  • The manufacture of riboflavine fortified Dwen-Jang has been tried with an exceedingly riboflavine productive Aspergillus oryzae #612 mutant which has been developed by the authors. Both the rice and barley koji of this mutant and Aspergillus sojae have been prepared. Their riboflavine production, saccharifying and protease activities have begin compared The riboflavine fortified Dwen-Jang has been manufactured using the barley koji of riboflavine productive mutant. Their riboflavine content and qualities have been studied comparing with an ordinary Dwen-Jang which has been prepared with the barley kojo of A. sojae strain. The following results have been obtained. (1) The baley koji was superior in riboflavine production and protease activity, inferior in saccharifying ability than rice koji both with A. oryzae #612 and A. sojae. (2) In barley koji, the mutant, A. oryzae #612, produces 1.5 times riboflavine than A. sojae and shows stronger saccharifying and protease activities than the latter. (3) The riboflavine fortified Dwen-Jaug manufactured contained $5.2{\gamma}/g$ of riboflavine, about 1.5 times that of A. sojae. The higher contents of free sugar and free amjno nitrogen have been observed than the ordinary Dwen-Jang manufactured with A. sojae.

Phenotypic and Genotypic Analysis of Rice Lesion Mimic Mutants

  • Lesion mimic mutant (LMM) plants display spontaneous necrotic lesions on their leaves without any pathogenic infection. Specific rice LMMs designated as spotted leaf (spl) including spl1, spl3, spl4, spl5 and spl6 are genetically known as lesion resembling disease (lrd) mutant. The inheritance patterns in the $F_1$ and $F_2$ progenies of these mutants are controlled by recessive genetic factors. Lesion development in the rice LMMs were controlled by both development stages and environmental factors. The rice LMMs exhibited higher numbers of spots under $45^{\circ}C$ temperature than those under $30^{\circ}C$. Contents of chlorophyll were drastically reduced at 60 days old LMM leaves when the spot formation was severe. The levels of endogenous hydrogen peroxide were highest at 45 days old mutants but reduced at 60 days old. Transcription levels of stress related genes including thioredoxin peroxidase and protein disulfide isomerase were reduced in spotted leaves than those of non spotted leaves. It could be suggested that scavenging system against reactive oxygen species induced by either stresses or innate metabolisms may not work properly in the rice LMMs. As these rice LMMs autonomously expressed clear lesions of lrd phenotype without pathogen infection, it could be useful to understand stresses responses in plants.

A Histone Deacetylase, MoHDA1 Regulates Asexual Development and Virulence in the Rice Blast Fungus

  • Interplay between histone acetylation and deacetylation is one of the key components in epigenetic regulation of transcription. Here we report the requirement of MoHDA1-mediated histone deacetylation during asexual development and pathogenesis for the rice blast fungus, Magnaporthe oryzae. Structural similarity and phylogenetic analysis suggested that MoHDA1 is an ortholog of Saccharomyces cerevisiae Hda1, which is a representative member of class II histone deacetylases. Targeted deletion of MoHDA1 caused a little decrease in radial growth and large reduction in asexual sporulation. Comparison of acetylation levels for H3K9 and H3K14 showed that lack of MoHDA1 gene led to significant increase in H3K9 and H3K14 acetylation level, compared to the wild-type and complementation strain, confirming that it is a bona fide histone deacetylase. Expression analysis on some of the key genes involved in asexual reproduction under sporulation-promoting condition showed almost no differences among strains, except for MoCON6 gene, which was up-regulated more than 6-fold in the mutant than wild-type. Although the deletion mutant displayed little defects in germination and subsequent appressorium formation, the mutant was compromised in its ability to cause disease. Wound-inoculation showed that the mutant is impaired in invasive growth as well. We found that the mutant was defective in appressorium-mediated penetration of host, but did not lose the ability to grow on the media containing H2O2. Taken together, our data suggest that MoHDA1-dependent histone deacetylation is important for efficient asexual development and infection of host plants in M. oryzae.

OsDOR1, a novel glycine rich protein that regulates rice seed dormancy

  • Regulation of seed dormancy is important in many grains to prevent pre-harvest sprouting. To identify and understand the gene related to seed dormancy regulation, we have screened for viviparous phenotypes of rice mutant lines generated by insertion of Ds transposon in a Korean Japonica cultivar (Dongjin) background. One of the mutants, which represented viviparous phenotype, was selected for further seed dormancy regulation studies and designated dor1. The dor1 mutant has single Ds insertion in the second exon of OsDor1 gene encoding glycine-rich protein. The seeds of dor1 mutant showed a higher germination potential and reduced abscisic acid (ABA) sensitivity compared to wild type Dongjin. Over-expression of Dor1 complements the viviparous phenotype of dor1 mutant, indicating that Dor1 function in seed dormancy regulation. Subcellular localization assay of Dor1-GFP fusion protein revealed that the OsDor1 protein mainly localized to membrane and the localization of OsDOR1 was influenced by presence of a giberelin (GA) receptor OsGID1. Further bimolecular fluorescence complementation (BiFC) analysis indicated that OsDOR1 interact with OsGID1. The combined results suggested that OsDOR1 regulates seed dormancy by interacting with OsGID1 in GA response. Additionally, expression of OsDOR1 partially complemented the cold sensitivity of Escherichia coli BX04 mutant lacking four cold shock proteins, indicating that OsDOR1 possessed RNA chaperone activity.

Isolation and Characterization of a Ds-tagged liguleless Mutant in Rice (Oryza sativa. L)

  • A liguleless mutant, which showed complete loss of lamina joint region at the junction between leaf blade and leaf sheath, was isolated from a Ds insertional mutants derived from the source cultivar, Dongjin. This mutant could not affect other developmental patterns like phyllotaxis. Southern blot analysis, using GUS as a probe, revealed that the liguleless mutant contained three Ds copies transposed in the rice genome. Among the four genomic sequences flanking the Ds, one was mapped in the intergenic region (31661640 - 31661759), and the other two predicted a protein kinase domain (12098980 - 12098667) as an original insertion site within a starter line used for massive production of Ds insertional mutant lines. Another predicted and inserted in first exon of liguleless 1 protein (OsLG1) that was mapped in coding region (LOC_Os04g56170) of chromosome 4. RT-PCR revealed that the OsLG1 gene was not expressed liguleless mutants. Structure analysis of OsLG1 protein revealed that it predicted transcription factor with a highly conserved SBP domain consisting of 79 amino acids that overlapped a nuclear localization signal (NLS). RT-PCR revealed that OsLG1 is mainly expressed in vegetative organs.

DNA Microarray and Gene Ontology Enrichment Analysis Reveals That a Mutation in opsX Affects Virulence and Chemotaxis in Xanthomonas oryzae pv. oryzae

  • Xanthomonas oryzae pv. oryzae (Xoo) causes bacterial leaf blight (BLB) in rice (Oryza sativa L.). In this study, we investigated the effect of a mutation in opsX (XOO1056), which encodes a saccharide biosynthesis regulatory protein, on the virulence and bacterial chemotaxis of Xoo. We performed DNA micro-array analysis, which showed that 63 of 2,678 genes, including genes related to bacterial motility (flagellar and chemotaxis proteins) were significantly downregulated ($<\;-2\;log_2$ fold changes) by the mutation in opsX. Indeed, motility assays showed that the mutant strain was nonmotile on semisolid agar swarm plates. In addition, a mutant strain (opsX::Tn5) showed decreased virulence against the susceptible rice cultivar, IR24. Quantitative real-time RT-PCR reaction was performed to confirm the expression levels of these genes, including those related to flagella and chemotaxis, in the opsX mutant. Our findings revealed that mutation of opsX affects both virulence and bacterial motility. These results will help to improve our understanding of Xoo and provide insight into Xoo-rice interactions.

Growth regulation of cow1 rice mutant seedlings by blue light

  • We assessed whether the cow1 mutant defects are associated with growth of Tos17 and T-DNA insertional rice in blue light (BL). Growth of oscow1 mutants which encoded a member of the YUCCA protein family was retarded in BL. Root to shoot ratios of the mutants were reduced about 2 times lower in the absence of NAA and about 2.5 times lower in the presence of NAA; the shoot growth was not significantly changed by NAA addition. Photosynthetic activity of the mutants was however inhibited in high light. Pigment analysis showed significant difference between wild-type (Chl a:b = 3.02) and mutants (3.84). Carotenoid contents of the mutants were also decreased considerably, implying the involvement of cow1 in pigment formation. These findings lead us to suggest that the growth retardation of oscow1 mutant plants by BL results from the difference of photosynthetic activity in part.

DEB 처리에 의해 유도된 벼 돌연변이 집단으로부터 도열병 감수성 돌연변이 분리 (Isolation of Mutants Susceptible to Rice Blast from DEB-treated Rice Population)

  • 도열병은 곰팡이 균(Magnaporthe grisea)에 의해 발병되는 것으로, 벼 수확량에 가장 큰 손실을 일으킨다. 본 연구에서는 벼 도열병 저항성 신호전달 체계에 관여하는 유전자를 분리하기 위하여 DEB(1, 3-Butadiene diepoxide) 처리를 통하여 벼 도열병 저항성 품종인 RIl260의 돌연변이 2,000여 종을 생산하고, 이들로부터 병 저항성 변이 개체를 조사하였다. 돌연변이 집단에서 백변종 돌연변이의 비율은 6.7%로 매우 높았으며, 이것은 DEB 처리에 의해서 생산된 집단 내에 돌연변이가 높은 빈도로 발생하였음을 보여준다. 돌연변이 집단의 병 저항성 분석을 통하여 완전히 혹은 부분적으로 벼 도열병에 저항성을 상실한 29개의 돌연변이체를 분리하였다. 이들 중에서 가장 심한 이병성 라인으로 확인된 M5465는 DNA 혼성화 반응 분석을 사용하여 분석하였으며, RIL260 품종에서 도열병 병 저항성과 밀접한 관련을 갖고 있는 것으로 보고된 Pi5(t) 유전좌위의 DNA 표지들이 실험에 사용되었다. 이 결과들은 M5465에서 Pi5(t) 유전좌위 내부에 DNA의 큰 결손 및 재배열이 있었음을 보여준다. 분리된 병 저항성 돌연변이 라인들은 Pi5(t)에 의해 매개된 도열병 저항성의 신호 전달 과정을 이해하는 데 유용하게 사용 될 수 있을 것이다.

Amylose, Tocopherol, Free Sugar and Fatty Acid Content in Selected Mutant Lines of Oryza sativa cv. Shindongjin

  • To assess the potential as biofortified rice varieties, new endosperm and grain mutant lines were selected from $M_4$ generation seeds of the rice cultivar Shindongjin, which were either $\gamma$-irradiated or treated with N-methyl-N-nitrosourea(MNU) and lipid, sugar, and tocopherol content were analyzed. Amylose content in non-waxy mutants ranged from 8.8% in SM-4, a dull-type mutant, to 29.5% in SM-51, compared to 18.9% in the parental variety, Shindongjin. SM-23, a floury-type mutant, contained 0.09 ${\mu}g/g$ $\alpha$-tocopherol(40.9% of total tocopherol), was three times higher than in the parental variety. SM-32, a giant embryo-type mutant, had a 2.2-fold higher total tocopherol content, 2.1-fold higher $\alpha$-tocopherol, and 5.5-fold higher $\delta$-tocopherol content(47.3% of total tocopherol) than the parental variety(0.13 ${\mu}g/g$). Total free sugar content was elevated in all selected mutants and 1.2-8.6 times higher than in the parental variety(11.38 ${\mu}g/g$). These increased sugar levels were due to increase in sucrose concentration. SM-23(floury-type mutant) and SM-51(high amylose-type mutant) had 4.6 and 7.0 times more sugar, respectively, than the parental variety(11.38 ${\mu}g/g$). With relatively high concentrations, most mutants showed elevated fatty acid content in the SM 32(giant embryo-type) and SM-51(high amylose-type) mutants, at 124.56 and 89.59 mg/g, respectively. All selected mutants displayed valuable characteristics for the development of new varieties in rice-breeding programs.
