A pot experiment was carried out to examined the effect of electrolyzed micronutrients (Fe, Mn, Zn, Sr, Se, Sn, Co, Ti, and V) solution treatments with nutrient solution and SCB slurry on the mineral content and growth of tomato in cherry tomato (Lycopersicon esculentum). The treatment of nutrient solution (NS)+micronutrients solution (MS) significantly increased the concentrations of Li, Zn, Sr, Se, Ti as compared with that of NS alone in the cherry tomato fruits, and SCB+MS solution treatment significantly increased Li, Zn, Se, Co, Sr, and Ti contents as compared with SCB treatment. The micronutrient contents of MN+SCB+MS treatment were significantly higher in Li, Zn, Se, Co and in Ti than those of SCB and NS treatment, respectively. The growth and yield of cherry tomato fruits was highest with NS treatment. The yield indices of cherry tomato treated with NS+MS treatment and SCB+NS+MS were 97% and 94% of NS treatment. In conclusion, it seems to be possible to produce micronutrient-fortified cherry tomato by the mixed treatment of electrolyzed micronutrients.
High rates of nitrogen fertilization dangerously increase the nitrate content of vegetable crops, and the accumulation of nitrate in edible crops is undesirable because of potential risks to human health. Micronutrient solution containing Cu, Mn, Mo, Zn was tested for the suppression of nitrate accumulation in lettuce grown in pots treated with Mg fertilizer under a greenhouse condition. The micronutrient solution was sprayed on leaves at 3 and 4 weeks after transplanting of 20-day old seedlings. Plants were harvested after 5-week growth, and yield, contents of chlorophyll, sugar, micronutrient and nitrate, and also nitrate reductase activity were measured. Fresh weight of lettuce was significantly increased by the application of Mg and micronutrients, and the effect was the most significant in the Mg+micronutrient treatment. Also contents of chlorophyll and micronutrients were higher in the plants of micronutrient treatments. Contents of nitrate were reduced by about 14-18% in lettuce with Mg and/or micronutrient applications. Compared to the plants of control treatment, nitrate reductase activity was also higher in those plants treated with micronutrients, and in the treatment of Mg+micronutrients the enzyme activity was six times as high as that of control treatment. Although the effect of mineral nutrients on the suppression of nitrate accumulation in lettuce was relatively small in this study, an appropriate supply of mineral nutrients could be one of the solutions for the nitrate accumulation in vegetables.
Suppression of nitrate accumulation in vegetables through foliar application of micronutrients was investigated. Spinach and lettuce were grown in pots under greenhouse condition. Micronutrient solutions containing Cu, Mn, Mo, and Zn were used; chitosan was added into one and the other contained chitosan oligomers. The micronutrient solutions were sprayed on the leaves at 3 and 4 weeks after transplanting of 20-day-old seedlings. Plants were harvested at 5-weeks after transplanting. Yield, contents of chlorophyll, Brix value, micronutrient, and nitrate, and nitrate reductase activity were measured. Fresh weights of lettuce and spinach were significantly increased by the foliar application of micronutrients. Contents of chlorophyll and micronutrients were higher in micronutrient-treated plants, while those of nitrate were reduced by about 10 and 14-23% in lettuce and spinach, respectively. Compared to the control plants, nitrate reductase activity was higher in plants treated with micronutrients. Results of this study indicate the effect of micronutrients on the suppression of nitrate accumulation was relatively small in comparison to the contents of nitrate in leaves of spinach and lettuce. To maximize the effect, nutrient composition in solution, application time, and frequency should be further examined, taking into consideration nitrogen level in soil and other environmental factors including light condition.
The objective of this study was to evaluate the effects of the application of compost on the availability of micronutrients in lettuce. Micronutrient contents of manure-based composts containing various other source materials were investigated. Total and extractable contents of micronutrients in the composts were analysed. Pots containing soil of relatively low micronutrient levels were treated with 1,000 and $2,000kg\;10a^{-1}$ of compost and used to grow lettuce plants under greenhouse conditions. Fresh and dry weights of lettuce and micronutrient uptake were determined after harvest. Manure-based composts of various other source materials contained very different amounts of total and extractable micronutrients. Total contents of B, Cu, Fe, Mn, Mo and Zn were in the range of 26-42, 27-160, 4,300-9,500, 290-790, 0-0.5 and $140-420mg\;kg^{-1}$, respectively. The contents of 0.1 N HCl extractable B, Cu, Fe, Mn and Zn were 23-32, 1.3-2.6, <1, 7-32 and 0.5-5% of total content, respectively. Contents of micronutrients extractable in DTPA solution were generally higher than those extractable in 0.1 N HCl. It was found that the fresh and dry matter productions of the plants were significantly higher in the compost treatment of $2,000kg\;10a^{-1}$. Lettuce grown in soil treated 1,000 and $2,000kg\;10a^{-1}$ of manure-based compost contained higher levels of B, Cu, Mo and Zn than lettuce grown without compost application. However, contents of Fe and Mn in lettuce were relatively lower in the compost treatments. In the compost treatments the proportions of micronutrients in soil and plant were all in the optimum ranges and below the toxicity levels. The results obtained allow us to establish that commercial composts could be used as soil amendment for plastic film house crop production with sufficient supply of micronutrients.

The present work aims to study the induction of somatic embryogenesis in cork oak (Quercus suber L.) from immature zygotic embryos and young apical leaves obtained from 2-month-old seedlings through acorn germination on sterilized peat. The immature zygotic embryos were grown for 1 month on the mineral solution of MS in the presence of 4.52 µM 2,4-D and 30 g/L sucrose. They were then transferred to the same mineral solution with no added growth regulators. In the third subculture, yellow somatic embryos, characterized by two voluminous cotyledons, were differentiated from the radicle of the immature zygotic embryos. The induction of somatic embryogenesis in young leaves required a series of transfers on different culture media containing 30 g/L sucrose and 100 mg/L myo-inositol. Secondary or recurrent somatic embryogenesis occurred within the immature somatic embryo radicles after 1 month of culture on growth regulator-free medium containing WPM macronutrients, MS micronutrients, and vitamins.
The objective of the present work was to determine the corresponding uptake and assimilation of ${NO_3}^-$ in roots and shoots of leafy radish by applying of mixed amino acids (MAA). The amino acids used in this experiment were alanine (Ala), ${\beta}-alanine\;({\beta}-Ala)$, aspartic acid (Asp), asparagines (Asn), glutamic acid (Glu), glutamine (Gln), and glycine (Gly). Leafy radish was grown by conventional fertilization with macro- and micronutrients under controlled conditions. The 15-day-old seedlings were treated 0, 0.3 and 3.0 mM of MAA containing 5 mM ${NO_3}^-$ in growth medium. Nitrate uptake was determined by following ${NO_3}^-$ depletion from the uptake solution. The activity of the enzymes related to the process of ${NO_3}^-$ reduction (NR: nitrate reductase; NiR: nitrite reductase; GS: glutamine synthetase) and the content of ${NO_2}^-\;and\;{ND_3}^-$ were analyzed in shoots and roots. The results of this study showed that ${NO_3}^-$ uptake was inhibited 38% with treatment of 0.3 mM of MAA. However, there was more than three times increase of N03- uptake in 3.0 mM MAA. In addition, the enzymatic activities were positively affected by the high MAA rate. Finally, the ${NO_3}^-$ content was increased slightly both in shoots and roots of leafy radish by MAA treatments.
Field and nutrient cultures of American ginseng (Panax quinquefolius L.) were used to establish foliar symptoms related to boron (B) concentration in leaves and soils, and to evaluate radish as a time-saving model system for B nutrition. Application of excess B, 8 kg/ha versus the recommended 1.5 kg/ha, to field plantings of 2-, 3-, and 4-yr-old American ginseng plants just prior to crop emergence caused, within 4 wk after crop emergence, leaf symptoms of chlorosis followed by necrosis starting at the tips and progressing along the margins. The B concentration in leaves of 2-4-yr-old plants receiving 1.5 kg/ha Bwas $30{\mu}g/g$ dry mass compared to $460{\mu}g/g$ dry mass where 8 kg/ha B was applied. Similarly, B concentration in soils receiving the lower B concentration was 1.8 mg/g dry mass and $2.2-2.8{\mu}g/g$ dry mass where the higher B concentration was applied. Application of 8 kg/ha B reduced the dry yield of 3rd-yr roots by 20% from 2745 kg/ha to 2196 kg/ha and 4th-yr roots by 26% from 4130 kg/ha to 3071 kg/ha. Ginseng seedlings and radish were grown under greenhouse conditions in nutrient culture with four B concentrations ranging from 0 mg/L to 10 mg/L. At 5 mg/L and 10 mg/L ginseng and radish developed typical leaf B toxicity symptoms similar to those described above for field-grown plants. Increasing B in the nutrient solution from 0.5 mg/L to 10 mg/L decreased, in a linear fashion, the root and leaf dry mass of ginseng, but not radish. Given the many similarities of ginseng and radish to B utilization, radish might be used as a timesaving model system for the study of B, and other micronutrients, in the slow-growing perennial ginseng.
Protoplasts of Pisum sativum L. were isolated and cultured from leaf mesophyll tissue. The successful yield of protoplast was obtained in an enzyme solution of 2% 'Onozuka R-10' and 2 % 'Macerozyme R-10' contained 6mM $CaCl_2{\cdot}2H_2O$ within 4 hours. They were divided in B5 culture medium supplemented with 2mg/l kinetin, 1mg/l 2, 4-D and 0.2% Difcobacto agar. Divisions of the protoplasts were continued and led to colony formation for 1 months. The colony from protoplasts of pea mesophyll tissue was formed to callus after subculture in a medium contained macronutrients and amino acids of BII medium and micronutrients and vitamins of B5 medium, and also supplemented with 2mg/l kinetin 2mg/l NAA.
This research was conducted to investigate the optimum level of nitrogen incorporated during formulation of root media as pre-plant fertilizer on the growth of plug seedlings of 'Bool-am No.3' Chinese cabbage. A root medium was formulated by blending peatmoss:coir dust:perlite at a ratio of 3.5:3.5:3.0 (v/v/v). The nitrogen was incorporated in the seven treatments at a rate of 0, 100, 250, 500, 750, 1,000, and $1,500mg{\cdot}L^{-1}$ during the root medium formulation. The concentrations of other essential nutrients except nitrogen were controlled to equal in all treatments. Then, the root medium was packed into 72-cell plug trays and seeds were sown. The growth measurements as well as tissue and soil solution analysis for nutrients were conducted 2 and 4 weeks after seed sowing. As seedlings grew, the pH in the extracted solution of all treatments tended to decrease. The decreases in the treatments of high N concentrations were more severe than those with low N, but the differences among treatments were not statistically significant. The differences of EC in extracted solution of root media among treatments were sizable until week 3, but the differences began to lessen and the EC decreased in all treatments after week 4. Growth of the aerial parts of plug seedlings at 2 weeks after sowing were highest in the $100mg{\cdot}L^{-1}$ and lowest in the $1,500mg{\cdot}L^{-1}$ treatments, but those at 4 weeks after sowing were highest in the $500mg{\cdot}L^{-1}$ and lowest in the $0mg{\cdot}L^{-1}$ treatments among all treatments tested. The tissue N content was highest and lowest in the treatments of 250 and $1,000mg{\cdot}L^{-1}$, respectively, when tissues were harvested at 4 weeks after sowing and analysed based on the dry weight of above-ground tissue. The contents of micronutrients were the highest in the 1,000 and $1,500mg{\cdot}L^{-1}$ treatments among all treatments. The results shown above indicate that the $250mg{\cdot}L^{-1}$ of pre-plant N and elevation of post-plant N concentration to above $100mg{\cdot}L^{-1}$ are suitable for raising plug seedlings of Chinese cabbage using inert media.
