mTOR Signal Transduction Pathways Contribute to TN-C FNIII A1 Overexpression by Mechanical Stress in Osteosarcoma Cells

  • Osteosarcoma is the most common primary malignant bone tumor with a very poor prognosis. Treating osteosarcoma remains a challenge due to its high transitivity. Tenascin-C, with large molecular weight variants including different combinations of its alternative spliced FNIII repeats, is specifically over expressed in tumor tissues. This study examined the expression of Tenascin-C FNIIIA1 in osteosarcoma tissues, and estimated the effect of mechanical stimulation on A1 expression in MG-63 cells. Through immunohistochemical analysis, we found that the A1 protein was expressed at a higher level in osteosarcoma tissues than in adjacent normal tissues. By cell migration assay, we observed that there was a significant correlation between A1 expression and MG-63 cell migration. The relation is that Tenascin-C FNIIIA1 can promote MG-63 cell migration. According to our further study into the effect of mechanical stimulation on A1 expression in MG-63 cells, the mRNA and protein levels of A1 were significantly up-regulated under mechanical stress with the mTOR molecule proving indispensable. Meanwhile, 4E-BP1 and S6K1 (downstream molecule of mTOR) are necessary for A1 normal expression in MG-63 cells whether or not mechanical stress has been encountered. We found that Tenascin-C FNIIIA1 is over-expressed in osteosar-coma tissues and can promote MG-63 cell migration. Furthermore, mechanical stress can facilitate MG-63 cell migration though facilitating A1 overexpression with the necessary molecules (mTOR, 4E-BP1 and S6K1). In con-clusion, high expression of A1 may promote the meta-stasis of osteosarcoma by facilitating MG-63 cell migration. Tenascin-C FNIIIA1 could be used as an indicator in metastatic osteosarcoma patients.

  • Expression of chinese cabbage was conducted in order to investigate its dewatering behavior. Chipped cabbage was packed into cylinder and pressed by piston up to the predetermined pressure on Instron-1000. The rates of dewatering were affected domintantly by the applied pressure, but not significantly by the packed amount of cabbage in the cylinder. The pressure effect was increased very abruptly at first, but the increase rate was very low at high pressure greater than 20 MPa, showing great deviation from linear dependence of flow rate on pressure in Darcy's Law. Therefore, water expression from cabbage was not Newtonian flow of water through cell wall. In fact, the squeezed water contained a lot of solid particles, showing destruction of cell wall. The content of solid particles in expressed water was only slightly lower than the dry matter content of fresh chinese cabbage, determined by drying oven method.

  • This study is a understanding on technological specific expression to reflect of a research institute in modern architecture. For this study, it separately reduce space program, plan program, design elements of elevation, structural system, mechanical system, building material program to its expressional elements of research institute. Also, it's arranged about a specificity that is expressed for technology each plan elements of a research institute. As a result, elements what is expressive of technology on research institute are influenced an aesthetic expression, expression of an arrangement system, structural expression, expression of architectural organization, and it influences more getting feels technological specificity expressed an aesthetic, structural system, architectural composition, mechanical distribution system in order of their magnitude. This is offered suggestion what must be an achievable complex both a simply technological expression and aesthetic expression.

  • This study examined how the characteristics of abstractive form among various contenporary housing architecture have been expressed. The conclusions were: First, abstractive characteristics and types related to from expression of contemporary housing architectures were minimal form and absolute form of geometrical abstraction, plastic form and atypical form of expressive abstraction and mechanical aesthetics of industrial abstraction. Second, the typological form expression characteristics in minimal expression related to geometrical abstraction were simplicity, purity, and the properties of matter, and the characteristics in absolute expression were overlapping, obliqueness and dispensability. On the other hand, plasticity and mobility of materials were distinctive in plastic form expression, and inclination, curve and asymmetry were distinctive in atypical expression. The distinctive nature of mechanical aesthetics related to industrial abstraction included transparency, simplicity and. the properties of matter Funhermore, the study aimed at the understanding of various from expressions showed up in contemporary housing architecture, revealing the aspects of abstractive form expression characteristics.

Effects of GaAsAl laser on the spinal neuronal activity induced by noxious mechanical stimulation (GaAsAl 레이저가 물리적 통증반응과 관련된 척수내 신경세포의 활성에 미치는 영향)

  • The present study was designed to investigate the effect of low power GaAsAl laser on Fos expression in the spinal cord induced by noxious mechanical stimulation. Noxious mechanical stimulation was applied to the right hind paw following 30min of low power laser treatment using different intensity and treatment point and the resulting Fos expression in the spinal cord dorsal horn was compared to that obtained in rats exposed only to the noxious mechanical stimulation. The results were summarized as follows: 1. In intact control rats, only a few Fos like immunoreactive(Fos-IR) neurons were evident in the lumbar spinal cord dorsal horn. Similarly, following prolonged inhalation anesthesia, Fos-IR neurons were absent in the dorsal horn of the lumbar spinal cord. In animals treated with noxious mechanical stimulation, neurons with nuclei exhibiting Fos immunostaining were distributied mainly in the medial half of ipsilateral laminae I-V at lumbar segments L3-5. These findings directly indicated that prolonged anesthesia used in this study did not affect the Fos expression in the spinal cord dorsal horn of intact animals and noxious mechanical stimulation treated animals. 2. In acupoint treated animals, 10mW of laser stimulation, not 3mW intensity, significantly reduced the number of Fos immunoreactive neurons in the spinal dorsal horn induced by noxious mechanical stimulation(P<.01). However, the supressive effect of low power laser stimulatin was not observed in 3m Wand 10m W of laser stimulation into non-acupoint. These data indicate that 10mW of low power laser stimulation into acupoint is capable of inhibiting the expression of Fos in the dorsal horn induced by noxious mechanical stimulation. In conclusion, these findings raise the possibility that low power laser stimulation into acupoint may be a promising alternative medicine therapy for the mechanical stimulation induced pain in the clinical field.

The effect of UNCL inactivation on the expression of mechanical stress related genes in cultured human PDL fibroblasts

  • A mutation of UNCL, an inner nuclear membrane RNAbinding protein, has been found to eliminate mechanotransduction in Drosophila. UNCL is expressed in human periodontal tissue including in periodontal ligament (PDL) fibroblasts. However, it is unclear how a mechanical stimulus is translated into cellular responses in PDL fibroblasts. The aim of this study was to evaluate the effect of UNCl on mechanical stress related genes in PDL fibroblasts in response to mechanical stress. The mRNA of TGF-$\beta$, COX-2, and MMP-2 was up-regulated after UNCL inactivation in PDL fibroblasts under the compression force. Under the tensile force, inactivation of UNCL decreased the expression of Biglycan, RANKL, MMP-2, and TIMP-2 mRNAs while it increased the expression of TIMP-1. p38-MAPK was expressed in PDL fibroblasts under compression forces whereas phospho-ERK1/2, p65-NFkB, and c-fos were expressed under tension forces. The expression and phosphorylation of the mechanical stress related genes, kinases, and transcription factors were changed according to the types of stress. Furthermore, most of them were regulated by the inactivation of UNCL. This suggests that UNCL is involved in the regulation of mechanical stress related genes through the signaling pathway in PDL fibroblasts.

Influence of Moisture Content and Seed Dimensions on Mechanical Oil Expression from African Oil Bean (Pentaclethra macrophylla Benth) Seed

  • Purpose: New low-cost oilseeds are needed to meet an ever-increasing demand for oil for food, pharmaceutical, and industrial applications. African oil bean seed is a tropical crop that is underutilized and has high oil yields, but there have been no studies conducted on its mechanical oil expression up to now. The objective of this work was to investigate the effect of moisture content and seed dimensions on mechanical oil expression from the seeds. Methods: Fresh oil bean seeds were procured, de-hulled, and cleaned. Initial seed moisture content, obtained in accordance with the ASAE standard, was 12% dry basis (db). The seeds were further conditioned by dehydration and rehydration prior to oil expression to obtain four other moisture levels of 8, 10, 14, and 16% db. The major diameter of the seeds was measured using digital vernier calipers, and the seeds were classified into size dimensions (< 40, 41-45, 46-50, 51-55, and > 55 mm). The oil yield and expression efficiency were obtained in accordance with standard evaluation methods. Results: The highest oil yield and expression efficiency (47.74% and 78.96%, respectively) were obtained for a moisture content of 8% db and seed dimensions of < 40 mm, while the lowest oil yield and expression efficiency (41.35% and 68.28%, respectively) were obtained for a moisture content of 14% db and seed dimensions between 51-55 mm. A mathematical model was developed to predict oil yield for known moisture content and seed dimensions, with a coefficient of determination $R^2$ of 95% and the confidence level of the predictive model of 84.17%. The probability of prediction F ratio showed that moisture content influence was more significant than seed dimensions. Conclusions: The higher the moisture content and larger the seed dimensions, the lower the oil yield from African oil bean seeds.

In vitro-growth and Gene Expression of Porcine Preantral Follicles Retrieved by Different Protocols

  • This study was conducted to determine how the isolation method of the porcine preantral follicles influenced the following follicular growth in vitro. Mechanical and enzymatical isolations were used for retrieving the follicles from prepubertal porcine ovaries, and in vitro-growth of the follicles and the expression of folliculogenesis-related genes were subsequently monitored. The enzymatic retrieval with collagenase treatment returned more follicles than the mechanical retrieval, while the percentage of morphologically normal follicles was higher with mechanical retrieval than with enzymatic retrieval. After 4 days of culture, mechanically retrieved, preantral follicles yielded more follicles with normal morphology than enzymatically retrieved follicles, which resulted in improved follicular growth. The mRNA expression of FSHR, LHR Cx43, DNMT1 and FGFR2 genes was significantly higher after culture of the follicles retrieved mechanically. These results suggest that mechanical isolation is a better method of isolating porcine preantral follicles that will develop into competent oocytes in in vitro culture.

Preemptive application of QX-314 attenuates trigeminal neuropathic mechanical allodynia in rats

  • The aim of the present study was to examine the effects of preemptive analgesia on the development of trigeminal neuropathic pain. For this purpose, mechanical allodynia was evaluated in male Sprague-Dawley rats using chronic constriction injury of the infraorbital nerve (CCI-ION) and perineural application of 2% QX-314 to the infraorbital nerve. CCI-ION produced severe mechanical allodynia, which was maintained until postoperative day (POD) 30. An immediate single application of 2% QX-314 to the infraorbital nerve following CCI-ION significantly reduced neuropathic mechanical allodynia. Immediate double application of QX-314 produced a greater attenuation of mechanical allodynia than a single application of QX-314. Immediate double application of 2% QX-314 reduced the CCI-ION-induced upregulation of GFAP and p-p38 expression in the trigeminal ganglion. The upregulated p-p38 expression was co-localized with NeuN, a neuronal cell marker. We also investigated the role of voltage-gated sodium channels (Navs) in the antinociception produced by preemptive application of QX-314 through analysis of the changes in Nav expression in the trigeminal ganglion following CCI-ION. Preemptive application of QX-314 significantly reduced the upregulation of Nav1.3, 1.7, and 1.9 produced by CCI-ION. These results suggest that long-lasting blockade of the transmission of pain signaling inhibits the development of neuropathic pain through the regulation of Nav isoform expression in the trigeminal ganglion. Importantly, these results provide a potential preemptive therapeutic strategy for the treatment of neuropathic pain after nerve injury.

  • The purpose of this study is to consider the changes of the early twentieth century fashion in pursuit of mechanical beauty. As for the research methodology, literature survey has been undertaken. The results of this study are as following. In early twentieth century, fashion concept on the expression of mechanical beauty was considered as abstractness, atypicality, and high-tech. The abstractness of fashion was expressed by geometrical purity, box silhouette, moderate color, and cheap materials in association with mechanical reasonability and efficiency. The atypicality of fashion was connected progressive taste motivated by mechanical dynamic effect. This tendency of dress was constructed technically in response with the movement of body and totally transgressed conventional notions of clothing. The high-tech of fashion was presented by geometrical forms and metal materials in relation with the future-oriented utopia and the harmony between human and machine.
