Effects of Germination Condition, Nursery Media and Nutrient Concentration on Seedling Growth Characteristics of Pak-choi and Lettuce in Plant Factory (발아조건과 배지종류 및 양액 농도에 따른 식물공장 재배 청경채와 상추의 묘 생육특성)

  • This experiment was conducted to study the effects of germination condition, nursery media and nutrient concentration in plant factory on the growth characteristics of pak-choi and lettuce seedlings. The results showed that temperature (15, 20, $25^{\circ}C$) and light conditions (lighting and no lighting) had no significant effect on germination rate of pak-choi and lettuce, whereas the interaction of growing media (rockwool and urethane sponge) and moisture management before and after sowing had significant effect on germination percentage. In case of no moisture management before and after sowing, germination percentage was much higher in rockwool medium than urethane sponge medium. But germination percentage of urethane sponge was higher than that of rockwool with applying moisture before and after sowing. At the nutrient concentration of EC $1.0dS{\cdot}m^{-1}$ the fresh weight of pak-choi was heavier than in EC 1.5 and $0.5dS{\cdot}m^{-1}$ by 1.8 and 6.0 times, respectively. At the nutrient concentration of EC $1.0dS{\cdot}m^{-1}$ the fresh weight of lettuce was also heavier than in EC 1.5 and $0.5dS{\cdot}m^{-1}$ by 1.7 and 3.5 times, respectively.

Performance of Acacia senegal (L.) Wild Seedlings Growth under some Tree Manures and NPK Fertilizers in Nursery Site

  • This study aimed to assess the effects of tree manures in comparison with NPK fertilizer on growth of Acacia senegal seedlings. It was conducted in the nursery of the Faculty of Forestry, University of Khartoum, Shambat (Lat.: $15^{\circ}$ 39' 387" N and Long.: $32^{\circ}$ 30' 871" E), during August 2008 and January 2009. The growing media were prepared by mixing ground foliage of trees with sandy soil (weight/volume) as follows, Albizia lebbeck (AL): 25 g, 50 g and 75 g; Azadirachta indica (AZ): 25 g, 50 g and 75 g; Khaya senegalensis (KH): 25 g, 50 g and 75 g; NPK fertilizer: 30 g per seedling. Albizia lebbeck and Azadirachta indica manures and NPK fertilizer have stimulated the Acacia senegal seed germination percentage in comparison with the control, with respective values of 89, 82.7, 81 and 71%. Khaya senegalensis manure has suppressed the Acacia senegal seed germination percentage with a value of only 49%. Effects of treatments on the seedlings growth parameters varied in the following percentages, in comparison to the control, shoot height: AZ 62.9%, AL 46%, KH 9.9% and NPK 27.8%; root length: AZ 25.8%, AL 31.5%, KH 30.6% and NPK 4.4%; diameter: AZ 75.2%, AL 37.1%, KH 34.3% and NPK 20%; Shoot biomass: AZ 319%, AL 195.2%, KH 57.1% and NPK 42.9%; root biomass: AZ 288.9%, AL 116.7%, KH 55.6% and NPK 16.7%%;. Thus, the effectiveness of the used substrates on the growth performance of Acacia senegal seedlings occurred in the following descending order: Azadirachta indica> Albizia lebbeck>Khaya senegalensis>NPK fertilizer. These results assert clearly the ameliorative and fertilizing characteristics of tree manures that can be reliably used for raising seedlings stocks in the nurseries.

  • 본 논문은 돈사 내부의 환기부를 연속적으로 작동한 상태에서 환기부의 입기팬 및 배기팬의 회전수를 감소시키거나 증가시킴으로서 축사 내부로 유입되는 환기량을 조절하고, 이에 따라 환기 운영 시에 축사 내부의 온도 편차를 최소화함과 동시에 유해가스에 의한 돈사 내 공기오염을 방지할 수 있는 지능형 돈사 환기 제어 시스템 설계에 관한 연구이다. 본 시스템에는 온도 측정 데이터를 일정간격으로 저장하고, 저장된 온도 변화의 특성을 추출하여 지능적으로 돈사의 상태를 최적으로 유지할 수 있는 지능형 환기 제어 모델, 사용자의 연령 및 상황을 고려한 가시적이고 직관적인 사용자 위주의 인터페이스 설계가 포함된다.

Effect of Artificial Soils and Aqueous Solutions for Plantlet Acclimatization of Somatic Embryos of Aralia elata (두릅나무 체세포배 유래 소식물체의 순화에 미치는 배양토 및 공급액의 효과)

  • In order to develop effective acclimatization methods for Aralia elata plantlets regenerated from somatic embryos, various acclimatizing conditions were compared regarding both survival rate and growth of the plantlets. The plantlets were transplanted into plastic boxes containing artificial soil in the presence of either several levels of MS liquid media, distilled water, 2% sucrose or 0.1% hyponex solution. They were then cultured by spraying of distilled water twice a week and maintained in the normal tissue culture room. Perlite was proved to be better than vermiculite on survival rate and growth of the plantlets. As the size of perlite (larger than 0.2 cm in diameter) increased, both the survival rate and growth of the plantlets improved. Among the various MS liquid media and different aqueous solutions tested, distilled water appeared to result in the best survival rate and growth. MS media were also effective in increasing survival rate and supporting growth when diluted to 1/4 and/or 1/8. The acclimatized plantlets could be transplanted directly onto the nursery bed and grown normally. The above results suggest that plantlets regenerated from somatic embryos of Aralia elata be effectively acclimatized using a plastic box containing perlite with distilled water treatment.

Types and Physicochemical Characteristics of the Nursery Media Used in Major Strawberry-Farming Regions (주요 딸기주산지에서 이용되는 육묘배지 종류 및 이화학적 특성)

  • In Danseong-myeon (myeon is a subdivision of), Sancheong-gun, and Sugok-myeon, Jinju City, pot seedling culture was used at the rates of 100% and 62%, respectively. Root restriction seedling culture was used relatively more frequently than pot seedling culture in Yangchon-myeon, Nonsan City, Guryong-myeon, Buyeo-gun, and Wolsan-myeon, Damyang-gun. Open-field seedling culture was used at the rates of 80% and 54% in Samnangjin-eup (eup is a subdivision of a gun), Miryang City and Ssangnim-myeon, Goryeong-gun, respectively. In Danseong, Sancheong, and Sugok, Jinju, granite soil was used as the medium for pot seedling culture at the rates of 90% and 80%, respectively. In Yangchon, Nonsan, Wolsan, Damyang, and Ssangnim, Goryeong, commercial bed soil and coir dust were used for seedling culture at the rates of 73%, 64%, and 60%, respectively. At the main production sites, the use of granite soil for seedling culture, highest rate in 2000-2005, continued to decrease; in 2011-2013, granite soil was used at the rate of 37%, while commercial bed soil and coir dust were used at the rates of 32% and 14%, respectively. The bulk density of commercial bed soil, expanded rice hulls, and coir dust was $0.11-0.16g/cm^{-3}$. Coir dust was the lightest material but had the highest and, 36.7% and 21.8%, respectively. In sandy loam soil and granite soil was low. The pH of all growth media was between 5.4 and 7.0, and the values were in the range of $0.15-0.66dS{\cdot}m^{-1}$. However it was necessary, to adjust the pH levels and enhance the of expanded rice hulls, sandy loam soil, and granite soil. Similarly, the fertilizer concentration and air permeability of coir dust need to be adjusted.

  • Selection of stress-tolerant ginseng lines in fields is very difficult because it is almost impossible to control properly the environmental conditions of soil. On the contrary, it can be studied with ease to search for stress-tolerant ginseng lines through in vitro culture because of easy manipulation of stress conditions. This study was conducted for the selection of ginseng pure lines tolerant to salt stress. Murashige & Skoog(MS) media with 2.5 folds of KNO$_3$, NH$_4$NO$_3$, MgSO$_4$.7$H_2O$, KH$_2$PO$_4$, and CaC1$_2$.2$H_2O$ was established for the selection of ginseng pure lines tolerant to salt stress in vitro. Among 88 ginseng pure lines bred by Korea Ginseng and Tobacco Research Institute, Punggi Hwangsuk, 78093, 82886, 78135, 86024 and KG104 lines was tolerant to salt stress. For the stable production of quality Korean ginseng, genetic tolerance to salt stress is one of important factors since relatively high salt concentrations in the ginseng nursery soil environment of Korea. Ginseng inbred pure lines were tested for their tolerance to salt stress through in vitro culture technique.

Effect of Mixture Rate of Used Media and Perlite on Physico-Chemical of Properties Root Media and Seedling Quality in Fruit Vegetables Plug Nursery System (공정육묘시 재활용 상토에 신규상토 및 펄라이트의 혼합비율이 상토의 이화학적 특성과 과채류 묘소질에 미치는 영향)

  • Improving the physico-chemical properties of used media by mixing ratio of new plug media (NPM), used plug media (UPM) and perlite is necessary to improve seedling quality. In this study, five treatments were designed to investigate mixing ratio of UPM and NPM by ratio of volume 0 : 100, 25 : 75, 50 : 50, 75 : 25, 100 : 0, respectively. On the other hand, nine treatments were designed to investigate of perlite volumes were added to UPM and mixed media (UPM : NPM (50 : 50)) with 0, 5, 10, and 20% of ratio volume. The physicochemical properties of all mediums and their effect on growth response of tomato and cucumber seedlings were determined. The result indicates that physical properties was improved when NPM was mixed with UPM and at mixed ratio of volume 50 : 50 (v : v) has similar pore spare, bulk density and water retention to NPM. Seedling quality of tomato and cucumber in mixed media (50 : 50) are better than other mixed ratio and similar to NPM. Addition perlite to UPM and mixed media 50 : 50 (v : v) increased the pore space and water retention. Physical properties such as particle density, pore space and bulk density were increased when perlite volume increased. However, the best of seedling quality was observed by the addition at 10% volume of perlite. These results suggested that optimum of mixed ratio for recycled used media is new media and used media 1 : 1 mixed.

  • One-year-old seedlings of Ginkgo biloba were treated with various simulated acid rains(pH 2.0, pH 3.0, pH 9.0 and pH 5.0) to examine the effects of simulated acid rain on the chemical properties of the tested soil. The seedlings were grown in a pot($4500cm^3$)containing one of three different soils(nursery soil, mixed soil and sandy soil). Simulated acid rain was made by diluting sulfuric and nitric acid solution($H^1SO^4$ : $HNO^3$ =3 : 1, V/V) with tap water and tap water(pH 6.4), and treated by 5mm each time for three minutes during the growing seasons(April to October 1985). Acid rain treatments were done three times per week to potted seedlings by spraying the solutions. The chemical properties of potting media were compared among three soil types as well as among the various pH levels. The results obtained in this study were as follows : 1. Exchangeable calcium and magnesium contents and base saturation of the soil decreased with decreasing pH levels of acid rain, and their decreasing rates were as follows : sandy soil was the highest, followed by mixed and nursery soils, However, exchangeable aluminum content rather increased as the pH levels decreased. 2. Available phosphate in the soil decreased as the pH levels of acid rain decreased. Its content increased in nursery soil, compared with those before acid gain treatment, Gut decreased in mined and sandy soils. 3. Soil sulfate and nitrate contents increased remarkably as the pH levels decreased, and the only significant difference in the sulfate was found among the pH levels. Soil sulfate content was the highest in nursery soil, followed by mixed and sandy soils.

Effect of Plug Cell Volume and Medium Composition on Rooting and Growth of Lateral Shoot Cuttings of Tomato Plant (플러그 셀의 크기와 배지(培地) 조성이 토마토 삽목묘(揷木苗)의 발근(發根)과 생육(生育)에 미치는 영향(影響))

  • This experiment was conducted to investigate the effect of plug cell volume and medium on rooting and growth of lateral shoot cuttings of a few tomato cultivars. Plug cell volume was varied from 23 to 300 mL or control (cutting bed) and media used were carbonized rice hull (CRH), CRH+perlite, CRH+peatmoss and perlite+peatmoss. Nursery plants were able to be transplanted in 15 to 20 days after lateral shoots cutting in tomato. In volume of cutting media, the pots of 23 to 300 mL were proper, although root growth was gradually limited in decreased media volume. Rooting and growth was not influenced by plug tray cell medium of 120 mL or more. Cherry tomato Pepe (c.v.) showed 100% rooting and better growth, while in Momotaro (c.v.) rooting was 90%. There were 100% rooting and no considerable changes grown in all media tested of cutting plug tray volume of 30 mL.

Classification of Fusarium moniliforme Isolates into Four Strains Based on Mycological Characteristics (벼키다리 병원균(病原菌)인 Fusarium moniliforme의 균주특성(菌株特性)에 의(依)한 Strain 분류(分類))

  • For the classification of 531 isolates of Fusarium moniliforme from infected rice plants, five criteria were employed for clarifying the mycological difference from I to IV based on host symptoms, conidial formation, septate of macroconidia, color of mycelium on the culture, and fungicide response. Four strains I, II, III, and IV were detected and Strain IV was the most prevalent one. Based on mycological characteristics, macroconidia with 6 or 7 septate were mostly produced by Strains I and II. Four septate macroconidia were mostly formed by Strain III while Strain IV mostly formed 3 septate macroconidia. Strain I was reddish purple (5RP 5/4) on both the bottom and the surface of the media. Strain II produced reddish purple(5RP 3/2) on both the bottom and the surface of the media. Strain I produced mostly macroconidia in pionnotes on the media surface while Strain II produced mostly microconidia in pionnotes on the media surface. Both of Strain III and IV produced mostly macroconidia in sporodochia on the surface of the mycelium. Strain IV was less reduction of germination and caused abnormal elongation of all parts of the plants, but did not kill the plants in nursery boxes. Strain III killed fewer plants than Strain IV. As above results, isolates of F. moniliforme were classified into four strains I, II, III, and IV, on the basis of five criteria.
