알스트로메리아는 절화 국제 시장에서 가장 중요한 작물 중 하나이다. 특히 긴 개화기간, 다양한 꽃 색깔, 그리고 저온적응성이 높고 배양기간 동안 낮은 에너지를 필요로 한다. 알스트로메리아는 낮은 증식률, 긴 시간이 걸리는 배양 과정, 높은 바이러스 감염율 때문에 기내 배양 기술이 오늘날까지 계속해서 발전해왔다. 캘러스 유도는 직접적인 식물체 발생 방법에 비해 다양한 배양 부위를 가지고 있고 캘러스만 유지를 잘한다면 캘러스를 유지하면서 식물체 분화를 동시에 할 수 있어 대량 증식에 많은 이용이 가능하다. 본 연구에서는 효율적인 캘러스 유도를 위한 다양한 호르몬과 품종을 비교하여 실험을 진행하였다. 마디 및 마디사이를 배양하여 실험을 진행한 결과, 8주 후부터 캘러스가 발생하기 시작하였고, 마디사이에 비해 마디에서의 캘러스 발생율이 높게 측정되었다. 또한 2,4-D와 picloram을 비교한 실험에서는 2,4-D를 처리한 배지에서 캘러스 발생율이 최대 2배 이상 높게 측정되었다. 이러한 결과를 이용하여 알스트로메리아 대량증식 시스템 체계 확립 및 신품종 육성에 도움을 줄 것이라 생각되어진다.
For the newly isolated $H_2$-producing chemoheterotrophic bacterium Citrobacter amalonaticus Y19, anaerobic glucose metabolism was studied in batch cultivation at varying initial glucose concentrations (3.5-9.5 g/l). The carbon-mass and energy balances were determined and utilized to analyze the carbon metabolic-pathways network. The analyses revealed (a) variable production of major metabolites ($H_2$, ethanol, acetate, lactate, $CO_2$, and cell mass) depending on initial glucose levels; (b) influence of NADH regeneration on the production of acetate, lactate, and ethanol; and (c) influence of the molar production of ATP on the production of biomass. The results reported in this paper suggest how the carbon metabolic pathway(s) should be designed for optimal Hz production, especially at high glucose concentrations, such as by blocking the carbon flux via lactate dehydrogenase from the pyruvate node.
본 연구는 아그로박테리움을 이용한 기능획득 일일초 모상근의 대량생산을 위한 조건 확립에 대한 것이다. 본 연구에서는, 효율적인 형질전환 일일초 모상근 생산에 있어서의 최적의 일일초 품종의 선발과 최적의 일일초 조직을 결정하였으며, 또한 다양한 배지에 있어서의 모상근 유도를 조사하였다. 최종적으로 약 2,500개의 독립적인 형질전환 일일초 모상근 line을 생산하였으며, 또한 이들을 이용하여, 대사체 연구를 위한 효율적 관리 시스템을 구축하였다. 이들 모상근 line은 일일초 인돌알칼로이드 생합성 관련 유전자의 발굴 및 기능해석에 유용하게 쓰일 것이다.
A soybean-sprout rot epidemic occurred in a mass production soybean sprout factory in 2000 and 2001 in Korea, which caused up to 20% production loss. Among the causal pathogenic bacteria and fungi, Pythium deliense was found to be the dominant pathogen of severe root and hypocotyls rot, particularly in recirculating water system. An average of 90% of the isolated fungi from the rotted sprout on potato dextrose agar were Pythium sp. The fungal density of Pythium in the sampled water was monitored in the recycled water system for 1 year using a selective medium (com meal agar with Pimaricin, 10 mg; Rifampicin, 10 mg; and Ampicillin, 100 mg per 1 liter). The drained water from the soybean-sprout cultivation always had a certain amount of fungus in it. The removal of Pythium from the recycling water system must be thorough, safe, and environment friendly. However, the pathogen in the water was easily found even after ozone and chlorine treatments, which were devised on the recycling system for the removal of microorganisms. 5-$\mu\textrm{m}$ pore size filter was applied and was able to successfully control the disease. As the sprout industry increasingly shifts into mass production, the demand for water will increase continuously. Recycling water for sprout production is eco-friendly. However, a process must be devised to be able to first decompose organic matters before Pythium zoospores are filtered.
효모 재순환계에 의한 당밀의 연속발효를 하였다. 단계적으로 YPD배지 농도를 10g/l에서 183.5g/l로 증가시킨 후 최종 발효배지는 당밀로 하였다. 최고의 효모농도는 25g/l였고, 운전중 평균 효모농도는 23.5g/l으로 종래 회분식보다 3.4배 높았다. 최적 발효조건은 180g/l의 당밀배지를 공급하였을 때 교반속도 500rpm, 희석을 0.037$h^{-1}$였다. 이 조건을 645시간 연속 운전하였으며 최고 알코올 생산성은 4.9g$l\cdot h$로서 이때의 희석율은 0.091$h^{-1}$, 알코올 농도는 53.9g/l이었다.
Objective: SanYangSam and SanYangSanSam are traditional Korea-medical herbs that are grown from Panax ginseng C.A. Meyer. In our previous studies, we found that the functional compounds in SanYangSam and SanYangSanSam were different and depended on the type and the cultivation environment of ginseng. This study aimed to profile the functional constituents in SanYangSam and SanYangSanSam. Methods: To profile the functional aspects of the many compounds that have therapeutic activities in SanYangSam and SanYangSanSam extracts, we used liquid chromatography tandem mass spectrometry and quadrupole orthogonal acceleration time-of-flight mass spectrometry. Results: A total of four major compounds were detected; two of which were the natural flavonoids kaempferol and quercetin. Among others, two polyacetylene compounds, including panaxydol and panaxynol, were detected. Conclusion: In this study, we found that panaxydol, one of the polyacetylene constituents of ginseng, is a candidate anti-cancer agent in SanYangSam and SanYangSanSam pharmacopuncture. In addition, we found that the panaxydol levels in the SanYangSanSam extract were over 30 times those in the SanYangSam extract.
Major progress has been made in the past decade towards understanding of the biosynthesis of red carotenoid astaxanthin and its roles in stress response while exploiting microalgae-based astaxanthin as a potent antioxidant for human health and as a coloring agent for aquaculture applications. In this review, astaxanthin-producing green microalgae are briefly summarized with Haematococcus pluvialis and Chlorella zofingiensis recognized to be the most popular astaxanthin-producers. Two distinct pathways for astaxanthin synthesis along with associated cellular, physiological, and biochemical changes are elucidated using H. pluvialis and C. zofingiensis as the model systems. Interactions between astaxanthin biosynthesis and photosynthesis, fatty acid biosynthesis and enzymatic defense systems are described in the context of multiple lines of defense mechanisms working in concert against photooxidative stress. Major pros and cons of mass cultivation of H. pluvialis and C. zofingiensis in phototrophic, heterotrophic, and mixotrophic culture modes are analyzed. Recent progress in genetic engineering of plants and microalgae for astaxanthin production is presented. Future advancement in microalgal astaxanthin research will depend largely on genome sequencing of H. pluvialis and C. zofingiensis and genetic toolbox development. Continuous effort along the heterotrophic-phototrophic culture mode could lead to major expansion of the microalgal astaxanthin industry.
본 연구에서는 새송이버섯 병재배방식과 느타리버섯 봉지재배방식의 재배단계별(최초 배지 단계, 균사체 배양 후 배지 단계, 자실체 수확 후 폐배지 단계)로 배지의 화학 성분의 흐름을 추적 구명하고, 독성 중금속(Cd, Pb, As)과 잔류 농약으로부터의 안전성을 평가하였다. 양 재배방식 공히 최초 배지와 비교해서 자실체 수확 후의 영양소의 양적 감소 정도에 있어서 총무게는 평균 29%, 건물과 유기물은 공히 21~25%, 섬유소는 19~22% 감소하였다. 재배단계에서 분해 이용된 유기물 량의 2/3~3/4은 섬유소(NDF)였으며, 이용된 섬유소의 주된 성분(50~70%)은 hemicellulose이었다. 균사체 성장이 배지의 화학적 성분에 미치는 효과는 새송이버섯의 병재배 시에는 미미하였으나(P>0.05), 느타리버섯의 봉지재배 시에는 ADF 성분을 제외한 거의 모든 유기물과 무기물이 분해 소실되는 것으로 나타났다(P<0.05). 자실체는 봉지재배 시보다는 병재배 시에 배지내의 영양소를 매우 활발하게 흡수하였다(P<0.05). 수확한 자실체는 폐배지보다 조단백질, 비섬유성탄수화물, 조회분 성분이 훨씬 높고(P<0.05), NDF 성분은 훨씬 낮아서(P<0.05), 폐배지의 화학적 조성에 바람직하지 못한 영향을 미치는 것으로 나타났다. 폐배지는 독성 중금속과 잔류농약의 오염이 거의 없어 위생적으로 안전하였다. 결론적으로 최초 배지와 비교해서 폐배지는 난분해성 섬유소 성분이 증가하고, 조단백질 중 순수단백질이 줄고, 비소화성단백질이 느는 등 질적으로 떨어져서 사료영양적 가치는 상대적으로 하락하는 것으로 나타났다.
Alcaligenes latus 유래의 phbC 유전자를 A. eutrophus에 재도입시킨 형질전환균주를 이용하여 높은 4HB 몰분율을 갖는 P(3HB-4HB)의 고농도 생산을 시도하였다. 형질전환균주는 총균체량, P(3HB-4HB) 농도, 그리고 축적률에서 모균주에 비해 다소 증가한 반면 P(3HB-4HB)내의 4HB 몰분율은 23.5 mol%로 모균주의 12.3 mol%에 비해 현저히 증가하였다. 이는 phbC유전자의 증폭으로 인해 해당과정에서 생성된 3HB와 전구물질인 ${\gamma}$-butyrolacton에서 전환된 4HB의 중합반응이 촉진되기 때문으로 사료된다. 또한 ${\gamma}$-butyrolacton의 농도 $Mg^{2-}$ 이온, 그리고 citrate 첨가량이 P(3HB-4HB)의 농도, 축적률, 그리고 4HB 몰분율에 미치는 영향을 검토하였다. P(3HB-4HB)내의 4HB 몰분율을 증대시키기 위하여 일반적으로 사용되는 2단계 배양법을 변형시켜 ${\gamma}$-butyrolacton과 citrate의 첨가시기를 늦춘 2단계 배양법을 활용하여 P(3HB-4HB)내의 4HB 몰분율을 61.0 mol%로 증가시킬 수 있었다. 또한 ${\gamma}$-butyrolacton의 첨가량을 조절하여 P(3HB-4HB)내의 4HB 몰분율이 92.0 mol%에 이르는 homopolymeric P(4HB)를 생산할 수 있었으며, 그 구조를 $^1$H-NMR을 통해 확인하였다.
Background: The geographical origin of Panax ginseng Meyer, a valuable medicinal plant, is important to both ginseng producers and consumers in the context of economic profit and human health benefits. We, therefore, aimed to discriminate between the cultivation regions of ginseng using the stable isotope ratios of C, N, O, and S, which are abundant bioelements in living organisms. Methods: Six Korean ginseng cultivars (3-yr-old roots) were collected from five different regions in Korea. The C, N, O, and S stable isotope ratios in ginseng roots were measured by isotope ratio mass spectrometry, and then these isotope ratio profiles were statistically analyzed using chemometrics. Results: The various isotope ratios found in P. ginseng roots were significantly influenced by region, cultivar, and the interactions between these two factors ($p{\leq}0.001$). The variation in ${\delta}^{15}N$ and ${\delta}^{13}C$ in ginseng roots was significant for discriminating between different ginseng cultivation regions, and ${\delta}^{18}O$ and ${\delta}^{34}S$ were also affected by both altitude and proximity to coastal areas. Chemometric model results tested in this study provided discrimination between the majority of different cultivation regions. Based on the external validation, this chemometric model also showed good model performance ($R^2=0.853$ and $Q^2=0.738$). Conclusion: Our case study elucidates the variation of C, N, O, and S stable isotope ratios in ginseng root depending on cultivation region. Hence, the analysis of stable isotope ratios is a suitable tool for discrimination between the regional origins of ginseng samples from Korea, with potential application to other countries.
