Phytochemical Constituents of the Root Bark from Morus alba and Their Il-6 Inhibitory Activity

  • Morus alba L., known as white mulberry, is a medicinal plant belongs to family Moraceae. It has long been used commonly in Ayurvedic for the treatment of lung-heat, cough, asthma, hematemesis, dropsy and hypertension. In the present study, seven prenylated flavonoids, along with four benzofuran compounds were isolated by means of repeated column chromatography. The structures of the known compounds were identified as kuwanon G (1), kuwanon E (2), kuwanon T (3), morusin (4), sanggenon A (5), sanggenon M (6), sanggenol A (7), moracin R (8), mulberofuran G (9), mulberofuran A (10) and mulberofuran B (11), by comparing their spectroscopic data with those reported in the literature. For these isolates, containing trace compounds, the inhibitory activity against IL-6 production in $TNF-{\alpha}$ stimulated MG-63 cells was examined. All isolated compounds (1 - 11) showed excellent inhibitory activity against IL-6 production in $TNF-{\alpha}$ stimulated MG-63 cells. Especially this study is first time to report that sanggenon A (5), sanggenon M (6), sanggenol A (7), mulberofuran G (9), mulberofuran A (10) and mulberofuran B (11) showed the inhibitory activity of IL-6 production. Our study suggested the possibility of anti-inflammatory regulation by compounds (1 - 11) isolated from M. alba.

강원도 일대에서 채취한 야생뽕의 시기별 뽕잎과 오디의 항산화능 분석 (Antioxidant Capacity Analysis of Water-Soluble Substances according to Maturity Stages in Yield-type Mulberry Leaves and Fruits Collected from Kang-Won Province)

  • 야생 뽕나무의 자원화 및 이용기술을 개발하기 위해 강원도 원주, 인제, 양양 3개 지역에서 성숙 단계별로 채취한 야생 뽕나무를 대상으로 뽕잎과 오디의 항산화능을 비교분석 하였다. 항산화능 측정장치(munilum L-100, ABCD GmbH) 및 ARAW-KIT(anti-radical ability of water-soluble substance)를 사용하였으며, ascorbic acid를 표준물질로 사용하여 calibration curve를 작성하였다. 각각의 지역에서 채취한 야생뽕의 형태학적 특성을 조사한 결과, 원주 지역 채집 개체는 백상형(Morus alba L.), 인제 지역 채집 개체는 산상형(Morus bombycis Koidz) 및 양양 지역 채집 개체는 노상형(Morus Lhou (Ser.) Koidz)의 뽕나무로 분류되었다. 강원도 원주, 인제 및 양양 지역에서 자생하고 있는 야생 뽕나무의 뽕잎에 대한 전체적인 항산화능을 분석한 결과 춘기 5개엽기의 어린 뽕잎의 항산화능이 가장 높았으며, 시기가 경과함에 따라 뽕잎의 항산화능은 점차 감소하는 경향이었다. 그러나 개체마다 각각의 특성을 나타냈다. 오디가 결실된 야생 뽕나무 중 오디의 수량성 및 크기 등 착과상태를 고려하여 이용가치가 있을 것으로 예상되는 개체는 No 7, 8, 10이었다. 산상형의 특징을 갖는 No. 7 개체 뽕나무는 뽕잎의 항산화능이 높을 뿐 만 아니라 뽕나무의 생육 상태 및 오디의 결실 상태도 양호했으므로 기능성 및 오디생산용 육종 유전자원으로 이용할 가치가 있는 것으로 판단되었으며, No. 8 및 No. 10 개체 뽕나무는 오디 생산을 위한 유전자원으로 이용할 수 있을 것으로 보인다.

뽕나무(Morus alba L.) 품종별 오디의 영양 및 기능성 성분과 이화학적 품질 특성 비교 (Comparison of Nutritional and Functional Constituents, and Physicochemical Characteristics of Mulberrys from Seven Different Morus alba L. Cultivars)

  • 현재 국내에서 재배되는 대표적인 뽕나무(Morus alba L.) 품종별 오디의 영양 및 기능성성분의 함량과 이화학적 품질 특성을 측정하여 비교하였다. 오디 일반성분의 함량은 수분 83.93~86.27%, 가용성 무질소물 8.63~11.76%, 조단백질 1.03~2.54%, 조지방 0.33~0.92%, 조섬유 0.89~2.87% 및 조회분 0.76~0.98% 범위로 익수뽕, 과상2호 및 청일뽕은 다른 품종(대성뽕, 용뽕, 대자뽕, 과상1호)보다 가용성 무질소물 함량이 높은 반면, 단백질, 지방 및 섬유소 함량이 낮았다. 총 당 및 환원당 함량은 과상2호, 청일뽕 및 익수뽕이 높은 반면, 과상1호 및 용뽕은 가장 낮게 나타났다. 총 폴리페놀 및 총 플라보노이드 함량은 과상1호, 대성뽕 및 용뽕이 다른 품종보다 높았으며, 총 안토시아닌 함량은 과상1호 및 2호, 용뽕, 대성뽕이 높았다. 또한, 오디의 anthocyanin 구성성분 즉, C3G 및 C3R 함량은 대성뽕, 과상1호 및 과상2호가 모두 높았다. 한편, 오디의 4가지 플라보노이드 즉, rutin, isoquercitrin, quercitrin, quercetin 함량은 품종에 따라 다소 차이가 있었으며, 과상2호가 2가지 주된 플라보노이드인 rutin 및 quercetin 함량이 높은 반면, 익수뽕 및 청일뽕은 낮았다. 오디 품종별 piceid 및 4-PM 함량을 측정한 결과, piceid 함량은 대성뽕 및 용뽕이 높았으며, 대자뽕, 익수뽕 및 청일뽕은 검출되지 않았다. 한편, 항당뇨 성분인 4-PM은 청일뽕 및 대성뽕이 높은 반면, 그 외 오디 품종들은 함량이 비슷하였다. 항고혈압성 GABA 함량은 청일뽕, 대성뽕 및 과상2호가 높은 반면, 익수뽕 및 용뽕은 낮았다. 오디 품종별 이화학적 품질 특성 중 과실의 중량 2.11~6.38 g, pH 4.08~4.80, 산도 0.39~0.73%, 당도 5.83~15.34oBrix, 색도 L값 26.31~31.49, a값 1.04~3.85, b값 -0.02~4.09 범위로 품종간에 유의적인 차이를 보였다. 특히 오디 품종 중 익수뽕 및 과상2호는 열매의 크기가 적당히 크고 다른 품종보다 산도는 다소 낮은 반면, 당도는 13~15%로 높았다.

품종이 다른 오디(Morus alba L.)의 성인병 관련 효소 억제효과 (Inhibitory Effects of Various Mulberry Fruits (Morus alba L.) on Related Enzymes to Adult Disease)

  • 생리활성을 갖는 새로운 기능성 물질을 개발할 목적으로 9종의 오디(Morus alba L.)를 이용하여 물과 60% 에탄올 추출물을 제조하고 이들의 angiotensin converting enzyme 활성억제효과, xanthine oxidase 활성억제효과, ${\alpha}$-amylase 활성억제효과 및 ${\alpha}$-glucosidase 활성억제효과를 탐색하여 본 결과, 페놀함량이 청일뽕과 강원3호 품종의 오디에서 물추출물에서 청일뽕과 강원3호 두품종이, 에탄올 추출물에서 대엽조생, 청일뽕, 강원3호, 희학 및 카타네오 등 5 품종이 2 mg/g 이상의 phenolic compound 함량을 나타내었으며, angiotensin converting enzyme 활성억제효과는 물 추출물에서 백운3호가 $90.9{\pm}4.5%$, 에탄올 추출물에서는 희학 품종이 $81.8{\pm}4.5%$의 활성억제효과를 나타내었다. 또한 xanthine oxidase 활성억제효과는 물 추출물에서 국상20호가 약 10% 정도, 에탄올 추출물에서는 카타네오가 $21.4{\pm}2.3%$의 활성억제효과를 나타내었다. 당분해활성에 관여하는 ${\alpha}$-amylase와 ${\alpha}$-glucosidase 활성억제효과를 측정해 본 결과 물추출물에서만 대엽조생, 수원뽕, 청일뽕, 강원3호, 희학 및 국상20호 등 6품종이 100%의 ${\alpha}$-amylase 저해효과가 관찰되었으며, 백운3호, 대엽조생, 청일뽕 및 희학은 물추출물에서, 대축면, 강원3호 및 국상20호는 에탄올 추출물에서 20% 이상의 ${\alpha}$-glucosidase 저해효과가 확인되었다. 이상의 결과, 높은 생리활성기능을 나타내는 여러 종의 오디의 물과 에탄올 추출물들은 항고혈압 및 항당뇨 효과가 있는 기능성 식품소재로 활용할 수 있을 것으로 판단되었다.

The Neuroprotective Potential of Cyanidin-3-glucoside Fraction Extracted from Mulberry Following Oxygen-glucose Deprivation

  • In this study, cyanidin-3-glucoside (C3G) fraction extracted from the mulberry fruit (Morus alba L.) was investigated for its neuroprotective effects against oxygen-glucose deprivation (OGD) and glutamate-induced cell death in rat primary cortical neurons. Cell membrane damage and mitochondrial function were assessed by LDH release and MTT reduction assays, respectively. A time-course study of OGD-induced cell death of primary cortical neurons at 7 days in vitro (DIV) indicated that neuronal death was OGD duration-dependent. It was also demonstrated that OGD for 3.5 h resulted in approximately 50% cell death, as determined by the LDH release assay. Treatments with mulberry C3G fraction prevented membrane damage and preserved the mitochondrial function of the primary cortical neurons exposed to OGD for 3.5 h in a concentration-dependent manner. Glutamate-induced cell death was more pronounced in DIV-9 and DIV-11 cells than that in DIV-7 neurons, and an application of $50{\mu}M$ glutamate was shown to induce approximately 40% cell death in DIV-9 neurons. Interestingly, treatment with mulberry C3G fraction did not provide a protective effect against glutamate-induced cell death in primary cortical neurons. On the other hand, treatment with mulberry C3G fraction maintained the mitochondrial membrane potential (MMP) in primary cortical neurons exposed to OGD as assessed by the intensity of rhodamine-123 fluorescence. These results therefore suggest that the neuroprotective effects of mulberry C3G fraction are mediated by the maintenance of the MMP and mitochondrial function but not by attenuating glutamate-induced excitotoxicity in rat primary cortical neurons.

Antioxidant Activities and Phenolic Compounds Composition of Extracts from Mulberry (Morus alba L.) Fruit

  • The objective of this research was to evaluate the ability of water and ethanol extracts from mulberry fruit (Morus alba L.) to influence the inhibitory activity of angiotensin converting enzyme (ACE) and xanthine oxidase(XOase). The total phenol contents and sixteen phenolic compounds were investigated in water and ethanol extracts. In order to understand the factors responsible for the potent antioxidant and antihypertensive ability of mulberry, it has been evaluated for anti-oxidative activity using Fenton's reagent/ethyl linoleate system and for free radical scavenging activity using the 1,1-diphenyl-2-picryl hydrazyl free radical generating system. The total phenol contents and total of phenolic compounds in ethanol extract showed higher levels than water extract in mulberry fruit six phenolic compounds (chlorogenic acid, narigin, syringic acid, quercetin, naringenin, kampferol) has a higher individual phenolic compound content in the 60% ethanol extraction than 80% ethanol extract. The inhibitory activity on angiotensin converting enzyme (ACE) were highest in 80% ethanol extract (9.0%). Also, activity of xanthine oxidase(XOase) inhibition appeared highest in 80% ethanol extracts and correlated well with the total phenolic content, which was modulated by the concentration of individual phenolic compounds. This result revealed, that strong biological activity was caused by specific phenol compound contents. Utilization of water and ethanol extracts from mulberry fruit are expected to be good candidate for development into source of free radical scavengers and anti-hypertentive activity

Quali-Quantitative Analysis of Flavonoids for Mulberry Leaf and Fruit of 'Suhyang'

  • BACKGROUND: Globally, mulberry (Morus sp.) is exploited for feeding leaf to silkworms in order to obtain silk fiber or for animal feedstock production. Also, mulberry fruit is known to a by-product that was produced from mulberry tree after harvesting leaves for silkworm rearing, as a yield and consumption of mulberry fruit was increased, it has been fixing to a newincome crop. Mulberry leaves and fruits are used for the health benefits of human beings. Mulberry contains various bioactive components, such as alkaloids and flavonoids. Mulberry flavonoids are an important part of the diet because of their effects on human nutrition. The flavonoids in mulberry leaf and fruit of 'Suhyang'(Morus alba L.) were determined. METHODS AND RESULTS: Flavonoids for mulberry leaf and fruit of 'Suhyang' were analysed using ultrahigh performance liquid chromatography coupled with diode array detection and quadrupole time-of-flight mass spectrometry (UPLC-DAD-QTOF/MS)technique. An UPLC-DAD-QTOF/MS system was used, and identification of mulberry leaves constituents was carried out on the basis of the complementary information obtained from LC spectra, MS ions, and MS/MS fragments. The mulberry leaf (16 flavonoids) and fruit (9 flavonoids) were isolated and analyzed from Suhyang using UPLC-DAD-QTOF/MS chromatogram. To the best of our knowledge, Quercetin 3-O-(6"-O-malonyl) glucoside and quercetin 3-O-rutinoside (rutin) was detected on the highest content in leaf and fruit, respectively and further research will be devoted to evaluate their biological activity. CONCLUSION: Obtaining information about the concentration of functional materials in mulberry leaves could contribute to the development and promotion of processed, functional products and offer possible industrial use of 'Suhyang', holding promises to enhance the overall profitability of sericulture.

상심자추출물 등 복합물의 퇴행성관절염 억제효과 (Inhibitory Effects of Complex of Mulberry Extract on Degenerative Arthritis)

  • Complex of mulberry extract (CME) is composed of extracts of mulberry (Morus alba L.) fruit, mulberry leaves and black beans (Glycine max (L.) Merr.). In this study, we investigated prevention effects of CME on degenerative arthritis. The $SC_{50}$ value of DPPH radical scavenging by CME was $158.49{\pm}11.35{\mu}/mL$. We found that CME significantly reduced the production of nitric oxide (NO) and protein expression of cyclooxygenase-2 (COX-2) in RAW 264.7 cells which were activated by LPS. Experiments using animal model of degenerative arthritis showed that CME (400 mg/kg body weight) inhibited the production of TNF-${\alpha}$ (77.5%) and IL-$1{\beta}$ (95.0%). Furthermore, it was observed that CME reduced to 85.9% of paw edema induced by carrageenan. These results suggest that CME could improve degenerative arthritis.
