  • 채소류(菜蔬類)의 시설재배(施設栽培)에서 수경재배(水耕栽培)를 대체(代替)할수있는 청정한 배양물질(培養物質)로서 왕겨훈탄의 활용방안을 구명(究明)하기 위하여 산의 종류를 달리하여 왕겨훈탄을 중화(中和)시킨후 몇가지 채소류를 재배하여 생장반응(生長反應)을 비교 검토 하였다. 염산(鹽酸)을 제외한 산(酸) 중화훈탄(中和燻炭)은 채소류(菜蔬類) 재배(栽培)를 위한 배양물질(培養物質)로 활용할수 있을만큼 정상적인 생육(生育)을 보였으며 산(酸) 중화(中和) 처리에 의해 질산태질소의 부가적 공급효과(供給效果)가 있는 질산중화훈탄 처리는 배추, 상치, 및 시금치의 생장량이 퇴비상토 처리보다 양호(良好) 하였다. 염산중화훈탄 처리에서는 특히 상치 및 시금치에서 생육후기(生育後期)에 장해증상(障害症狀)을 보이며 가장 부진한 생육을 나타냈다. 산 중화훈탄 처리에서 자란 식물체중(植物體中)의 화학성분(化學成分)들은 퇴비상토의 경우와 거의 비슷한 함양분포(含量分布)를 보였으며 단백태질소(蛋白態窒素)함량은 오히려 산(酸) 중화훈탄(中和燻炭) 처리들에서 더 높았다. 따라서 채소류(菜蔬類)의 생장과 수확기의 화학성분(化學成分)등을 고려(考慮)할때 질산 중화훈탄은 시설(施設) 채소재배(菜蔬栽培)를 위한 청부(淸浮)한 배양물질(培養物質)로서 활용 가능한 것으로 생각된다.

Agastache rugosa Leaf Extract Inhibits the iNOS Expression in ROS 17/2.8 Cells Activated with TNF-$\alpha$ and IL-$\beta$

  • It has been suggested that nitric oxide (NO) derived from inducible nitric oxide synthase (iNOS) may act as a mediator of cytokine-induced effects on bone turn-over. NO is also recognized as an important factor in bone remodeling, i.e., participating in osteoblast apoptosis in an arthritic joint. The components of Agastache rugosa are known to have many pharmacological activities. In the present study, we investigated the effects of Agastache rugosa leaf extract (ELAR) on NO production and the iNOS expression in ROS 17/2.8 cells activated by a mixture of inflammatory cytokines including TNF-$alpha$ and IL-1$\beta$. A preincubation with ELAR significantly and concentration-dependently reduced the expression of iNOS protein in ROS 17/2.8 cells activated with the cytokine mixture. Consequently, the NO production was also significantly reduced by ELAR with an IC$_{50}$ of 0.75 mg/mL. The inhibitory mechanism of iNOS induction by ELAR prevented the activation and translocation of NF-$\kappa$B (p65) to the nucleus from the cytosol fraction. Furthermore, ELAR concentration-dependently reduced the cellular toxicity induced by sodium nitroprusside, an NO-donor. These results suggest that ELAR may be beneficial in NO-mediated inflammatory conditions such as osteoporosis.

파밤나방(Spodoptera exigua)에 대한 곤충병원세균류 배양액의 곤충면역억제활성 및 비티 생물농약과 혼합효과 (Immunosuppressive Activity of Cultured Broth of Entompathogenic Bacteria on the Beet Armyworm, Spodoptera exigua, and Their Mixture Effects with Bt Biopesticide on Insecticidal Pathogencity)

  • 곤충병원선충으로부터 분리된 곤충병원세균들(Xenorhabdus nematophila, X. sp. and Photorhabdus temperata subsp. temperata)은 곤충혈강에서 높은 살충효과를 보유하고 있다. 이들 세균들은 또한 phospholipase $A_2$ 효소 작용을 억제하여 면역중개반응을 무력화하여 곤충기주의 면역억제를 유도하게 한다. 본 연구는 이들 세균이 성장된 배양액이 이러한 면역억제 활성을 보유하는 지를 분석하였다. 이를 위해 $0.2\;{\mu}m$ 공극 크기의 여과막을 이용하여 세균배양액으로부터 세균을 제거시켰다. 이렇게 멸균된 세균배양액은 배양액 농도에 따라 뚜렷하게 파밤나방(Spodoptera exigua) 5령충의 혈구세포 활착행동을 억제시켰다. 또한 이러한 억제효과는 세균별로 차등을 보였으며, 이 가운데 X. nematophila의 배양이 가장 높았다. 이 세균의 면역억제 활성 성질을 Bacillus thuringiensis (Bt)의 감염력 제고에 적용하였다. 세 세균 모두의 배양액(세균 포함)들은 낮은 농도의 Bt와 혼합하여 섭식 처리할 경우 어린 파밤나방 유충에 대해서 뚜렷하게 Bt 감염력을 제고시켰다. 다시 이 세균을 제거한 멸균된 배양액을 낮은 Bt농도 처리와 혼합하여 섭식 처리하였다. 이때 X. nematophila의 배양액만이 Bt의 감염력을 제고시켰다. 본 연구는 이들 세 곤충병원세균의 배양액에 곤충의 면역억제물질이 포함되었음을 보였으며, 이들 물질을 Bt와 혼용하였을 때 Bt의 살충력을 제고시킬 수 있음을 나타냈다.

광산배수 내 황산염 저감을 위한 생물반응기의 설계인자 도출 연구 (Study on Determination of Design Factor of Bioreactor for Sulfate Reduction in Mine Drainage)

  • 황산염 함량이 높은 광산배수를 대상으로 황산염환원균(Sulfate reducing bacteria, SRB)을 이용한 생물반응기의 설계인자를 도출하고자 컬럼실험을 수행하였다. 생물반응기의 기질물질로 우분을 기본으로 하여 버섯퇴비를 혼합하고, 보조제로써 볏짚, 석회석을 각각 혼합하여 적용하였다. 우분(70%), 버섯퇴비(10%), 볏짚(20%)을 기질물질로 사용하였을 때 최대 82%의 용존된 총 황 제거효율을 보였고, 체류시간은 2일 일 때 높은 효율을 보였다. 생물반응기 내 광산배수의 흐름이 상향류 일 때 배출수 내 환원형태의 황이 산소와 직접 접촉하면서 재산화되어 제거효율이 감소하는 것으로 나타났다. 황산염환원에 따른 금속 황화물 형성을 유도하기 위한 무기성 슬러지의 주입은 비소의 환원성 용해 및 SRB에 독성을 발현하여 생물반응기의 장기적 운영을 저해할 수 있다. 연구결과, 우분을 포함한 혼합기질물질을 활용한 하향류의 생물반응기가 황산염 및 용존된 총 황을 효과적으로 제거하였고, 이는 황산염 함유량이 높은 염수의 광산배수에서 적용이 가능할 것으로 판단된다.

종양이식 생쥐모델에서 도두(刀豆), 우방근(牛蒡根) 추출물의 대장암 억제 효과 (Suppression of colon cancer by administration of Canavalia gladiata D.C. and Arctium lappa L., Redix extracts in tumor-bearing mice model)

  • Objective : In the present study, we examined whether Canavalia gladiata D.C. (CG) and Arctium lappa L., Redix (AL) mixture (CGAL), their components, lupeol and chicoric acid, regulate immune system and suppress the tumor in vitro and in vivo. Methods : LPS-induced reactive oxygen species (ROS) and nitric oxide (NO) were measured after treatment with CG extract (CGE), CGAL, lupeol, chicoric acid and lupeol and chicoric acid mixture (lupeol+CA) in Raw264.7 cell. To determine the effect of CGE on immune responses, immune cell population and IgG production were assessed in mice. To investigate the effect of CGAL and their component on anti-tumor activity, tumor volume and weight were measured, cell cycles and immune cell population were analyzed in MC38 injected tumor bearing mice. Also, NK cell activity was determined in splenocyte isolated from tumor bearing mice. Results : CGE, CGAL, lupeol, chicoric acid and lupeol+CA decreased the LPS-induced ROS and NO production without cell toxicity in RAW264.7 cells. CGE increased the immune cell populations of $CD4^+T$, $CD8^+T$ and macrophages in various immune organ of mice. In tumor bearing mice, CGAL, lupeol, chicoric acid and lupeol+CA suppressed tumor volume and weight. In cell cycle analysis, they decreased the percentages of S phase. In addition, CGAL, lupeol, chicoric acid and lupeol+CA immune cell populations of $CD4^+T$, $CD8^+Tcell$, NK cell and macrophage in tumor as well as NK cell activity. Conclusion : CGAL and its compounds may enhance immune responses and suppress tumor growth, and may be capable of developing health functional foods.

The Estrogenicity and Reproductive Toxicity by Combined Treatment of Bisphenol A and Benzyl butyl phthalate during Gestation, Lactation Period in Rats

  • The co-administration of BPA and BBP induced slow weight gain compared with single administration in dams. Also, such mixture induced low neonatal body weights in next generation. The dams treated with BPA and BBP showed significant organ weight changes in liver, spleen exposed during lactational periods. But the dams exposed during lactational periods showed significant organ weight changes not only in liver, spleen but also in kidney, uterus and ovary. The F1 female rats exposed during lactation periods showed significant organ weight changes in liver, spleen, ovary. The F1 male rats showed significant organ weight changes in liver, kidney, epididymis, vesicular glands, prostate. However no clear synergistic effects of BPA and BBP could be found. Estrogen receptor ${\alpha}$ expression by BPA and BBP in the uterus(dam, F1 female) and testis(F1 male) were studied. There was no significant different $ER{\alpha}$ expression pattern between control and treated groups. But $ER{\alpha}$ expression were increased in F1 male testis and female uterus. F1 male showed distinct $ER{\alpha}$ expression, especially in the group of lactational combined exposure. Synergistic $ER{\alpha}$ expression was found by combined treatment of BPA and BBP.

콩 함유 올리고당의 기능적 특성 (Functional Characteristics of Soybean Oligosaccharide)

  • To enjoy a healthy life, it is important to have a well-balanced diet. However, in today's society, there is an increase in the consumption of preprocessed foods and frequency of eating out. Also the western diet, which is becoming move popular worldwide, contains relatively high levels of protein and fat, and a low amount of fiber, Furthermore, the increased availability of favorite foods has created a condition were the individual diet is less variable. With these conditions, it is difficult to maintain a diet that is nutritionally balanced. With these unbalanced diets, which are difficult to change, there has been an increase in adult disease and health problems, such as colon and breast cancer, It is speculated that metabolites for carcinogens are produced from diet components and that intestinal bacteria contribute to the production of these metabolites. Therefore, it is necessary to evaluate the relationships between health, diet, and intestinal microflora. Soybean oligosaccharide is composed of water-soluble saccharides that have been extracted from soybean whey, a by-product from the production of soy protein. This is mainly a mixture of mono-, di-, tri-, and tetrasac-charides, with the principle components being the oligosaccharide raffinose and stachyose. When consumed by humans, the oligosaccharides cannot be digested in the human duodenal and small intestinal mucosa, and these are selectively utilized by beneficial bifidobacteria in intestines. The results of acute and subacute toxicity tests, soy-bean oligosaccharides were nonpoisonous. Soybean oligosaccharides promote the growth of indigenous bifido-bacteria in the colon which by their antagonistic effects, suppress the activity of putrefactive bacteria. Also, they reduce toxic metabolites, detrimental enzymes and plasma lipid, and increase in the frequency of bowel evacuation and fecal quantities. Consequently, soybean oligosaccharides as functional foods components have potential roles in the prevention and medical treatment of chronic adult diseases. The study of processing property and physiological function of soybean oligosacchavides and development of high oligosaccharide variety allow the creation of new and exciting foodstuffs that aye functional healthy.

Genotoxicity on Structural Derivatives of Sophoricoside, a Component of Sophora Japonica, in Bacterial and Mammalian Cells

  • To develop the novel anti-allergic drug, many sophoricoside derivatives were synthesized. Among these derivatives, JSH-II-3, VI-3, VII-3, VIII-3, VII-20 and VII-20 (sodium salt) were selected and subjected to high throughput toxicity screening (HTTS) because they revealed strong IL-5 inhibitory activity and limitation of quantity. Single cell gel electrophoresis (Comet) assay, mouse lymphoma thymidine kinase ($tk^{+/-}$) gene assay (MOLY), chromosomal aberration assay in mammalian cells and Ames reverse mutation assay in bacterial system were used as simplified, inexpensive, short-term in vitro screening tests in our laboratory. Through the primary screening using the comet assay, we could choose the first candidates of sophoricoside derivatives with no genotoxic potentials as JSH-VI-3, VII-3, VII-20 and VII-20 (sodium salt). Also JSH-VII-3, VII-20 and VII-20 (sodium salt) are non-mutagenic in MOLY assay, while JSH-II-3 is mutagenic at high concentration with the presence of metabolic activation system in both comet assay and MOLY assay. The selected derivatives (JSH-VI-3, VII-3, VII-20 and VII-20 (sodium salt) are not mutagenic in S. typhimurium TA98 and TA100 strains both in the presence and absence of metabolic activation. From results of chromosomal aberration assay, 6 h treatment of JSH-VI-3, VII-3 and VII-20 (sodium salt) were not revealed clastogenicity both in the presence and absence of S-9 mixture. Therefore, we suggests that JSH-VI-3, VII-3, VII-20 and VII-20 (sodium salt), as the optimal candidates with both no genotoxic potential and IL-5 inhibitory effects must be chosen. To process the development into new anti-inflammatory drug of these derivatives, further investigation will need.

Evaluation of the Genetic Toxicity of Synthetic Chemicals [XII] -in vitro Chromosomal Aberration Assay with 11 Chemicals in Chinese Hamster Lung Fibroblast-

  • The validation of many synthetic chemicals that may pose a genetic hazard in our environment is of great concern at present. Since these substances are not limited to the original products, and enter the environment, they have become widespread environmental pollutants, thus leading to a variety of chemicals that possibly threaten the public health. In this respect, the regulation and evaluation of the chemical hazard playa very important role to environment and human health. The clastogenicity of 11 synthetic chemicals was evaluated in Chinese hamster lung (CHL) fibroblast in vitro. Benzoyl chloride (CAS No. 98-88-4) induced chromosomal aberrations with statistical significance at the concentration of 31-123 $\mug/ml$ and 43 $\mug/ml$ in the absence and presence of S-9 metabolic activation system, respectively. 2-Propyn-l-o1 (CAS No. 107-19-7) and 2-Phenoxy ethanol (CAS No. 122-99-6) revealed clastogenicity only at the highest concentration in the presence of S-9 mixture. However, 1-naphthol (CAS No. 90-15-3) which is one of the most cytotoxic chemical among 11 chemicals tested revealed no clastogenicity both in the presence and absence of S-9 metabolic activation system. From the results of chromosomal aberration assay with 11 synthetic chemicals in CHL fibroblast in vitro, Benzoyl chloride (CAS No. 98-88-4), 2-Propyn-l-01 (CAS No. 107-19-7) and 2-Phenoxy ethanol (CAS No. 122-99-6) revealed positive clastogenic results in this study.

Clostridium botulinum의 분포 및 수종 식품에서의 botulinum toxin 생성능 비교연구 (Incidence of Clostridium botulinum in Natural Reservoirs and Toxin Production in some foodservice)

  • 토양을 비롯하여 어류, 패류, 조류, 포유류의 소화기관으로부터 Clostridium botulinum 분리를 시도하였다. 총 158개 분리원을 screening한 결과 10개 시료로부터 Clostridium botulinum 분포 가능성을 확인하였으며 6개 시료로부터 Clostridium botulinum을 순수 분리하여 3.8% 분리율을 나타내었다. 분리 균주의 형태적 특징, 배양상의 특성 및 생화학적 특성 등을 표준 균주의 특성과 비교하고 항혈청에 의한 중화시험을 실시하여 분리균주를 동정하였다. Egg york agar에서의 opalescence 생성, 탄수화물 이용성, Egg york GAM 배지상에서의 pearly layer 생성 등으로부터 Clostridium botulinum으로 동정할 수 있었으며 trypsin에 의한 toxicity 활성화, type E 항혈청에 의한 opalescence 생성억제 및 mouse 방어효과가 인정되어 type E 로 동정하였다. 국내에서 시판되고 있는 수 종의 식품을 대상으로 Clostridium botulinum 의 toxin 생성능을 비교하였던 바 식품의 종류, 사용균주에 따라 toxin 생성량에 현저한 차이가 있었다. 분리균주 type E 의 경우 어패류통조림, ham 식품에서 많은 양의 toxin 이 생성되었으며 sausage, 과일통조림 식품에서는 비교적 적었다. 그러나 type A 의 경우에는 어패류, ham , sausage 식품에서 상당량의 toxin 이 생성되었으며 과일통조림에서도 비교적 맣은 양의 toxin이 생성되었다 .
