This study aimed to replace the imported Chinese complete medium for Lentinula edodes with a newly-developed complete medium that is suitable for export as well. Three media compositions that supported fast mycelium growth and higher density, compared to that in the control, were chosen. Culture in the T13 medium reduced the incubation period for 'Nongjingo' by 12 days and for 'Cham-aram' by 10 days, compared to that required for the control; in addition, the number of days required for browning was greatly reduced for both varieties. The quantity of each mixed medium was increased according to the composition from the 1st to the 5th cycle by 5.9% for 'Nongjingo' and 12.6% for 'Cham-aram' in T13, compared to that in the control. A mixed medium comprising oak sawdust + Douglas fir sawdust + corn flour (40:40:20, v/v) was selected as the most suitable complete medium.
An Immunostimulating polysaccharide was produced from the suspension culture of Angelica gigas H4, plant cells. In order to enhance the polysaccharide production by the A. gigas cell culture, medium composition and physical conditions were optimized. Schenk and Hildebrandt (SH) medium was selected as an optimal basal medium for the growth of A. gigas. The maximum cell and polysaccharide concentration obtained in SH medium were 15.8 g DCW/l and 0.85 g polysaccharide/l, respectively, at $25^{\circ}C$ under dark condition. For the enhanced polysaccharide production, a polysaccharide production medium (PPM) was established by modifying Gamborg B5 medium with optimized carbon sources, growth regulators, organic and inorganic elements. Optimal initial pH and temperature were 6.0-6.6 and $20^{\circ}C$, respectively, and the dark condition was better than the light condition. The maximum polysaccharide concentration of 1.5 g/l could be obtained through the optimization of the medium composition and physical conditions.
This study was carried out to investigate the optimum composition and concentration of medium for the acclimation of the shoots derived from bioreactor culture of Scrophularia buergeriana. Rooting and growth of the shoots cultured in bioreactor for 4 weeks were better on liquid full strength MS medium while one-tenth of MS medium was good for rooting and growth of the shoots cultured in bioreactor for 6 weeks. Leaf expansion was more effective in the shoots cultured for 6 weeks on solid MS medium than in those cultured for 4 weeks. No leaf expanded in liquid medium. Vitrification rate was higher in full strength solid MS medium than in half strength MS medium. It is concluded that acclimation of the shoots is more effective on the half strength solid MS medium containing 1.2% agar.
The MRS medium is widely used as an optimized medium for the growth of Lactobacillus spp. and also used for the growth of Leuconostoc spp. Leuconostoc mesenteroides shows quite different physicochemical properties compared to Lactobacilli spp. and it is one of the major strain of kimchi fermenting microorganisms with its usefulness in our traditional foods and availability in biotechnology in the future, specifically tailor-made medium is necessary for the growth of Leuconostoc mesenteroides. Sequential experimental designs (Plackett-Burman, fractional factorial, steepest ascent, central composite design and response surface methodology) were introduced to optimize and improve the Leuconostoc medium. Fifteen medium ingredients were investigated and fructose, sodium acetate and ammonium citrate were determined to give a critical and positive effect for cell-growth. The yield of biomass using the optimal medium was improved more than that of the MRS medium and the result of fed-batch culture showed the capability of the improvement in cell mass similar to the E.coli system.
Mucor plumbeus FRI 0007 was grown on media containing starch solely as carbon source, urea as nitrogen source and minerals including magnesium, calcium and iron of different concentration. The ratio of nonpolar and polar lipid of the total lipid produced by the Muror plumbeus FRI 0007 changed by minerals added in the medium and incubation period. The nonpolar lipid content was higher on the medium containing only one mineral rather than 5 minerals and the nonpolar lipid consisted mainly of trig1yceride, free fatty acid and free sterol. The triglyceride content was higher on the medium containing one mineral and decreased with the incubation time lapse. The major fatty acid composition of total, nonpolar and polar lipid were oleic, palmitic and linoleic acid which comprised about 90% of total fatty acids and their compositions changed slightly depending on the minerals added in the medium.
We investigated factors affecting primary cultures of Pacific abalone Haliotis discus hannai ovary-dissociated cells to identify general aspects of their early-phase culture. Ninety-seven cell populations derived from 30 individuals were cultured in different media with varying compositions of medium supplements, and initial attachment, subculture, and survival for ${\geq}10$ weeks were assessed according to medium composition and individual. We also examined the time required for subculture and the rate of cell death according to both culturing period and passage number within 10 weeks. A lack of fetal bovine serum (FBS) and hemolymph significantly inhibited the growth of cultured cells, while we detected no significant effect of medium composition on initial cell attachment. Through data reallocation, with the omission of data from cell populations cultured in FBS-free and hemolymph-free media, we showed that growth inhibition was also affected by individual differences among the abalones used. During the culture, we observed four different types of cell morphology. Moreover, considerable time was required for subculture-18.4 and 19.5 days for first and second subcultures, respectively-and cell death did not occur within 30 days or for passage 0. Our results will provide valuable information for developing universal cell culturing guidelines in abalone species and suggest the feasibility of culturing abalone ovary-dissociated cells.
Selective adsorption of plant metabolites from a polar dilute solution onto a polycarboxyl ester sorbent (XAD-7) was investigated. Experimental results demonstrated that neutral resins could selectively concentrate specific flavonoids from dilute aqueous mixtures. Adsorption was dependent on the pH of medium, dosage of the resins and medium composition. Especially the medium composition was a key factor for the selective adsorption and it was found that the selective adsorption was dependent on specific sorbent-sorbate-medium characteristics. Under the optimum condition, selectivity increased up to 85% and the yield of recovery was approached to 98%. It was also found that XAD-7 adsorbed flavonoids in the order of hydrophobicity.
The effects of variation in composition of the medium on the conversion of Gluconacetobacter hanseii PJK cells producing cellulose ($Cel^+$) to non-cellulose producing ($Cel^-$) mutants and the production of bacterial cellulose (BC) in an agitated culture were investigated. The impeller speed greater than 500 rpm was required to decrease the population of $Cel^-$ mutants to minimum in a basal medium containing $1.5\%$ ethanol because the optimum impeller speed to minimize the population of $Cel^-$ mutants increased with the concentration of ethanol added to a basal medium. Ethanol fed-batch culture could not increase the BC production in an agitated culture unlike that of a shaking culture. The amount of BC produced in a basal medium containing $1\%$ ethanol was $39\%$ more than that of the same medium with $0.27\%\;Na_{2}HPO_4$. Increase in the concentration of acetic acid in a basal medium decreased the BC production. The pH control of the culture broth increased the cell mass in the batch culture and improved the production yield of water-soluble polysaccharide (WSPS), but did not affect the production of BC.
Bifidobacterium strain was isolated from the feces of brast fed infants. The isolated strain was identified as Bifidobacterium longum by 16S rRNA sequence analysis and named as Bifidobacterium SH2. The MRS medium was modified to obtain high density cells of Bifidobacterium SH2. The optimal medium was determined to be 50 g/L lactose, 10 g/L beef extract, 10 g/L peptone, 5 g/L yeast extract, 7 g/L sodium acetate, 2 g/L ammonium citrate, 2 g/L disodium phosphate,1 g/L tween 80, 0.2 g/L MnSO$_4$ and 0.5 g/L L-cysteine. The pH and temperature were optimized as 5.0 and $37^{\circ}C$, respectively. Through out the optimization of medium composition and culture conditions, the dry cell weight and viable cell count were 2.5 times and 1.8 times higer than those in MRS medium, respectively.
Rhizopus japonicus had the highest chitosan productivity compared with the chitosan productivity among Rhizopus sp. strains. To increase the productivity of microbial chitosan from Rhizopus faponicus, production medium and incubation conditions were optimized. The composition of the medium and the incubation conditions were as follows : starch 2%, yeast extract 2.5%, KH2PO4 0.05%, MgSO4 0.01%, FeSO4 0.002%, MnSO4 0.002%, ZnSO4 0.002%, CaC12 0.002%, PH 5.5, incubation temperature medium compared with chitosan productivity.
