Studies on the canine ringworm by Microsporum canis and carrier state of dermatophytes in canine and feline (Microsporum canis에 의한 개의 피부사상균증 및 개, 고양이의 피부사상균의 보균상황)

  • This study was performed to examine the distribution of dermatophytes in asymptomatic 454 dogs and 106 cats and in 19 dogs with skin lesions in Taegu The results obtained were summarized as follows ; The causative agent was identified a Microsporum(M) canis in 4 dogs with skin lesions. The isolation rates of dermatophytes were 10.1%(46/454) in asymptomatic dogs and 33.0%(35/106) in asymptomatic cats. The isolation rates of dermatophytes in dogs and cats were lower at house than that at pet cannel and animal care center, and that in females were higher than that in males. From dogs, 43(93.5%) of M canis and 3(6.5%) of M gypseum, and from cats, 33(94.2%) of M canis, 1(2.9%) of M gypseum and M nanum were isolated.

Investigation of Active Antifungal Compounds of Essential Oil from Chamaecyparis obtusa Against Dermatophytes, Microsporum canis and Trichophyton Mentagrophytes (피부사상균 Microsporum canis 및 Trichophyton mentagrophytes에 대한 편백정유의 항진균활성물질 탐색)

  • The present study was conducted to evaluate the application of Chamaecyparis obtusa and to investigate potential utilization of essential oil from C. obtusa as plant-based medicine. The antifungal activity of essential oil from leaves and twigs of C. obtusa (Sieb. Et Zucc) was determined and the major components of active fractions against Microsporum canis (KCTC6591) and Trichophyton mentagrophytes (KCTC6077) were identified by GC/MS analysis. In treatment of essential oil from C. obtusa, the strain M. canis was more resistant than the other, T. mentagrophytes. In the agar diffusion assay, essential oil from C. obtusa inhibited hyphal growth of M. canis and T. mentagrophytes at the concentration of more than 5,000 ppm. The zones named B and C in the TLC assay of essential oil from C. obtusa showed antifungal activities. Among four sub-fractions of n-hexane extract from B and C zones, named as B-1, B-2, C-1 and C-2, the C-2 showed the highest antifungal activity. Instrumental GC/MS analysis for sub-fractions showed that a major component of C-1 was ${\alpha}$-terpineol as terpene alcohol, while C-2 contained sesquiterpenes such as elemol, cedrol and eudesmol.

Effects of Chinae Rhizoma Extracts on the Growth of the Dermatophytes (토복령알코홀추출물의 항진균작용)

  • The majority of drugs used in the treatment of superficial fungal infections has limited values due to its low efficacy or development of resistance. For the purpose of searching efficacious agent on the superficial fungal infections induced by dermatophytes which is regarded as the most malicious one, authors examined whether Chinae Rhizoma Extracts have significant on it. Extracts from Smilax china Linne used for the study are water extract (CRWE), ethanol extract (CREE) and methanol extract (CRME). In in vitro studies, the spores of the dermatophytes were inoculated on Sabouraud's glucose agar media which contained three extracts of Chinae Rhizoma in each concentration of $500\;{\mu}g/ml$, $1,000\;{\mu}g/ml$ and $5,000\;{\mu}g/ml$ respectively, and also $1,000\;{\mu}g/ml$ of salicylic acid and undecylenic acid $1,000\;{\mu}g/ml$ as comparable drugs. The growth of the dermatophytes were observed for 3 weeks. The species of the dermatophytes used in this experiment were Epidermophyton floccosum, Microsporum canis, Microsporum cookei, Microsporum gypseum, Microsporum nanum, Trichophyton mentagrophytes, Trichophyton rubrum, Trichophyton tonsurans and Trichophyton verrucosum distributed from The Institute of Tropical Medicine in Belgium. The results of the studies were as follows: 1. The growth of M. canis, M. nanum, T. mentagrophytes, T. rubrum & T. tonsurans were slightly inhibited in CRWE $1,000\;{\mu}g/ml$ and CRWE $5,000\;{\mu}g/ml$, and only slight inhibition on the growth of E. floccosum, M. canis and M. gypseum were observed in CRWE $5,000\;{\mu}g/ml$. 2. Complete inhibition of T. rubrum, moderate inhibition of M. nanum & T. tonsurans, and slight inhibition of E. floccosunl, M. canis, M. cookei & T. mentagrophytes in growth were observed in concentration of CREE $500\;{\mu}g/ml$. The growth of M. gypseum was slightly inhibited, moderate inhibition on the growth of M. canis, M. cookei & T. mentagrophytes, and complete inhibition of E. floccosum, M. nanum, T. rubrum & T. tonsurans in growth were observeed by CREE $1,000\;{\mu}g/ml$. With $5,000\;{\mu}g/ml$ of CREE, the growth of E. floccosum, M. canis, M. cookei, M. gypseum, T. mentagrophytes, T. rubrum & T. tonsurans were completely inhibited except T. verrucosum being showed slight inhibition. 3. In CRME $500\;{\mu}g/ml$, slight inhibition of T. verrucosum, moderate inhibition of M. gypseum and complete inhibition of E. floccosum, M. canis, M. cookei, T. mentagrophytes, T. rubrum & T. tonsurans in growth were observed. The growth of E. floccosum, M. canis, M. cookei, M. gypseum, M. nanum, T. mentagrophytes, T. rubrum & T. tonsurans were completely inhibited except T. verrucosum being showed moderate inhibition in both CRME $1,000\;{\mu}g/ml$ and CRME $5,000\;{\mu}g/ml$. 4. In $1,000\;{\mu}g/ml$ of undecylenic acid, slight inhibition of T. verrucosum and complete inhibition of E, floccosum, M. canis, M. cookei, M. gypseum, M. nanum, T. mentagrophytes, T. rubrum & T. tonsurans in growth were observed. From the above results, it was found that Chinas Rhizoma Alcoholic Extracts(CREE & CRME) exerted significant antifungal activity, and their effects were probably derived from the pharmacological actions of triterpenoidal saponin and steroidal saponin.

  • Although numerous drugs are available for the treatment of superficial fungi infections of skin, the clinical effects of the majority of such drugs are not satisfactory. In the hope of searching the effective drugs for superficial fungi infections, authors studied whether Veratrum rhizoma extracts had any effect on fungi, with water extract (VRWE), ethanol extract (VREE) and methanol extract (VRME) from Veratrum album L. var. grandiflorum Max. In in vitro studies, the spores of fungi were inoculated on Sabouraud's glucose agar media which contained three extracts of Veratrun rhizoma in each concentration of $500\;{\mu}g/ml$, $1,000\;{\mu}g/ml$ and $5,000\;{\mu}g/ml$ respectively, and the growth of the fungi were observed for 3 weeks. The species of the fungi used in these experiments were Epidermophyton floccosum, Microsporum canis, Microsporum nanum, Microsporum gypseum, Microsporum cookei, Trichophyton rubrum, Trichophyton mentagrophytes, Trichophyton tonsurans and Trichophyton verrucosum. The growth of M. nanum and M. gypseum were moderately inhibited, and significant inhibition on the growth of E. floccosum, M. canis, M. cookei, T. mentagrophytes, T. rubrum and T. tonsurans were observed by VREE $1,000\;{\mu}g/ml$. By VREE $5,000\;{\mu}g/ml$, the growth of all tested fungi were significantly inhibited except T. verrucosuia being showed slight inhibition. 3. Significant inhibition on the growth of M. canis, T, mentagrophytes, T. rubrum and T. tonsurans were noted, and moderate inhibition of M. nanum, slight inhibition of E. floccosum and M. gypseum in growth were observed by VRME $500\;{\mu}g/ml$. The growth of E. floccosum, M. canis, M. nanum, M. cookei, T. mentsgrophytes, T. rubrum and T. tonsurans were significantly inhibited by VRME $1,000\;{\mu}g/ml$, and that of M. gypseum was moderate. With $5,000\;{\mu}g/ml$ of VRME, significant inhibition on the growth of E. floccosum, M. canis, M. nanum, M. gypseum, M. cookei, T mentagrophytes, T. rubrum and T. tonsurans were observed, and T. verrucosum was showed only slight inhibition. From the above results, it was found that the extracts of organic solvents from Veratrum rhizoma (VREE & VRME) exerted significant antifungal activity, and their effects were probably derived from the pharmacological action of steroidal alkaloids.

A Case of Feline Ringworm by Microsporum canis in Korea (Microsporum canis에 의한 고양이의 백선증)

  • Attempts were made to determine the clinical features and the causative organism of ringworm occurred in a cat in Chinju city, Korea. Alopecia and greyish white crusty lesions were observed around the eye, nose and posterior limb while irregular, partial loss of hairs and white scales were in the ear, neck and abdomen lesions. Direct microscopic and cultural examination were carried out for the hairs and crusts taken from the skin lesions, and the causative organism was identified as M. canis.

The Antidermatophytic Potential of the Marine Isolate of Aspergillus sp. Collected from South Coast of Korea

  • This study was carried out to assess the antidermatophytic potential of the ethyl acetate(EtOAc) extract of the marine isolate of Aspergillus sp.. The fungus was isolated by serial dilution, and was identified Aspergillus sp.. The EtOAc extract of the fungus was examined to evaluate the antidermatophytic efficacy against the fungal pathogens infecting human skin using the disc diffusion and MIC(minimum inhibitory concentration) determination methods. The EtOAc extract($5{\mu}l\;disc^{-1}$) was considered to have the antidermatophytic activity based on the inhibition percentage of the mycelial growth of the fungi tested such as Trichophyton mentagrophytes KCTC 6085, Microsporum canis KCTC 6591, Microsporum canis KCTC 6348, Trichophyton rubrum KCTC 6352, Microsporum canis KCTC 6349 and Trichophyton mentagrophytes KCTC 6316. The percentage of the inhibition ranged from 54% to 81, and the MIC obtained was 62.5, 62.5, 250, 125, 125, and $125{\mu}g\;ml^{-1}$, respectively. The extract had a strong detrimental effect on the spore germination of the tested skin infectious pathogens. These findings strongly support the role of the ethyl acetate extract as a potential antidermatophytic agent.

Antifungal Activity of Extracts from Pyrethrum Cinerariaefolium V. (제충국화(除?菊花) 추출물(抽出物)의 항진균작용(抗眞菌作用))

In the hope of searching for effective drugs for superficial fungi infections, the authors observed fungistatic effects of Pyrethri Flos, a common herb in Korea, with water extract (PFWE), ethanol extract (PFEE), and methanol extract (PFME) from Pyrethrum cinerariaefolium V. In in vitro studies, the spores of fungi were inoculated on Sabouraud's glucose agar media which contained Pyrethri Flos extracts in each concentrations of $500\;{\mu}g/ml$, $1,000\;{\mu}g/ml$ and $5,000\;{\mu}g/ml$ respectively, and the growth of the fungi was observed for 3 weeks. The species of the fungi used in these experiments were Epidermophyton floccosum, Microsporum canis, Microsporum nanum, Microsporum gypseum, Microsporum cookei, Trichophyton rubrum, Trichophyton mentagrophytes, Trichophyton tonsurans and Trichophyton verrucosum. The results of these studies were as follows; 1. The growth of M. nanum & T. rubrum was slightly inhibited by PFWE $500\;{\mu}g/ml$, and the growth of M. nanum, M. cookei & T. rubrum was slightly inhibited by PFWE $1,000\;{\mu}g/ml$. The growth of E. floccosum, M. gypseum & M. cookei was slightly inhibited, however the growth of M. canis, M. nanum, T. mentagrophytes, T. rubrum & T. tonsurans was significantly inhibited by PFWE $5,000\;{\mu}g/ml$. With $500\;{\mu}g/ml$ of PFEE, the growth of M. canis, M. nanum, T. mentagrophytes, T. rubrum & T. tonsurans was significantly inhibited, and moderate inhibition of M. cookei growth and slight inhibition E. floccosum & M. gypseum were observed. The growth of M. canis, M. nanum, T. mentagrophytes, T. rubrum & T. tonsurans was significantly inhibited, and the growth of E. floccosum, M. gypseum & M. cookei was moderately inhibited by PFEE $1,000\;{\mu}g/ml$. Significant inhibitions of the growth of E. floccosum, M. canis, M. nanum, M. gypseum, M. cookei, T. mentagrophytes, T. rubrum & T. tonsurans were observed by PFEE $5,000\;{\mu}g/ml$. 3. The growth of E. floccosum & M. cookei was moderately inhibited, and the growth of M. canis, M. nanum, M. gypseum, M. mentagrophytes, T. rubrum & T. tonsurans was significantly inhibited by PFME $500\;{\mu}g/ml$. But the growth of E. floccosum, M. canis, M. nanum, M. gypseum, M. cookei, T. mentagrophytes, T, rubrum & T. tonsurans was significantly inhibited, and the growth of T. verrucosum was slightly inhibited in both PFME $1,000\;{\mu}g/ml$ and $5,000\;{\mu}g/ml$.

Survey of the canine dermatophytes in eastern area of Kyongbuk (경북 동부지역 사육견의 피부사상균 보균실태 조사)

  • The dermatophytes was isolated from skin of the 175 healthy dogs and 22 dogs with pathological skin lesions at Pohang and Kyongju. The isolates were identified by the morphological appearance after cultivation and lactophenol cotton blue staining. 1, The isolation rates of dermatophytes were 8.5%(15/175) in dog with healthy skin and 27% (6/22) in dogs with pathological skin lesions. 2. From asymptomatic dogs, the isolation rates of dermatophytes in female dogs were higher than those in male dogs and those in young dogs were higher than those in old dogs. 3. Isolation rates of microsporum canis and trichophyton spp from asymtomatic dogs were 14 (93%) and 1(6.7%), respectively. 4. The causative agents of 6 dogs with fermatophytosis were identified as all M canis.

  • The antifungal activity of salicylaldehyde-o-phenylenediimine, a derivative of Schiff base, was studied in vitro against various species of fungi, especially, dermatophytes. The antifungal tests were performed according to the tube-dilution method, and all subcultures were incubated at room temperature for 14 days. Salicylaldehyde-o-phenylenediimine was effective against fungi such as Trichophyton mentagrophytes, Trichophyton rubrum, Microsporum canis and Fonsecaea compacta. Trichophyton mentagrophytes was inhibited at $20{\mu}g$ per ml, Trichophyton rubrum at $40{\mu}g$ per ml, Microsporum canis at $6{\mu}g$ per ml, and Fonsecaea compacta at $2{\mu}g$ per ml.

Efficacy of the Rhinacanthus nasutus Nees Leaf Extract on Dermatophytes with Special Reference to Trichophyton mentagrophytes var. mentagrophytes and Microsporum canis

  • The effect of Rhinacanthus nasutus leaf extract on the growth of dermatophytes had been investigated. In vitro the extract exhibited high activity against various species of dermatophytes (Trichophyton mentagrophytes var. mentagrophytes, T. mentagrophytes var. interdigitale, T. rubrum, Microsporum canis and M. gypseum). The minimal inhibitory concentration (MIC) values of the extract revealed that all the dermatophytes tested had MIC values of 13.6 mg/ml. The extract exhibited fungistatic activity at lower concentrations $({\leq}13.6\;mg/ml\;or\;below\;the\;MIC\;value)$ and fungicidal activity at higher concentrations $({\geq}13.6\;mg/ml$\;or\;above\;the\;MIC\;value)$. The results suggested that the extract acted on the cell wall of the dermatophytes which subsequently leading to the formation of cytopathological and membrane structural degeneration and finally leading to cell lysis and death.
