Effects of Methylcellulose on Fibrolytic Bacterial Detachment and In vitro Degradation of Rice Straw

  • Two in vitro experiments were conducted to evaluate the effect of methylcellulose (MC) on i) bacterial detachment from rice straw as well as ii) inhibition of bacterial attachment and fiber digestibility. To evaluate the effect of MC on fibrolytic bacterial detachment (Exp 1), in vitro bacterial cultures with 0.1% (w/v) MC solution were compared with cultures without MC after 8 h incubation. The effect of MC on inhibition of bacterial attachment was determined by comparing with real-time PCR the populations of F. succinogenes, R. flavefaciens and R. albus established on rice straw pre-treated with 0.1% MC with those on untreated straw after incubation for 0, 6 and 12 h (Exp 2). The major fibrolytic bacterial attachment on rice straw showed significantly lower populations with either the addition of MC to the culture or pre-treated rice straw compared to controls (p<0.05). Also, the digestibility of rice straw with MC was significantly lower compared with control (p<0.05). The F. succinogenes population did not show detachment from rice straw, but showed an inhibition of attachment and proliferation on rice straw in accordance with a decrease of fiber digestion. The detachments of Ruminococcus species co-existed preventing the proliferations with subsequent reduction of fiber degradation by MC during the incubation. Their detachments were induced from stable colonization as well as the initial adhesion on rice straw by MC in in vitro ruminal fermentation. Furthermore, the detachment of R. albus was more sensitive to MC than was R. flavefaciens. These results showed the certain evidence that attachment of major fibrolytic bacteria had an effect on fiber digestion in the rumen, and each of fibrolytic bacteria, F. succinogenes, R. flavefaciens and R. albus had a specific mechanism of attachment and detachment to fiber.

  • The purpose of present study is to compare the effect of treatment using $Guidor^{(R)}$ as a barrier membrane in conjunction with platelet-derived growth factor and insulin like growth factors on experimental dehiscence defects. Following the resection of premolar crowns, roots were submerged. After 12 weeks of healing period, experimental dehiscence defects of 4mm in height and 4mm in width were surgically created on the mid-facial aspect of the lower premolar roots in each of 4 adult dogs. After root planning and demineralization of the root surface with citric acid, the control groups received 4% methylcellulose gel only, the test group I received 4% methylcellulose gel and were covered by $Guidor^{(R)}$ and the test group II were treated with PDGF and IGF and 4% methylcellulose gel with $Guidor^{(R)}$ coverage. Histological and histomorphometric analysis following 8 weeks of healing revealed the following results. 1. The new bone formation showed no statistically significant difference in all groups with $0.59{\pm}0.82mm$($14.03{\pm}19.60%$) for control, $0.70{\pm}0.39mm$($16.30{\pm}9.01%$) for group I, $0.87{\pm}0.76mm$($18.74{\pm}16.03%$) for group II. 2. The new cementum formation showed no statistically significant difference in all groups with $0.54{\pm}0.48mm$($l6.38{\pm}14.57%$) for control, $0.95{\pm}0.38mm$($23.43{\pm}9.30%$) for group I, $1.01{\pm}0.75mm$($22.10{\pm}16.ll%$) for gorup II. 3. The root resorption showed statistically significant differences betweenthe control group and all test groups(p<0.05) with $2.11{\pm}0.53mm$($52.93{\pm}12.32%$) for control, $0.63{\pm}0.27mm$($15.32{\pm}7.05%$) for group I, $0.89{\pm}0.33mm$ ($19.26{\pm}7.11%$) for group II. On the bases of these results, there were no statistically difference between treatment using resorbable membrane and resorbable membrane in conjunction with PDGF and IGF in the dehiscence defects, where it was difficult to maintain space. The use of membrane seemed to be more effective in the inhibition of root resorption.

The Mitigation of Bitterness of Zipeprol Solution (Zipeprol 내용액제의 고미교정에 관한 연구)

  • Ziperol, anti-tussive, is considerably bitter. Therefore, it is necessary to mitigate the bitterness in ziperol syrup for children. In this experiment, it was attempted to mitigate the bitterness of zipeprol by means of polymers such as $\beta$-cyclodextrin, arabic gum, HPMC(hydroxypropyl methylcellulose), PEG 2000(polyethyleneglycol 2000), PVP(polyvinylpyrrolidone). Caffeine was used as the reference standard of bitterness. In the result of this experiment, $\beta$-cyclodextrin, would mitigate the bitterness of zipeprol more largely than any other polymers. Arabic gum was the second choice which would mitigate the bitterness of zipeprol.

  • 이 글에서는 호수생태계에서 일어나는 변화 중 .betha.-glucosidase 활성도를 중심으로 생화학적인 분석과정을 통하여 유기물질의 분해, 생성, 소비등에 대한 과정을 설명하고자 한다. .betha.-glucosidase효소는 비교적 기질특이성이 넓고, glucose, celluhexose, carboxy-methylcellulose등의 .betha.고리를 가수분해시켜 monomer로 분해시키는 효소로서 수환경에서는 매우 중요하다. 즉 cellobiohydrolase에 의해 분해된 oligomer나 dimer를 세균의 세포막을 통과할 수 있는 크기로 분해시키는 효소이고, 이 효소의 분해 산물인 포도당은 세균에 의하여 바로 이용되고, catabolite inhibitor로 작용하므로, 이 효소 활성도의 높고 낮음은 수중생태계에서 탄소의 흐름과 세균 군집의 성장을 이해하는데 매우 중요하다(3).

Effect of Hydrocolloids on the Quality of Rice Dumpling Skins (Hydrocolloids가 쌀만두피 특성에 미치는 영향)

  • This study investigated the formation of rice dumpling skins with various food additives to improve their texture. Rice (Millyang260 and Hanarum) was obtained from the Rural Development Administration. Rice was milled through an air classifying mill (ACM), and hydroxypropyl methylcellulose (HPMC), xanthan gum, guar gum, carrageenan, or propylene glycol alginate (PGA) was used as food additives. The effects of hydrocolloids on textural were evaluated and rice dumpling skin prepared with additives showed significantly higher values than the control for cooking properties and texture. The presence of hydrocolloids in rice dumpling skin appeared to decrease its weight and volume after cooking. Textural properties of rice dumpling skin with hydrocolloids were similar to the control regardless of cooking condition.

Study on Improving Properties of Tile Cement Mortar by Mixing of Additives (Additives의 혼합에 의한 Tile Cement Mortar 물성향상 연구)

  • In this study, to improve the required properties of tile cement mortar such as excellent water retention capacity (WRC), workability, open time, sag resistance, and tile adhesive strength, tile cement mortars containing the several additives with different ratio were compared and analyzed. By adding small amount of synthesized starch to hydroxypropyl methylcellulose (HPMC) which is used for improving WRC, the decrease of moisture evacuation from mortar surface was observed and the workability of mortar was improved with long open time. Polyacrylamide (PAAm) and ethylencvinyl acetate (EVAc) were also added in order to increase the adhesion of tile. As a results, the saggings of mortar itself and tile were decreased and the adhesive strength of mortar between base and tile was enhanced. By adding melment, the workability was improved by increasing the fluidity of mortar. It is postulated that the properties of tile cement mortar was improved by adding 0.80~1.20% of HPMC, 0.10~0.15% of starch, 0.001~0.015% of PAAm, 0.05~0.10% of EVAc and 0.003~0.005% of melment to the cement mortar.

  • Electrically conducting composite films were prepared by a vapor phase in situ polymerization of pyrrole in the methyl cellulose film containing a copper(II) perchlorate. Methylcellulose had high affinity to pyrrole and was used as a matrix polymer. Conducting polypyrrole was embedded in the methylcellulose film forming a conducting network and the conductivity of the composite films ranged $10^{-1}$ to $10^{-7}S/cm$. The conductivities of conducting composite films were dependent on the nature of the matrix polymers, concentration of oxidant and polymerization time. In situ polymerization of pyrrole was observed in the matrix polymer and confirmed by UV-vis spectra. From the results of the thermogravimetric analysis, the chemical oxidative polymerization of pyrrole in the matrix polymers did not give any negative effects on the thermal stability of the composite films. Electron micrograph of composites indicated good penetration of PPy in the matrix polymer. DMA suggested a certain degree of incompatibility of the polypyrrole in the composites.

The Effects of Coating Treatments on Enteric Coating of the Soft Capsules Containing Omega-3 Fatty Acids (오메가-3 연질캡슐의 코팅 조건에 따른 장용성 코팅품질에 미치는 영향)

  • This article presents an evaluation of the effects of coating conditions on the enteric coating quality of soft gelatin capsules containing Omega-3 fatty acids. Three conditions were controlled: concentration of hydroxypropyl methylcellulose phthalate (6, 8, and 10 wt% in solution), temperature of the inlet air (32, 35, and $38^{\circ}C$), and the coating solution feed rate (7.5, 11.25, and 15.0 g/min). The transparency of the enteric coated soft gelatin capsules was evaluated by measuring the degree of whiteness of the surface using a spectrophotometer. Results showed that the most important parameter in the enteric coating process was the coating solution feed rate. As the coating solution feed rate decreased and inlet air temperature increased, the degree of whiteness of coating surfaces decreased. We also evaluated the disintegration properties of the enteric coated capsules in accordance with the Korea Health Functional Food Code.
