited the passive cutaneous anaphylaxis (PCA) induced by anti-dinitrophenyl (DNP)-IgE and DNP-human serum albumin (HSA) in vivo. In addition, KJS dose-dependently inhibited the release of histamine from peritoneal mast cells in rat. Also, KJS inhibited the delayed type hypersensitivity (DTH) induced by SRBC and the contact dermatitis induced by dinitrofluorobenzene (DNFB). KJS inhibited the proliferation of splenocytes, the subpopulation of B220+ cells and CD4+CD8-(Th) cells in splenocytes and the production of γ-interferon in serum and splenocytes. These findings suggest that KJS prevented the type I allergy by the inhibition of histamine release from mast cells and the type IV allergy by the inhibition of γ-interferon production and B lymphocytes subpopulation. These results indicate that KJS may be useful for the prevention and treatment of type I and type IV allergy related disease.

  • The effect of aqueous extract of Salvia plebeia R. Br. (Labiatae) (AESP) on immunoglobulin (lgE) antibody mediated allergic reactions in rats was investigated. AESP inhibited passive cutaneous anaphylaxis (PCA) when intravenously, intraperitoneally, and orally administered. AESP dose-dependently inhibited histamine release from rat peritoneal mast cells activated by anti-DNP IgE antibody. Moreover, AESP had an inhibitory effect on anti-DNP IgE antibody induced $TNF-{\alpha}$ production from RPMC. These results suggest that AESP inhibit the IgE-mediated allergic reaction in rats.

  • Antigenicity of DA-3285, human recombinant erythropoietin which was produced from mammalian cells, was examined in guinea pigs and mice. In active systemic anaphylactic test, mild anaphylactic signs were observed in guinea pigs sensitized subcutaneously with DA-3285 or DA-3285 incorporated with complete Freund's adjuvant(CFA) when challenged with high dose(1000 IU/Kg) of DA-3285. Other groups showed negative responses. In mouse-rat passive cutaneous anaphylaxis test, 20% sera of mice immunized with DA-3285 or DA-3285 mixed with aluminum hydroxide(alum) showed mild positive responses. In the case of indirect haemagglutination reaction(IHA) test, when sheep red blood cells coated with DA-3285 was incubated with mouse serum, all the serum samples were showed negative responses. These results suggest that DA-3285 has a very weak antigenic potential and probably would not induce systemic allergic reactions in clinical uses.

  • I experimented on commercial infant formula in passive cutaneous anaphylaxis inhibition methods. I used several guinea pigs for this experiment. The results obtained from this study were as follows : 1) Commercial infant formula showed positive reactions, while soy-based formula showed negative reactions. 2) The products from different companies showed different reactions respectively The products of "A" company showed low allergenicity compared to the products of "B" company. The Products of "C" company showed the lowest allergenicity. 3) It seemed to be that the differences of allergenicity among the products by various companies were caused by the differences of the materials used, the combination ratio of materials, the heat treatment level, and manufacturing processes., and manufacturing processes.

  • Effect of Compound-A, a phenylpropanoid isolated from Arctium lappa fruit, on heterologous passive cutaneous anaphylaxis (HPCA), the release of histamine, and Phospholipase $A_2$ (PLA2) and phosphadiesterase (PDE) activities were studied by the method of Levine and Vaz. Anti-serum was prepared from ovalbumin (OA)-sensitized male Balb/c mouse at two weeks after the last challenge of OA and alumina gel. Heterologous PCA test in rats were carried out to determine the contents of leakaged pigment in the dorsal skin 30 minutes after i.v. injection of 0.2 ml of 1 % OA and 1 % Evans blue mixture (1:1). Rumex crispus Suppresses Type I Hypersensitive Immune Response (소리쟁이(Rumex crispus) 추출물의 제1형 알레르기 반응 억제 효과)

  • Rumex crispus is known to have anticancer, antioxidant, antibacterial, and bone loss inhibitory activities. Mast cells are critical immune cells that induce a type 1 IgE-mediated allergic reaction. However, there are no reports of inhibitory effects of Rumex crispus on mast cells and allergic reactions. In this study, we performed some experiments to investigate whether Rumex crispus ethanol extract(RCE) has any inhibitory effect on antigen-induced type I allergic response in vitro and in vivo. RCE inhibited degranulation of IgE-mediated mast cells(IC50, ~57 ㎍/ml) and cytokine production such as TNF-α and IL-4 in a dose-dependent manner. In vivo, RCE significantly inhibited passive cutaneous anaphylaxis(PCA)(ED50, ~198 mg/kg) in mice. Furthermore, RCE inhibited degranulation of MCs in ear tissue of mice with PCA. Mechanism studies showed that RCE inhibited the activation of Syk and Syk-dependent pathway such as LAT, PLC-γ, Akt, and MAP Kinase. Our results demonstrate for the first time that RCE inhibits type I hypersensitive response by suppressing the activity of Syk in mast cells, thereby reducing degranulation and cytokine production. Taken together, RCE could be used as a novel therapeutic material to suppress allergic diseases.
