Effects of Akebia quinata extract on alcohol-induced damage of liver, spleen and thymus in rats (목통(木通)이 알코올 투여로 유발된 흰쥐의 간, 비장 및 가슴샘의 손상에 미치는 영향)

  • Objective : This study was conducted to investigate the effects of Effects of Akebia quinata (AQ) extract on alcohol-induced damage of liver, spleen and thymus in rats. Method : Experimental animals were divided in to 4 groups; Normal group, Alcohol group, AQ50 group and AQ200 group. All rats, except for Normal group, were fed 25 % ethanol for 55 days. During experimental period, Normal group and Alcohol group were administrated saline, and AQ50 group and AQ200 group were administrated AQ extract at dose of 50 and 200 mg/kg/day, respectively. We measured organ weight, liver triglyceride contents, and alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP) and triglycerid levels in serum. Also, we conducted histomorphometry and histopathological observation of liver, spleen and thymus. Results : AQ significantly decreased the level of AST, ALT and triglyceride in serum and the liver triglyceride contents induced by ethanol. Also, AQ significantly inhibited lipid droplets accumulation in hepatocytes. The decreased relative organ weight of spleen and thymus by ethanol were increased by AQ administration. In histopathological analysis of spleen, the rats administrated AQ 200 mg/kg presented significantly increased mean diameters of white pulps, numbers of white pulps and splenic thicknesses. The administration of 200 mg/kg AQ improved decreased lobular thickness and cortex thickness of thymus, which were decreased by ethanol. Conclusions : The results of present study indicated that AQ has an ameliorating effect for fatty degeneration of liver and damage of spleen and thymus.

Influence of fat levels and types of dietary fiber on lipid metabolism of rats (식이중 지방 수준과 Fiber 종류가 흰쥐의 지방대사에 미치는 영향)

  • The influence of fat level and dietary fiber on serum and liver lipid concentration of the rat was investigated. Groups of rats were fed respectively 3%, 5%, 20% fat containing and free fiber diet until 5th week. From 5th week to 9th week high fat diet groups of rates were devided high fat diet group, high fat pectin supplemented diet group and high fat celulose supplemented diet group. Pectin and cellulose at 10% was supplemented to high fat diets. After rats were fed for 9 weeks, all rats were sacrificed to collect the liver and blood samples by heart puncture. Serum and liver lipids were determined on all rats and compared the histochemical lipid staining method with the biochemical analysis of serum and liver lipids. A high fat diet caused increased level of the total lipids, total cholesterol and free cholesterol contents in the liver and the serum. Rats fed pectin reduced body weight, werum lipid and liver lipid but cellulose had no effect to reduce serum and liver lipid concentration. These results indicate that dietary fat level led to changes in the lipid metabolism of rats and that pectin was more effective in lowering serum and liver lipid than cellulose.

Antifibrotic Effects of Oriental Herbs Extraction on Liver Cirrhosis (한약재(자하거, 백출, 차전자) 추출물의 간경화 억제효능에 관한 연구)

  • Hepatic cirrhosis is an important feature of chronic liver disease. Liver cirrhosis is characterized by hyperaccumulation of fibrous tissue components and is commonly observed in latter or terminal states of chronic hepatic disease. The antifibrotic effects on liver cirrhosis by oriental herbs extraction material were examined in bile duct ligated rats. Oriental herbs extraction (0.99 mg/kg rat weight/day) was administrated to cirrohotic rats for 4 weeks. Liver collagen content of bile duct ligated rats was significantly increased. And liver histology showed collagen fiber deposition was increased as well as the normal architecture was lost with large zone of necrosis being observed. Herbs extraction administrated rats showed significantly decreased liver collagen content, accumulation of collagen fiber in histological analysis, and biochemical markers of hepatic diseases. Those results demonstrate the usefulness of herbs extraction materials as an antifibrotic agent for liver cirrhosis.

Hepatoprotective Effect of Alnus japonica and Portulaca oleracea Complex on Alcohol-induced Liver Injury Mice Models by Anti-oxidation Activity

  • The effectiveness of the extracts of Alnus japonica and Portulaca oleracea, which are effective in improving alcohol-induced liver damage, was confirmed using acute and chronic alcoholic liver injury animal models. In the acute alcoholic liver injury model, dieting Alnus japonica and Portulaca oleracea complex (ALPOC) at a dose of 50 mg/kg showed no significant change in liver or body weight, while measured plasma ALT activity to be deficient (28.12 U/ml) compared to the alcohol intake group (42.5 U/ml), and confirmed that restored it to an average level. It showed an improvement of 34.9% compared to the alcohol intake group. AST activity confirmed that it showed a very effective liver protection activity by showing a gain of 12.6%. The chronic alcoholic liver damage animal model demonstrated that ALT showed an improvement effect of 25%, and AST showed an effect similar to that of the positive control group, Hovenia extract. In addition, through H&E staining analysis, observed that the ALPOC improved the necrosis and bleeding of the liver. And the ALPOC group showed intense antioxidant activity of 127% or more compared to the alcohol intake group, and this was confirmed to have a very high activity, which is more than 20% higher than that of the hovenia fruit extract.

Hydrolysis of Esters and Amides of 20R- and 20S-Dihydroprednisolonic Acid in Rat Serum and Liver Homogenate

  • The hydrolysis rates of ester and amide derivatives of 20-dihydroprednisolonic acid were measured in rat serum and liver homogenate. The hydrolysis rate of the esters in serum was found to be faster than that in liver homogenate on the basis of blood volume and liver weight, while the amide derivatives showed much slower change. And it is also found that the size of substituents at C-21 and C-20 configuration expressed considerable effects on the hydrolysis rate of these derivatives.

  • To investigate the deaging effects of introperitoneally injected Chondroitin Sulfate (CS) on various enzyme activity (AST, ALT, MDA (Malon dialdehyde), SOD (Superoxide dismutase), GPx (Glutathione peroxidases) and histophathology of liver tissue, ovariectomized rats were used. The antioxidative effects of chondroitin sulfate (100 mg/kg and 200 mg/kg body weight) were investigated at the antioxidative enzyme activities of liver homogenate fractions (liver total homogenate, mitochondrial, and microsomal fractions) and sera. In addition, the rat liver was histologically examined. Intraperitoneally injected CS, depend on dosage, indicated a protective effect against ovariectomy-inducted aging. Moreover, inflammation and cirrhosis in liver tissue of CS treated group were significantly decreased. Based on these results, intraperitoneally injected CS is a useful material to delay aging.

Clinical and Ultrasonographic Studies for the Liver Lesion Induced by Tetrachlorethylene in Dogs (개에서 Tetrachlorethylene에 의해 유발된 간장 병변의 임상 및 초음파학적 연구)

  • This study was performed to examine the clinical, serological, ultrasonographic and pathological findings in dogs with acute liver disease induced by tetrachlorethylene at 4 times of anthelminthic oral dose. The results obtained through this experiment could be summarized as follows: 1. The dogs administered with tetrachlorethylene, revealed decreased body weight, and showed lethargy and depression. 2. In serological findings, bilirubin values slightly increased, AST and ALT was decreased at 1∼3 days, and after that time increased according to the lapse of days, and revealed the highest at 5 days, and decreased to normal values at 6 days. 3. In ultrasonographic findings, branches of the portal vein were increased, the echodensity of the liver parenchyma was decreased at early stage, and increased at mid stage, and decreased at last stage. 4. In histopathological findings, necrosis of parenchymal cell, and perivascular hemorrhage were observed more severely at 6 days, as compared with 3 days. There results suggest that ultrasonographic examination is considered to be a more simple, rapid, non-invasive and useful diagnostic method for acute liver parenchymal lesion.

  • The present experiment was performed to investigate the acute effects of cadmium on ultrastructural and biochemical changes in mouse kidney and compare these changes with liver damage. Mouse were injected with cadmium chloride at a dose of 5 mg/kg body weight. After treatment, mouse were sacrificed at time intervals of 6, 12, 24 and 48 hours. It was observed that ultrastructural changes in mouse kidney were composed of swelling of mitochondria, dilation in endoplasmic reticulum, wrinkling at basal infolded membrane, formation of autophagosome and partial loss of microvilli in brush. border, and that ultrastructural changes in liver were mitochondrial change, dilation and deterioration of rough endoplasmic reticulum and proliferation of smooth endoplasmic reticulum. Biochemical effects of cadmium were more severe on liver than kidney. Therefore, acutely injected cadmium caused not only liver damage, but also kidney damage.

Gene Expression Analysis of Acetaminophen-induced Liver Toxicity in Rat (아세트아미노펜에 의해 간손상이 유발된 랫드의 유전자 발현 분석)

  • Global gene expression profile was analyzed by microarray analysis of rat liver RNA after acute acetaminophen (APAP) administration. A single dose of 1g/kg body weight of APAP was given orally, and the liver samples were obtained after 24, 48 h, and 2 weeks. Histopathologic and biochemical studies enabled the classification of the APAP effect into injury (24 and 48 h) and regeneration (2 weeks) stages. The expression levels of 4900 clones on a custom rat gene microarray were analyzed and 484 clones were differentially expressed with more than a 1.625-fold difference(which equals 0.7 in log2 scale) at one or more time points. Two hundred ninety seven clones were classified as injury-specific clones, while 149 clones as regeneration-specific ones. Characteristic gene expression profiles could be associated with APAP-induced gene expression changes in lipid metabolism, stress response, and protein metabolism. We established a global gene expression profile utilizing microarray analysis in rat liver upon acute APAP administration with a full chronological profile that not only covers injury stage but also later point of regeneration stage.

Effect of Hovenia dulcis THUNBER var. koreana Nakai Fruits Extracts on Glucose, Lipid Metabolism and Antioxidant Activities in Streptozotocin Induced Diabetic Rat

  • This investigation was performed to study the antioxidant activities of Hovenia dulcis THUNBER var. koreana Nakai fruits extracts and the effect of Hovenia dulcis fruits extracts on glucose, lipid metabolism in diabetic rats. DPPH free radical scavanging activitiy and superoxide anion radical Scavenging of Hovenia dulcis fruits $80\%$ methanol extracts were $0.06\pm0.002mg$ polyphrnol/ml and $0.l2\pm0.00lmg$ polyphmol/ml, respectively. Hovenia dulcis fruits $80\%$ methanol extracts were partitioned into hexan, dichloro methane, ethyl acetate and butanol, successively. Ethyl acetate fraction were good antioxidant activity. Streptozotocin (45 mg/kg body weight, i.p.) induced diabetic rats showed a significant increases of plasma glucose, triglyceride and total cholesterol. Concomitantly significant decrease of plasma high density lipoprotein. Glutathione level were decrease in cytosol of liver. Lipid peroxide were increase in microsome of liver. Group 1 and 2 were treated with Hovenia dulcis fruits ethyl acetate extracts 50 mg/kg body weight and 20mg/kg body weight for 24 days, individually. Group land 2 rats showed decreased plasma glucose, triglyceride, total cholesterol and lipide peroxide in microsome of liver tissue of rats, and increased plasma high density lipoprotein and glutathione in cytosol of liver tissue rats. The result suggest that Hovenia dulcis THUNBER var. koreana Nakai fruits extracts may effectively normalize the impaired antioxiants status in streptozotocin induced diabetic rats. Hovenia dulcis fruits ethyl acetate extracts were used to improve the imbalance between free radicals and antioxidant system due to the diabetes.
