항산화제로서 비타민 C가 적출된 쥐심장에서 허혈 및 재관류후 좌심실 기능회복에 미치는 영향 (Effects of vitamin C as antioxidant on recovery of left ventricular function after ischemia and reperfusion in isolated rat heart)

  • 과거에 심근보호에 대한 많은 조사는 저온의 고칼릅 심정지액 및 국소적 냉각에 의한 방법으로는한 계가 있다는 지적이 있었다. 따라서 최근의 실험들은 재관류의 방법에 따른 허혈 후 심근회복에 대하여 촛점이 맞춰 지고 있다. 재관류 시 산소에 의한 심근손상이 밝혀 짐으로써 oxygen free radical scavenger에 대한 관심이 높아지고 있다. 따라서 본 교실에서는 쥐에게 항산화제로서 비타민 C를 먹인 후 Langendorf'r system을 이용하여 허혈 및 재관류 시 좌심실 기능의 변화를 관찰하였다 대상은 체중 190-))Og의 Sprague-Dawley쥐를 암수 구별없이 사용하였다. 편의상 비타민을 먹이지 않 은 대조군을 Group A (n=10)라 하였고 200mg의 비타민을 먹인 실험군을 Group R (n=10)라 하였다. 실 험군의 경우는 비타민 C 200mg을 경구투여한 후 24시간에 시행하였다. 방법은 언저 복강을 통해 헤파 린과 펜토탈을 주입한 후 심장을 적출하여 Langendorff system에 거치하고 비운동성 역관류를 시켰다. 관류액은 변형된 Krebs-Henseleit solution을 사용하였다. 좌심실내에 풍선을 삽입하여 polygraph를 통해 좌심실의 혈역학적 기능을 관찰하였다. 먼저 20분간 심 揚\ulcorner안정될 때까지 기다린 다음 51. Thomas심 정 지액으로 심정 지를 시킨후 30분간 허 혈시키고 다시 20분간 재관류시켰다. 각 Group에서 허혈전후의 좌심실압의 비, dp/dt의 비,박동수의 비, 재관류 후 첫 박동 및 안정될 때 까지의 시간을Group간에 비 교하였다. 좌심실압의 비는 Group A가 평균 88.9%, Group B가 114%로 실험군이 의의있게 높았으며 역시 dp/dt도 Group A가 89.6%, Group B가 1)2.9%로 실험군이 의의 있게 높았다 그러나 박동수의 비, 재관류 후 첫 박동 및 안정될 때 까지의 시간의 비교는 통계 학적 의의가 없었다. 결론적으로 항산화제로서 비타민 C는 허혈 및 재관류 시 좌심실 기능회복에 도움이 되었다. 그러나 향후 임상적 적용을 위해서는 더욱 자세하고 많은 실험 이 요구된다.

Native plants (Phellodendron amurense and Humulus japonicus) extracts act as antioxidants to support developmental competence of bovine blastocysts

  • Objective: Phellodendron amurense (P. amurense) and Humulus japonicus (H. japonicus) are closely involved in anti-oxidative response and increasing antioxidant enzymes activities. However, the effects of their extracts on development of preimplantation bovine embryos have not been investigated. Therefore, we investigated the effects of P. amurense and H. japonicus extracts on developmental competence and quality of preimplantation bovine embryos. Methods: After in vitro fertilization, bovine embryos were cultured for 7 days in Charles Rosenkrans amino acid medium supplemented with P. amurense ($0.01{\mu}g/mL$) and H. japonicus ($0.01{\mu}g/mL$). The effect of this supplementation during in vitro culture on development competence and antioxidant was investigated. Results: We observed that the blastocysts rate was significantly increased (p<0.05) in P. amurense ($28.9%{\pm}2.9%$), H. japonicus ($30.9%{\pm}1.5%$), and a mixture of P. amurense and H. japonicus ($34.8%{\pm}2.1%$) treated groups compared with the control group ($25.4%{\pm}1.6%$). We next confirmed that the intracellular levels of reactive oxygen species (ROS) were significantly decreased (p<0.01) in P. amurense and/or H. japonicus extract treated groups when compared with the control group. Our results also showed that expression of cleaved caspase-3 and apoptotic cells of blastocysts were significantly decreased (p<0.05) in bovine blastocysts derived from both P. amurense and H. japonicus extract treated embryos. Conclusion: These results suggest that proper treatment with P. amurense and H. japonicus extracts in the development of preimplantation bovine embryos improves the quality of blastocysts, which may be related to the reduction of ROS level and apoptosis.

고밀도 배양에 있어서 영양강화 방법 및 종류에 따른 rotifer의 지방산 조성의 변화 (Change of fatty acid compositions of rotifer according to enrichment diets and methods in the high density culture)

  • 본 연구는 고밀도 배양시 rotifer의 영양강화 방법 및 영앙강화제에 따른 rotifer의 개체밀도와 지방산 조성을 조사하였다. 첫째 방법은 담수산 Chlorella로 rotifer를 고밀도로 배양한 후 영양강화제인 유지효모, Algamac, Super Selco, 해수산 Chlorella로 2차 영양강화 하였다. 두 번째 방법은 rotifer 반 연속 고밀도 배양중 18시간 동안 담수산 Chlorella를 공급한 후 6시간동안 영양강화제를 공급하였고 대조구로 담수산 Chlorella을 공급하지 않고 24시간 동안 해수산 Chlorella를 공급하였다. 배양수조는 5$\ell$를 이용하여 수온 $28^{\circ}C$를 유지하였고 산소가스를 공급하였다. 반 연속 고밀도 배양 방법에서 24시간 동안 해수산 Chlorella를 공급하였을 때 rotifer 밀도와 용존산소는 안정적으로 유지되었고 rotifer의 n-3 HUFA의 함량이 다른 실험구보다 가장 높게 나타났다. 그러나 2차 영양강화 방법과 반 연속 고밀도 배양 방법에서 유지효모, Algamac 및 Super Selco로 영양강화하였을 때 rotifer의 밀도와 용존산소는 급격히 감소하는 경향을 보였고 이들 영양강화제 가운데 Super Selco가 비교적 높은 n-3 HUFA의 함량i글 보였다. 따라서 본 실험을 통해서 rotifer 고밀도 배양시 안정적인 rotifer 성장과 rotifer의 질적 개선을 위해서는 24시간 해수산 Chlorella를 공급하는 것이 효율적인 것으로 판단된다.

말티즈 견에서 발생한 원발성 폐동맥 고혈압증 (Primary Pulmonary Hypertension in a Maltese Dog)

  • 중성화된 1년령 말티즈 개(2.4kg)가 간헐적인 기침, 호흡곤란, 운동 불내성을 주요 증상으로 내원하였다. 신체검사 시 제 2음 모순 분열, 확장기 역류성 잡음이 관찰되었으며, 심장초음파에서 주폐동맥의 확장, 심실 중격 비대를 동반한 우심실 편심성 비대, 심각한 삼첨판 역류증과 폐동맥 역류증(5.4m/sec, 3.4m/sec)이 관찰되었다. 이러한 진단 검사 결과를 바탕으로, 본 증례를 선천성 심장 내 단락, 폐쇄성 폐질환, 그리고 우심계의 압력 과부하 또는 폐혈관계과관류와 관련된 전신질환과 감별하여 원발성 폐동맥 고혈압증으로 잠정진단 하였다. 환자는 furosemide와 aspirin을 투여하여, 산소요법을 실시하였다. 본 증례는 드물게 발생하는 개의 원발성 폐동맥 고혈압이다.

Comet Assay를 이용한 케일, 명일엽, 당근, 돌미나리 녹즙의 Chinese Hamster Lung 세포 DNA 손상 보호 효과 (Protective Effect of Yellow-Green Vegetable Juices on DNA Damage in Chinese Hamster Lung Cell Using Comet Assay)

  • The present study was attempted to investigate the antioxidant capacity of popular yellow-green vegetable juices (kale, Angelica keishei, carrot, small water dropwort) and to investigate the effect of vegetable juices on protecting oxidative damage to DNA in cultured Chinese hamster lung (CHL) cells. Antioxidant capacity was analyzed by TRAP assay (Total radical-trapping antioxidant potential). Cellular DNA dmamage was measured by SCGE (single-cell gel electrophoresis, also known as comet assay. Cells incubated in medium with PBS (negative control) or with various concentration of the freeze dried green juices (25, 50, 100, 250 $\mu\textrm{g}$/$m\ell$) resuspended in PBS were treated with $H_2O_2$ (200 ${\mu}{\textrm}{m}$) as an oxidative stimulus for 5 min at 4$^{\circ}C$. The physiological function of each vegetable juice on oxidative DNA damage was analyzed and expressed as tail moment (tail length X percentage migrated DNA in tail) . Kale juice had the highest TRAP value suggesting that kale has the highest antioxidant capacity followed by Angelica keishei, small water dropwort and carrot. Cells treated with $H_2O_2$ had extensive DNA damage compared with cells treated with PBS or pre-treated with vegetable juice extracts. All green juices inhibited $H_2O_2$-induced DNA damage with kale being the most effective juice among the tested juices. These results indicate that green juice supplementation to CHL cells followed by oxidative stimulus inhibited damage to cellular DNA, supporting a protective effect against oxidative damage induced by reactive oxygen species.   • To evaluate anesthetic effecto of propofol infusion after premedication with xylazine, 20 days were randomly assigned 4 groups. Propofol was infused (group 1: 0.2 mg/kg/min, group 2 : 0.4 mg/kg/min, group 3 : 0.6 mg/kg/min, group 4 : 0.8 mg/kg/min) for a period of 90 minute immediately after premedication with xylazine(1 mg/kg) and atropine(0.05mg/kg) under oxygen supplementation. Induction of anesthesia was rapid and smooth providing satisfactory conditions for intubation in all the dogs. No vomiting and cyanosis were observed after induction and during propofol infustion. There was pain reflex in group 1 but not in group 3 and 4. Mean arousal times (mins) were $6.18{\pm}3.65(group 1), 13.07{\pm}5.05(group 2), 22.06{\pm}6.48(group 3) and 23.33{\pm}9.28 (group 4) and Mean walking times were 16.20{\pm}6.15(group 1), 15.80{\pm}4.73(group 2), 28.27{\pm}7.55 (group 3), 39.10{\pm}13.75$ (group 4) respectively. In group 4, body temperature during total infusion period in group 3, 4. Hematologic values (WBC, RBC, PCV) and serum chemistry values(ALT, AST, BUN, creatinite) were monitored before anesthesia, 1 hour and 1 day after termination of infusion postanestesia. No significant changes were monitored in all experimental group. Although propofol infusions of 0.2 mg/kg/min and 0.4mg/kg/min were considered too low to maintain a suitable depth of anesthesia, but that of 0.6mg/kg/min were considered too low to maintain a suitable depth of anesthesia, but that of 0.6mg/kg/min proper to provide a light planes for minor surgical procedure during 90 minutes with xylazine premedication.

  • A dog (neutered male Schnauzer, 11 years old, 8 kg) presented with recurrence of immune-mediated hemolytic anemia. Rapid whole blood transfusion was initiated, and then sudden tachycardia and dyspnea were observed. Invasive arterial blood pressure, arterial blood gas analysis, and thoracic radiograph indicated transfusion-associated circulatory overload (TACO). Persistent high blood pressure of 160-205 mmHg was observed; the thoracic radiograph revealed interstitial infiltration and a fissure line, which suggested pulmonary edema and pleural effusion. Despite furosemide administration and nasal oxygen supplementation, hypertension and respiratory distress were not completely controlled. Finally, cardiac arrest occurred and the patient expired due to TACO 24 hours after the transfusion.

Effect of L-carnitine on sperm quality during liquid storage of boar semen

  • Objective: This study was conducted to investigate the effect of L-carnitine on the pig semen characteristics during storage. Methods: Spermatozoa samples were examined for spermatozoa quality and then randomly divided into 5 groups: 0 (control), 12.5, 25, 50, and 100 mM L-carnitine. Sperm motility, plasma membrane integrity and antioxidant parameters (total reactive oxygen species, total antioxidant capacity, and malondialdehyde) were evaluated after 0, 3, 5, and 10 day cooled-storage at 17℃. Moreover, ATP content, mitochondria activity as well as sperm-binding and in vitro fertilizing ability of preserved boar sperm were also investigated. Results: Supplementation with 50 mM L-carnitine could effectively maintain boar sperm quality parameters such as sperm motility and membrane integrity. Besides, we found that L-carnitine had positive effects on boar sperm quality mainly through improving antioxidant capacities and enhancing ATP content and mitochondria activity. Interestingly, by assessing the effect of L-carnitine on sperm fertility and developmental potential, we discovered that the extender containing L-carnitine could improve sperm quality and increase the number of sperms bounding to zona pellucida, without improving in vitro fertility and development potential. Conclusion: These findings suggested that the proper addition of L-carnitine to the semen extender improved boar sperm quality during liquid storage at 17℃.

Protective effect of the standardized green tea seed extract on UVB-induced skin photoaging in hairless mice

  • BACKGROUND/OBJECTIVES: Ultraviolet B (UVB) irradiation on skin can induce production of reactive oxygen species (ROS), which cause expression of matrix metalloproteinases (MMPs) and collagen degradation. Thus, chronic exposure of skin to UVB irradiation leads to histological changes consistent with aging, such as wrinkling, abnormal pigmentation, and loss of elasticity. We investigated the protective effect of the standardized green tea seed extract (GSE) on UVB-induced skin photoaging in hairless mice. MATERIALS/METHODS: Skin photoaging was induced by UVB irradiation on the back of Skh-1 hairless mice three times per week and UVB irradiation was performed for 10 weeks. Mice were divided into six groups; normal control, UVB irradiated control group, positive control (UVB + dietary supplement of vitamin C 100 mg/kg), GSE 10 mg/kg (UVB + dietary supplement of GSE 10 mg/kg), GSE 100 mg/kg (UVB + dietary supplement of GSE 100 mg/kg), and GSE 200 mg/kg (UVB + dietary supplement of GSE 200 mg/kg). RESULTS: The dietary supplement GSE attenuated UVB irradiation-induced wrinkle formation and the decrease in density of dermal collagen fiber. In addition, results of the antioxidant analysis showed that GSE induced a significant increase in antioxidant enzyme activity compared with the UVB irradiation control group. Dietary supplementation with GSE 200 mg/kg resulted in a significant decrease in expression of MMP-1, MMP-3, and MMP-9 and an increase in expression of TIMP and type-1 collagen. CONCLUSIONS: Findings of this study suggest that dietary supplement GSE could be useful in attenuation of UVB irradiation-induced skin photoaging and wrinkle formation due to regulation of antioxidant defense systems and MMPs expression.

Phaleria macrocarpa Suppresses Oxidative Stress in Alloxan-induced Diabetic Rats by Enhancing Hepatic Antioxidant Enzyme Activity

  • Oxidative stress is caused by an imbalance between the production of reactive oxygen and an ability of a biological system, to readily detoxify the reactive intermediates or easily repair the resulting damage. It has been suggested that developmental alloxan-induced liver damage is mediated through increases in oxidative stress. The anti-diabetic effect and antioxidant activity of Phaleria macrocarpa (PM) fractions were investigated in alloxan-induced diabetic rats. After two weeks administration of PM, the liver antioxidant enzyme and hyperglycemic state were evaluated. The results showed that oral administration of PM treatments reduced blood glucose levels in diabetic rats by oral administration (P < 0.05). Serum glutamic-oxaloacetic transaminase (sGOT) and serum glutamic-pyruvate-transaminase (sGPT) were also diminished by PM supplementation. The superoxide dismutase (SOD), catalase (CAT) and glutathione-peroxidase (GPx) activities, and glutathione (GSH) level in the alloxan-induced diabetic rats were significantly decreased (P < 0.05) compared to those in the normal rats but were restored by PM treatments. PM fractions also repressed the level of malondialdehyde (MDA) in the liver. Glutathione reductase (GR), glutathione-S-transferase (GST) and $\gamma$-glutamylcysteine synthase (GCS) were also reduced in alloxan-induced diabetic rats. PM fractions could restore the GR and GST activities, but the GCS activity was not affected in rat livers. From the results of the present study, the diabetic effect of the butanol fraction of PM against alloxan-induced diabetic rats was concluded to be mediated either by preventing the decline of hepatic antioxidant status or due to its indirect radical scavenging capacity.
