Antioxidant properties of tidal pool microalgae, Halochlorococcum porphyrae and Oltamannsiellopsis unicellularis from Jeju Island, Korea

  • In this study, we assessed the antioxidant properties of tidal pool microalgae, Halochlorococcum porphyrae and Oltamannsiellopsis unicellularis, from Jeju Island, Korea. Specifically, the antioxidant activity of fractions isolated from 80% methanol extract, and digests produced from five proteases and carbohydrases, were investigated. Almost all the fractions and the 80% methanol extract exhibited higher effects on 1,1-Diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging. The ethyl acetate fraction showed the highest superoxide anion scavenging activity, while both n-hexane and chloroform fractions exhibited higher $H_2O_2$ scavenging activity. Among the enzymatic digests from H. porphyrae and O. unicellularis, all the digests exhibited remarkable DPPH scavenging activities. In nitric oxide inhibition, all the digests recorded significantly higher effects than those of the commercial antioxidants (p < 0.05). Flavozyme and Neutrase digests from H. porphyrae, and Termamyl and Alcalase digests from O. unicellularis, showed significant effects in metal chelating. Lipid peroxidation was significantly inhibited in the ethyl acetate fraction, in the Celluclast and Protamex digests from H. porphyrae, and in the chloroform fraction from O. unicellularis. These findings suggest that the two tidal pool microalgae tested in this study are rich in potential antioxidative compounds, the specific properties of which can be considered for use in the food and pharmaceutical industries.

Antibacterial Activity of Dielectric Barrier Discharge Plasma against Main Food-borne Bacteria in Suspensions (유전체장벽방전 플라즈마에 의한 주요식중독세균의 살균 효과)

  • Dielectric barrier discharge (DBD) plasma is one of the promising next generation non-thermal technologies for food sterilization. The present study investigated the effects of DBD plasma on the reduction of most common food-borne pathogenic bacteria (Staphylococcus aureus, Bacillus cereus, Vibrio parahaemolyticus, Salmonella enterica) and sanitary indicative bacteria (Escherichia coli) in the suspension (initial inoculum of approx. 9 log CFU/mL). The bacterial counts were significantly (P<0.05) reduced with the increase in the treatment time (1-30 min) of DBD plasma in the suspension. The D-values (time for 90% reduction) of DBD plasma by first-order kinetics for S. aureus, B. cereus, V. parahaemolyticus, S. enterica, and E. coli were 17.76, 19.96, 32.89, 21.55, and 15.24 min, respectively (R2>0.90). These results specifically showed that 30 min of DBD plasma treatment in > 90% reduction of seafood-borne pathogenic and sanitary indicative bacteria. This suspension study may provide the basic data for use in seafood processing and distribution.

  • The antibacterial activity of marine bacterium(Pseudomonas aeruginosa JYMB1-3) was assayed against Vibrio anguillarum with the aim of evaluating the possible use for biocontrolling fish disease as probiotic strain. Inhibition test on the solid medium showed that vibrios were especially sensitive to the JYMB1-3. Edwardsiella tarda, Streptococcus sp. and Staphylococcus epidermidis were also sensitive to that strain, however the antibacterial abilities were varied to the pathogens. The vibriostatic activity of antibacterial substance produced from $10^7$ cells of the strain for 24 hours was equivalent to $2.5{\mu}g$ of chloramphenicol.

  • Dual frequency acoustic pinger(AP) was manufactured to reduce study effect by long-term use of developed single frequency AP to prevent cetacean bycatch. Directivity characteristic of transducer was the omnidirectional pattern which showed less than ${\pm}3dB$ the change range of sensitivity on the beam pattern of right and left. Source power level(SPL) was 1384311pa with epoxy window before casing however after casing 1170B11Pa at sea. Dual frequency Af was tested to identify the avoidance behavior of bottlenose dolphin by its working. However the efficiency of dual frequency AP about the study effect was verified experiment repeatedly using single and dual frequency AP.

  • Recently, with the rapid development in the industries such as mechanical plants, automobiles, ships and marine structures, it is enlarged by the use of the SS 41 steel. This mechanical plants and marine structures are exposed m corrosion because of Cl-under marine environments. To protect their accidents, mainly applied anti-corrosion epoxy coating and various protective its structures. In this study, corrosion control characteristics on the epoxy coating were investigated by the galvanic corrosion of impressed voltage tester under marine environments The main results obtained are as follows; 1. Corrosion current density of amine-epoxy coating becomes more increased than that of other epoxy coating and the time area rate of pin hole and pit until 5% becomes most rapid. 2. The potential of SUS 304 stainless steel(cathode) for Al-epoxy coating is nearly zero potential. 3. Corrosion current density of Amine-epoxy by shot blast becomes more decreased than that of not shot blast and cathodic potential becomes more noble. 4. As distance of anode and cathode is more decreased, corrosion current density of epoxy coating is more increased and cathodic potential becomes less noble.

Genetic Diversity in Cultured and Wild Populations of the Ascidian Halocynthia roretzi Inferred from Mitochondrial DNA Analysis

  • Nucleotide sequences of about 500 bp from the 5' end of mitochondrial (mt) DNA Cytochrome Oxidase I (COI) were analyzed to estimate the genetic variation between wild and cultured populations of the ascidian Halocynthia roretzi from two sites along the coast of Korea. A total of 25 haplotypes were defined by 21 variable nucleotide sites in the examined COI region. Genetic diversity (haplotype diversity and nucleotide divergence) of wild populations was higher than that of the cultured population. These data suggest that reduced genetic variation in the cultured population may have results from bottleneck effect caused by the use of a limited number of parental stock and pooling of gametes for fertilization. Pairwise population $F_{ST}$ estimates inferred that wild and cultured populations were genetically distinct. The combined results suggest that sequence polymorphism in the COI region would be preferable for estimating the genetic diversity of ascidian populations.

  • In these days, there are increasing demands for weight reduction in many industrial fields including marine industries. Therefore, magnesium thin films for lightweight materials were prepared on magnesium alloy substrate. The influence of gas pressure and substrate bias voltages on the crystal orientation and morphology of the films was determined by using X-ray diffraction and FE-SEM, respectively. And the effect of crystal orientation and morphology of the magnesium thin films on corrosion behavior was estimated by measuring electro-chemical anodic polarization curves in deaerated 3% NaCl solution. From the results, corrosion resistance of Mg thin films was improved by controlling the crystal orientation and morphology of the films with effective use of plasma ion plating technique.

  • In order to substitute incandescent lamps, a power supply device for 7W class LED lamps which are environmentally friendly and energy saving is designed LED lamps consist of a multitude of chip LED connected in parallel and series. 11ms it is necessary to supply LED lamps with DC voltage and current. However, when LED lamps are in use, they are connected directly to AC 220V. This is why we need to have AC/DC, DC/DC power converters including a control system of voltage and current. For this, a transformerless and simple LED lamp driver is designed 조ich can control the current and output voltage for LED string of LED lamp.

  • Bio-energy offers the opportunity to lessen fossil fuel consumption. Energy derived from solar, wind, hydroelectric, geothermal, and biomass sources are considered renewable. Because most forms of bio-energy are derive deither directly or indirectly from the sun, there is an abundant supply of renewable energy available, unlike fossil fuels. The use of bio-energy also provides environmental, economic and political benefits. Bio-energy can be produced from a marine source such as biomass provides a $CO_2$ neutral, non-polluting form of energy. In this paper, the potential of marine biomass is increasingly discussed, given the size of the resource in that more than three quarters of the surface of planet earth is covered by water.

Survival of Anisakis species larvae of chub mackerel (Scomber japonicus) in different kinds of condiments

  • Anisakiasis is a well-known zoonosis caused by ingestion of raw or thermally undercooked seafood product contaminated with live Anisakis nematode third stage larvae (L3). Several traditional processing techniques have been used to kill or remove the Anisakis larvae worldwide, but thermal processing or deep freezing are the most effective treatments to kill the Anisakis larvae. In this study, we investigated the survival of Anisakis larvae in several condiments (soy bean sauce, wasabi, vinegar, red pepper paste) commonly consumed when eating raw fish in Korea. We also examined several different media (NaCl solution, absolute alcohol, soju) to investigate their larvicidal effect. When directly exposed to various condiments, the most effective larvicidal effect was observed in the mixture of wasabi and soy bean sauce. When exposed to different NaCl solutions, the larvicidal ability became more effective as the concentration increased, but did not show 100% killing effect. In soju, the L3 were killed under less than 4 hr. We observed the larvicidal effects of several condiments in this study, but these results are thought to be carefully interpreted for actual use because all the condiments in this study showed the effect in hours and in general, the L3 are exposed to these condiments only for seconds before ingested in real situation.
