Obesity is caused from an imbalance between energy intake and expenditure, which may lead to pathologic growth of adipocytes and accumulation of fat in tissue. We examined the inhibitory effects of Eriobotrya japonica leaf and seed extracts on lipid absorption in vitro and fat accumulation during the differentiation of 3T3-L1 to adipocytes. 3T3-L1 preadipocytes were stimulated with DMEM media containing 10% FBS, 0.5 mM 3-isobuthyl-1-methyxanthine (IBMX), $5\;{\mu}g/ml$ insulin, and $1\;{\mu}g/ml$ dexamethasone for differentiation to adipocytes. E. japonica leaf extract at concentration of 0.5 or 1 mg/ml inhibited pancreatic lipase activity. The cell viability of 3T3-L1 adipocytes slightly reduced about 3% by treatment of E. Japonica leaf and seed extracts. The leaf and seed extracts of E. japonica effectively inhibited the accumulations of lipid droplet and expression of $C/EBP{\alpha}$ promoting adipogenesis. Thus, this data suggest that E. japonica leaf and seed extracts inhibit fat accumulation through regulation of $C/EBP{\alpha}$, and leaf extract is more effective in lipid absorption and adipogenesis than seed extract.
The objective of this study was to solidify the simvastatin self-microemulsifying drug delivery system (SMEDDS) and to improve the encapsulation efficiency of solidified alginate beads using sodium alginate. Typical simvastatin SMEDDS was composed of various oils, surfactants and cosurfactants. Also solidified-alginate beads was prepared by crosslinking liquid emulsion mixtures containing sodium alginate and other excipients (cetylpyridinum chloride (CP-Cl), hydroxypropyl methylcellulose, starch and so on). in $CaCl_2$ solution, it has been investigated that the drug release pattern and encapsulation efficiency were varied with the ratio of cationic lipid (CP-Cl). Solidified sodium alginate beads containing simvastatin SMEDDS were redispersed into media without re-aggregation. Oil droplet size of redispersed solidified-beads in media produced smaller than the initial size. The density of beads and drug loading amount were increased with increasing cationic lipid content. These systems have advantages of storage stability and predictability of drug release rate.

Chitosan is the deacetylated product of chitin. Chitosan and its derivatives have many properties that make them attractive for a wide variety of health applications. This study was performed to investigate some effects of water soluble chitooligosaccharides on liver function in the mouse. The animals given the sterol diet containing 3% cholesterol for 6 weeks showed increase in plasma cholesterol level, which were lowered by 23% when they were fed on 1% chotooligosaccharide. While there was no significant change in liver cholesterol and plasma HDL-cholesterol levels. Continuous administration of 15% ethanol via drinking water to mice for 8 weeks elicited pathological alterations such as inflammation, necrosis, accumulation of lipid droplets in the liver and increase in GPT activity, while simultaneous administration of ethanol and chitooligosaccharide prevented remarkedly ethanol-induced liver injury; there was no observable lipid droplet and GPT activity was decreased by 25% in the liver. These results suggest that chitooligosaccharide play some roles in liver function, such as reducing the plasma cholesterol level and preventing alcoholic liver disease.
BACKGROUND/OBJECTIVES: It has been shown that vitamin A supplementation has different effects on skeletal health and the antioxidant system. Deficiency or excess of this vitamin can lead to health problems. Vitamin A can work as either an antioxidant or prooxidant depending on its concentration. The present study was conducted to investigate the effects of different doses of vitamin A supplementation on the antioxidant system in rats. MATERIALS/METHODS: Forty Spargue-Dawley male rats were divided into four groups according to the dose of vitamin A received: 0 (A0), 4,000 (A1), 8,000 (A2), and 20,000 (A3) IU retinyl palmitate/kg diet. After a feeding period of 4 wks, lipid peroxide levels, glutathione concentration, antioxidant enzyme activities, and vitamins A and E concentrations were measured. Histopathological changes were observed in rat liver tissue using an optical microscope and transmission electron microscope. RESULTS: Lipid peroxide levels in plasma were significantly decreased in the A1 and A2 groups compared to the A0 rats. Erythrocyte catalase and hepatic superoxide dismutase activities of the A2 group were significantly higher than those of the A0 group. Hepatic glutathione peroxidase activity was significantly lower in the A3 group compared to the other groups. Total glutathione concentrations were significantly higher in the A1 and A2 groups than in the A0 group. Histological examination of liver tissue showed that excessive supplementation of vitamin A might lead to lipid droplet accumulation and nuclear membrane deformation. CONCLUSIONS: These results indicate that appropriate supplementation of vitamin A might have a beneficial effect on the antioxidant system in rats.

It has been reported that Rosa acicularis has anti-obesity activity by inhibiting the digestive lipase activity. However, there is a lack of clear in vitro studies regarding the anti-obesity activity of Rosa acicularis. Therefore, in this study, we aimed to verify the anti-obesity activity of Rosa acicularis leaves (RAL) and elucidate its mechanism of action in 3T3-L1 preadipocytes. RAL dose-dependently inhibited the accumulation of lipid droplets and triacylglycerol. RAL had no effect on cell proliferation and survival in undifferentiated 3T3-L1 cells, but it inhibited cell proliferation in differentiating 3T3-L1 cells. RAL increased ATGL, p-HSL, and HSL, and decreased perilipin-1 in differentiating 3T3-L1 cells. In addition, RAL reduced lipid droplet accumulation and increased free glycerol content in differentiated 3T3-L1 cells. RAL increased ATGL and HSL in differentiated 3T3-L1 cells. Also, RAL increased p-AMPK, PPARγ, UCP-1, and PGC-1α in differentiating 3T3-L1 cells. AMPK inhibition by Compound C attenuated RAL-mediated increase of ATGL, HSL, PPARγ, and UCP-1 in 3T3-L1 cells. Taken together, it is thought that RAL may inhibit lipid accumulation through lipolysis and thermogenesis via the activation of AMPK in adipocytes.
This study was carried out to observe the functional ultrastructures of photoreceptor cells in frog(Rana catesbeiana) retina using transmission electron microscope. The photoreceptor cells are divided into two types-rod and cone cells-consist of outer and inner segment. The long outer segment of rod cell contains dense stacks of membrane and formed vertical and horizontal folds. The outer segment of cone cell is small, and vertical and horizontal folds are not exist. The electron dense cytoplasm of rod cell contains compact mitochondria, Golgi complexes, and endoplasmic reticula. The inner segment of cone cell shows low electron density and contains a large lipid droplet in the upper part of inner segment. In addition, cone cell contains many mitochondria, Golgi complexes. rough endoplasmic reticula, ribosomes and numerous glycogen particles. It is believed that these ultrastructural characteristics are closely associated with photoreceptive function of photoreceptor cells in frog retina.
The purpose of this research was to determine the effect of phenol compounds from green tea leaves and surfactant micelles on lipid oxidation in com oil-in-water emulsion (O/W). The concentration of phenol and surfactant in continuous phase of the O/W with exceed Brij 700 and phenol compounds was measured. The particle size of O/W with phenol (100 ppm) increased with increasing added exceed surfactant $(0{\sim}2.0%)$ and the concentration of surfactant and phenols in the continuous phase higher than these of control. Lipid oxidation rates, as determined by the formation of lipid hydroperoxides and headspace hexanal, in the O/W emulsions containing phenol compounds (100 ppm) and exceed surfactant $(0{\sim}2.0%)$ decreased with increasing concentration of exceed surfactant. The ability of the phenol compounds and exceed surfactant to inhibit hydroperoxide and headspace hexanal producing as lipid oxidation in O/W was BHT>procyanidin B3-3-O-gallate> (+)-gallocatechin > (+)-catechin and 2% > 1 % > 0% of exceed surfactant. These results indicate that phenol compounds and exceed surfactant could alter the physical location of hydroperoxide in O/W.

Myelophycus simplex Papenfuss is distributed over the northern Pacific and southern coast of Korea, and is a member of the brown algae family. The objective of this study was to investigate the effect of M. simplex methanol extract on adipocyte differentiation and adipogenesis in 3T3-L1 preadipocytes. Treatment with M. simplex methanol extract significantly suppressed terminal differentiation of 3T3-L1 preadipocytes in a dose-dependent manner, as confirmed by a decrease in lipid droplet content observed by Oil Red O staining. Also, the M. simplex methanol extract significantly suppressed the triglyceride content of 3T3-L1 preadipocytes in a dose-dependent manner. Treatment with 300 and $500{\mu}g/ml$ of M. simplex methanol extract caused a 42% and 76% reduction in lipid droplet content, respectively. In order to understand the anti-adipogenic effects of M. simplex methanol extract, the changes in the expression of several adipogenic transcription factors, including peroxisome proliferator-activated receptor (PPAR) ${\gamma}$-cytidine-cytidine-adenosine-adenosine-thymidine (CCAAT)/enhancer binding protein (C/EBP) ${\alpha}$ and ${\beta}$, were investigated using immunoblotting. M. simplex suppressed the expression of $PPAR{\gamma}$, $C/EBP{\alpha}$, and $C/EBP{\beta}$ proteins compared with control. Therefore, the results of this study suggest that M. simplex methanol extract inhibits adipocyte differentiation and thus may have applications as a potential source for an anti-obesity functional food agent.

Oenanthe javanica has been used as a food source and also in traditional folk medicine for its detoxifying properties and anti-microbial effects since ancient times. In this study, we evaluated the effect and mechanism of O. javanica seed methanol extract (OJSE) on adipocyte differentiation by 3T3-L1 preadipocytes. Under non-toxic conditions, OJSE treatment resulted in a dose-dependent inhibition of lipid droplet generation and triglyceride accumulation by suppressing adipocyte differentiation, which are associated with the decreased expression of key proadipogenic transcription factors including CCAAR/enhancer binding protein ${\alpha}$, ${\beta}$ ($C/EBP{\alpha}$, $C/EBP{\beta}$) and peroxisome proliferator-activated receptor ${\gamma}$ ($PPAR{\gamma}$). OJSE also significantly inhibited proliferation and differentiation of 3T3-L1 preadipocytes through G1-phase arrest, indicating that OJSE blocked mitotic clonal expansion during adipocyte differentiation. Investigation of the alteration of G1 phase arrest-related proteins indicated a dose-dependent increase in the expression of p21 and reduction in expression of cyclin E, Cdk2, E2F-1 and phospho-Rb by OSJE. Taken together, these results suggest that OJSE inhibits adipocyte differentiation by blocking the mitotic clonal expansion, which is accompanied by preadipocyte cell cycle arrest.

In this study, we aimed to determine the various effects of Osmanthus fragrans (O. fragrans) flower extract on the skin in order to utilize it as a cosmetic material. For this purpose, Osmanthus fragrans flower extract (OFFE) of Jeju Island was prepared and used in the experiment. The experiments were evaluated by the quantitative real-time polymerase chain reaction (qRT-PCR) and lipid droplet staining assay. First, the OFFE decreased the gene expressions of three representative pro-inflammatory cytokines (IL-8, IL-6, and IL-1α) and an inflammation-related enzyme, PTGS2 induced by poly I:C in epidermal keratinocytes. In addition, the OFFE increased the gene expression levels of collagen (COL1A1) and elastin (ELN) in dermal fibroblasts. Further, the OFFE showed the inhibitory effect in sebum production by linoleic acid in sebocytes. Therefore, from this study, it is expected that OFFE can be used as a natural cosmetic material for anti-inflammatory, anti-aging, and sebum inhibitory efficacy.
