  • A furanquinone substance(methyl 5-hydroxy-dinaphtho [1,2-2',3'] furan-7,12-dione-6-carboxylate) which has been isolated from the methanol extract of Paulownia tomentosa stem was synthesized by condensing 2,3-dichloro-1,4-naphthoquinone with methyl 1,4-dihydroxy-naphthalene-2-carboxylate in pyridine and identified by NMR, MS, UV, IR etc.

  • The 2,3-dichloro-1,4-naphthoquinone was reacted with diethyl methylmalonate and sodium amide in the toluene to yield 3-chloro-2-(1-methyl-1-diethoxycarbonyl)-methyl-1,4-naphthoq uinone(I). When this compound I was reacted with some alkylamines (methylamine, ethylamine, ethanolamine, 2-bromoethylamine, propylamine, isopropylamine, cyclohexylamine, benzylamine, 4-piperidylmethylamine), 3-methyl-2,3-dihydrobenz(f)indole-2,4,9-trione derivatives(IIa-i) were obtained via intramolecular cyclization.

  • 알킬화제로 작용할 수 있는 naphthoquinones인 fibrostatin이 Streptomyces catanulae subsp. griseospora 배양액으로부터 추출되었다. 이들 quinone 화합물의 합성은 이 화합물들의 생물학적 반응성 연구에 매우 중요하다. 본 논문에서는 효과적으로 치환체들의 위치를 조절할 수 있는 방법인 Hooker oxidation을 이용하여 fibrostatin B를 합성하였다.

  • Activity-guided fractionation of a hexane-soluble extract of the roots of Lithospermum erythrorhizon, using a mouse brain monoamine oxidase (MAO) inhibition assay, led to the isolation of two known naphthoquinones, acetylshikonin and shikonin, and a furylhydroquinone, shikonofuran E. These compounds were shown to inhibit MAO with $IC_{50}$ values of 10.0, 13.3, and $59.1 {\mu}M$, respectively. Although no specificity for MAO-A and MAO-B was shown by acetylshikonin and shikonin, a Lineweaver-Burk plot analysis indicated that the inhibition was competitive for both MAO-A and MAO-B activity.

  • Various analogues of 5,8-dimethoxy-1,4-naphthoquinone (DMNQ) such as 2- or 6-(1-hydroxyiminoalkyl)-DMNQs were prepared and evaluated for the antitumor action. (1 -Hydroxyiminoalkyl)-DMNQs derivatives expressed greater antitumor action than (1-hydroxyalkyl) - or acyl-DMNQ derivatives. Moreover, 6-(1-hydroxyiminoalkyl)-DMNQ derivatives expressed higher antitumor action than 2-substituted ones, suggestive of a steric effect. Some of 6-(1-propyloxyalkyl)-DMNQ derivatives with an alkyl group of butyl to octyl moiety showed T/C values of >400%

  • We identified a 1,134-bp putative type III polyketide synthase from the sequence analysis of Streptomyces peucetius ATCC 27952, named Sp-RppA, which is characterized as 1,3,6,8-tetrahydroxynaphthalene synthase and shares 33% identity with SCO1206 from S. coelicolor A3(2) and 32% identity with RppA from S. griseus. The 1,3,6,8-tetrahydroxynaphthalene synthase is known to catalyze the sequential decarboxylative condensation, intramolecular cyclization, and aromatization of an oligoketide derived from five units of malonyl-CoA to give 1,3,6,8-tetrahydroxynaphthalene, which spontaneously oxidizes to form 2,5,7-trihydroxy-1,4-naphthoquinone (flaviolin). In this study, we report the in vivo expression and in vitro synthesis of flaviolin from purified gene product (Sp-RppA).

  • The angular and planar heterocyclic compounds containing nitrogen, sulfur and oxygen were synthesized by reaction of 6,7-dichloro-5,8-quinoxalinedione with aromatic and aliphatic dinucleophiles. Nucleophilic reactivity was somewhat different between 2,3-dichloro-1,4-naphthoquinone and 6,7-dichloro-5,8-quinolinedione with dinucleophiles. The distribution of electron in heterocycle appeared to contribute to this difference. The intercalation comple of planar heterocyclic compound between GC/GC base pairs showed the optimum intercalation but the intercalation of angular heterocyclic compound was not good. Thus, the planar compound was expected to have antitumor activity.
