암환자에게 반코마이신의 집단약물동태학 모델연구 (Population Pharmacokinetic Modeling of Vancomycin in Patients with Cancer)

  • The purpose of this study was to determine pharmacokinetic parameters of vancomycin using peak and trough plasma level (PTL) and Bayesian analysis in 20 Korean normal volunteers, 16 gastric cancer and 12 lymphoma patients and also using the compartment model dependent (nonlinear least squares regression: NLSR) and compartment model independent (Lagrange) analysis in 10 ovarian cancer patients. Nonparametric expected maximum (NPEM) algorithm for calculation of the population pharmacokinetic parameters was used, and these parameters were applied for clinical pharmacokinetic parameters by Bayesian analysis. Vancomycin was administered as dose of 1.0 g every 12 hrs for 3 days by IV infusion over 60 minutes in normal volunteers, gastric cancer and lymphoma patients. Population pharmacokinetic parameters, K and Vd in gastric cancer and lymphoma patients using NPEM algorithm were $0.158{\pm}0.014{\;}hr^{-1},{\;}0.630{\pm}0.043{\;}L/kg{\;}and{\;}0.131{\pm}0.0261{\;}hr^{-1},{\;}0.631{\pm}0.089{\;}L/kg$ respectively. The K and Vd in gastric cancer and lymphoma patients using Bayesian analysis were $0.151{\pm}0.027,{\;}0.126{\pm}0.056{\;}hr^{-1}{\;}and{\;}0.62{\pm}0.105,{\;}0.63{\pm}0.095{\;}L/kg$. The K and Vd in ovarian cancer patient using the NLSR and Lagrange analysis were $0.109{\pm}0.008,{\;}0.126{\pm}0.012{\;}hr^{-1}{\;}and{\;} 0.76{\pm}0.08,{\;}0.69{\pm}0.19{\;}L/kg$, respectively. It is necessary for effective dosage regimen of vancomycin in cancer patients to use these population parameters.

p관리도의 불량률의 변화 탐지 (Detection of Changes of the Population Fraction Nonconforming in the p Control Chart)

  • In this paper we calculate the subgroup size necessary for detecting the change of percent defective with several detection probabilities for orginal population fraction nonconforming p, changed population fraction nonconforming $p^*$, and the ratio k=$p^*$/p in the usage of p control charts. From our calculation we can know the error level of normal a, pp.oximation in detection probability calculation and recommend the subgroup size with lower error levels of normal a, pp.oximation, and then we show the reasonable subgroup size necessary for p, $p^*$, k, and the detection probability of the change of fraction nonconforming in a process. The information that we here show in tables will be useful when p control chart users decide the subgroup size in the p control chart users decide the subgroup size in the p control chart.

Two Polymorphisms of Interleukin-4 Gene in Korean Adult Periodontitis

  • Adult periodontitis is a multifactorial disease characterized by multple genetic and environmental factors. In view of the importance of interleukin-4 (IL-4) gene as a genetic factor for adult periodontitis, we investigated the relationship between two polymorphisms (-590 C $\rightarrow$ T polymorphism and 70 bp repeat polymorphism) of the human IL-4 gene and adult periodontitis in the Korean population. Genomic DNA was extracted from white blood cells of 32 adult periodontitis patients and 150 normal controls, respectively. There were no significant differences in the allele, genotype and haplotype distributions of two polymorph isms between normal controls and adult periodontitis group. Therefore, our results suggest that IL-4 gene locus contributes little to the interindividual susceptibility for adult periodontitis in Korean population.

Comparison Analysis of Immune Cells between CT26 Tumor Bearing Mice and Normal Mice

  • It has well studied that immune cells are strongly related to tumor progression and tumor suppression. To identify the difference of immune cell between tumor bearing mice and normal mice, we examined systemically the immune cell of CT26 tumor bearing mice on 21 days after tumor cell administration. As previously reported, CD4+ and CD8+ T cells population of tumor bearing mice significantly decreased 38% and 30% on day 21 compared to that of normal mice, respectively. All subpopulation of CD4 and CD8+ T cell significantly decreased, except CD49b+ T cell subpopulation. But, myeloid cell population ($CD11b^{high}$ and all Gr-1+ subpopulation) of tumor bearing mice significantly increased on day 21. Especially, all subpopulation of CD11b+Gr-1+ cell of tumor bearing mice significantly increased on day 21. Also, Foxp3+$CD25^{high}$ CD4 T cell (regulatory T cells) population significantly increased on day 21. These results suggest that tumor can induce the decline of T lymphocyte and the expansion of myeloid cells and regulatory T cells, and provide the basic information for the study of tumor immunology.

The CTG repeat polymorphisms of myotonic dystrophy (DM) gene in Korean population

  • Myotonic dystrophy (DM) is caused by the expansion of CTG trinucleotide repeat near the 3' end of the gene encoding for a member of protein kinase gene family (DMPK). The normal range of the CTG repeat was determined in 178 normal individuals (141 unrelated individuals and 37 of 9 families) by polymerase chain reaction (PCR), polyacrylamide gel electrophoresis and silver staining method. And the expansion of the CTG repeats in a DM family was analyzed with Southern analysis. In normal population, the range of CTG repeat is between 5 and 34 and 19 different alleles were observed in that range, and $(CTG)_{11-14}$ alleles were predominant. 4 members of an affected family showed the 0.5-2.0 kb size expansion of CTG repeats. In this study we could predict the incidence of DM in Korea as 1 in 20,000 and we could establish the diagnostic procedure for myotonic dystrophy.

ESTIMATING VARIOUS MEASURES IN NORMAL POPULATION THROUGH A SINGLE CLASS OF ESTIMATORS

  • This article coined a general class of estimators for various measures in normal population when some' a priori' or guessed value of standard deviation a is available in addition to sample information. The class of estimators is primarily defined for a function of standard deviation. An unbiased estimator and the minimum mean squared error estimator are worked out and the suggested class of estimators is compared with these classical estimators. Numerical computations in terms of percent relative efficiency and absolute relative bias established the merits of the proposed class of estimators especially for small samples. Simulation study confirms the excellence of the proposed class of estimators. The beauty of this article lies in estimation of various measures like standard deviation, variance, Fisher information, precision of sample mean, process capability index $C_{p}$, fourth moment about mean, mean deviation about mean etc. as particular cases of the proposed class of estimators.

A Sequential Approach for Estimating the Variance of a Normal Population Using Some Available Prior Information

  • Using some available information about the unknown variance $\sigma$$^2$ of a normal distribution with mean $\mu$, a sequential approach is used to estimate $\sigma$$^2$. Two cases have been considered regarding the mean $\mu$ being known or unknown. The mean square error (MSE) of the new estimators are compared to that of the usual estimator of $\sigma$$^2$, namely, the sample variance based on a sample of size equal to the expected sample size. Simulation results indicates that, the new estimator is more efficient than the usual estimator of $\sigma$$^2$whenever the actual value of $\sigma$$^2$ is not too far from the prior information.

Normative Measurements of Grip and Pinch Strengths of 21st Century Korean Population

  • Background Measuring grip and pinch strength is an important part of hand injury evaluation. Currently, there are no standardized values of normal grip and pinch strength among the Korean population, and lack of such data prevents objective evaluation of post-surgical recovery in strength. This study was designed to establish the normal values of grip and pinch strength among the healthy Korean population and to identify any dependent variables affecting grip and pinch strength. Methods A cross-sectional study was carried out. The inclusion criterion was being a healthy Korean person without a previous history of hand trauma. The grip strength was measured using a Jamar dynamometer. Pulp and key pinch strength were measured with a hydraulic pinch gauge. Intra-individual and inter-individual variations in these variables were analyzed in a standardized statistical manner. Results There were a total of 336 healthy participants between 13 and 77 years of age. As would be expected in any given population, the mean grip and pinch strength was greater in the right hand than the left. Male participants (137) showed mean strengths greater than female participants (199) when adjusted for age. Among the male participants, anthropometric variables correlated positively with grip strength, but no such correlations were identifiable in female participants in a statistically significant way. Conclusions Objective measurements of hand strength are an important component of hand injury evaluation, and population-specific normative data are essential for clinical and research purposes. This study reports updated normative hand strengths of the South Korean population in the 21st century.

인간정자의 처리에 있어서 Percoll과 Sil-Select 방법의 비교 (Comparison between Percoll and Sil-Select Methods on the Human Spermatozoa Treatment)

  • Objective: To evaluate silane-coated silica particles (Sil-select) as an alternative to polyvinylpyrrolidone-coated particles (Percoll) for gradient separation of spermatozoa, for use in assisted reproduction. Methods: 20 normal semen based on WHO criteria were included in this study. Recovery of motile and morphologically normal spermatozoa after using two-layer Percoll and Sil-select gradient respectively was recorded. Motility, HOST (hypoosmotic swelling test) and the detection of malondialdehyde for LPO (lipid peroxidation) after 24 h of incubation at $37^{\circ}C$ in a 5% $CO_2$ incubator were compared. Results: Percoll (78.5%) and Sil-select (79.1%) showed a significant increase in the motility compared to ejaculate (60.9%) but no difference between Percoll and Sil-select. Normal sperm morphology significantly increased after Percoll (57.6%) and Sil-select (53.7%) compared to ejaculate (35.8%) but no difference between Percoll and Sil-select. No differences in the recovery of motile spermatozoa and motility, HOST and the production of malondialdehyde after 24 h incubation were found when comparing the use of Percoll and Sil-select. Conclusion: Sil-select seems to be an attractive alternative to Percoll for sperm separation in assisted reproduction.

SSR 분자마커를 이용한 찰옥수수 및 종실용 옥수수 자식계통들의 핵심집단에 대한 유전적 다양성 및 집단구조 분석 (Analysis of Genetic Diversity and Population Structure for Core Set of Waxy and Normal Maize Inbred Lines using SSR Markers)

  • 본 연구는 총 50개의 SSR 마커를 이용하여, 찰옥수수 및 종실용 옥수수 핵심집단(찰옥수수 40계통, 종실용 옥수수 40계통)의 자식계통들의 유전적 다양성, 집단구조 및 계통유연관계를 분석하였다. 1. 그 결과 65bp에서 225bp 크기의 범위로 총 242개의 대립단편들을 증폭시켰다. SSR primer들에서 증폭된 대립단편의 수는 최소 2개에서부터 최대 9개까지의 범위로 나타났고, 평균 4.84개가 증폭되었다. 그리고 GD값은 0.420에서 0.854의 범위로 나타났고, 평균 0.654의 값을 나타내었다. 2. 80개의 옥수수 자식계통들의 집단구조를 분석한 결과, 13개의 찰옥수수 자식계통은 group I에 포함되었고, Group II는 7개의 찰옥수수 자식계통과 38개의 종실용 옥수수 자식계통들이 포함되었다. 나머지 22개의 자식계통들은 admixed group에 포함되었으며, 20개 찰옥수수 자식계통과 2개의 종실용 옥수수 자식계통으로 구성되어있다. 3. UPGMA법에 의한 계통유연관계 분석 결과, 80개 옥수수 자식계통들은 유전적 유사성 31.7% 수준에서 크게 3개의 그룹으로 나누어졌다. Group I은 40개의 찰옥수수 자식계통과 11개의 종실용 옥수수 자식계통을 포함하고 있었고, Group II는 27개의 종실용 옥수수 자식계통을, 그리고 Group III은 단지 2개의 종실용 옥수수 자식계통을 포함하고 있었다. 따라서 본 연구의 결과는 앞으로 강원도 농업기술원 옥수수시험장에서 육성한 찰옥수수 및 종실용 옥수수 자식계통들에 대한 유전자원 관리 및 선발 그리고 교배조합 구성 및 예측 등에 유용한 정보를 제공할 것으로 기대된다.
