The anticomplementary activity of stilbenes from medicinal plants in Korea was investigated in vitro. 3,5-Dihydroxy-4'-methoxystilbene (3) was most potent with $IC_{50}$ value of $1.5{\times}10^{-4}M$ followed by rhapontigenin (4), oxyresverastrol (2), 2,3,4',5-tetrahydroxystilbene-2-O-beta-glucoside (9), rhaponticin (8), resverastrol (1), and piceid (7). The activity was found to be increased by a methylation on a hydroxy group of C-4' of 1, but decreased by further methylation on hydroxy groups of C-3 and C-5 and glucosylation on any hydroxy group of 1. Addition of hydroxy group on C-2' of 1 or C-3' of 3 was little affected on the anticomplementary activity but the activity was increased by O-glucosylation on C-2 of 1.
The methanol extracts from 30 species of oriental medicinal plants belonging to 24 families were tested for their lhicidal and antifeeding activit~es against diamondback moth (Plutello xylostella L) and tobacco cutworn (Spodoptera litura F.) by a leaf-dipping method at a concentration of 5, 000 ppm. The methanol extract from Copti chinensis only showed a potent larvicidal activity against P xylostello. Strong antifeeding activity against P. xylostello was observed from the extmds from Platycodon grandiflorurn, Codonopsis pilosula, Asomm sieboldii, Rhus chinensis and Uthospermum erythrorhizon And a potent antifeeding activity against S. liturn was obtained from Akebia quinata and Equlsetum hyemale extracts. A significant antifeeding activity against both species was obtained from R chinensis and C. chinensis extracts.

According to previous reports, antioxidant activities of Codonopsis lanceolata could be increased by a steaming process. This study was performed to improve its antioxidant activity and skin whitening activities of C. lanceolata by high pressure and stepwise steaming complex process. The complex processed C. lanceolata showed highest free radical scavenging acitivity as 45.21%, and for phenol and flavonoid contents, complex processed C. lanceolata contained higher than those from conventional extraction process or steaming process alone. The Cytotoxicity of all C. lanceolata extracts also showed low cytotoxicity against human fibroblast cell (CCD-986sk) as 4.49 ~ 10.40%. In whitening activity, high inhibition of tyrosinase activity was estimated as 25.08% by adding the extracts from complex process. We found that whitening and antioxidant activity of complex processed C. lancolata extract was higher than those obtained from conventional extraction and a steaming process because various kinds of antioxidant compounds could be easily released by combined process, compared to one of each process.
Phospholipase D is a ubiquitous enzyme that plays an important role in various lipid mediated cellular signaling pathways and produces rare phospholipids, phosphatidylethanol or phosphatidylbutanol, instead of phosphatidic acid with unique catalytic activity transphosphatidylation in the presence of primary alcohols. The reaction products, phosphatidylethanol or phosphatidylbutanol are used as markers of in vitro phospholipase D activity in many studies. For the sensitive detection of the phospholipase D products, we developed an advanced lipid extraction method that facilitates recovery of the compounds. With the new method, the activity change of phosaholipase D by agonists could be detected more easily and the recovery rate was also increased. The increase of detected enzyme activity change was about double fold compared to the conventional lipid extraction method. This method provides selective force for the phospholipase D products in the extraction procedure.
This study was conducted to investigate the change of antioxidant activity and ingredient in 4 species medicinal plant by processing. As a result, EtOAc extract and n-BuOH extract obtained from Cistanche deserticola Y. C. Ma and processed Cistanche deserticola Y. C. Ma showed difference on the experiment of antioxidant activity by DPPH method. Change of ingredient also was confirmed by TLC and HPLC analyses. Two main compounds of antioxidant activity change were isolated by column chromatography from the EtOAc extract and n-BuOH extract of Cistanche deserticola Y. C. Ma. The chemical structure of the compound 1 isolated from EtOAc extract of Cistanche deserticola Y. C. Ma was elucidated as 2'-acetylacteoside by means of IR and NMR analyses. Also, the chemical structure of the compound 2 isolated from n-BuOH extract of Cistanche deserticola Y. C. Ma was confirmed as acteoside. The antioxidant activity of processed Cistanche deserticola Y. C. Ma. was declined by 3 time decrements of 2'-acetylacteoside such as 1.89 mg/g to 0.62 mg/g and 2 time decrements of acteoside such as 5.81 mg/g to 2.67 mg/g by processing.
This study was carried out to investigate the antimicrobial activities of organic extracts obtained from the fruit of Thuja orientalis L. The native fruits in Korea were collected and extracted by 80% ethanol, and the extract was sequentially fractionated with n-hexane, chloroform, ethylacetate, and butanol. The fraction yields of n-hexane, chloroform, ethylacetate, butanol and water of ethanol extract were 10.15%, 10.05%, 1.45%, 45.35% and 27.55%, respectively. n-Hexane-soluble fraction showed the highest antibacterial activity against gram positive bacteria, while the chloroform, ethylacetate, butanol and aqueous fractions did not show any antibacterial activity. Minimum inhibitory concentration (MICs) on Staphyloycoccus aureus, Bacillus subtilis and Sateptococcus pneumoniae, n-hexane-soluble fraction were $100\;{\mu}g$, $500\;{\mu}g$ and $50\;{\mu}g$/disc, respectively. The antibacterial activity was not destroyed by heating at 80, 100, $120^{\circ}C$ for 30 min and was not affected by pH. In the inhibitory test against the Staphylococcus aureus, Bacillus subtilis and Sateptococcus pneumoniae, n-hexane-soluble fraction showed potent growth inhibition at the concentration of 0.1 and $0.5\;{\mu}g/mL$ for 12~24 hours and n-hexane-soluble fraction did not show any mutagenic activity.
The antimicrobial activity of 10 different traditional medicinal herbs extracts against E. coli was determined using paper disc method. Caesalpinia sappan and Rhus javanica, extracts in 5 mg/ml, and Paeonia suffruticosa and Seutellaria baicalensis extracts in 10 mg/ml showed significant antimicrobial activity against E. coli. Minimum inhibitory concentrations of medicinal herbs extracts were in the range of $1.4{\sim}8mg/ml\;and\;1.2{\sim}12mg/ml$, for MeOH extracts and EtOH extracts, respectively. In addition, the antimicrobial activity of each solvent fraction was most significant with the EtOAc layer. Optical density at 620nm after 24 hours incubation of E. coli in the presence of 100, 300 or 500 ppm of Caesalpinia sappan extract ranged from 0.02 to 0.1 compared to 0.6 in the absence of Caesalpinia sappan extract, indicating that growth of E. coli was significantly inhibited within 24 hours by the addition of at least 300 ppm of Caesalpinia sappan extract. Optical density at 620 nm after 24 hours incubation of E. coli in the presence of 300 ppm of Rhus javanica extract ranged from 0.02 to 0.2 compared to 0.5 in the absence of Rhus javanica extract, indicating that growth of E. coli was also significantly inhibited within 24 hours by the addition of at least 500 ppm of Rhus javanica extract. In conclusion, these findings suggest that extracts from medicinal herbs may play important roles in antimicrobial activities against E. coli.
We previously reported that the novel isoflavone-free peptide mixture (black soybean peptide, BSP) had several beneficial effects like antiobesity and hypotriglyceridemic effect. However, there are no reports for BSP on anti-hypertensive activity. BSP induced heme oxygenase-1 (HO-1) in HUVECs, thus investigated the HO-1-induced activity in HUVECs and the anti-hypertensive effects in SHR animal model. BSP significantly induced HO-1 expression both at transcriptional and protein levels in a time- and dose-dependent manner as measured by RT-PCR and Western blot analysis, respectively. These inductions were abolished by pretreatment of N-acetyl-cystein (NAC, 1~10 mM), but not by employing Tempol, a superoxide dismutase (SOD) mimetic (1~5 mM). As expected, enzymatic activity (~2 fold) determined by bilirubin formation assay and cGMP concentration (~6 fold) were significantly increased in BSP-treated cells. Based on the numerous evidences on the beneficial effects of HO-1 and our results, we investigated in vivo effects of BSP on the antihypertensive activity. The administration of BSP (1% in drinking water) significantly decreased mean blood pressure (BP) (from $218.6{\pm}6.99$ to $190.0{\pm}3.42$ mm Hg, p<0.01). This result indicates that BSP is strong inducer of HO-1 expression, which may be triggered by oxidative stress, and has anti-hypertensive activity.

We investigated the effects of heat treatment and extraction method on the antioxidant activities of five medicinal plants: Cyperus rotundus, Eucommia ulmoides, Bupleurum falcatum, Achyranthes japonica Nakai, and Akebia quinata. Extraction was performed with only ultrasound, ultrasound after heating at $130^{\circ}C$ for 2 hours, and reflux extraction with distilled water. The phenolic contents of reflux extraction and ultrasound extraction after heating were higher than only ultrasound extraction, and ultrasound extraction after heating samples was higher than reflux extraction except for Eucommia ulmoides and Cyperus rotundus. Total flavonoid content was higher in reflux and ultrasound extraction after heating samples than only ultrasound extraction, except for Cyperus rotundus. ABTS radical scavenging activity was higher in reflux extraction and ultrasound extraction after heating a sample, than only ultrasound extraction. DPPH radical scavenging activity was higher in reflux extraction except for Achyranthes japonica Nakai and Akebia quinata. The reducing power of ultrasound extraction after heating was higher with Achyranthes japonica Nakai. From the results of this study, we can expect to increase the antioxidant activity of medicinal plant extracts by applying suitable extraction and pretreatment conditions on the type of medicinal plant.
This study was conducted to investigate traditional medicinal plants (TMP) 1 and 2, two different multi-herbal mixtures consisting of 24 herbs. Regarding the contents of flavonoid compounds, the ethanol extract (EE) of TMP2 yielded the highest content of flavonoid compounds (40.981 mg/g), followed by EE of TMP1 (28.23 mg/g), hot water extract of TMP2 (WE, 10.046 mg/g), and WE of TMP1 (6.59 mg/g). Antioxidant activities of EE and WE of TMP1 and TMP2 were measured based on DPPH radical scavenging activity. At 1 mg/ml, the highest DPPH radical scavenging activity was shown in EE of TMP2 (96%), followed by EE of TMP1 (94%). Nitric oxide (NO) production by RAW264.7 macrophage cells stimulated with lipopolysaccharide (LPS) was reduced to 82, 77, 86, and 47% by addition of $500\;{\mu}g/ml$ of EE and WE of TMP1 and TMP2, respectively. These results were not due to the cytotoxicity of the extracts. NO synthesis was increased by 2.3, 3.6, 3.0, and 1.8 fold by addition of $500\;{\mu}g/ml$ of EE and WE of TMP1 and TMP2, respectively. These results show that medicinal plants play a significant role in antioxidative activity and activation of the immune system in the pathogenesis of inflammatory diseases, and different mixtures of the medicinal plants showed different effects.
