Effect of Dietary Marine Microalgae (Schizochytrium) Powder on Egg Production, Blood Lipid Profiles, Egg Quality, and Fatty Acid Composition of Egg Yolk in Layers

  • Two hundred and sixteen Institut de S$\acute{e}$lection Animale (ISA) brown layers (40 wks of age) were studied for 6 wks to examine the effect of microalgae powder (MAP) on egg production, egg quality, blood lipid profile, and fatty acid concentration of egg yolk. Dietary treatments were as follows: i) CON (basal diet), ii) 0.5% MAP (CON+0.5% Schizochytrium powder), and iii) 1.0% MAP (CON+1.0% Schizochytrium powder). From 44 to 46 wks, egg production was higher in 1.0% MAP treatment than in control treatment (linear, p = 0.034); however, there was no difference on the egg production from 40 to 43 wks (p>0.05). Serum triglyceride and total cholesterol were significantly reduced in the groups fed with MAP, compared to those in groups fed with control diets (Quadratic, p = 0.034 and p = 0.039, respectively). Inclusion of 0.5% MAP in the diet of layers improved egg yolk color, compared with hens fed with basal diet at 46 wks (quadratic, p = 0.044). Eggshell thickness was linearly increased in MAP-fed treatments at 46th wk (p<0.05). Concentration of yolk docosahexaenoic acid (DHA; C22:6n-3) was increased in treatment groups fed with MAP (linear, p<0.05). The n-6 fatty acids, n-6/n-3 fatty acid, and unsaturated fatty acid/saturated fatty acid were decreased in treatment groups fed with MAP (linear, p<0.05). These results suggest that MAP improved the egg production and egg quality, and may affect serum lipid metabolites in the layers. In addition, MAP increases yolk DHA levels, and deceases n-6/n-3 fatty acid ratio.

Methods for sampling and analysis of marine microalgae in ship ballast tanks: a case study from Tampa Bay, Florida, USA

  • Ballasting and deballasting of shipping vessels in foreign ports have been reported worldwide as a vector of introduction of non-native aquatic plants and animals. Recently, attention has turned to ballast water as a factor in the global increase of harmful algal blooms (HABs). Many species of microalgae, including harmful dinoflagellate species, can remain viable for months in dormant benthic stages (cysts) in ballast sediments. Over a period of four years, we surveyed ballast water and sediment of ships docked in two ports of Tampa Bay, Florida, USA. Sampling conditions encountered while sampling ballast water and sediments were vastly different between vessels. Since no single sample collection protocol could be applied, existing methods for sampling ballast were modified and new methods created to reduce time and labor necessary for the collection of high-quality, qualitative samples. Five methods were refined or developed, including one that allowed for a directed intake of water and sediments. From 63 samples, 1,633 dinoflagellate cysts and cyst-like cells were recovered. A native, cyst-forming, harmful dinoflagellate, Alexandrium balechii (Steidinger) F. J. R. Taylor, was collected, isolated, and cultured from the same vessel six months apart, indicating that ships exchanging ballast water in Tampa Bay have the potential to transport HAB species to other ports with similar ecologies, exposing them to non-native, potentially toxic blooms.

Nannochloris eucaryotum growth: Kinetic analysis and use of 100% CO2

  • Microalgae are receiving an increasing attention because of their potential use as $CO_2$ capture method and/or as feedstock for biofuels production. On the other hand the current microalgae-based technology is still not widespread since it is characterized by technical and economic constraints that hinder its full scale-up. In such contest the growth kinetics of Nannochloris eucaryotum (a relatively unknown marine strain) in batch and semi-batch photobioreactors is quantitatively investigated with the aim of obtaining the corresponding kinetic parameters suitable for process engineering and its optimization. In particular the maximum growth rate was evaluated to be 1.99 $10^{-3}\;h^{-1}$. Half saturation concentrations for nitrates ($K_N$) and phosphates uptake ($K_P$) were evaluated as 5.4 $10^{-4}\;g_N\;L^{-1}$ and 2.5 $10^{-5}\;g_P\;L^{-1}$, respectively. Yield factors for nitrogen ($Y_N$) and phosphorus ($Y_P$) resulted to be 5.9 $10^{-2}\;g_N\;g^{-1}$ biomass and 6.0 $10^{-3}\;g_P\;g^{-1}{_{biomass}}$, respectively. The possibility of using 100% (v/v) $CO_2$ gas as carbon source is also evaluated for the first time in the literature as far as N. eucaryotum is concerned. The strain showed a good adaptability to high concentrations of dissolved $CO_2$ as well as to low pH. The lipid content under 100% $CO_2$ is about 16.16 %wt $wt^{-1}$ and the fatty acid methyl esters composition of the extracted oil is in compliance with the European regulation for quality biodiesel.

Application of FITC-conjugated lectin probes for the recognition and differentiation of some Korean coastal red tide microalgae

  • Harmful micro algae isolated from Korean coastal waters, were tested with FITC-conjugated lectins and observed by epifluorescent microscopy to distinguish each other. Strain-specific sugar composition at the cell surface was suggested by the affinity of lectins to different microalgae. The micro algae Cochlodinium polykrikoides (CP-1) and Gymnodinium $A_3\;(GA_{3-1}\;1)$, are morphologically similar, but exhibited different binding activity with the lectins ECA, HPA and WGA. In Peridiniales, the micro alga Alexandrium tamarense (AT) bound HPA and WGA, but Scrippsiella trochoidea (ST-1) did not bind those lectins. Three species of Prorocentrum also exhibited different binding specificity with HPA, PHA and SBA. A non­toxic Korean isolate of Heterosigma akashiwo (HA-2) bound ConA, PEA and UEA. These results suggest that lectins are useful in discriminating morphologically similar species, as well as different species or strains within the same genus.

Gene Expression Analysis of Zeaxanthin Epoxidase from the Marine Microalga Dunaliella tertiolecta in Response to Light/Dark Cycle and Salinity

  • Zeaxanthin is an important pigment in the photo-protection mechanism of microalgae. However, zeaxanthin epoxidase, an enzyme involved in the accumulation and conversion of zeaxanthin, has not been extensively studied in microalgae. In this work, we report the expression pattern of zeaxanthin epoxidase in Dunaliella tertiolecta (DtZEP) at different light and diverse salinity conditions. To confirm the responsiveness to light conditions, the ZEP expression pattern was investigated in photoperiodic (16 h of light and 8 h of dark) and continuous (24 h of light and 0 h of dark) light conditions. mRNA expression levels in photoperiodic conditions fluctuated along with the light/dark cycle, whereas those in continuous light remained unchanged. In varying salinity conditions, the highest mRNA and protein levels were detected in cells cultured in 1.5 M NaCl, and ZEP expression levels in cells shifted from 0.6 M NaCl to 1.5 M NaCl increased gradually. These results show that mRNA expression of DtZEP responds rapidly to the light/dark cycle or increased salinity, whereas changes in protein synthesis do not occur within a short period. Taken together, we show that DtZEP gene expression responds rapidly to light irradiation and hyperosmotic stress. In addition, ZEP expression patterns in light or salinity conditions are similar to those of higher plants, even though the habitat of D. tertiolecta is different.

Characterization of Heterochlorella luteoviridis (Trebouxiaceae, Trebouxiophyceae) isolated from the Port of Jeongja in Ulsan, Korea

  • A unicellular green alga was axenically isolated from the Port of Jeongja, Ulsan, Korea. Morphological, molecular, and biochemical analyses revealed that the isolate belonged to Heterochlorella luteoviridis. This is the first report of this species in Korea. The microalgal strain was named as H. luteoviridis MM0014 and its growth, lipid composition, and biomass properties were investigated. The strain thrived over a wide range of temperatures ($5-30^{\circ}C$) and withstood up to 0.5 M NaCl. The results of gas chromatography/mass spectrometry analysis showed that the isolate was rich in nutritionally important polyunsaturated fatty acids. Its major fatty acids were linoleic acid (35.6%) and ${\alpha}$-linolenic acid (16.2%). Thus, this indigenous marine microalga is a potential alternative source of ${\omega}3$ and ${\omega}6$ polyunsaturated fatty acids, which are currently obtained from fish and plant oils. Ultimate analysis indicated that the gross calorific value was $19.7MJ\;kg^{-1}$. In addition, the biomass may serve as an excellent animal feed because of its high protein content (51.5%). Therefore, H. luteoviridis MM0014 shows promise for applications in the production of microalgae-based biochemicals and biomass feedstock.

  • Global warming has affected the distribution of organisms for decades and has displayed rapid ascent recently. Research into the effects on tropical organisms are vital. Padina boryana is a resourceful marine microalgae in the Maldives Sea in the Laccadive region. A 70% ethanol extraction (PBE) of this seaweed was used to investigate its antioxidant potential. Both in vitro and in vivo models were implemented. PBE exhibited protective potential against H2O2 induced apoptosis. ROS levels were suppressed due to PBE. PBE expressed a cytoprotective nature. In vivo experiments involving the zebra fish model conformed its validity. The antioxidant efficacy of PBE was dose dependent. Study outcomes suggest PBE has potential as a novel and valuable marine resource to aid the functional food and cosmeceutical industries.

  • Gonad maturation of Manila clam, Ruditapes philippinarum was induced in this study using a recirculation system over 8 weeks in early spring. Clams used in the experiment were collected in $15^{th}$ April 2010 from the west coast of Korea, as the surface water temperature remained $11^{\circ}C$. To induce gametogenesis and subsequent maturation seawater temperature was elevated $1^{\circ}C$ per day over 10 days to reach $20^{\circ}C$. For the experiment, clams were raised in 120 L quadrangle tank maintained with re-circulated seawater system over 57 days. Water quality parameters including the water temperature, salinity dissolved oxygen, ammonium ion and nitrate levels in the tanks were monitored daily. Mixture of concentrated microalgae including Tetraselmis, Isochrysis, Pavlova and Thalassiosira weissflogii was supplied to clams twice a day, and quantity of the daily ration was adjusted as 3% of clam body dry weight. Histology was applied to examine gonad maturation. Daily monitoring of the water quality parameters indicated that the recirculation system supplied suitable environment to Manila clam; the nitrogenous components stayed below toxic levels (< 0.2 mg/L). At the beginning of the study, clams were mostly in early developing stage. As the seawater temperature reached $20^{\circ}C$, 10 days after the experiment, 20% of clams reached late development at 12 days. First ripe clams were observed at 42 days and 40% of clams were in ripe and ready for spawning at the end of study, 57 days after the experiment. In this study, gametogenesis of Manila clam was successfully induced by elevating water temperature and supplying commercially produced microalgae in a recirculation tank system.

Phaeodactylum tricornutum으로부터 항산화성 물질의 분리 및 구조 (Structure and Isolation of Antioxidative Substance Derived from Phaeodactylum tricornutum)

  • 본 연구에서는 비교적 성장이 빠르고 배양이 용이한 황갈편조 식물문 중 규조강에 속하는 P. tricornutum으로부터 선정하여 항 산화성 물질을 분리$\cdot$정제하여 물질의 구조를 밝혔다. P. tricornutum의 각 유기용매 추출물의 항산화성은 chloroform획분이 가장 좋았으며, 이 획분을 시료로 하여 silica gel column chromatography, PTLC 및 HPLC를 사용하여 항산화성 물질을 분리$\cdot$정제하였다. Silica gel column을 사용하여 분리된 획분의 항산화성은 dichloromethane: methanol (5: 1)로 용출시킨 획분에서 가장 높았으며, 이 획분의 PTLC 획분은 항산화성이 $20\%$ 증가하였다. 최종 정제된 물질의 $IC_50$값은 $8.3{\mu}g/mL$으로 화학적으로 합성된 항산화제인 BHT 및 BHA보다는 낮았지만 천연 항산화제인 $\alpha-tocopherol$보다는 다소 높게 나타났으며, 이 물질의 화학적 구조는 carotenoid류인 zeaxanthin으로 동정되었다.

Increasing the Triacylglycerol Content in Dunaliella tertiolecta through Isolation of Starch-Deficient Mutants

  • The production cost of biodiesel from microalgae is still not competitive, compared with that of petroleum fuels. The genetic improvement of microalgal strains to increase triacylglycerol (TAG) accumulation is one way to reduce production costs. One of the most promising approaches is the isolation of starch-deficient mutants, which have been reported to successfully increase TAG yields. To date, such a stable mutant is not available in an oleaginous marine microalga, despite several advantages of using marine species for biodiesel production. Algae in the genus Dunaliella are known to tolerate high salt concentration and other environmental stresses. In addition, the cultivation processes for large-scale outdoor commercialization have been well established for this genus. In this study, Dunaliella tertiolecta was used to screen for starch-deficient mutants, using an iodine vapor-staining method. Four out of 20,016 UV-mutagenized strains showed a substantial reduction of starch content. A significantly higher TAG content, up to 3-fold of the wild-type level, was observed in three of the mutants upon induction by nitrogen depletion. The carotenoid production and growth characteristics of these mutants, under both normal and oxidative stress conditions, were not compromised, suggesting that these processes are not necessarily affected by starch deficiency. The results from this work open up new possibilities for exploring Dunaliella for biodiesel production.
